Abstract:OBJECTIVE To study the acute and mutagenic risk of the extract from the fermentative products of Tremella aurantialba(FTAE),and provide experimental basIS for safety evaluation of the extract.METHODS According to the terms from"technical standards for test& toxicologieal assessment of health food"issued by Healthy Ministry of PRC,acute toxicity,and Ames,mouse marrow cell micronucleus,mouse sperm aberration and feeding test for 30 days were performed.After the FrAE was intragastrically administered three times at the maximal volume to mice in range of 1 day,the number of anmials poisoned and died in each group were noted daily for 14 consecutive days.The maximum dose was determined.The Ames tests using the Salmonella typhimurium strain TA97,TA98,TA100 and TA102 at five concentrations of 0.1,0.5,1.0,2.5 and 5.0 mg/utensil of the FTAE were carried out.The tests of sperm aberration bone and marrow cell micronucleus in mice were assayed at three intragastrical doses of 2.5,5.0 and 10.0 g·kg~(-1) of the FTAE,respectively.The feeding test for 30 days in rats was performed at three intragastrical doses of 2.5,5.0 and 10.0 g·kg~(-1) of the FTAE.RESULTS The oral acute toxicity study of the FTAE in mice revealed that the maximum dose should be no more than 24.0 g·kg~(-1),which was 240 times the maximum recommended human dose in the clinic practice.The results of genotoxicity tests of the FTAE were all negative,including Ames,mice marrow cell micronucleus and mice sperm aberration tests.No significant differences were observed in blood routine examination and chemieal indexes,hepatic and renal functions indexes,and organ coefficients among the 3 different dosage groups of the FrAE and compared with normal control group throughout the 30-day test period of rats.In addition,there was no abnormal condition for the pathologic examination of main organs.CONCLUSION The maximum dosage of FTAE should be no more than 24.0 g·kg~(-1) belongings to non-toxicity on the acute toxicity rating criteria and showed no genotoxicity in the expermiental condition.
