Abstract:With the improvement of life quality,there has been an increasing demand on high quality and high safety food sweeteners. Sucralose is exactly such an ideal artificial sweetener,which has the features of non-nutritive,low-calorie,high sweetness,high safety,and good flavor. Therefore,it is extremely necessary to study the synthetic methods of sucralose. This paper mainly summarizes the recent progress in the synthetic methods of sucralose,and comprehensive evaluation is given. It is concluded that,while the omni-group protection method contains too many synthetic steps and the mono-group protection method of the acetic anhydride method and trimethyl orthoacetate method are still in development, the organotin method has fewer synthetic steps and the organotin can be recycled, therefore it is considered to be the most applicable in industrial production so far. On the other hand, chemical-enzyme synthetic method,especially the sucrose method,is believed to be the most promising one in the future due to its environment friendly and high selectivity aspect. In addition, there are some other synthetic methods,such as enzymatic hydrolysis of sucralose-6-ester,etc.,worthy of further research. Finally,the research trends are also discussed and several development suggestions are proposed.
An in Vivo Bioelectronic Tongue for Sweetness Detection Using Rat Gustatory Perception Based on Brain-Computer Interface

Abstract:The detection of sweetness is of great significance due to flavor and health concerns. In this study, an in vivo bioelectronic tongue for sweetness detection is constructed using rat gustatory system and implantable microelectrode array. By recording extracellular potentials in “sweet hotspot” within rat gustatory cortex, sweetener-evoked response patterns are generated by neural decoding algorithms. The results show that sweetener stimulation can cause a significant increase in the power spectral density of the local field potentials signal at 50-80 Hz, and its dynamic response in the time domain can be used for identification and classification of sweetness. Meanwhile, quantitative study shows that power of response signal is dose-dependent and the detection range covers sugar contents of daily beverages. Comparisons with results of human taste panelists also confirm that this in vivo bioelectronic tongue can work across species and has the potential to mimic human taste and evaluate sweetness in practical applications.

Abstract:We discussed the biological mechanism of taste, studied the artificial taste system through mimicking of biological taste structure as well as the sensitive characteristics and selectivity of artificial lipid membranes. We designed a multi-channel taste sensor array based on biological lipid materials. Abstract:Sweet taste detection is of great significance in fields of disease prevention, food safety inspection, and drug regulation. In this study, a novel biomimetic sensor was constructed for sweet taste detection by compositing graphene oxide, hemin, and 3-aminophenylboronic acid (APBA). First, optimal fabrication of the sensor was investigated by comparing different composition proportions of graphene oxide and APBA functionalized hemin in saccharide measurements. Then, typical substances of natural sugars (i.e., glucose and fructose) and artificial sweeteners (i.e., saccharin and aspartame) were measured with the sensor through optical absorption spectroscopy. The spectra displayed apparent but different absorbance changes in the presence of those two kinds of saccharides, showing different sensing mechanisms of the biomimetic sensor for sugars and sweeteners like sweet receptors. Finally, dose-dependent and specific responses of the sensor were observed by comparing with other taste substances. The results suggested that the sensor can detect and distinguish natural sugars and artificial sweeteners in a biomimetic way. The research provided a low-cost, long-lifetime, and efficient approach for sweet taste detection, indicating its promising applications in fields such as food and beverage analysis. Abstract:The intrinsic perception process of taste is obviously far less known than those of vision, audition, touch and olfaction. Despite that taste cells utilize a variety of sensory mechanisms to translate plenty of gustatory sensations such as sour, sweet, bitter, salty and umami into cellular signals, gustatory perception mechanisms are still under exploration due to the lack of effective methods on cellular and molecular level. Recently the development of molecular biological and electrophysiological studies has promoted exploration of olfactory and gustatory transduction and coding mechanisms dramatically. Based on the studies of artificial olfaction, artificial taste and cell-based biosensor in our laboratory, this paper reviews the current research on taste transduction mechanism. We introduce the recent advances in cell chip that combined biology with microelectronics, discuss taste cell chip as well as its potential of prospective application in taste transduction mechanism in detail and propose the research trends of taste chip in future.
ChemSweet: An AI-driven computational platform for next-gen sweetener discovery

Abstract:Nowadays, the overconsumption of artificial sweeteners and their related adverse health impacts have proposed an urgent need to develop safe and healthy alternatives. Herein, we introduce ChemSweet, an AI-based platform for the rapid discovery of potential sweet molecules (http://chemsweet.ddai.tech) with the consideration of their physicochemical properties, sweetness profile, and health risks at the same time. Machine learning prediction models of four important physicochemical and four toxicity properties were established and integrated with the platform to evaluate the candidate molecules' biosafety and stability during the processing processes. Then, a new sweet taste prediction system was developed which ensures the sweet evaluation of six specific kinds of sweeteners. To facilitate the practical application of ChemSweet, the SuperNatural database was integrated for the rational screening of promising new sweeteners. We successfully identified 294 potential sweeteners that simultaneously meet the multiple anticipated criteria. We believe that ChemSweet will serve as a useful tool for identifying safe and healthy sweeteners while reducing the timeframe and high experimental costs.
Effects of the Artificial Sweetener Neotame on the Gut Microbiome and Fecal Metabolites in Mice

Abstract:Although artificial sweeteners are widely used in food industry, their effects on human health remain a controversy. It is known that the gut microbiota plays a key role in human metabolism and recent studies indicated that some artificial sweeteners such as saccharin could perturb gut microbiome and further affect host health, such as inducing glucose intolerance. Neotame is a relatively new low-caloric and high-intensity artificial sweetener, approved by FDA in 2002. However, the specific effects of neotame on gut bacteria are still unknown. In this study, we combined high-throughput sequencing and gas chromatography-mass spectrometry (GC-MS) metabolomics to investigate the effects of neotame on the gut microbiome and fecal metabolite profiles of CD-1 mice. We found that a four-week neotame consumption reduced the alpha-diversity and altered the beta-diversity of the gut microbiome. Firmicutes was largely decreased while Bacteroidetes was significantly increased. The Phylogenetic Investigation of Communities by Reconstruction of Unobserved States (PICRUSt) analysis also indicated that the control mice and neotame-treated mice have different metabolic patterns and some key genes such as butyrate synthetic genes were decreased. Moreover, neotame consumption also changed the fecal metabolite profiles. Dramatically, the concentrations of multiple fatty acids, lipids as well as cholesterol in the feces of neotame-treated mice were consistently higher than controls. Other metabolites, such as malic acid and glyceric acid, however, were largely decreased. In conclusion, our study first explored the specific effects of neotame on mouse gut microbiota and the results may improve our understanding of the interaction between gut microbiome and neotame and how this interaction could influence the normal metabolism of host bodies.

Insights into Chemical Structure-Based Modeling for New Sweetener Discovery

Abstract:The search for novel, natural, high-sweetness, low-calorie sweeteners remains open and challenging. In the present study, the structure-based machine learning modeling and sweetness recognition mechanism were investigated to assist this process. It was found that whether or not a compound was sweet was closely related to molecular connectivity and composition (the number of hydrogen bond acceptors and donors), tpsaEfficiency, structural complexity, and shape (nAtomP and Fsp3). While the relative sweetness of sweet compounds was more determined by the molecular properties (tpsaEfficiency and Log P), structural complexity and composition (nAtomP and ATSm 1). The built machine learning models exhibited very good performance for classifying the sweet/non-sweet compounds and predicting the relative sweetness of the compounds. Moreover, a specific binding pocket was found for sweet compounds, and the sweet compounds mainly interacted with the VFT domain of the T1R2-T1R3 through hydrogen bonds. In addition, the results indicated that among the sweet compounds, those that were sweeter bound to the VFT domain stronger than those that had low sweetness. This study provides very useful information for developing new sweeteners.

Abstract:Sweeteners play an irreplaceable role in daily life and have been found in multitudinous food products. However, excessive or unreasonable intake of sweeteners as food additives brings about untoward problems due to the accumulation in the human body. Therefore, a comprehensive review of different sweeteners' pretreatment and determination methods is urgently needed. In this review, we comprehensively reviewed the progress of different pretreatment and detection methods for sweeteners in various food, focusing on the latest development since 2015. Current state-of-the-art technologies, such as headspace single-drop microextraction, ultrasound-assisted emulsification microextraction, solid-phase microextraction, two-dimensional liquid chromatography, and high-resolution mass spectrometry, are thoroughly discussed. The advantages, disadvantages, critical comments, and future perspectives are also proposed. This review is expected to provide rewarding insights into the future development and broad application of pretreatment and detection methods for sweeteners in different food samples.

Abstract:Sweetness in food delivers a delightful sensory experience, underscoring the crucial role of sweeteners in the food industry. However, the widespread use of sweeteners has sparked health concerns. This underscores the importance of developing and screening natural, health-conscious sweeteners. Our study represents a groundbreaking venture into the discovery of such sweeteners derived from egg and soy proteins. Employing virtual hydrolysis as a novel technique, our research entailed a comprehensive screening process that evaluated biological activity, solubility, and toxicity of the derived compounds. We harnessed cutting-edge machine learning methodologies, specifically the latest graph neural network models, for predicting the sweetness of molecules. Subsequent refinements were made through molecular docking screenings and molecular dynamics simulations. This meticulous research approach culminated in the identification of three promising sweet peptides: DCY(Asp-Cys-Tyr), GGR(Gly-Gly-Arg), and IGR(Ile-Gly-Arg). Their binding affinity with T1R2/T1R3 was lower than −15 kcal/mol. Using an electronic tongue, we verified the taste profiles of these peptides, with IGR emerging as the most favorable in terms of taste with a sweetness value of 19.29 and bitterness value of 1.71. This study not only reveals the potential of these natural peptides as healthier alternatives to traditional sweeteners in food applications but also demonstrates the successful synergy of computational predictions and experimental validations in the realm of flavor science.

Abstract:A 15-year-old female collared peccary (Tayassu tajacu) was presented for ophthalmic examination following sudden onset of blindness. Bilateral retinal detachment was diagnosed, neoplasia suspected, and euthanasia performed. Widespread tumor dissemination was apparent at autopsy, and transitional cell carcinoma was diagnosed histologically. The tumor was identified as arising from the ovary. Epidemiologic features of this case are discussed.

Abstract:In recent years, the worldwide increase in lifestyle diseases and metabolic disorders has been ascribed to the excessive consumption of sucrose and added sugars. For this reason, many approaches have been developed in order to replace sucrose in food and beverage formulations with alternative sweetening compounds. The raising awareness concerning the synthetic sweeteners due to their negative impact on health, triggered the need to search for alternative substances. Natural sweeteners may be classified in: (i) non-nutritive (e.g., neohesperidine dihydrochalcone, thaumatin, glycyrrhizin mogroside and stevia) and (ii) bulk sweeteners, including both polyols (e.g., maltitol, mannitol, erythritol) and rare sugars (e.g., tagatose and allulose). In this review we discuss the most popular natural sweeteners and their application in the main food sectors (e.g., bakery, dairy, confectionary and beverage), providing a full understanding of their impact on the textural and sensory properties in comparison to sucrose. Furthermore, we analyze the use of natural sweeteners in blends, which in addition to enabling an effective replacement of sugar, in order to complement the merits and limits of individual compounds. Finally, microencapsulation technology is presented as an alternative strategy to solving some issues such as aftertaste, bitterness, unpleasant flavors, but also to enhance their stability and ease of use.

Abstract:Growing demand for the tasty and healthy food has driven the development of low-calorie sweeteners, sweet taste modulators, and bitter masking compounds originated from natural sources. With the discovery of human taste receptors, increasing numbers of sweet taste modulators have been identified through human taste response and molecular docking techniques. However, the discovery of novel taste-active molecules in nature can be accelerated by using advanced spectrometry technologies based on structure-activity relationships (SARs). SARs explain why structurally similar compounds can elicit similar taste qualities. Given the characterization of structural information from reported data, strategies employing SAR techniques to find structurally similar compounds become an innovative approach to expand knowledge of sweeteners. This review aims to summarize the structural patterns of known natural non-nutritive sweeteners, sweet taste enhancers, and bitter masking compounds. Innovative SAR-based approaches to explore sweetener derivatives are also discussed. Most sweet-tasting flavonoids belong to either the flavanonols or the dihydrochalcones and known bitter masking molecules are flavanones. Based on SAR findings that structural similarities are related to the sensory properties, innovative methodologies described in this paper can be applied to screen and discover the derivatives of taste-active compounds or potential taste modulators.

Abstract:Human sensation for sweet tastes and the thus resulting over-consumption of sugar in recent decades has led to an increasing number of people suffering from caries, diabetes, and obesity. Therefore, a demand for sugar substitutes has arisen, which increasingly has turned towards natural sweeteners over the last 20 years. In the same period, thanks to advances in bioinformatics and structural biology, understanding of the sweet taste receptor and its different binding sites has made significant progress, thus explaining the various chemical structures found for sweet tasting molecules. The present review summarizes the data on natural sweeteners and their most important (semi-synthetic) derivatives until the end of 2019 and discusses their structure-activity relationships, with an emphasis on small-molecule high-intensity sweeteners.

Abstract:Sugar is crucial as an essential nutrient for humans as well as for providing texture, sweetness and so on to food. But with the rise in people's pursuit of health, it is becoming increasingly clear that excessive consumption of sugar can locate a load on the body. It has been that excessive sugar is associated with many diseases, such as dental caries, obesity, diabetes, and coronary heart disease. Therefore, researchers and industries are trying to reduce or substitute sugar in food without affecting the sensory evaluation. Substituting sugar with sweeteners is alternatively becoming the most traditional way to minimize its use. So far, the sweeteners such as stevia and xylitol have been are commercially applied. Several studies have shown that technological innovation can partially compensate for the loss in sweetness as a result of sugar reduction, such as cross-modal interactions that stimulate sweetness with aroma, nanofiltration that filters disaccharides and above, enzyme-catalyzed sugar hydrolysis, and microbial fermentation that turns sugar into sugar alcohol. This review summarizes these studies to enhance the safety and quality of sugar-reduced products, and will provide some theoretical frameworks for the food industry to reduce sugar in foods, meet consumers' needs, and promote human health. Sugar reduction in beverages: Current trends and new perspectives from sensory and health viewpoints

Abstract:Sugar, as an essential component of beverages, not only provides sweetness in beverages but also plays a significant role in their flavor, texture, and preservation. In recent years, global sugar consumption has continued to increase, causing a variety of health concerns. Currently, there is growing awareness of the adverse effects of high-sugar consumption. Since beverages are the primary source of daily sugar intake, sugar reduction in beverages is imperative. In this work, the necessity of sugar reduction in beverages was first introduced. Furthermore, four primary sugar reduction strategies (direct sugar reduction, multi-sensory integration, sweeteners, and sweetness enhancers) employed in the beverage industry were systematically summarized. Each sugar reduction strategy was critically compared, while the current research progresses as well as challenges were discussed. The application of sweeteners is the most effective and widely used strategy for sugar reduction in spite of flavor and health concerns of sweeteners. Meanwhile, multi-sensory integration is also a promising strategy for sugar reduction. In addition, different evaluation methods (chemical, cell-based and sensory methods) for sweetness were overviewed. Given the current challenges of sugar reduction, the prospects of sugar reduction in beverages were also discussed. The present work can provide the current progress for sugar reduction in the beverage industry.

Abstract:Sugar widely exists in the nature. It plays a key role in various molecular recognition processes. Aiming at probing sugar derivatives and their synthetic study, four series of sugar derivatives were designed and synthesized. The first chapiter described the synthesis of C glycosyl analogues of arbutin, a natural product for whitening skin. The second chapiter is devoted to the synthesis of C glycosyl analogues of vitamin E, with the aim to improve their anti oxidant activity. The third chapiter described the synthesis of photolabile sugars as biochip's basic blocks. By regioselective opening of 4,6 0 (o nitro)benzylidene ring of methyl glycopyranoside, followed by C 4 epimerisation, a series of 6 0 (o nitro)benzyl protected glycopyranosides were synthesized. In the Chapiter 4, a series of glycosyl amino acids have been designed and synthesized as potential inhibitors of PTP 1B, a potentiel target to treat diabetes. Preliminary biological result showed that these compounds inhibited PTP 1B in micromolar range.
