Abstract:An approximate 700 bp fragment of porcine circovirus(PCV2) ORF2 gene was inserted into the site between 1215th and 1216th bp of the 3′ end of VP2 gene of porcine parvovirus(PPV) SC-1 strain.The VP2-ORF2 gene was cloned into the eukaryotic expression vector pEGFP-C1 to construct the recombinant plasmid pEGFP.VO.After identification by digestion with restriction enzyme and PCR amplification,the plasmid pEGFP.VO was transfected into the COS-7 cells by liposome mediate.Then,the expression product was analysed by inversion fluorescence microscope,electron microscope,immunofluorescence and bioinformatics.The results showed that the recombinant plasmid pEGFP.VO was expressed in the COS-7 cells but virus-like particles were not observed by electron microscope.By analysis of the three-dimensional structure of PPV capsid protein and through immunofluorescence test,it was confirmed that the insertion position of the exogenous gene was located at the 3′ end of PPV VP2 gene,and the expression product was correspondingly located and packed in the multi-peptide C end of VP2 possibly affecting the formation of virus-like particles.This result indicated that the 3′ end of PPV VP2 gene was not suitable for the insertion of exogenous gene to construct the virus-like particles.
