Abstract:Systematic phytochemical investigation on the stems of Kadsura coccinea resulted in the isolation of eleven new lanostane-type triterpenoids, named kadcoccinones G-Q (1-11), together with two known structural analogues, kadpolysperin M (12) and abiesatrine D (13). Their structures were characterized on the basis of comprehensive spectroscopic methods, and the absolute configuration of 1 was ascertained by single-crystal X-ray diffraction. Structurally, compounds 1/2, 4/5, and 6/7 were three pairs of C-12 epimers, respectively. Additionally, compounds 1-11 were evaluated for their in vitro cytotoxicity against the human tumor HL-60, SMMC-772, A-549, MCF-7, SW-480, and HeLa cell lines using the MTS viability assay. Abstract:Eleven triterpene acids including 10 new compounds (kadcoccinic acids A-J, 1-10) were isolated from the stems of Kadsura coccinea. Except for 10, these compounds feature a rearranged lanostane skeleton with a 6/6/5/6 tetracyclic ring system, and compounds 1 and 2 are the first examples of 2,3-seco-6/6/5/6-fused tetracyclic triterpenoids. Their structures were established primarily by spectroscopic and spectrometric methods. Additionally, the absolute configuration of 3 was determined by single-crystal X-ray diffraction. Several of the compounds isolated were tested for their anti-HIV-1 and cytotoxic activities.

Abstract:Three new triterpenoids, kadcotriones A-C (1-3), together with the biogenetically related lanostane-type triterpenoid (4), were isolated from Kadsura coccinea. Compound 1 features a 12,14β-dimethyl 6/6/6-fused tricyclic skeleton, while 2 and 3 are characterized by a 6/6/5-ring system. Their structures were determined by NMR and electronic circular dichroism spectroscopic methods. Compounds 2 and 4 exhibited anti-HIV-1 activities with EC50 values of 30.29 and 54.81 μM, respectively.

Abstract:Fourteen new rearranged 6/6/5/6-fused triterpenoid acids, namely, kadcoccine acids A-N (1-14), were isolated from an EtOAc-soluble extract of the stems of Kadsura coccinea. Their structures were characterized mainly by analyzing 1D and 2D NMR and HRESIMS data and were shown to feature a rare 14(13 -> 12)-abeo-lanostane skeleton. Compounds 7 and 8 represented the first examples of a 5-substituted 2(5H)-furanone motif on the C-17 side chain of this skeleton. The absolute configurations of C-23 for compounds 1, 7, and 8 were determined by comparison of their experimental electronic circular dichroism spectra. All the isolates were screened for their in vitro cytotoxicity against six human tumor cell lines (HL-60, SMMC-7721, A-549, MCF-7, SW-480, and HeLa), and compounds 2 and 8 exhibited weak inhibitory effects with IC50 values ranging from 3.11 to 7.77 mu M.

Abstract:An investigation of EtOAc extracts of Kadsura coccinea (Lem.) A. C. Smith, has led to the isolation of two new compounds characterized as 3-hydroxy-12-hydroxyl coccinic acid ( 1 ) and 3-hydroxy-neokadsuranic acid A ( 2 ). Their structures were established by 1D and 2D NMR techniques and mass spectroscopy. Antiproliferative effects of the isolated compounds were evaluated against four human tumor cell lines (A549, HCT116, HL-60 and HepG2), and it was found that compound 1 exhibited antiproliferative effects with IC 50 values ranging from 3.01 to 18.08 μg/mL.

Abstract:A pair of new triterpenoid epimers, kadcoccitones A (1) and B (2), together with a new biogenetically related compound kadcoccitone C (3), were isolated from Kadsura coccinea. The epimers featured an unprecedented carbon skeleton with a 6/6/5/5-fused tetracyclic ring system unit and a C(9) side chain. Their structures were determined by spectroscopic data, ECD calculation, and single-crystal X-ray diffraction. Compounds 1 and 3 showed anti-HIV-1 activity with an EC(50) value of 47.91 and 32.66 μg/mL, respectively.

Abstract:Phytochemical investigation on the stems of Kadsura coccinea led to the isolation of 8 new triterpenoids, kadcoccilactones K–R (1–8), and 10 known analogues. Their structures were elucidated on the basis of extensive spectroscopic analysis. Compounds 1–3 characterized with an aromatic ring E in their molecules are rarely naturally occurred kadlongilactone derivatives. Moreover, all compounds were evaluated for their inhibitory activity against K562, Bel-7402, and A549 human tumor cells. Compounds 9 and 10 exhibited potent cytotoxicity against K562, Bel-7402, and A549 cell lines with IC50 values less than 0.1, 0.1, and 1.0μm, respectively.

Abstract:Two new bicyclic sesquiterpenes, namely 6,9-dihydroxycadinan-4-en-3-one (1) and guai-3-en-10-ol (2), together with one known sesquiterpene were isolated from the stems of Kadsura heteroclita, which is a Tujia ethnomedicine called Xue Tong'. Their structures were elucidated by using spectroscopic analyses and by comparing the NMR data with those reported previously.

Abstract:AIM:To study the chemical constituents of the roots and stem bark of Kadsura coccinea.METHOD:Compounds were isolated by column chromatography on silica gel and Sephadex LH-20, and finally purified by prep-HPLC. Their structures were elucidated by extensive spectroscopic methods, including 1D- and 2D-NMR, and HR-ESI-MS.RESULTS:Two compounds were determined as (7'S,8'S,8R)-(8β,8'α)-dimethyl-4,4'-dihydroxy-5,3'-dimethoxy-5'-cyclolignan glucoside (1) and micrandiactone H (2), respectively.CONCLUSION:Compunds 1 and 2 are new and neither showed inhibitory effects on nitric oxide (NO) production in lipopolysaccharide-induced RAW264.7 macrophages.

Abstract:An extensive study of the triterpenoids produced by the stems of Kadsura ananosma, has led to the isolation of eleven new ones, kadnanolactones A-I (1–4, 7–11) and kadnanosic acids A (5) and B (6), and six known analogues. Their structures were elucidated mainly by comprehensive spectroscopic analysis, and DFT computational methods were applied to validate the stereochemistry of an epoxide in compound 7. All triterpenoids were evaluated for their cytotoxicity against HL-60, SMMC-7721, A-549, PANC-1, and SK-BR-3 human cancer cells.

Abstract:In the present work, we reported the triterpenoids isolated from n-butanol fraction of Kadsura heteroclita which is a Tujia ethnomedicine with trivial name "Xuetong". This effort resulted in the isolation of six unpresented triterpenoids xuetongsu A-F (1-6), along with five known triterpenoids (7-11). The structures of the reported compounds were established on the 1D, and 2D NMR and HRESIMS spectra, along with CD spectroscopic analysis. Moreover, the absolute stereochemistry of compound 7 was determined by X-ray diffraction analysis. Antioxidant and cytotoxic activities were evaluated for all isolated compounds, compound 7 shown weak cytotoxic activity against HL-60 with IC50 value of 50.0 mu M.

Abstract:Two new triterpene lactones, polysperlactones A (2) and B (3), were isolated from the stems of Kadsura polysperma, together with the known compounds heteroclitalactone D (1) and schisanlactone E (4). Their structures were elucidated by spectroscopic methods, including 2D-NMR and HR-MS techniques. The configuration of 1 was confirmed by X-ray analysis. Compounds 2 and 3 are members of a rare class of 3,4-secolanostane metabolites with ring-expanded or cyclized structures, respectively.

Their structures were elucidated by analysis of spectroscopic evidence including extensive 2D‐NMR data.

Abstract:Phytochemical investigations of the stem of Kadsura heteroclita (Roxb) Craib (Schizandraceae) resulted in the isolation and structure elucidation of six new triterpenoidal compounds named heteroclitalactones A - E (1-5) as well as heteroclic acid (6) and heteroclitalactone F (7), which was isolated for the first time from a natural source, and the six known compounds schisanlactone E (8), cycloartenone (9), schisandronic acid (10), nigranoic acid (11), changnanic acid (12) and schisanlactone B (13), respectively. The structures of these compounds were characterized by extensive 1D and 2D NMR spectral analyses. The majority of these triterpenoids showed moderate cytotoxic activity against the human tumor cell lines Bel-7402, BGC-823, MCF-7 and HL60. Among the compounds tested, heteroclitalactone D (4) showed the strongest cytotoxic activity against the HL-60 cells with an IC50 of 6.76 mu M.
