Learning and memory impairment in rat model of Parkinson disease

Abstract:AIM: To evaluate the learning and memory impairment in rat model of Parkinson disease at different stages with Morris maze in order to provide evidence for the early diagnosis and treatment of memory disorder in Parkinson disease. METHODS: The study was finished in the Stem Cell Transplantation Laboratory of Anhui Provincial Hospital and the Department of Neurobiology of Anhui Medical University from June 2004 to April 2005. ① Selection: Rats with memory impairment and behavior of circle were excluded before model establishment of Parkinson disease.② Model establishment: Rat models of Parkinson disease were induced by 6-hydroxydopamine (6-OHDA) with stereotactic technique. ③Measured indexes: The latency for the rats to find platform, swimming distance, swimming speed and the percentage of swimming distance in each quadrant within 120 s to the total distance were detected with Morris water maze at 1 and 3 months. RESULTS: Totally 74 rats were used in the study. ① Rats with memory impairment, circling at the same place and the unsuccessful rat models(n=33) were excluded. Finally, 41 rats were involved, including 15 in the 1-month Parkinson disease group, 16 in the 3-month Parkinson disease group and 10 in the control group. ② The swimming speed of rats in the 1-month Parkinson disease group was significantly different from that in the control group (P < 0.05), but insignificantly different from that in the 3-month Parkinson disease group (P > 0.05). ③ The swimming distance for the rats with Parkinson disease was significantly increased as compared with that in the control group (P < 0.05). ④ In the control group, the swimming distance in the quadrant in which the platform located was significantly different from those in the other quadrants (P < 0.05). The swimming distances were insignificantly different among the quadrants in the 1 and 3-month Parkinson disease groups. CONCLUSION: Rat models of Parkinson disease manifest gradual spatial memory and exploratory strategy impairment, which are resemble to patients with early Parkinson disease.
