Multipotent differentiation potential of bone marrow adipocyte

Abstract:Objectives Bone marrow contains multipotent stromal cells that can differentiate into osteogenic, adipogenic, and reticular cells. The purpose of this study was to isolate the adherent cell from bone marrow adipose, and to investigate the differentiation potential of these cells. Methods Bone marrow adipocytes (BMAs) were obtained from the floating fat layer of bone marrow by adhesion culture methods. CD marker profiles of BMAs were examined by flow cytometry. Transcription factor PPAR-γ, C/EBP-α, and leptin were analyzed by RT-PCR. The multipotent differentiation potential of BMAs was investigated in vitro. Results Flow cytometry analysis showed that BMAs had similar CD marker profiles with bone marrow stromal cells (BMSCs). Mesenchymal lineage markers including CD105 and CD106 were observed in both types of cells, while CD49d was only expressed in BMAs. RT-PCR analysis showed that PPAR-γ2 , C/EBP-α and leptin, which are characteristic phenotype markers in post-adipogenesis period, were observed in BMAs. In vitro differentiation study showed that BMAs could differentiate into osteoblasts, and mature adipocytes. Conclusion In vitro study showed that bone marrow adipocytes had very similar phenotype characteristics with BMSCs. The multipotent differentiation potential of BMAs suggested that it might be an alternative cell source for tissue engineering.
