Establishment of the Suspension Culture and Regeneration System of Carrot

Abstract:The tap roots of carrot were used as the material to induce the calli and to establish the suspension cell culture system,then the calli and the cells of suspension culture were transferred on differentiation media for regeneration. The results showed that the inductivity of calli of carrot tap roots was 100 %,the best hormone and its concentration was 2,4-D and 0.1 mg/L.The calli could be approximately divided into four types by the external appearance: the type I was the optimal material for the establishment of the suspension cell culture,and the type Ⅲ could be used to establish the suspension cell culture after subculture.The absolute-rest precipitation method was found to be the simple and effective way to subculture the cell suspension system. The suitable medium for regeneration was MS or B5 basic medium without any hormone and the regenerated plant on MS could simultaneously develop the roots and shoot,the however,the rooting of the seedlings on B5 medium was very difficult on the original medium and must be transferred on MS medium to develop root.
