Abstract:A novel tandem one-pot method for the synthesis of polysubstituted pyridine derivatives has been developed via Knoevenagel-type reaction and subsequent 675 electrocyclization of the Blaise reaction intermediates and 3-formylchromones. Abstract:An efficient route to synthesize 4-chloro-7H-pyrrolo[2,3-d]pyrimidine (6) and 2,4-dichloro-7H-pyrrolo[2,3-d]pyrimidine (10) starting from diethyl malonate in five steps with the total yields of 45.8% and 44.8% was disclosed in the presented paper. The chemical structures of the objective products as well as the related intermediates were confirmed.

Abstract:Fused pyridazino[4,5-b][1,4]oxazepine-diones were easily obtained in moderate to excellent yields via Smiles rearrangement. The synthetic approach is supported by a one-pot coupling-Smiles rearrangement-cyclization process. This process has potential applications in the synthesis of biologically and medicinally relevant compounds.

Abstract:A series of novel 1,3,5-triarylpyrazoles were synthesized from flavanones, arylhydrazines, and trimethyl phosphate in an one-pot procedure. Facile reaction process, easy after-reaction workshop, and good yields are the distinct characteristics of the developed protocol. The target compounds were characterized by element analysis, infrared ray (IR), 1H NMR spectra, and electrospray ionization-mass spectrometry. The structure of representative compound (C23H20N2O3, Mr = 372.42) was further confirmed by X-ray diffraction. It crystallizes in monoclinic, space group P 21/c, a = 8.9720(5), b = 24.5523(13), c = 8.9687(6) angstrom, = 90.0000, = 102.6417(17), = 90.0000 degrees, V = 1927.76(20) angstrom 3, Z = 4, (MoK) = 0.086, F(000) = 784, Dc = 1.283 g/cm3, the final R = 0.0349 and wR = 0.0844 for 1668 observed reflections (I > 2 sigma(I))

Abstract:An efficient procedure for the synthesis of 7‐(aryl)‐8‐nitro‐2,3,6,7‐tetrahydroimidazo[1,2‐a]pyridinones, 8‐(aryl)‐9‐nitro‐3,4,7,8‐tetrahydropyridone[1,2‐a]pyrimidines and 9‐(aryl)‐10‐nitro‐2,3,4,5,8,9‐hexahydropyridone[1,2‐a]diazepine via one‐pot three component reaction of diamine, nitroketene dithioacetal (1,1‐bis(methylsulfanyl)‐2‐nitroethene), and coumarine‐3‐ carboxylic acid derivatives in EtOH under reflux conditions is reported. The advantages of this procedure are simplicity, easy purification, good yields, and catalyst‐free conditions. All products were confirmed by 1H‐ and 13C‐NMR, IR, MS, and X‐ray crystal structure analyses.

Abstract:Direct synthetic methods of 6,7-chphenylpyrazolo[1,5-a]pyrimidine derivatives have been developed. Cyclocondensation of isoflavones with 3-aminopyrazole in the presence of sodium methoxide as alkali promoter gave 6,7-diphenylpyrazolo[1,5-a]pyrimidines in moderate to good yields.

Abstract:Abstract: Novel hydrazones 4, 5, and oxime 6 were produced from the reaction of ketone 3 with hydrazine, phenylhydrazine, and hydroxylamine HCl, respectively. One pot multicomponent reaction of chalcones 7a-c, 3-(1H-indol-3-yl)-3-oxopropanenitrile 8, and ammonium acetate in MeOH at reflux temperature gave 2-(1H-indol-3-yl)-nicotinonitrile derivatives 10-12. Additionally, a one-pot reaction of chalcone 7a, malononitrile or ethyl cyanoacetate, and ammonium acetate in AcOH at reflux temperature afforded 6-aminopyridine derivatives 13 and 14, respectively.

Abstract:A one-pot synthesis of pyrrolo[3,2-d]pyridazine derivatives via zirconocene-mediated four-component coupling of one Si-tethered diyne, two nitriles, and one azide is reported. When TMSN3 was used, pyrrole-2,3-diones were formed in good yields. Further condensation of these highly functionalized pyrrole-2,3-diones with hydrazine and hydroxylamine afforded useful pyrrole-fused heterocycles.

Abstract:5,6,7,8-tetrahydro-4H-pyrazolo[1,5-a] diazepine carboxylates are valuable scaffolds for drug design and medicinal chemistry. In this paper we disclose a short scalable synthesis and chemical reactivity patterns of the pyrazolo-diazepine pharmacophore. Commercial and cheaply available Methyl pyrazole 3,5-dicarboxylate was alkylated with 3-bromo-N-Boc propyl amine and the resulting derivative underwent concomitant cyclization upon deprotection of the Boc protecting group to yield the pyrazolo-diazepine skeleton. Selective reduction of the lactam was accomplished cleanly using borane and the resulting amine was pro-tected using a tert-butyloxycarbonyl protecting group. The free N-terminal of the diazepine underwent smooth Buchwald and Chan arylations among various standard chemistry applications examined on this pharmacophore.

Abstract:Reactions of easily accessible chloropyrimidinyl hydrazones with bromine were investigated. Oxidative cyclization followed by concomitant bromination led to the formation of [1,2,4]triazolo[4,3-c]pyrimidines, which then underwent the Dimroth rearrangement to afford highly functionalized 2-substituted [1,2,4]triazolo[1,5-c]pyrimidines.

Abstract:In the presence of trifluoroacetic acid and methanesulfonic acid (MSA), a series of monohydric and polyhydric phenols readily undergo a domino Pechmann-dehydration reaction with various 4-hydroxy-6-oxocyclohexane-1,3-dicarboxylates to give 7,8-dihydro-6H-dibenzo[b,d]pyran-6-ones. Subsequent dehydrogenation of the 7,8-dihydro-6H-dibenzo[b,d]pyran-6-ones was performed expediently with DBU/BrCCl3 to give various 6H-dibenzo[b,d]pyran-6-one derivatives. This strategy has the advantages of inexpensive and easily available raw materials, metal-free procedure, mild reaction conditions, good compatibility with various functional groups, and satisfactory yields.

Abstract:This paper presents an efficient one-pot approach for synthesizing hydroxylated 1,3-dithiane-pyrazolone hybrids. The process involves a reaction between pyrazolone derivatives, carbon disulfide, and epichlorohydrin (ECH). The structures of the produced 1,3-dithian-pyrazolone hybrids are determined based on their IR, 1 H NMR, 13 C NMR, and Mass spectroscopic data. Analysis of the 1 H NMR spectra of compounds 4b and 4d revealed first-order splitting of the methylene hydrogens of the 1,3-dithian moiety, indicating that the OH group occupies an axial position in the preferred conformation of these derivatives. Despite the use of optically pure ECH as a starting material, racemization of products occurs during the reaction. The proposed reaction mechanism provides a rational explanation for this observation.

A mixture of N-isocyaniminotriphenylphosphorane, an aldehyde, and Meldrum's acid undergo a 1:2:1 addition reaction under mild conditions to afford the title compounds in good to excellent yields.

Abstract:A facile and efficient one-pot synthesis of substituted thieno[2,3-b]pyridines has been developed. Treatment of enaminones, such as 2-acetyl-3-(dimethylamino)propenamides and -propenoates, with 2-cyanothioacetamide in the presence of potassium carbonate in N,N-dimethylformamide at 80 degrees C followed by addition of methylene-active bromides at room temperature provided, via intramolecular cyclization, 2,3,5,6-tetrasubstituted thieno[2,3-b]pyridines in yields of 78-90%. This protocol, which combines construction and modification of the thieno[2,3-b]pyridine ring, increases the structural diversity of the final products from readily available materials.

Abstract:An efficient and concise method is hereby reported for the synthesis of pyrido[3,4- c ]pyridazin-8(7H)-ones from the commercially available material, methyl 4,6-dichloropyridazine-3-carboxylate. The four-step reaction process involves Suzuki coupling, Buchwald coupling, ammoniation, and intramolecular cyclization. The advantage of this route represents the development of a novel C5 site-selective Suzuki monocoupling of 3,5-dichloropyridazine, with an overall yield of over 73 %.
