Expression of Inhibitor of Apoptosis Protein Survivin in Clear Cell Renal Cell Carcinoma and It's Clinical Significance

Abstract:Objective To assess the expression of survivin,a member of the inhibitor of apoptosis protein family,in clear cell renal cell carcinoma(ccRCC)and to determine its clinical significance.Methods Immunohistochemical staining for survivin was performed using paraffin-embedded tissues from 48 cases of ccRCC and 20 cases of normal kidney tissues.We also assessed the expression of survivin in 10 cases of ccRCC by Western blot.Results Of the 48 ccRCC specimens,43(89.58%)showed positive staining of survivin immunohistochemically.In contrast,no expression of survivin was detected in nomal kidney tissues.A significant increase in survivin expression was associated with increased tumor stage,increased tumor grade and distant metastasis(P 0.05).However,there was no significant correlation between the expression of survivin and patient age,sex,tumor size and cancer embolus in inferior vena cava.Western blot analysis in 10 cases of ccRCC also demonstrated the high expression of survivin.Conclusion Survivin-mediated inhibition of apoptosis is associated with progression of renal cell carcinoma.This may be relevant for prognosis and alternative therapeutic approaches of ccRCC.
