Abstract:Objective To optimize the ultrasonic extraction technology and evaluate the antioxidant activity of total flavonoids from Verbena officinalis L. by response surface methodology. Methods On the basis of single factor experiments, the ultrasonic time, the ratio of liquid to material and ethanol volume fraction was used as influencing factors, the ultrasonic extraction process of total flavonoids from Verbena officinalis L was optimized by response surface methodology. The antioxidant capacity of total flavonoids from Verbena officinalis L. was studied by DPPH free radical and H2O2 scavenging activity. Results The optimum extraction process of total flavonoids from Verbena officinalis L. was as follows: the ultrasonic time was 40 min, the ratio of liquid to material was 40:1 and the ethanol volume fraction was 60 ％. Under these conditions, the predicted content value of total flavonoids in the extract was 3.31 ％, the measured value was 3.26 ％, and the error was 1.51 ％. The total flavonoids from Verbena officinalis L. possess strong DPPH free radical and H2O2 scavenging activity. Conclusion The extraction technology by ultrasonic wave has the advantages of high extraction efficiency, convenient operation and wide adaptability. The optimum extraction process obtained by response surface method is of small error, good accuracy and strong feasibility. The total flavonoids from Verbena officinalis L. show remarkable antioxidant activity.
