Abstract:A new aristololactam, 5-methoxyaristololactam I (1), was isolated from an ethanol extract of the whole plant of Asarum ichangense C. Y. Cheng et C. S. Yang, together with twenty known compounds (2-21). The structure of 1 was elucidated by spectroscopic methods. Compounds 11, 13, 17, and 19 were isolated from Aristolochiaceae for the first time. Compounds 2, 14, and 15 are considered as common constituents of the genus Asarum.

Abstract:To study the chemical constituents of Asarum himalaicum, fifteen compounds were isolated from a 70% ethanol extract by using a combination of various chromatographic techniques including column chromatography over silica gel, Sephadex LH-20, and semi-preparative HPLC. By spectroscopic techniques including 1H NMR, 13C NMR, and HR-ESI-MS, these compounds were identified as 4-demethoxyaristolochic acid BII (1), aristolochic acid I (2), aristolochic acid Ia (3), 7-hydroxyaristolochic acid I (4), aristolochic acid IV (5), aristolic acid II (6), debilic acid (7), aristololactam I (8), 9-hydroxyaristololactam I (9), 7-methoxyaristololactam IV (10), (2S)-narigenin-5, 7-di-O-beta-D-pyranosylglucoside (11), 4-hydroxybenzoic acid (12), 3, 4-dihydroxybenzoic acid (13), 4-hydroxycinnamic acid (14), and beta-sitosterol (15). All of these compounds (1-15) were obtained from A. himalaicum for the first time. Among them, 1 was identified as a new compound, and compounds 3-6, 9, 12-14 were isolated from Asarum genus for the first time. Since the kidney toxicity of aristolochic acids and aristololactams has been reported, the result of this investigation suggests that it should be cautioned to use A. himalaicum as a medicine.

Abstract:Two novel phenanthrene derivatives, aristololactam IVa (1) and 9-hydroxy aristolochic acid I (2) were isolated from the stem and leaves of Anstolochia contorta Bunge, together with 17 known compounds (3-19). The structures of these compounds were determined by spectroscopic analysis. The phenanthrenes obtained were tested for cytotoxicity against renal proximal tubular epithelial cell line (HK-2). Aristololactam IVa and 7-methoxy aristololactam IV were found to have strong cytotoxic activity against HK-2 cells with a potency similar to or even stronger than those of aristolochic acid I and aristololactam I.

Abstract:More than 80 aristolochic acids (AAs) and aristololactams (ALs) have been found in plants of the Aristolochiaceae family, but relatively few have been fully studied. The present study aimed at developing and validating a liquid chromatography tandem mass spectrometry (LC/MSn) for the analysis of these compounds. We characterized the fragmentation behaviors of 31 AAs, ALs, and their analogues via high performance liquid chromatography coupled with electrospray ionization mass spectrometry. We summarized their fragmentation rules and used these rules to identify the constituents contained in Aristolochia contorta, Ar. debilis, Ar. manshurensis, Ar. fangchi, Ar. cinnabarina, and Ar. mollissima. The AAs and ALs showed very different MS behaviors. In MS1 of AAs, the characteristic pseudomolecular ions were [M + NH4](+), [M + H](+), and [M + H - H2O](+). However, only [M + H](+) was found in the MS1 of ALs, which was simpler than that of AAs. Distinct MSn fragmentation patterns were found for AAs and ALs, showing the same skeleton among the different substituent groups. The distribution of the 31 constituents in the 6 species of Aristolochia genus was reported for the first time. 25 Analogues of AAs and ALs were detected in this genus. A hierarchical schemes and a calculating formula of the molecular formula of these nitrophenanthrene carboxylic acids and their lactams were proposed. In conclusion, this method could be applied to identification of similar unknown constituents in other plants.

Abstract:Five new phenanthrene derivatives: 9-ethoxy-7-methoxy-aristololactam IV (1), norcepharadione A N-beta-D-glucopyranoside (2), aristololactamoside I (3), aristololactamoside II (4) and aristothiolactoside (5) together with eleven known phenanthrene derivatives (6-16) were isolated from the ethanol extract of the roots and rhizomes of Asarum heterotropoides var. mandshuricum. The aristololactams with substitution of ethoxy at C-9 position (1, 9, and 10) and the sulfur-containing phenanthrene derivative (5) were reported in the genus Asarum for the first time. Furthermore, six phenanthrene glucoside derivatives (2-5, 13 and 14) were also found in this genus for the first time and compounds 7 and 9-15 were isolated from the genus Asarum for the first time. Six of them (1, 2, 9, 10, 13 and 14) were submitted to cytotoxicity test against human renal proximal tubular epithelial cell lines (HK-2) using mu and LDH assays. Compounds 1 and 10 showed significant cytotoxic activity against HK-2 cell lines with IC50 values of 18.18 and 20.44 mu mol/L in MIT assay and 8436 and 35.06 mu mol/L in LDH assay, respectively. Compound 9 showed moderate cytotoxicity in MIT assay with IC50 values of 95.60 mu mol/L, but no cytotoxicity in LDH assay. Compounds 2, 13 and 14 showed cytotoxic effect in neither MIT assay nor LDH assay. Considering the other nephrotoxic phenanthrene derivatives (6, 8, 12, 15 and 16) previously tested, the results implied the potency of renal toxicity of this herb used as a medicine. Studies on Morphology and Aristolochic Acid Analogue Constituents of Asarum Campaniflorum and a Comparison with Two Official Species of Asari Radix Et Rhizoma

Abstract:Dried whole herbs or roots and rhizomes of Asarum campaniflorum have been sold under the trade name Xixin and used as folk remedies in its producing areas for a long time. In order to avoid the misuse of A. campaniflorum as official species of Xixin (Asari Radix et Rhizoma), a comparative study based on the morphological and phytochemical analysis of the aerial and underground parts was carried out. The usual morphological methods and a microscopic imaging system were used. The results show that A. campaniflorum could be easily distinguished from two official species (Asarum sieboldii and A. heterotropoides var. mandshuricum) by the diameter of thicker roots (1.3–2.7 mm), distinct large parenchymatous cells in phloem of roots, and the size of oil cells in upper leaf epidermises [(40)80–140(174) µm in diameter, where numbers in parentheses are for rare cases], etc. Nine aristolochic acid analogues (AAAs) were identified and estimated by high-performance liquid chromatography–diode array detection (HPLC-DAD). The aerial and underground parts of A. campaniflorum contained 3–4 AAAs, i.e., aristololactam-II-N-β-d-glucoside (AL-II-Glc), aristololactam I (AL-I), aristololactam II (AL-II), and aristolochic acid I (AA-I), while only AL-I was detected in underground parts of A. sieboldii and no AAAs were detected in underground parts of A. heterotropoides var. mandshuricum. The respective contents (in mg/g) in aerial and underground parts of A. campaniflorum, were as follows: AL-I, 0.06–0.12, 0.05–0.10; AL-II, 0.03–0.04, 0.01–0.03; AA-I, 0.01–0.02, 0.0–0.0. These data suggest A. campaniflorum has a high risk of causing aristolochic acid nephropathy. All these discoveries can contribute to not only the better understanding of this new resource species, but also the safe use of the crude drug Xixin.

Abstract:Two aristololactams, aristololactam GI (1) and aristololactam GII (2), and three aporphines, fissistigamide A (3), fissistigamide B (4) and fissistigmine (5), together with nineteen known alkaloids, one flavone and one anthraquinone were isolated from the ethanol extracts of the stems of Fissistigma oldhamii (Annonaceae). Their structures were elucidated primarily by analysis of NMR, IR, UV, MS and CD data. Alkaloid 1 is a chiral aristololactam formed from a phenylpropanoid derivative attached to a 3,4-dihydroxy aristololactam scaffold. The absolute configuration of 1 was determined by comparing experimental and calculated ECD spectra. The anti-inflammatory activity of the crude extracts and the five alkaloids were tested by measuring the amount of TNF-α and IL-6 released from LPS stimulated RAW264 cell via ELISA. The results demonstrated that the CHCl(3)-soluble part and alkaloid 2 exhibited significant anti-inflammatory activity in vitro in both assays.

Abstract:Twelve compounds, including two new aristolochic acid analogues with a formyloxy moiety (9–10) and 10 known aristolochic acid derivates (1–8 and 11–12), were obtained from the roots of Aristolochiacontorta. Their structures were elucidated using extensive spectroscopic methods. Their cytotoxic activity in human proximal tubular cells HK-2 was evaluated by the MTT method, which has been widely used to assess cell viability. Among these molecules, compounds 3 and 9 were found to be more cytotoxic. Furthermore, molecular modeling was used to evaluate, for the first time, the interactions of compounds 3 and 9 with the target protein organic anionic transporter 1 (OAT1) that plays a key role in mediating aristolochic acid nephropathy. Structure–activity relationships are briefly discussed.

Abstract:From the ethyl acetate extract of Asarum forbesii Maxin, four new constituents, asarumin A(I), B(II), C(III) and D(IV), were isolated along with elemicin (V), trans-asarone(VI) and linoleic acid(VII). The structures of the new compounds were elucidated as methyl 3 S-benzoyloxy-2 S-hydroxy-2-isopropylbutyrate for I, methyl 2 R-benzoyloxyisopentanoate for II, methyl 2 R-trans-cinnamoyloxyisopentanoate for III and methyl 2 R-piperonyloyloxyisopentanoate for IV. Compounds I, II, III and VII showed weak inhibition of PCA in rats, but the other compounds were inactive.

Abstract:Abstract Antidesma ghaesembilla is an important medicinal and food plant in many Asian countries. Ten substances could be isolated from the dichloromethane and methanol extract: sitostenone (3), daucosterol (4), chavibetol (5), asperphenamate (6), protocatechuic acid (7), vanillic acid-4-O-β-D-glucoside (8), 1-O-β-D-glucopyranosyl-3-O-methyl-phloroglucinol (9), and aristolic acid II-8-O-β-D-glucoside (10), and two new aristolic acid derivatives, 10-amino-5,7-dimethoxy-aristolic acid II (= 6-amino-9,11-dimethoxyphenanthro[3,4-d]-1,3-dioxole-5-carboxylic acid; 1) and 5,7-dimethoxy-aristolochic acid II (= 9,11-dimethoxy-6-nitrophenantro[3,4-d]-1,3-dioxole-5-carboxylic acid; 2). Exposure to humans of some of these compounds is associated with a severe disease today known as aristolochic acid nephropathy. Therefore, the traditional usage of this plant has to be reconsidered carefully.

[Study on preparation method of reference substance of aristolactam Ⅰ-deoxyriboside adducts]

Abstract:A two-step synthetic process of bromination and cross-coupling with aristololactam Ⅰ as raw material was successfully developed. Three aristolactam Ⅰ-deoxyriboside adducts, namely AAⅠ-dA, AAⅠ-dG, and AAⅠ-dC were obtained after a sequential procedure of impurity removal and purification in four different solvents. The yield of the two-step reaction can reach 90%, and the purity of the product is more than 98%, which can meet the requirements of qualitative and quantitative analyses as traditional Chinese medicine chemical reference products. The process has been proven to have good repeatability and scalability, and it features a concise preparation procedure, efficient purification, and high yield and purity, requiring no chromatographic separation. Compared with pre-vious methods, the newly developed process has significant advantages and is suitable for the preparation of chemical reference products of aristolactam Ⅰ-deoxyriboside adducts. This process provides technical support for the preparation of reference products of aristolactam Ⅰ-deoxyriboside adducts and a solid material basis for the related toxicological research.

Abstract:A new antimicrobial eudesmanolide, 1-oxo-8 alpha-hydroxy-11 alpha H-eudesm-4-en-12,6 alpha-olide (1), was isolated from a medicinal plant Artemisia giraldii and its structure was elucidated by spectroscopic methods (IR, MS, H-1 NMR, C-13 NMR, H-1-H-1 COSY and NOESY). Antimicrobial bioassay indicated that this compound inhibited the growth of human pathogenic bacteria Staphylococcus auerus, Bacillus subtilis and Escherichia coli as well as human opportunistic pathogenic fungi Candida tropicalis, Gecotrichum candidun, Aspergillus flavus and A. nigers.

Abstract:A new sesquiterpenoid derivative, named aspergiketone (1), along with seven known compounds (2-8) were isolated from the coastal saline soil fungus Aspergillus fumigatus. Their structures were elucidated by spectroscopic analysis, and by comparison of experimental and reported data. The absolute configuration of compound 1 was defined by X-ray diffraction analysis. Compound 1 was cytotoxic towards HL-60 and A549 cell lines with IC50 values of 12.4 and 22.1 mu M, respectively.

Abstract:Aristolochic acid analogues (AAAs) are well-known toxins. We performed the first comprehensive screening on AAAs in Asari Radix et Rhizoma (underground part of Asarum heterotropoides Schmidt), the only Aristolochiaceae plant widely used in clinical practice. LC-HRMS revealed 70 trace AAAs using polygonal mass defect filtering and precursor ion list strategies, 38 of which were newly discovered in A. heterotropoides. UHPLC-QTrap-MS/MS was then utilized for quantitative/semiquantitative analysis of 26 abundant compounds. Seventeen AAAs were detected from 91 batches of A. heterotropoides and 20 AAAs from 166 consumable products. For 141 Asari-containing proprietary products, aristolactam I and aristolactam II-glucoside exhibited the widest distribution, present in 98% products. AA IVa was the most abundant, detected in 91%. Notably, 60% of the products contained AA I (0.03-0.79 ppm). The safety was assessed using linear extrapolation, permitted daily exposure, cumulative amount, and the margin of exposure. It is recommended that AA I content be limited to 3 ppm.

Abstract:Marine-derived fungi have been reported to have great potential to produce structurally unique metabolites. Our investigation on secondary metabolites from marine-derived fungi resulted in the isolation of seven new polyketides (phomopsiketones D–G (1–4) and letendronols A–C (5–7)) as well as one known xylarinol (8) in the cultural broth of Letendraea sp. Their structures and absolute configurations were elucidated using a set of spectroscopic and chemical methods, including HRESIMS, NMR, single-crystal X-ray diffraction, ECD calculation, and a modified version of Mosher’s method. Compound 2 showed weak inhibition against nitric oxide production in lipopolysaccaride-activated macrophages with an IC50 value of 86 μM.

Abstract:Elansolid A is a structurally complex polyketide macrolactone natural product that exhibits promising antibacterial properties. Its challenging asymmetric total synthesis was achieved by a convergent strategy, in which the tetrahydroindane core of the molecule and an eastern vinyl iodide moiety were combined as the main fragments. The central tetrahydroindane motif was constructed with high stereoselectivity by a bioinspired intramolecular Diels–Alder cycloaddition, generating four stereogenic centers in a single step. The stereocontrol of this key step could be achieved by virtue of a 1,3-allylic strain generated by the temporary introduction of a steric-directing iodine substituent on the substrate. Abstract:Artesin A (1), a rare symmetrical dimeric guaianolide, was isolated from the aerial parts of Artemisia sieversiana. Its structure was determined by analysis of the spectroscopic data, including HRESIMS, 1D, and 2D NMR and the absolute configuration was resolved by single crystal X-ray diffraction. Artesin A exhibited a moderate inhibition effect on the LPS-induced nitric oxide production in BV-2 microglial cells with an IC50 value of 38.78μM.
