Abstract:Two series of N-containing heterocycles were synthesized by one-step intramolecular or intermolecular annulations of N-substituted pyrrole-2-carbaldehydes and pyrrole-2-carbonitriles. These facile and atom-economic synthetic routes involved the inherent three potential electrophilic and nucleophilic reactive sites of the synthons and the reaction solvent plays a crucial role leading to these different types of products. Abstract:AbstractA new triethylamine‐catalyzed cascade reaction of aromatic aldehydes with terminal conjugated acetylenes has been developed. This serial multiple bond forming process furnishes diverse polycyclic aromatic hydrocarbons including naphthalenes, phenanthrenes, benzofurans, and tetrahydronaphtho[2,3‐b]furans. The chemical outcome of the process depends on the reaction temperature and can therefore be tailored selectively by an appropriate choice of experimental conditions. The resulting tetrahydronaphthofurans can be aromatized further to give strongly fluorescent naphtho[2,3‐b]furans (fluorescent quantum yield, Φf, up to 0.94) in a one‐pot procedure. Furthermore, polysubstituted furans are obtained when the reaction is performed in the presence of dialkyl acetylenedicarboxylates. Abstract:Me OTf-catalyzed formal [4 + 2] annulation of styrene oxides with alkynes to afford polysubstituted naphthalenes has been realized, which undergoes sequential electrophilic cyclization/ring expansion. A range of substrates were tolerated in the formation of naphthalene derivatives with high regioselectivity in satisfactory yields. The reaction could also be carried out on gram scale.

Abstract:The FeCl3-promoted annulation reaction of aryl acetaldehydes with alkynes has been established, which provides a new and versatile straightforward procedure for the regioselective synthesis of mono-, di-, and polysubstituted naphthalenes under mild conditions. Interestingly, the present catalytic system not only differentiates between internal and terminal alkynes but also shows unprecedented complete Me3SiOH elimination selectivity for silyl alkyne substrates. Furthermore, the synthesis of a series of substituted phenanthrenes via reactions of nathphyl acetaldehydes with internal alkynes is also achieved for the first time in good yields with excellent regioselectivity.

Abstract:Enantioselective [3 + 2] annulation of N-heteroarenes with alkynes has been developed via a cobalt-catalyzed dearomative umpolung strategy in the presence of chiral ligand and reducing reagent. A variety of electron-deficient N-heteroarenes, including quinolines, isoquinolines, quinoxaline, and pyridines, and internal or terminal alkynes are employed in this reaction, showing a broad substrate scope and good functionality tolerance. Annulation of electron-rich indoles with alkynes is also developed. This protocol provides a straightforward access to a variety of N-spiroheterocyclic molecules in excellent enantioselectivities (76 examples, up to 99% ee).

Abstract:A tandem insertion of thiocyanate to enamine was performed for the regioselective synthesis of multisubstituted benzoimidazo[2,1-b]thiazoles. This method was shown to be effective in addressing the issue of isomerization encountered in common strategies. With a change made to the leading group on the aniline fragment of enamine, the reaction achieved different transformations, thus enabling multisubstituted benzo[4,5]imidazo[2,1-b]thiazoles and thiazoles in satisfactory yields.

Abstract:One-pot synthesis of tetracyclic naphthoxazole derivatives from electron-deficient naphthoquinones and alkynes was achieved via Rh(III)-catalyzed C-H activation and C-sp3-H bond cleavage for the first time. This approach proceeds through a tandem cascade process involving substrate tautomerization, C-H activation, oxidative addition, cyclization, and aromatization. Abstract:The current review describes different annulation strategies reported with electrophilic benzannulated heterocycles for accessing heteroacenes. For the past two decades, the chemistry of electrophilic benzannulated heterocycles was extensively investigated, and several dipolar cycloadditions, metal and organo-catalyzed transformations were introduced for the generation of fused heterocycles. In this review, we have collected all the reports where the annulation of electrophilic benzannulated heterocycles results in a fully aromatic system, viz. heteroacenes with tri-, tetra-, and pentacyclic rings. We reviewed every paper on the synthesis of fused heterocycles that was accessible and categorized the review into several parts based on the electrophilic benzannulated heterocycle used in the heteroacene synthesis such as electrophilic indole, electrophilic benzothiophene, and so forth. The generality and mechanistic postulates of each methodology are highlighted. In addition, we have also tried to feature the advantages or shortcomings of each method and have mentioned the possible applications of these methodologies for accessing heteroacenes for material applications.

Abstract:A novel Ag-catalyzed radical cascade bicyclization/aromatization of C-linked 1,7-enynes with 1,3-dicarbonyls has been achieved, providing a step- and atom-economy approach for the straightforward construction of 2,3-dihydro-1H-cyclopenta[a]naphthalenes, an important structural scaffold existed in biologically active compounds. From this transformation, structurally diverse 2,3-dihydro-1H-cyclopenta[a]naphthalenes were obtained in moderate to good yields with excellent regioselectivity. Moreover, the further product derivatizations in facile ways were also exemplified.

Abstract:Using palladium‐catalyzed reaction of 5‐(2‐bromophenyl)pent‐3‐en‐1‐ynes with arylboronic acids was studied. Reaction proceeds with C‐Br bond activation, migratory insertion, metathesis with arylboronate, and reductive elimination to yield the desired naphthalene derivatives. Highly substituted naphthalene derivatives were prepared via palladium‐catalyzed reaction of 5‐(2‐bromophenyl)pent‐3‐en‐1‐ynes with arylboronic acids. Typically, reaction of 3‐bromo‐4‐(3‐(4‐chlorophenyl)‐5‐phenyl‐1‐(p‐tolyl)pent‐2‐en‐4‐yn‐1‐yl)‐N‐methylaniline (1a) with PhB(OH)2 in the presence of Pd(PPh3)4 as the catalyst under basic conditions provided 8‐benzhydryl‐7‐(4‐chlorophenyl)‐N‐methyl‐5‐(p‐tolyl)naphthalen‐2‐amine (2a) quantitatively. Overall, 19 new naphthalene compounds were obtained by this methodology and their structures were characterized by spectroscopic methods. In addition, crystal structure of 2j was determined to confirm the structural details. Reaction pathway leading to the desired product is also discussed.

Abstract:Nitrogen-containing heterocycles are present in most approved drugs, reflecting the significance of their synthetic strategies. By utilizing oxadiazolone as a nitrenoid (nitrene-like) precursor, we have developed a general strategy for the annulation with nucleophilic heterocycles to access various polycyclic aminoheterocycles. We have discovered that 2-pyrryl-substituted substrates undergo a rearrangement, which indicates a spirocyclization-migration pathway. Given their fluorescence and biological activity as kinase hinge binders, these fragment-like aminoheterocycles represent potential starting points for applications in chemical biology and drug discovery.

Abstract:Alkynylcyclopropanes have been used for the first time as coupling partners in transition-metal-catalyzed C-H functionalization. Specifically, a Cp*Rh-III-catalyzed regioselective annulation of alkynylcyclopropanes with N-aryloxyamides via redox-neutral C-H/C-C activation has been developed, which affords highly functionalized 2,3-dihydrobenzofurans bearing an (E)-exocyclic carbon-carbon double bond and a tetra-substituted carbon center in moderate to good yields with a broad substrate scope.

Abstract:The NBS-mediated organocatalytic cascade synthesis of 4,5-dihydronaphtho[2,1- b ]furan-1-carbaldehydes from α,β-unsaturated aldehydes and 2-tetralones have been developed. In this synthesis strategy, the C1 of in situ generated brominated 2-tetralone was used as identical twin electron donor–acceptor, and the Cβ and Cα of enals were used as the heterogeneous twin electron acceptor–donors.
