Abstract:Astilbotriterpenic acid (1), a novel ursane-type triterpenoid from the rhizomes of Astilbe chinensis, has cytostatic properties in several cancer cell lines and induces apoptosis in HeLa cell. The aim of this study was to investigate the mechanisms by which such properties are exerted, with special reference to the anti-proliferative and apoptotic potential on HeLa cells. Compound 1 showed a marked concentration- and time-dependent inhibition of HeLa cell proliferation, and induced G(0)/G(1) phase arrest of HeLa cell in a dose-dependent manner. There was a quick attenuation of mitochondrial membrane potential (Ay)m) with the alterations of Bcl-2/Bar mRNA express value in 1-treated HeLa, indicating the participation of a mitochondria-related mechanism. Pretreatment of a pan-caspase inhibitor (benzyloxycarbonyl)-Val-Ala-Asp-(fluoromethyl) ketone (z-VAD-fmk) significantly increases the viability of 1-treated HeLa cells implied that the participation of caspase; Western-blot analysis showed the activation of initiator caspase-8 and caspase-9, and executor caspase-3. Meanwhile, treatment with 1 stimulates the generation of reactive oxygen species (ROS) in HeLa cell. Taken together, our data showed that compound 1 induced growth arrest and apoptosis in HeLa cells through mitochondria-related pathways and ROS production, and may be further evaluated as a chemotherapeutic agent for human cancer.
Apoptosis in HeLa cells caused by the inducible expresion of human granzyme B fusinon protein gene

Abstract:To investigate apoptosis in HeLa cells induced by expresion of human granzyme B fusion protein. Granzyme B fusion protein gene, which consisted of portion of transmembrane sequence of Pseudomonae exotoxin A (PE) and active granzyme B gene, was generated by recombinant PCR and cloned into the inducible exprsion vetor pIND. The HeLa cels were co-transfected with the resulting exprssion vector and assistant plasmid pVgRXR, and subjected to G418 and zeocin selection. Expresison of traget protein on ponasterone A induction was detectd by indirect immunofluoresce, and the effect on morphology and growth of HeLa cells was observed by electronic microscopy. TUNEL staining and cell counting. The HeLa cels inducibly exprssing granzyme B fusion protein were obtained. The traget rotein was detected in the cytoplasm of the cells after ponasterone A ndcution, resultuin gin increased cell size and multinucleation, and growth inhibiton was observed. Elecutronic microscopy and TUNEL analysis ashowed that the inducible expression of granzyme B ffsion protein gene casued typeicla features of apoptosis. Inductible expression of granzyme B fusion protein gene ieads to apoptosis of HeLa cells.

Abstract:3beta-Hydroxy-12-oleanen-27-oic acid (ATA) was an antitumor active oleanane-type triterpenoid from the rhizomes of Astilbe chinensis. ATA was structurally very similar to oleanolic acid, with the only difference being interchange of the carboxyl and methyl group at the C-14 and C-17 positions. Oleanane-type triterpene with a carboxyl group at C-14 is present in a limited number of natural resources. ATA was found to exhibit more distinctive cytotoxicity toward human cervical squamous carcinoma HeLa cells than oleanolic acid, which suggested that the position of the carboxyl group significantly affects the cytotoxicity of oleanane-type pentacyclic triterpenes with a carboxyl group. The biological mechanism responsible for the cytotoxicity of ATA is not yet well understood. In this study, we investigated the induction of apoptosis in HeLa cells by ATA and the putative pathways of its actions. ATA induced a marked concentration- and time-dependent inhibition of HeLa cell proliferation, and reduced the protein content in HeLa cells. ATA-treated HeLa displayed typical morphological apoptotic characteristics and formation of DNA ladders in agarose gel electrophoresis. Flow cytometric analysis showed that the HeLa cell cycle was arrested in the G(0)/G(1) phase by ATA, and the apoptotic rate of HeLa cells treated with ATA 20 microg/mL for 48 h was 22.63 +/- 1.65%. Meanwhile, ATA increased the expression of Bax, decreased the expression of Bcl-2, and lowered the DeltaPsi(m). DEVD-CHO 2 microM could increase the viability of ATA-treated HeLa cells. These results indicate that ATA could significantly induce cell apoptosis through down-regulating Bcl-2 expression, up-regulating Bax expression, lowering DeltaPsi(m), and activating the caspase-3 pathway, and should be useful in the search for new potential anti-tumor agents and for developing semisynthetic oleanane-type triterpene derivatives with anti-tumor activity.

Abstract:Objective Phosphatidylethanolamine (PE) is an important phospholipid component in the cell membrane and is involved in the formation of membrane asymmetry. PE is exposed on the cell surface with phosphatidylserine during apoptosis. However, the effects of PE on cell apoptosis are not clear. In this study, we investigated effects of PE on apoptosis in human cervical cancer HeLa cells. Methods HeLa cells were used as the experiment material, and were divided into five groups: blank PE, respectively. The cell growth was tested by MTT assay; the cell cycle and apoptosis were analyzed using flow cytometry. Results Compared with the control group, PE inhibited the growth of HeLa cells in all the treatment groups in dose- and time-dependent manners, and induced the apoptosis, but did not change the cell cycle. Conclusion PE inhibits the growth of HeLa cells by inducing the apoptosis.

Abstract:9-0xo-10, 11-dehydroageraphorone (euptox A), a cadenine sesquiterpene, is the main toxin from Eupatorium adenophorum. The aim of the present study was to examine the induction and mechanism of HeLa cell apoptosis by euptox A. The apoptosis-inducing effect of the euptox A on HeLa cells was examined by MTT assay. The underlying mechanism was analyzed by flow cytometry and quantitative PCR. Flow cytometry results suggested that euptox A effectively inhibited the proliferation of HeLa cells, arrested the cell cycle transition from S to G2/M phase, did not continue to complete the cell cycle activity (mainly from 4 times and mitosis), and induced cell proliferation. The RT-qPCR detection results showed that euptox A induced apoptosis by improving the gene expression level of apoptotic proteases such as caspase-10 in HeLa cells. Its mechanism of action was associated with the upregulation of apoptotic gene expression and arresting of the cell cycle.

Abstract:To study the apoptosis induced by cisplatin in cervical cancer cell line HeLa and its mechanism,cell growth inhibition of cisplatin on HeLa cells was analyzed by MTT assay.Cell apop-tosis was examined by cytometry and Hoechst33258 staining after treatment with cisplatin.The ef-fects of cisplatin on transcription of E6 were analyzed by RT-PCR.The protein expressions of E6,p53,p21,Bax and Bcl-2 were studied by Western blotting.Cisplatin inhibited proliferation in a time-and dose-dependant manner.Cytometically,sub-G1 peak showed higher apoptosis rates in the ex-perimental group than those in the control.Hoechst33258staining exhibited apoptosis induced by cis-platin.RT-PCR revealed that cisplatin decreased transcription of E6.Western blotting showed that cisplatin decreased protein expression of E6 and increased protein expression of p53,p21and Bax.It had no effect on protein expression of Bcl-2.It is concluded that cisplatin can induce apoptosis in HeLa cells by suppressing HPV E6 and thereby restoring the function of p53.

Ursane-type triterpene with a COOH group at C(14) is present in a limited number of natural resources. Compound I was found to exhibit more distinctive cytotoxicity toward human cervical squamous carcinoma (HeLa) cells than ursolic acid, suggesting that the position of the COOH group significantly affects the cytotoxicity of ursane-type pentacyclic triterpenes with a COOH group. To elucidate the underlying biological mechanism responsible for the cytotoxicity of 1, we investigated its growth-inhibitory and apoptosis-inducing effect on HeLa cells. Compound 1 induced a marked concentration- and time-dependent inhibition of cell proliferation with an IC50 value of 6.80 +/- 0.88 mu g/ml following 48 h incubation. The drug-treated HeLa cells displayed typical morphological apoptotic characteristics and formation of DNA ladders in agarose-gel electrophoresis. Flow cytometric analysis showed that the cell cycle was arrested in G(0)/G(1) phase by 1, and the apoptotic rate of HeLa cells treated for 48 h with 20 mu g/ml of 1 was 21.08 +/- 2.14%. Also, 1 increased and decreased the expression of Bax and Bcl-2 proteins, respectively, and lowered the mitochondrial transmembrane potential (Delta(Psi m)). The peptidic capase-3 inhibitor DEVD-CHO (NH2-Asp-Glu-Val-Asp-CHO, at 2 mu m) could increase the viability of HeLa cells previously treated with 1. These results indicate that 1 induces efficient cell apoptosis through down-regulating Bcl-2 expression, up-regulating Bax expression, lowering Delta psi(m), and by activating the caspase-3 pathway.
A natural product enhances apoptosis via mitochondria/caspase-mediated pathway in HeLa cells.

Abstract:Cervical cancer is the fourth most lethal human malignancy and the leading cause of death among females around the world. Many antitumor agents have microbial origins. 5 '-epi-SPA-6952A is a new 24-membered macrolide isolated from the cultured broth of Streptomyces diastatochromogenes. Therefore, we studied the activity and molecular mechanism of 5 '-epi-SPA-6952A in human cervical carcinoma HeLa cell. The results showed that 5 '-epi-SPA-6952A significantly inhibited cell proliferation and migration. In addition, 5 '-epi-SPA-6952A obviously increased the production of intracellular reactive oxygen species and DNA damage in HeLa cells. Moreover, nuclear shrinkage of cells, decrease in mitochondrial membrane potential, and upregulation of Bax/Bcl-2 ratio resulted in the release of cytochrome c, and activation of caspase-9/3 was observed in HeLa cells treated with 5 '-epi-SPA-6952A, which means it enhanced the intrinsic mitochondrial apoptosis. Besides, DNA-damage associated proteins poly (ADP-ribose) polymerase (PARP) and p53 were also studied, and the expressions of cleaved-PARP and p53 were drastically increased in HeLa cells treated with 5 '-epi-SPA-6952A. Furthermore, we confirmed that 5 '-epi-SPA-6952A affected the survival of HeLa cells by blocking cell cycle progression in the G1 phase. Taken together, the results shows that 5 '-epi-SPA-6952A significantly inhibited HeLa cells proliferation via intrinsic mitochondrial apoptosis, cell cycle arrest, and blocking cell migration.

Abstract:To investigate the apoptotic effects of an extract of red-tide algae, Protoceratium reticulatum, on HeLa cells, we used liquid chromatography-tandem mass spectrometry, optical microscopy, Hoechst 33342/propidine iodide staining, and DNA gel electrophoresis to analyze its constituents and toxicity, as well as rhodamine 123 staining to investigate changes in mitochondrial membrane potential. Analysis showed that the P. reticulatum extract contained the yessotoxins (YTXs) homo-YTX, 45-OH-YTX and 45-OH-homo-YTX. The results indicated that P. reticulatum extract negatively influences HeLa cells and induces their apoptosis.

Abstract:BACKGROUND. We validated the induction of apoptotis in human prostate cancer PC3 cells by apoptosis-inducing nucleosides (AINs) released from the CD57(+) HLA-DRbright-natural suppressor (57.DR-NS) cell line. We analyzed the molecular signaling pathway during AINs-induced apoptosis in PC3 cells. METHODS. Direct and indirect co-cultures between 57.DR-NS and PC3 cells were performed. AINs were isolated by high-performance liquid chromatography ((HPLC) from 57.DR-NS cell cultures, Apoptosis in PC3 cells was analyzed by DNA fragmentation, sub-G, DNA content with flow cytometry, and terminal deoxynucleotide transferase-mediated dUTP nick end-labeling (TUNEL) method. The DNA strand breaks and activation of caspase-3 in PC3 cells were measured by DNA unwinding and flow cytometry assay. RESULTS. The 57.DR-NS cell line generated apoptosis in PC3 cells via AINs. AINs isolated from 57.DR-NS cell cultures induced apoptosis in PC3 cells. Furthurmore, we found DNA strand breaks followed by activation of caspase-3 during AINs-induced apoptosis in PC3 cells. CONCLUSIONS. The data obtained here indicated that AINs could induce apoptosis in PC3 cells through DNA strand breaks and activation of caspase-3. (C) 2002 Wiley-Liss, Inc.

Abstract:Cervical cancer continues to be a threat to female health globally. In the present study, the potential anticancer activity of 2-[2-hydroxyl-1-(4-methoxy phenyl) ethyl]-3-(4-benzyloxy phenyl) isoindolin-1-one (CDS-1548), was evaluated in HeLa cells. CDS-1548 is an organic small-molecule compound characterized by two chiral centers, with the nuclear parent 1H-isoindolin-1-one. CDS-1548 administration significantly elevated the transcriptional activity of p53 and its downstream target genes in a dose-dependent manner. Additionally, CDS-1548 treatment increased the expression levels of p53 and mouse double minute 2 homolog, as well as inducing apoptosis in HeLa cells. Furthermore, CDS-1548 treatment downregulated the expression of B-cell lymphoma 2, upregulated Bcl-2 homologous antagonist killer, promoted the release of cytochrome c from mitochondria to cytoplasm, and activated the production of caspase 3 and 9. Collectively, these results suggested that CDS-1548 inhibited HeLa cell proliferation by promoting G2/M cell cycle phase arrest and inducting of mitochondria-mediated apoptosis.
Proteomic Investigation into Betulinic Acid-Induced Apoptosis of Human Cervical Cancer HeLa Cells

Abstract:Betulinic acid is a pentacyclic triterpenoid that exhibits anticancer functions in human cancer cells. This study provides evidence that betulinic acid is highly effective against the human cervical cancer cell line HeLa by inducing dose- and time-dependent apoptosis. The apoptotic process was further investigated using a proteomics approach to reveal protein expression changes in HeLa cells following betulinic acid treatment. Proteomic analysis revealed that there were six up- and thirty down-regulated proteins in betulinic acid-induced HeLa cells, and these proteins were then subjected to functional pathway analysis using multiple analysis software. UDP-glucose 6-dehydrogenase, 6-phosphogluconate dehydrogenase decarboxylating, chain A Horf6-a novel human peroxidase enzyme that involved in redox process, was found to be down-regulated during the apoptosis process of the oxidative stress response pathway. Consistent with our results at the protein level, an increase in intracellular reactive oxygen species was observed in betulinic acid-treated cells. The proteins glucose-regulated protein and cargo-selection protein TIP47, which are involved in the endoplasmic reticulum pathway, were up-regulated by betulinic acid treatment. Meanwhile, 14-3-3 family proteins, including 14-3-3β and 14-3-3ε, were down-regulated in response to betulinic acid treatment, which is consistent with the decrease in expression of the target genes 14-3-3β and 14-3-3ε. Furthermore, it was found that the antiapoptotic bcl-2 gene was down-regulated while the proapoptotic bax gene was up-regulated after betulinic acid treatment in HeLa cells. These results suggest that betulinic acid induces apoptosis of HeLa cells by triggering both the endoplasmic reticulum pathway and the ROS-mediated mitochondrial pathway.

Cervical Cancer HeLa Cell Autocrine Apoptosis Induced by Coimmobilized IFN-γ Plus TNF-α Biomaterials.

Abstract:Using external methods to induce the death of cancer cells is recognized as one of the main strategies for cancer treatment. Research indicated that TNF-α is frequently used in tumor biotherapy while IFN-γ can directly inhibit tumor cell proliferation. In our study, TNF-α and IFN-γ were coimmobilized on polystyrene material (PSt) or Fe3O4-oleic acid nanoparticles (NPs). Then the structural change of these two proteins can be observed. Meanwhile, the expressions of both TNF-α and IFN-α increased significantly, as determined by gene microarray analysis; however, in the presence of TNF-α plus IFN-α inhibitors, TNF-α and IFN-α did not increase in HeLa cells induced by coimmobilized IFN-γ plus TNF-α. Our results indicate that such change can stimilate HeLa cells to secrete more TNF-α and IFN-α, by which the apoptosis of HeLa cells could be further induced. This study is the first report of autocrine-induced apoptosis of HeLa cells. In addition, we performed ELISA, RT-PCR, flow cytometry, and Western blot analyses, as well as a series of analytical tests at the animal level. our data also indicate that the PSt-coimmobilized IFN-γ plus TNF-α has apparent effects for cancer treatment in vivo, which is of great significance for translation into clinical medicine.

Abstract:Hypocrellin A(HA), a photosensitive perylenequinone compound of Hypocrella bambusae, inhibited the proliferation of several tumor cell fines. Human cervical cancer cells, HeLa cells, were used as a model to elucidate the molecular mechanisms of HA-induced tumor cell death. The results show that HA can induce the oligonucleosomal fragmentation of DNA in HeLa cells and also can increase the expression of apoptosis inducer Bax mRNA and that it decreases the expression of apoptosis suppressor, Bel-2 mRNA, in mitochondria. It can be concluded from the data that HA-induced apoptosis is related to the balance between Bel-2 and Bax gene expressions.

Study on the resveratrol induced apoptosis in hunman cervical cancer cell line HeLa

Abstract:Objective:To explore the effect of resveratrol on the apoptosis of cervical cancer HeLa cell.Methods:HeLa cells were cultured and treated with different concentrations(0,12.5,25,50,100,200,300,400,600μmol/L) of Res for 24,48,72h.The growth inhibition rate was detected by MTT method,and the cell apoptosis was analyzed with flow cytometry(FCM) using annexinV/propidium iodide(PI) double staining.Morphologic configuration of apoptotic cells were observed with fluorescence microscope,and caspase-3 activity was assessed by colorimetric assay.Results:Res inhibited the proliferation of HeLa cells in a time- and dosage- dependent manner (P<0.05).After Res treatment at 0,200,300μmol/L for 48h,the apoptosis rates of HeLa cells were 2.2%,7.1%,6.23%,while the necrosis rates were 7.7%,16.04%,15.43%,respectively.The cells showed significant apoptotic morphological changes under fluorescence microscope.After 200μmol/L Res treatment for 8h,the activity of caspase-3 activity increased,and reached the peak at the 24h,then declined gradually.After HeLa cells were treated with 50,100,200μmol/L Res for 24h,the activity of caspase-3 was found increased by 1.92,2.51,4.53 folds compared with that of the control group(P＜0.05).Conclusion:Res can inhibit the proliferation and induce apoptosis of HeLa cells in a timeand dosage-dependent manner by increasing caspase-3 activity.

Abstract:Human cervical cancer is potentially lethal, and therefore the development of effective and tolerable therapeutic options is vital. In the present study, the in vitro effect of the synthetized compound JOT01006 (C21H20C1NO4) on human cervical epithelioid carcinoma cell line (HeLa) was examined. The results demonstrated that JOT01006 induced morphological changes and cytotoxicity (decreased the percentage of viable cells) in a dose-dependent manner. JOT01006 induced apoptosis which was analyzed by flow cytometric methods and confirmed by DAPI staining and DNA fragmentation analyzed by DNA gel electrophoresis. JOT01006 also induced reactive oxygen species (ROS) overproduction before causing endoplasmic reticulum (ER) stress which was also confirmed by the increased levels of Grp78 and Gadd153. Western blotting was selected to demonstrate that JOT010006 promoted p53, Bak, PARP, caspase-3 levels and decreased the levels of Bcl-2 and Bcl-xL. Our results also showed that JOT01006 also promoted caspase-12 production followed by apoptosis. The results also showed that JOT01006 inhibited the migration of HeLa cells potentially through inhibition of MMP-2 and -9.
Dimethylfumarate Induces Cell Cycle Arrest and Apoptosis Via Regulating Intracellular Redox Systems in HeLa Cells

Abstract:Dimethylfumarate (DMF) is cytotoxic to several kinds of cells and serves as an anti-tumor drug. This study was designed to investigate the effects and underlying mechanism of DMF on cervical cancer cells. HeLa cells were cultured and treated with 0, 50, 100, 150, and 200 μM DMF, respectively. After 24 h, cell growth was evaluated using Cell Counting Kit-8 (CCK-8) assay and the cell cycle was examined using flow cytometry. In addition, cell apoptosis was detected by Annexin V/propidium iodide (PI) staining and the expressions of caspase-3 and poly-ADP-ribose polymerase (PARP) were detected using western blotting. The redox-related factors were then assessed. Furthermore, all of the indicators were detected in HeLa cells after combined treatment of DMF and N-acetyl-l-cysteine (NAC, an oxygen-free radical scavenger). The cell number and cell growth of HeLa were obviously inhibited by DMF in a dose-dependent manner, as the cell cycle was arrested at G0/G1 phase (P < 0.05). The apoptotic HeLa cells were markedly increased, and the expression levels of caspase-3 and PARP were significantly increased in a DMF concentration-dependent way (P < 0.05). Meanwhile, loss of △Ψm, increase in reactive oxygen species and O2 ·−, and the decrease in catalase activity and glutathione (GSH) level were found after DMF treatment (P < 0.05). All these changes were significantly attenuated and even completely disappeared by adding NAC (P < 0.05). In conclusion, the cytotoxicity of DMF on cell proliferation and apoptosis of HeLa cells was mainly related to the intracellular redox systems by depletion of intracellular GSH.

Abstract:The aim of this study is to investigate effects of DHEA on the viability and apoptosis of Jurkat cells,in which the effect of DHEA on apoptosis in Jurkat cells was determined by DNA ladder,Hoechst33258,PI staining and Annexin-V- FITC/PI double staining.RT-PCR method was used to analyze Fas and FasL mRNA expression and the Fas/FasL proteins were evaluated by flow cytometry.Changes in mitochondrial membrane potential were detected with JC-1 fluorescence.It was demonstrated viability of Jurkat cells was down regulated,and numbers of Annexin V-positive and propidium iodide-negative cells were increased significantly after DHEA treatment.The sub-diploid peak of cells with fragmented DNA were also in creased vs control.Agarose gel electrophoresis of DHEA-treated Jurkat cells showed DNA characteristic fragmentation of ap- optotic cells.JC-1 staining showed cells with green fluorescence increased significantly vs control.Fas and FasL mRNA and protein expression of Jurkat cells was also increased in DHEA treated classes than that of controls.The results revealed that DHEA could induce apoptosis in Jurkat cells in vitro and the Fas/FasL pathway and mitochondrion membrane potential muta- tion play roles in these regulations.
