Cloning and Its Molecular Characteristics Analysis of the B2L Gene of Orf Virus Isolated in Guizhou

Abstract:According to the published B2L gene sequence of ORFV strain in Genbank,one pair of primer was designed.The B2L gene of orf virus was amplified with the primer by PCR and then cloned into pMD18-T plasmids and sequenced.The acquired sequences contained 1 137 bp.The homology analysis revealed that the homology of the strain Guizhou strain and the Hubei strain(GU320351)was 99.1%,homology of the Guizhou strain and the North American orf virus isolated(AY278208)in 2003,and isolated strain from various ruminant species of a zoo(AY424971) in 2003 was 97.1%.The homology of deduced amino acid sequence shared 96.3% to 99.5%.There were a little variation between Guizhou strain and the reference virus strains.The test provided a theoretical basis of selection the Orf virus vaccine for Guizhou province.
