CLINICO-PATHOLOGIC SIGNIFICANCE OF herg1 GENE AND HERG1 PROTEIN OVEREXPRESSION IN GASTRIC CANCER

Abstract:Objective To investigate the expression of herg1 gene in gastric carcinoma,and the relation of HERG K~+ channels with clinico-pathologic features.Methods Reverse transcription and PCR(RT-PCR),Western blot(WB) and immunohistochemistry(IHC) were used to detect the expression of herg1 gene in 64 cases of gastric carcinoma,and the patients were followed-up.Results The positive expression of herg1 gene and HERG1 protein was 64 in 64 cases(100%) of gastric cancers,but no expression was found in normal tissues by using RT-PCR and WB. The positive expression was 52 in 64 cases(81.25%) of gastric cancer,but no expression was found in normal tissues by using IHC.The positive rate was 10 in 10 cases(100%) of gastric cancers with lymph node and liver metastasis but 42 in 54 cases(78%) of gastric cancers with no metastasis.The positive rate of herg1 gene and HERG1 protein was related to lymph node and liver metastasis but not to pathologic type(P<0.01).Conclusion HERG K~+ channels were probably expressed positively in almost all kinds of gastric carcinoma and play an important role in tumor genesis and development.We could predict tumor invasion and metastasis by detecting herg1 gene and HERG1 protein.We have provided evidence in theory and experiment to establish the molecular marker for tumor metastasis.
