Selective Activation of Nuclear Factor- κB Nuclear Translocation by Intermittent Hypoxia in Human Umbilical Vein Endothelial Cells

Abstract:Objective:To construct a novel in vitro endothelial cell system for the simulation of intermittent hypoxia/reoxygenation(IH/ROX) or continuous hypoxia(CH),and explore the translocation of nuclear factor-κB(NF-κB) from cytosol to nuclear and changes of inflammatory status associated.Methods:A gas control delivery system was developed to permit the exposure of ECV304 cell cultures to IH/ROX cycles and CH mode.Prepared nuclear,cytoplasmic extraction and culture media were analyzed for standardized NF-κB optical density levels and interleukin(IL)-6 concentrations in this model with enzymelinked immunosorbent assay(ELISA).Results:This model resulted in cyclic fluctuations of cellular pO2 values between(76.28±1.29) mm Hg and(54.94±1.05) mm Hg during each hypoxic episode.With the model we could control gas tension and simulate IH/ROX and CH mode.Standardized nuclear NF-κB optical density levels in IH group were higher than those in correlated CH group(P < 0.01) which was the same in hypoxic extent and collected hypoxic duration.Both the nuclear NF-κB and IL-6 levels in IH group were higher than those in CH added to IH group(P < 0.01).With rehabilitation of 2 hrs from IH/ROX exposure,IH group still had higher nuclear NF-κB levels than control group(P < 0.01),whereas those in CH group did not change throughout the procedure(P > 0.05).Cytoplasmic NF-κB levels did not have obvious difference in all these groups.Conclusion:Our data indicated a selective activation of NF-κB translocation from cytosol to nuclear department by IH,which had greater extent than by CH,and the translocation could cause dramatic inflammatory reaction on cellular level.Though long after the termination of IH,ECV304 cells still had a substantial residual inflammation.
