Abstract:Objective To study the steroidal saponins from Allii Macrostemonis Bulbus.Methods The silica gel and ODS column chromatographies were used for the isolation and purification of compounds from Allii Macrostemonis Bulbus.Their structures were identified through comparing with references and spectral analyses.Results Three steroidal saponins were isolated from the 70% ethanol extract from Allii Macrostemonis Bulbus and identified as 5β-spirostane-25(27)-en-3β,12β-diol-3-O-β-D-glucopyranosyl(1→2)-β-D-galactopyranoside(1),(25R)-5β-spirostane-3β,12β-diol-3-O-β-D-glucopyranosyl-(1→2)-β-D-galactopyranoside(2),and 5β-spirostane-25(27)-en-2β,3β-diol-3-O-β-D-glucopyranosyl-(1→2)-β-D-galactopyranoside(3).Conclusion Compound 1 is a new compound named macrostemonoside S.Compound 2 is isolated from Allii Macrostemonis Bulbus for the first time.

Abstract:Seven new triterpenoid saponins (1-7), have been isolated and elucidated from the roots of Gypsophila oldhamiana together with five known triterpenoid saponins (8-12). These saponins which could be classified into three series: 3-O-monoglucosides (1, 8, 9), 28-O-monoglucosides (24, 12) and 3, 28-O-bidesmosides (5-7, 10, 11), have been evaluated for their a-glucosidase inhibition activity. As a result, the preliminary structure-activity relationships were discussed based on the position of sugar linkage attached to the aglycone, and 28-O-monoglucosides 2-4 and 12 showed significant inhibitory activities on a-glucosidase. Abstract:AIM:To investigate chemical constituents of Gypsophila paniculata.METHODS:The compounds were isolated by silica gel,Sephadex LH-20,Rp-C18 column chromatography and preparative liquid chromatography.Their structures were elucidated by ESI-MS,NMR techniques (1H,13C,HMQC,HMBC,TOCSY) and acid hydrolysis.Moreover their α-glucosidase inhibition activity was evaluated compared with that of their aglycone.RESULTS:Two triterpenoid saponins,caryophyllacosides A (1) and B (2),were isolated from the roots of Gypsophila paniculata,without having recourse to chemical degradation or modification.Both of them were found to have potent α-glucosidase inhibitiory activity.CONCLUSION:The two triterpenoid saponins are new boactive compounds.

Abstract:Two new steroidal saponins, chinenoside VI (1) and neomacrostemonoside D (8b), together with ten known saponins were isolated from the bulbs of Allium chinense. On the basis of chemical evidence and spectral analysis (IR, H-1-NMR C-13-NMR H-1-H-1 COSY, HMQC, HMBC and SI-MS), chinenoside VI was established as (25R)-24-O-beta-D-glucopyranosyl-3 beta,24 beta-dihydroxy-5 alpha-spirost 3-O-alpha-arabinopyranosyl-(1-->6)-beta-D-glucopyranoside, and neomacrostemonoside D was elucidated as neotigogenin 3-O-beta-D-glucopyranosyl-(1-->2)[beta-D-glucopyranosyl-(1-->3)]-(6-acetyl-beta-D-glucopyranosyl)-(1-->4)-beta-D-galactopyranoside, respectively.

Abstract:Two new steroidal saponins (1 and 2) were isolated from the dried bulbs of Allium macrostemon Bunge. Their structures were elucidated by the spectral data as 26-O-beta-D-glucopyranosyl-5 alpha-furost-25 (27)-ene-3 beta, 12 beta, 22, 26-tetraol-3-O-beta-D-glucopyranosyl (1 -> 2) [beta-D-glucopyranosyl (1 -> 3)]-beta-D-glucopyranosyl (1 -> 4)-beta-D-galactopyranoside (1) and 26-O-beta-D-glucopyranosyl-5 beta-furost-20 (22)-25 (27)-dien-3 beta, 12 beta, 26-triol-3-O-beta-D-glucopyranosyl (1 -> 2)-beta-D-galactopyranoside (2), respectively. Their cytotoxic activities on several cancer cell lines (MCF-7, NCI-H460, SF-268 and HepG2) were tested. 1 showed special cytotoxity on SF- 268, while 2 showed cytotoxity on NCI-H460 and SF-268 cell lines, respectively.

Chemical structures of these compounds were elucidated by spectroscopic techniques. Anti-inflammatory activities of these new compounds were evaluated.

Abstract:Two new triterpenoid saponins (1, 2) and a known sapogenin (3) were isolated from the dried carpophore of Xanthoceras sorbifolia Bunge. (Sapindaceae), and their structures were established as 3-O-beta-d-glucopyranosyl, 28-O-[alpha-l-rhamnosyl (1 --> 2)]-beta-d-glucopyranosyl 16-deoxybarrin-gtogenol C (1), 3-O-[beta-d-glucopyranosyl(1 --> 6)] 3'-angeloyl)-beta-d-glucopyranosyl, 28-O-[alpha-l-rhamnosyl(1 --> 2)]-beta-d-glucopyranosyl 16-deoxybarringtogenol C (2), 16-deoxybarringtogenol C (3), by spectral and chemical methods. Compounds 1 and 2 were found to have activity of inhibiting the proliferation of two human tumour cell lines while compound 3 was inactive.

Abstract:AIM: To study the chemical constituents of the roots of Achyranthes bidentata B1.. METHODS: The chemical constituents were isolated and purified by macroporous adsorptive resin D101, silica gel, and ODS column chromatographies and preparetive HPLC. Their structures were elucidated on the basis of ID and 2D NMR analyses. RESULTS: Two feruloyl tyramine glycosides and seven triterpenoid saponins were obtained and identified as N-trans-feruloyl-3-methoxytyramine-4'-O-beta-D-glucopyranoside (1), N-trans-feruloyl-3-methoxytyra mine-4-O-beta-D-glucopyranoside (2), PJS-1 (3), chikusetsusaponin IVa (4), oleanolic acid 3-O-[beta-D-glucuronopy ranoside-6-O-methyl ester]-28-O-beta-D-glucopyranoside (5), oleanolic acid 3-O-[beta-D-glucuronopyran-oside-6-O-ethyl ester]-28-O-beta-D-glucopyranoside (6), oleanolic acid 3-O-[beta-D-glucuronopyranoside-6-O-butyl ester]-28-O-beta-D-glucopyranoside (7), ginsenoside R-0(8) and hederagenin-28-O-beta-D-glucopyranosyl ester (9). CONCLUSION: Compound I is a new feruloyl tyramine glycoside, while compounds 2 and 9 are reported from A. bidentata for the first time.

Abstract:Aim To isolate and structurally elucidate the steroidal saponins of Allium macrostemon Bunge and evaluate the anti-cancer bioactivities of these compounds in vitro.Methods Solvent extraction and column chromatography were used to isolate the steroidal saponins and physic-chemical constants and spectroscopic analysis were employed for the structural elucidation.MTT were used to evaluate the bioactivities in vitro.Results Eight steroidal saponins were isolated and elucidated as macrostemonoside B(1),macorstemonoside C(2),macorstemonoside E(3),(22S)-cholest-5-ene-1β,3β,16β,22-tetraol-1-O-α-L-rhamnopyranosyl-16-O-β-D-glucopyranoside(4),(25R)-26-O-β-D-glucopyranosyl-22-hydroxy-furost-5(6)-ene-3β,26-diol-3-O-β-D-glucopyranosyl(1→2)[β-D-glucopyranosyl(1→3)]-β-D-glucopyranosyl(1→4)-β-D-galactopyranoside(5),macorstemonoside F(6),macorstemonoside G(7),(25R)-26-O-β-D-glucopyranosyl-22-hydroxy-5β-furost-3β,26-diol-3-O-β-D-glucopyranosyl(1→2)-β-D-galactopyranoside(8).Compounds 2,5 and 8 could extremely inhibit the growth of cancer cell SF-268 and NCI-H460 at the concentration of 25 μg·mL -1.Conclusion Compounds 4,5 and 8 are isolated from Allium macrostemon Bunge for the first time and compounds 2,5 and 8 show anticancer bioactivities in vitro.

Their structures were determined by spectroscopic analysis and chemical method. All the isolated compounds were evaluated for their activities against ATP-citrate lyase (ACLY) and nuclear factor kappa B (NF-κB).

Abstract:Three new steroidal saponins, (25R)-ruscogenin-3-yl alpha-L-rhamnopyranosyl-(1 -> 2)-[beta-D-xylopyranosyl-(1 -> 4)]-beta-D-glucopyranoside (1). diosgenin-3-yl 2-O-acetyl-alpha-L-rhamnopyranosyl-(1 -> 2)-[beta-D-xylopyranosyl-(1 -> 4)]-beta-D-glucopyranoside (2), and pennogenin-3-yl 2-O-acetyl-alpha-L-rhamnopyranosyl-(1 -> 2)-[beta-D-glucopyranoside-(1 -> 4)]-beta-D-glucopyranoside (3) were isolated from the fibrous roots of Ophiopogon japonicus (THUNB.) KER-GAWL. Their structures were determined by spectroscopic methods including IR, HR-ESI-MS, and 1D- and 2D-NMR. All of these three steroidal saponins exhibited weak cytotoxicities against Hela and Hep2 cell lines.

Abstract:Much attention has been paid to cholestane-type steroidal glycosides because of their importance from the perspectives of both chemical diversity and significant biological activities. A phytochemical investigation of the rhizomes of Polygonatum odoratum (Liliaceae) resulted in the isolation of three novel cholestane-type steroidal glycosides (1-3) with unique Δ(14,16)-unsaturated D-ring structures as well as two novel spirostane-type steroidal saponins (4 and 5) and three known steroidal glycosides (6-8). Their structures were determined by various spectroscopic methods and chemical reactions. Steroidal saponin 7 showed significant antifungal activity against Candida albicans JCM1542 (MIC 3.1 μg/mL) and Aspergillus fumigatus JCM1738 (MIC 6.3 μg/mL).

Abstract:Fructus Tribuli is a traditional Chinese medicine used clinically for many years. Crude Fructus Tribuli and stir-fried Fructus Tribuli are recorded in the Pharmacopoeia of the People's Republic of China. However, the differences between steroidal saponins in crude Fructus Tribuli and stir-fried Fructus Tribuli have not been compared. In this study, ultra-high-performance liquid chromatography quadrupole time-of-flight mass spectrometry along with multivariate statistical analysis was developed to discriminate the chemical profiles and identify the steroidal saponins of crude Fructus Tribuli and stir-fried Fructus Tribuli. Additionally, an ultra-high-performance liquid chromatography triple-quadrupole mass spectrometer was used for the simultaneous quantification of nine major steroidal saponins to analyze the variations between crude Fructus Tribuli and stir-fried Fructus Tribuli. Finally, a total of 30 steroidal saponins whose structures or contents changed significantly after processing were found and identified. The mechanism of structural transformations deduced indicated that during the stir-frying of Fructus Tribuli, C-22 hydroxy furostanol saponins were converted to the corresponding furostanol saponins containing C-20-C-22 double bonds by dehydroxylation and deglycosylation reactions that occurred in the spirostanol saponins causing the generation of steroidal sapogenins. This study was successfully applied to the global analysis of crude Fructus Tribuli and stir-fried Fructus Tribuli. The results of this research will be beneficial to explore the processing mechanism of Fructus Tribuli.
