Abstract:This article summarizes the studies of transglutaminase home and broad and explains its mechanism,resource and application in diary products,especially its effects to protein,whey protein and casein in liquid milk and the amelioration to the quality of yoghourt.

Abstract:Cross-linking of casein and gelatin catalyzed by transglutaminase was studied in this work.When the ratio of casein to gelatin was fixed at 4∶1(w/w) and the content of hydroxyproline in the obtained product was used as index,the addition level of transglutaminase,reaction temperature and time were studied by single factor experiments.The optimal cross-linking conditions obtained were that protein concentration was fixed at 5%(w/v),enzyme addition level was 20 U/g proteins,reaction time was 4 h and reaction temperature was 45 ℃.SDS-PAGE analysis demonstrated that some protein polymers existed in the obtained product.Compared to casein and transglutaminase-induced cross-linked casein,the dispersion of the obtained product exhibited markedly improved apparent viscosity and viscoelastic behaviors.Our result indicated that cross-linking of casein and gelatin could improve their rheological properties.

Abstract:BACKGROUNDTransglutaminases catalyze post-translational modification of proteins by epsilon-(gamma-glutamyl) links and covalent amide bonds. Research on properties and applications of plant transglutaminases is less developed than in animals and micro-organisms. In a previous study, optimized Zea mays transglutaminase was purified from recombinant Pichia pastoris strain. The main objective of the present study was to characterize this enzyme and assess its effect on the properties of yoghurt.RESULTSThe purified recombinant transglutaminase presented a K-m of 3.98 mu mol L-1 and a V-max of 2711 min(-1) by the fluorometric method. The enzyme was stable after incubation for 30 min below 50 degrees C and over a broad pH range of 5-8 at -20 degrees C for 12 h. The results showed that the crosslinking reaction catalyzed by this enzyme could effectively improve the properties of full and non-fat yoghurts. Also, the properties of non-fat yoghurt could be improved similar to the full-fat product by recombinant transglutaminase.CONCLUSIONThe application of recombinant transglutaminase in yoghurt indicated that this enzyme could be used as a substitute for microbial transglutaminase in the production of yoghurt, thus providing experimental evidence for the future application of plant transglutaminases in the food industry. Abstract:Transglutaminase can generate covalent cross-links between protein molecules or intramolecular covalent cross-links to change functional properties of proteins, and is applied in industries of food processing, medicine, cosmetic and textile. Microbial transglutaminase has aroused much more attention because of its calcium-independent and broader substrate specificity. This article summarized the gene expression of microbial transglutaminase, reviewed the gene characteristics and enzymatic characteristics, analyzed the relationship between crystal structures and function, and introduced the advantages and problems of microbial transglutaminase production using genetic engineering technique. In order to improve the yield of the enzyme, the activity and thermo-stability, the suggestions about expression strategies and molecular modification of microbial transglutaminase were also provided.

Abstract:Using transglutaminase(TG)to cross-link the casein,the effect on the composition and textural characterize of resulted cheeses was studied.The results indicated that TG promoted the fermentation process and increased the recovery of protein and fat.The protein in whey decreased significantly with TG treated.The TG had important effect on the textural properties of resulted cheese.But there was minor influence of TG on the composition and textural properties when rennet presented.

Abstract:A gene from Zea mays coding a TGase was expressed in E. coli and identified by Western blotting. Under optimal expression conditions, the production and specific activity of refolded TGZ were 1.41 mg/L and 0.34 U/mg. The activated TGZ was employed to cross‐link milk proteins. The yoghurt treated with TGZ showed a lower syneresis, higher apparent viscosity and texture than untreated yoghurt. The properties of TGZ‐treated sample were better than those of MTG‐treated samples, so TGZ may have potential as an additive in yoghurt manufacture.

Abstract:As a biocatalyst,transglutaminase can catalyze cross-linking reaction between γ-carboxyamide groups of peptide-bound glutamine residues and the ε-amino group of lysine residues in proteins,then forming the covalent bonds of ε-(γ-glutamyl)-lysine in proteins.The cross-linking reaction had advantages of mild reactive conditions and substrate specialties,and markedly improved functional properties of various proteins,such as gel strength,water-holding capacity and thermal stability.Thus,transglutaminase has been widely used in the modification of various food proteins.In the paper,characteristics of transglutaminase from different sources and its applications in different food systems were introduced,and the studies on its catalysis in soy proteins,were summarized as well.

Abstract:Keeping the stability of yogurt during the fermentation, storage and transportation process is necessary to reduce the syneresis. To achieve this goal, the combination of the acid treatment and transglutaminase (TG) was proposed to improve the textural properties of milk acid gels. We investigated the synergistic effect of acid treatment and TG on the water holding capacity (WHC) and physical stability of milk acid gels. As the pH of the skimmed milk decreased from 6.7 to 5.2, the casein micelles dissociated into smaller particles, exposing more site for the crosslinking of TG. The WHC and stability of the TG-treated acid gels were significantly enhanced as the particle size of casein micelles decreased. Our results suggested that the combination of the treatment of acid and TG could significantly improve the textural properties of milk acid gels by the dissociation of casein micelles induced and increase in the covalent-crosslinking. This study provided a simple and novel method to improve the stability of the milk gels, which showed enormous potential for dairy processing.

Abstract:This paper mainly studied the characteritics of TG-K and TG-N apparently. The crude enzyme activity of TG-K and TG-N were 11.32U/ml and 9.41U/ml respectively. The results showed that the more separated and purification steps, the less of enzyme activity and protein contents, but the loweling level of TG-N was less than TG-K. The molecular weight of TG- K is 41.5 kDa and while that of TG-N 39.6kDa by SDS-PAGE. The different ionics have different effects on cross-linking casein of TG-K or TG-N. It has proved that TG-K is better to cross-linked casein observing the ultra structure of yoghourt by EM.
Effect of microbial transglutaminase crosslinking on the functional properties of yak caseins: a comparison with cow caseins

Abstract:Microbial transglutaminase (mTGase) crosslinking is a useful method for significantly improving the physical functionality of proteins. This study focused on the effect of mTGase modification on the functional properties of yak and cow caseins, including solubility, emulsifying property, water and oil absorption capacity, foaming property, and stability in the presence of Na + , Ca 2+ , and Mg 2+ . The results showed that the crosslinking degree of yak and cow caseins differed at the same mTGase concentration, but the change in functional properties of cow and yak caseins was similar. After mTGase crosslinking, the solubility of caseins decreased, while the emulsifying properties increased because of casein polymerization. MTGase crosslinking was very effective in improving water and oil absorption properties, stability in the presence of Na + , Ca 2+ , and Mg 2+ , and foaming activity, but did not affect the foam stability of cow acid caseins (caseins content of 94.22 %, w / w ) and yak acid caseins (caseins content of 94.28%, w / w ). The results could be used as a reference for selecting the proper modification method to improve the predetermined functional properties of caseins according to industrial processing requirements.

Abstract:Finding new crosslinking enzymes and elucidating internal cross-linking mode are beneficial to meeting the requirement of various protein substrates and processing conditions in food industry. Transglutaminase (TG) can lead to the covalent crosslink of ε-(γ-glutamine)-lysine isopeptidic bonds in proteins through acyl-transfer reaction. This work reported the characterization of transglutaminases (TGs) from four Bacillus strains with obvious differences in amino acid sequences and enzymatic properties. Among them, TG from B. clausii (BCLTG) displayed the highest optimal temperature (70 °C) and specific activity (282 U/mg). The molecular dynamics simulation indicated that enlargement of substrate accessible tunnel in BCLTG with the increased temperature might result in the easy approach of substrate molecule to the active center. Moreover, BCLTG could cause the crosslinking of bovine serum albumin and almost all protein constituents in whey protein and soy protein isolate, and the crosslinking path of BCLTG towards three proteins was revealed. This study provides an ideal candidate for proteins crosslinking and theoretical foundation and preliminary information for applying TG in food processing.
