Abstract:Objective To investigate the radiosensitization by yCDglyTK fusion suicide gene/prodrug system in human nasopharyngeal carcinoma cells (CNE-2). Methods The plasmid vector of pcDNA3.1(-)CMVe·Egr-1yCDglyTK was introduced into CNE-2 cells by electroporation. Then the cells were exposed to various doses of radiation and prodrug. Observation was made for the change of expression and function of fusion suicide gene in yCDglyTK-expressing cells through immunohistochemistry, HPLC, MTT and clonogenic assay. The influence in growth of CNE-2 cells was observed too. Results Ionization radiation can induce an increased expression of yCDglyTK gene in suicide-gene positive cells; Combined with ionization radiation and prodrug showed greater toxicity to yCDglyTK-expressing CNE-2 cells than disposed with ionization radiation or prodrug alone. Conclusion The yCDglyTK fusion suicide gene/prodrug system can enhance the radiosensitivity of CNE-2 cells, and show synergistic killing effect when combined with ionization radiation.

Abstract:Gene-radiotherapy is a new strategy in the treatment for cancer recently.However,solid tumors are low radio sensitivity due to their regular hypoxia,thus affecting the transcriptional activity of radiation-regulated sequence.Therefore,this therapy fails to produce the desired effects.Introduction of hypoxic promoter into the upstream of the therapeutic gene can overcome the decrease on the transcriptional activity of radiation-regulated sequence due to the hypoxic state of tumor and thus enhance the killing effect on tumor cells.This paper reviews the research status,existing problems and prospect of radiotherapy combined with radiation-induced,hypoxia-induced and hypoxia/radiation double-induced promoters with suicide gene.

Abstract:Objective As generally known that non-cytotoxic pro-drug can be transformed into cytotoxic drug by suicide gene,this work is to investigate the effect of Coli cytosine deaminase/5-fluorocytosine suicide gene(CD/5-FC) used alone or combined with irradiation in esophageal carcinoma cell line(EC).Methods CD gene was amplified from Coli DNA genome library with PCR technique,with the eukaryotic vector pcDNA3.1-CD then constructed.EC109 cells were transfected with pcDNA3.1-CD by liposome method.The cytotoxic effect,by-stander effect and radiosensitization effect of CD/5-FC in EC109 was analyzed.Results The transfection of CD gene into EC109 and its transcription was confirmed by RT-PCR method.In vitro,5-FC showed significantly cytotoxic effect on the EPC cell transfected with CD gene.After adding 5-FC,the survival rate of cultured cell containing 5% transfect CD gene cell was 41.8%±14.2% while that in the control group was(94.6)±4.3%,(t=3.14,P0.05).The survival rate of cultured cell containing 10% transfected CD gene cell was 37.8±(4.4%) compared to 95.6%±5.4% in the control group,(t=9.75,P0.01).CD/5-FC showed significant radiosensitization effect,the survival fraction of CD transfected cell was much lower in 5-FC combined with irradiation,when compared with 5-FC alone and radiotherapy alone group together,(F=11.50,P0.01).When it was compared with 5-FC alone group and irradiation alone group separately,the difference was also significant(F=(4.11),P0.05 and F10.53,P0.01,respectively).Conclusions Suicide gene CD/5-FC shows conspicuous by-stander effect and radiosensitization effect.
Radiosensitization of NSCLC cells to X-rays and carbon ions by the CHK1/CHK2 inhibitor AZD7762, Honokiol and Tunicamycin

Abstract:Although radiotherapy, especially carbon-ion radiotherapy, is an effective treatment modality against non-small-cell lung cancer (NSCLC), studies using radiation combined with sensitizer for improving the efficacy of radiotherapy are still needed. In this work, we aimed to investigate in NSCLC A549 and H1299 cell lines the effects of different linear energy transfer (LET) radiations combined with diverse sensitizing compounds. Cells pretreated with the CHK1/CHK2 inhibitor AZD7762, Honokiol or Tunicamycin were irradiated with low-LET X-rays and high-LET carbon ions. Cell survival was assessed using the clonogenic cell survival assay. Cell cycle distribution and apoptosis were measured with flow cytometry, and DNA double strand break (DSB) and repair were detected using γ-H2AX immunofluorescence staining. Our results revealed that AZD7762, Honokiol and Tunicamycin demonstrated low cytotoxicity to NSCLC cells and a pronounced radiosensitizing effect on NSCLC cells exposed to carbon ions than X-rays. Unrepaired DNA DSB damages, the abrogation of G2/M arrest induced by irradiation, and finally apoptotic cell death were the main causes of the radiosensitizing effect. Thus, our data suggest that high-LET carbon ion combined with these compounds may be a potentially effective therapeutic strategy for locally advanced NSCLC.

Abstract:PURPOSE:To investigate whether hypoxia targeted bifunctional suicide gene expression-cytosine deaminase (CD) and uracil phosphoribosyltransferase (UPRT) with 5-FC treatments can enhance radiotherapy.MATERIALS AND METHODS:Stable transfectants of R3327-AT cells were established which express a triple-fusion-gene: CD, UPRT and monomoric DsRed (mDsRed) controlled by a hypoxia inducible promoter. Hypoxia-induced expression/function of CDUPRTmDsRed was verified by western blot, flow cytometry, fluorescent microscopy, and cytotoxicity assay of 5-FU and 5-FC. Tumor-bearing mice were treated with 5-FC and local radiation. Tumor volume was monitored and compared with those treated with 5-FC or radiation alone. In addition, the CDUPRTmDsRed distribution in hypoxic regions of tumor sections was visualized with fluorescent microscopy.RESULTS:Hypoxic induction of CDUPRTmDsRed protein correlated with increased sensitivity to 5-FC and 5-FU. Significant radiosensitization effects were detected after 5-FC treatments under hypoxic conditions. In the tumor xenografts, the distribution of CDUPRTmDsRed expression visualized with fluorescence microscopy was co-localized with the hypoxia marker pimonidazole positive staining cells. Furthermore, administration of 5-FC to mice in combination with local irradiation resulted in significant tumor regression, as in comparison with 5-FC or radiation treatments alone.CONCLUSIONS:Our data suggest that the hypoxia-inducible CDUPRT/5-FC gene therapy strategy has the ability to specifically target hypoxic cancer cells and significantly improve the tumor control in combination with radiotherapy.

Abstract:The aim of study was to investigate the killing effect of double suicide gene system mediated by retroviral vector on K562 cells in vivo and ex vivo. CDglyTK gene was transfected into PA317 cells by using lipofectamine. K562 cells were infected with viral supernatant. K562/CDglyTK cells were treated with 5-fluorocytosine (5-FC) and/or ganciclovir (GCV). Mice were randomly divided into three groups: tumor formation, tumor inhibition and tumor therapy. Each mouse was implanted with K562/CDglyTK cells or K562 cells. The results indicated that the killing effect of 5-FC in combination with GCV on K562/CDglyTK was more significant than using 5-FC or GCV alone. In vivo study showed that after being injected subcutaneously with K562 cells and K562/CDglyTK cells, there was not obvious difference in tumor formation rate of mice, 5-FC + GCV could suppress tumor formation of the K562/CDglyTK cells. After being treated with 5-FC and GCV, the median tumor volume of mice implanted with K562/CDglyTK cells decreased obviously, compared with the control group. Their median survival was significantly prolonged. It is concluded that double suicide genes are more effective for killing effect on K562 cells in vivo and in ex vivo. It may be applicable to clinical gene therapy.

Abstract:Background To investigate the selective killing efficacy of the double suicide genes driven by KDR promoter. Materials and methods A double suicide gene system with the KDR promoter, pcDNA3-KDRp-CDglyTK, was constructed and transfected into lung cancer cell lines L9981 and NL9980, and human hepatocellular carcinoma cell line HepG2. The efficiency and specificity of the double suicide gene system were assayed by in vitro cellular proliferation and apoptosis, as well as in vivo xenograft studies. Results The transgenic CD and TK genes were only expressed in L9981 and NL9980 but not in HepG2 cells. Pre-treating transfected cells with 5-Fc and GCV significantly reduced proliferation, enhanced apoptosis in L9981 and NL9980 but not in HepG2 cells. The tumor formed by L9981 and NL9980 cells with the double suicide gene system was much smaller in vivo. Conclusion Tumor targeted expression of CDglyTK gene driven by KDR promotor represents a novel strategy for effective gene therapy of tumor with intrinsic KDR.
Activated CD4+ T Cells As Radiosensitizer: Enhancement of Irradiation-Induced Cytotoxicity by Pretreatment of Tumor Cell with Activated CD4+ T Cells

Abstract:3116 Background: Radiotherapy is one of the standard modalities for the treatment of cancer. Irradiation causes not only tumor cell death, but also can up-regulate tumor antigen and MHC class I molecule expression, thereby promoting the accumulation and activation of immune cells in tumor tissue. We hypothesize that activated CD4+ T cells (aCD4) can presensitize tumor cells and enhance gamma-irradiation -induced cytotoxicity. The rationale is that aCD4 release multiple cytokines, which may regulate gene-expression in tumor cells, thereby affecting their proliferation and apoptosis. Methods: Human cervical carcinoma cell line Hela was used as our initial tumor model. Hela cells were irradiated with 4 Gy after presensitization with aCD4 for 48 hours. The cells were cultured for 2 or 5 more days for apoptosis and cell proliferation assays, respectively. Transwell system was used to determine whether cell contact, soluble factors released by aCD4, or both were essential in this model. Several cytokines including IFN- γ were used to replace aCD4 to determine the contribution of each cytokine. Other cancer cell lines used to validate the strategy included cervical carcinoma cell line Caski, glioma cell lines LN229 and U373, and prostate carcinoma cell line DU145. Results: For Hela cells, growth inhibition was 25.8 ±3.3% in group treated with irradiation alone, 31.5±11.3% in group treated with aCD4 alone, and 60.5±1.3% in the combination group; while the percentage of apoptotic cells as determined by flow cytometry assay using Annexin V was 7.15±0.6%, 9.25±0.4% and 15.65±4.6%, respectively. Similar results were also obtained with other cancer cell lines. For example, for DU145, cell growth inhibition was 35.4±4.0 % in group treated with irradiation alone, 32.65±2.7% in group treated with aCD4 alone, and 70.3±0.14% in the combination group. This effect was aCD4 and irradiation dose dependent. Transwell experiments showed that soluble factors secreted by aCD4 were responsible for the observed activity. Of the cytokines tested, IFN-γ was found to play major role in our model. Growth inhibition was 10.15±1.0% in group treated with IFN- γ alone and 57.03±1.9% in the IFN- γ and gamma-irradiation combination group. Conclusion: We have demonstrated that aCD4 can act as a radiosensitizer to enhance the cytotoxic effect of gamma irradiation in multiple cancer cell lines. Soluble factors released by aCD4 were essential in this effect. IFN- γ was found to play a major role. We are currently working on the mechanism underlying the observed activity. In a separate project, we have also demonstrated great synergism when chemotherapy was given after tumor cells had been presensitized with activated immune cells.
The combination of suicide gene therapy and radiation enhances the killing of nasopharyngeal carcinoma xenographs

Abstract:Nasopharyngeal carcinoma (NPC) is very common in Southern China and Southeast Asian countries. To explore a novel and more effective approach to NPC therapy, a combined strategy of suicide genes and radiation was designed in this study. Five suicide gene expression cassettes, yeast CD, yeast CD/UPRT, and yeast CDglyTK gene controlled by CMV, and Egr-1 and a synthetic CMV-enhanced Egr-1 promoter (CE) were constructed in an expression vector p11MS. The expression of suicide genes in NPC CNE-2 cells were detected by RT-PCR and Western blot. The cytotoxicity of suicide gene therapy and radiation were analyzed by MTT assay. An animal study in which yeast CD/UPRT-expressing CNE-2 tumors in nude mice were treated with 5-FC and radiation was also developed. Our results revealed that p11MSCEyCD/UPRT and p11MSCEyCDglyTK are superior over three other constructs in the killing of NPC cells in vitro. We combined suicide gene-expressing tumors, 5-FC treatment, and radiation in vivo and found that the tumors greatly regressed, some disappeared completely in 3 nude mice in the yCD/UPRT group, and a significant difference of tumor volumes was observed between this group and the other four groups (p < 0.05). Our results indicated that suicide gene therapy and radiation have a synergic effect on NPC therapy, and the combined strategy of radiogene therapy is of great potential as a substitute for the traditional method, radiation alone, in NPC therapies.
The Effect of RAC2 on Radiosensitivity of Melanoma Cells

Abstract:Malignant melanoma is characterized by high degree of malignancy, early metastasis and high mortality. However,it is indicated by clinical experience that malignant melanoma is resistant to conventional radiotherapy. There-fore, it is of great significance to radio-sensitize melanoma cells in the treatment of this kind of malignancy. In the pres-ent study, the melanoma cell lines with inducible RAC2 over-expression or RAC2 knockdown were established. The effect of RAC2 on radiosensitivity of melanoma cells were evaluated by colony formation assay and γH2AX assay. Mean-while, the ROS yield and the NADPH oxidase activity were analyzed in the irradiated melanoma cells. We find that RAC2 over-expression sensitizes the melanoma cells by enhancing the NADPH oxidase activity and the subsequent ROS production.

Abstract:Background: To construct a recombinant adenovirus containing CDglyTK double suicide genes and evaluate the antitumor efficacy of the double suicide genes driven by kinase domain insert containing receptor (KDR) promoter on lung cancer cells. Methods: A double suicide gene system with the KDR promoter, pcDNA3-KDRp-CDglyTK, was constructed and transfected into lung cancer cell lines L9981 and NL9980, and human hepatocellular carcinoma cell line HepG2. The efficiency and specificity of the double suicide gene system were assayed by in vivo xenograft studies. Different injection techniques in vivo antitumor effects had also been compared. Results: The transgenic CD and TK genes were only expressed in L9981 and NL9980 but not in HepG2 cells. It also showed the evident tumor inhibiting ability by intratumoral injection than tail vein injection in the highly metastatic human large cell lung cancer cell line L9981, which displayed higher inhibitory rate than in the less metastatic human lung cell line NL9980. But, no effect was shown on the tumors from the hepatic carcinoma HepG2 cells. Conclusions: the KDR promoter is capable of regulating a double suicide gene system in human lung cancer cells, thus providing laboratory evidence to develop a gene therapy approach against various cancers. expression of CDglyTK genes under the control of KDR promotor represents a new strategy for effective gene therapy of tumors expressing intrinsic KDR.

Abstract:Cytolethal distending toxin (CDT) produced by Campylobacter jejuni is a genotoxin that induces cell-cycle arrest and apoptosis in mammalian cells. Recent studies have demonstrated that prostate cancer (PCa) cells can acquire radio-resistance when DOC-2/DAB2 interactive protein (DAB2IP) is downregulated. In this study, we showed that CDT could induce cell death in DAB2IP-deficient PCa cells. A combination of CDT and radiotherapy significantly elicited cell death in DAB2IP-deficient PCa cells by inhibiting the repair of ionizing radiation (IR)-induced DNA double-strand break (DSB) during G2/M arrest, which is triggered by ataxia telangiectasia mutated (ATM)-dependent DNA damage checkpoint responses. We also found that CDT administration significantly increased the efficacy of radiotherapy in a xenograft mouse model. These results indicate that CDT can be a potent therapeutic agent for radio-resistant PCa.
Improved Effects of a Double Suicide Gene System on Prostate Cancer Cells by Targeted Regulation of Prostate-Specific Membrane Antigen Promoter and Enhancer

Abstract:Objective: To explore the specific killing effect on prostate cancer cells of a dual cytosine deaminase (CD) and uracil phosphoribosyltransferase (UPRT) expression plasmid system controlled by the prostate-specific membrane antigen (PSMA) promoter and enhancer. Methods: The CD gene was used to construct the recombinant plasmid prostate-specific membrane antigen(promoter/enhancer)-CD (pPSMA(E/P)-CD). The specific regulatory function of the pPSMA(E/P) promoter was demonstrated by detection of enhanced green fluorescent protein (EGFP) expression in the LNCaP cell line. Survival of cells transfected with different plasmids and treated with 5-fluorocytosine (5-FC) was measured by microculture tetrazolium assay. Cell cycle changes were measured by flow cytometry. Results: Target-specific expression of PSMA(E/P) was observed in the prostate cancer cell line. Cytotoxicity of 5-FC was greater against LNCaP cells transfected with pPSMA(E/P)-CD and UPRT and pPSMA(E/P)-CD than control groups. Percentages of cells in S phase were 37.5% (LNCaP) and 30.6% (5-FC treatment) in the un-transfected groups, whereas they were 23.9% and 12.4% in the double and single suicide gene groups, respectively. Conclusions: Our findings confirm the cytotoxic efficacy of the pPSMA(E/P)-CD + 5-FC and pPSMA(E/P)-CD and UPRT + 5-FC suicide gene systems. The CD and UPRT gene system quickly and directly converted 5-FC into 5-FU, and then into toxic metabolites. The CD and UPRT double suicide gene system was more effective in inducing tumor cell apoptosis with 5-FC than the single suicide gene system. Thus, this construct can specifically target prostate cancer cells and might have a role in gene therapy against prostate cancer.

Abstract:Objective To investigate the different killing effect to human pulmonary adenocarcinoma cell line cells GLC-82 with coexpressed double suicide genes compared with single gene. Methods Recombinant expression vectors containing CD (cytosine deaminase) and/or TK(thymidine kinase) gene under CMV promoter were constructed successfully. The vectors were transfected to GLC-82 tumor cell lines by use of lipofectamine. The clones were picked out after G418 selection. The cytotoxicity to these transgenic cells under treatment with 5-Fc and GCV were measured by MT1 assays. Results Double and single suicide gene transfer were both stably expressed in GLC-82 cells . The cytotoxic effects of co-expressed TK-CD genes were superior than that of the single gene. Conclusion The CD + TK/5-Fc + GCV coexpression system is more effective for killing effect of tumor cells than CD/5-Fc or TK/GCV system alone.
Repressing Effects of Double Suicide Genes System on Human Cholangiocarcinoma in Vivo and in Vitro

Abstract:Objective To investigate the repressing effects of cytosine deaminase(CD) and herpes simplex(virus) thymidine kinase(HSV-tk) double suicide genes coexpression system on human cholangiocarcinoma QBC939 cells and QBC939 cells transplantated tumor in nude mice.Methods CD and HSV-tk double suicide genes were transfered into QBC939 cells using liposomes.After G418 selection,the positive clones of QBC939/CD+tk cells were picked up and cultured.The expression of CD and HSV-tk genes was(confirmed) by RT-PCR.In vitro,the QBC939/CD+tk cells were treated with 5-Fc and/or GCV,and the cytoxicity efficacy was evaluated by microculture tetrajolium test(MTT) method.The QBC939/CD+tk cells were inoculated subcutaneously into nude mice,and when the tumors were palpable,5-Fc and GCV were(injected) intraperitoneally,and the volumes of transplantated tumors were measured before and after medication.Results Double suicide genes were stably expressed in QBC939/CD+tk cells.The repressing capability of combination of 5-Fc and GCV on QBC939/CD+tk cells was more effective than that of using either 5-Fc or GCV alone.The increase of cell-repressing was assaciated with increase the concentration of the prodrug.The repressing effect of combination of the 2 prodrugs on early period of transplantation tumor was obvious,even complete abolition of tumor was noted,and moreover a marked local bystander effect was observed.(Conclusions) In vitro and in vivo,the cell-repressing efficacy of double suicide gene system on cholongiocarcinoma cells and the tramsplanted tumor of nude mice was significant,and the bystander effect was obvious.
Targeted Killing Effects of Double Suicide Genes Controlled by the MDR1 Promoter on C6/ADR Cells

Abstract:Objective To investigate the targeted killing effects of a double suicide gene system controlled by the MDR1 promoter with the prodrug GCV+5-FC on C6/ADR cells.Methods The recombinant vector pcDNA3.MDR1P.CD.TK containing CD and TK double suicide genes controlled by the MDR1 promoter was transfected into C6/ADR cells by using liposome.PCR and RT-PCR were used to identify the integration and expressions of the CD and TK genes.This recombinant vector-transfected C6 cells(C6/CDTK) and normal C6/ADR cells were set as controls.After adding the prodrug,the effects of double suicide genes on cell growth,cell cycle and proliferation were determined by cell growth curve,flow cytometry(FCM) and plate colony formation assay.Results PCR proved double suicide genes were integrated into C6/ADR and C6 cells.RT-PCR revealed that the CD and TK genes were expressed in C6/ADR/CDTK cells whereas not in C6/CDTK cells.After using GCV+5-FC,the growth of C6/ADR/CDTK cells was inhibited.G1 phase proportion was significantly higher in C6/ADR/CDTK cells(99.93%) than in C6/ADR cells and C6/CDTK cells(32.68% and 47.57%,P0.05);the formation rate was significantly lower in C6/ADR/CDTK cells [(16.7±0.9)%] than in C6/ADR cells and C6/CDTK cells [(96.7±2.1)% and(86.7±1.9)%,P0.05]. Conclusion The pcDNA3.MDR1P.CDTK/GCV+5-FC system has targeted killing effects in C6/ADR cells.

Abstract:Radiotherapy is an essential curative treatment modality for colorectal cancer. Apoptosis is the major mechanism of IR-induced cell death and aberrant apoptotic signaling results in radioresistance, which is a hallmark of most, perhaps all, types of human cancers. Potentiating the induction of apoptosis is an emerging strategy for cancer radiotherapy. Here, we determined that targeting CDK8 selectively radiosensitized colorectal cancer through the mitochondria-dependent intrinsic apoptotic signaling, which was mediated through the induction of the transcription of apaf1 that was e2f1- and not p53-dependent. Importantly, the enhanced transcriptional activity of e2f1 was dependent on the kinase activity of CDK8 itself and not on the assembling of the mediator complex. In addition, clinical inhibitor, and in vivo studies confirmed the radiosensitizing effect of CDK8. Our results provide a new targeting strategy to improve the radiotherapy of CRC.

Abstract:OBJECTIVE:To explore the therapeutic efficacy of double suicide gene system driven by carcinoembryonic antigen (CEA) promoter (Cp-CDglyTK) on colorectal carcinoma xenograft in nude mice. METHODS:The plasmid pcDNA3.1(-)Cp-CDglyTK was transfected into the CEA-positive SW480 and CEA-negative HeLa cells respectively. The expression of suicide gene was detected by RT-PCR. And the transfected cells were treated with 5-fluorocytosine (5-FC) and ganciclovir (GCV) at different concentrations and the cell-killing and bystander effects assayed by methyl thiazolyl tetrazolium (MTT). By a transplantation of cultivated cells, SW480 or HeLa cell lines were injected subcutaneously into right axillary of nude mice to establish 96 SW480 and 72 HeLa tumor animal models. Nude mice were completely randomized with statistical software according to tumor volume. For prodrug therapy, 48 SW480-bearing mice were divided equally into 4 groups of I-IV. At the same time, 48 HeLa-bearing mice were divided equally into 4 groups of V-VIII. Groups I & V received an intratumoral injection of PBS, groups II & VIGCV and 5-FC intratumorally, groups III & VII PBS intraperitoneally and groups IV & VIII GCV and 5-FC intraperitoneally. Forty-eight SW480-bearing mice were divided equally into 4 groups of IX∼XII and 24 Hela-bearing ones into groups of & in therapy experiment by suicide gene plus prodrug. Six groups received an intratumoral injection of liposome Lipofectamine and plasmid CP-CDglyTK and then an intraperitoneal injection of drug. The groups of IX and received an injection of PBS, group X GCV, group XI 5-FC and groups XII & GCV and 5-FC. The observation parameters included tumor bulk, tumor weight, survival time and treatment effect in each group. RESULTS:SW480 cells transfected by plasmid pcDNA3.1(-)Cp- CDglyTK expressed CDglyTK gene. The inhibition rates of GCV and 5-FC were significantly higher than those of HeLa cells (59.87% ± 0.21% vs 9.90% ± 0.09%, P < 0.01). And higher inhibition rates and stronger bystander effect existed in double versus single produg (all P < 0.05). Tumor size, final tumor weight and survival time of nude mice in groups ofII, IV, VI & VIII had no significant difference with groups ofI, III, V & VII (all P < 0.05). Final tumor size and weight of group XII was significantly smaller than those of groups of IX, X and XI ((150.0 ± 3.2) vs (522.5 ± 1.9) and (256.8 ± 10.4) and (260.7 ± 2.2) mm(3), (54.1 ± 10.4) vs (682.0 ± 12.0) and (251.8 ± 15.1) and (271.6 ± 17.7) mg, all P < 0.05). Meanwhile, the tumor inhibition rate and survival time of group XII(92.1% and (25.7 ± 0.8)d) were significant higher and longer than group X (63.1% and (21.8 ± 0.5) d) and group XI (60.2% and (18.0 ± 0.9) d) (all P < 0.05). However, no significant difference existed in tumor size, final tumor weight and survival time between groups and (all P > 0.05). The inhibition rate of group was merely 0.9%. CONCLUSION:CDglyTK double suicide gene system driven by CEA promoter may inhibit CEA positive colorectal cancer xenograft in prodrug-treated nude mice.
