Optical Molecular Imaging of Lipopolysaccharides-induced NO Production in Plant Cells

Abstract:Lipopolysaccharides ( LPS),which are the main components of Gram-negative bacterial cell wall,could be sensed by plants to initiate plant defense responses.Lsing fluorescent probes molecular imaging and laser confocal scanning microscopy,the temporal and spatial characteristics of nitric oxide ( NO) production induced by LPS in Arabidopsis protoplasts were investigated in vivo and in real-time.After 90 min treatment with 100 μg/ml LPS,a strong NO accumulation at the cell membrane of the living Arabidopsis protoplasts could be detected at the population or single cell level. Moreover,the LPS-induced NO production was sensitive to L-monomethyl-L-arginine,which is a widely used inhibitor of NO synthase,indicating that the NO production in response to LPS is NO synthase-dependent.The results positively contribute to better understanding the mechanisms of LPS action and the NO signal transduction pathway,and may provide some reference about the application of biophysical techniques in related plant physiology research.
