Abstract:Direct C-H functionalization of aryl ethers remains challenging owing to their low reactivity and selectivity. Herein, a novel strategy for nondirected C-H alkenylation of aryl ethers promoted by a dual ligand catalyst was demonstrated. This catalytic system readily achieved the highly efficient alkenylation of alkyl aryl ethers (anisole, phenetole, n-propyl phenyl ether, n-butyl phenyl ether and benzyl phenyl ether), cyclic aryl ethers (1,4-benzodioxan, 2,3-dihydrobenzofuran, dibenzofuran), and diphenyl oxides. Moreover, the proposed methodology was successfully employed for the late-stage modification of complex drugs containing the aryl ether motif. Interestingly, the compounds developed herein displayed fluorescent properties, which would facilitate their biological applications.

Abstract:Starting from commercially available simple olefms, alkene isomerization could realize the efficient construction of multi-substituted alkenes, which might be difficult to access with other synthetic methods, in an atom-economic manner via the stereoselective or positional-selective modulation of the carbon-carbon double bonds. The palladium catalyzed stereoselective (ZIE) and the positional selective isomerization reactions of alkenes are summarized. The mechanistic perspectives, and their synthetic applications in the synthesis of drug molecules and natural products are discussed in detail.

Abstract:A series of poly(2-oxazoline) (POX) derivatives bearing prolinamide pendants were designed as organocatalysts and evaluated in the direct asymmetric aldol reaction between aromatic aldehydes and cyclic ketones. The structural variation of the alkyl spacer connecting the polymer backbone with the catalytic unit was applied so as to deduce structure–performance relationships combined with comparable experiments from model catalysts. Results showed that the POX-bound prolinamides can promote the aldol reaction more effectively as compared to their small-molecular and non-POX-bound analogs. The catalyst P3 containing the pyrrolidine moiety closer to the tertiary amide backbone exhibited the overall best catalytic efficiency, affording anti-products in 84% yield with 89% ee in the representative aldol addition of cyclohexanone to 4-nitrobenzaldehyde at a 10 mol.% catalyst loading. Furthermore, the influence of trifluoroacetic acid as an additive on the asymmetric transformation was investigated. Theoretical calculations revealed that the protonation of the aldehyde carbonyl group switched the activation mode of the aldol acceptor through hydrogen bond interactions, thereby changing the relative energy barrier of the enamine/aldehyde reaction transition states, which accounted well for the significant improvement in the enantioselectivity of the acidic additives observed experimentally.

Abstract:Abstract Recently, the asymmetric bifunctionalization of alkenes has received much attention. However, the development of enantioselective alkoxyalkenylation has posed a considerable challenge and has lagged largely behind. Herein, we report a new palladium-catalyzed enantioselective alkoxyalkenylation reaction, using a range of primary, secondary, and tertiary γ-hydroxy-alkenes with alkenyl halides. By employing newly identified Xu-Phos ( Xu8 and Xu9 ) with a suitable side-arm adjacent to the PCy 2 motif, a series of allyl-substituted tetrahydrofurans were obtained in good yields with up to 95% ee . Besides ( E )-alkenyl halides, ( Z )-alkenyl halide was also examined and provided the corresponding ( Z )-product as a single diastereomer, supporting a stereospecific oxidative addition and reductive elimination step. Moreover, deuterium labeling and VCD experiments were employed to determine a cis -oxypalladation mechanism. DFT calculations helped us gain deeper insight into the side-arm effect on the chiral ligand. Finally, the practicability of this method is further demonstrated through a gram-scale synthesis and versatile transformations of the products.

Abstract:DFT calculations have been performed on the mechanism for the alkyloxycarbonylation of internal alkene with HCOOH catalyzed by palladium complex with P,N hemilabile ligands. Four different cycles have been probed in detail, and a plausible catalytic cycle involves decomposition of HCOOH/HCOOMe to CO, internal alkene isomerization, terminal alkene insertion, CO migratory insertion and methanolysis. It is shown that decomposition and isomerization processes exploit a cooperative P,N hemilabile ligand and Pd(0) (NH-Pd) rather than Pd(II) hydride (Pd-H) mechanism. Intriguingly, the simultaneous presence of PTSA acts as a hydrogen shuttle (H-shuttle), assisting CO-generation and methanolysis. With such a mechanism, the rate-determining transition state corresponds to internal alkene isomerization, which is congruent with the experimental observation that isomerization was the slow step in this process. The back-bonding between palladium and olefin and rapid hydrogen transfer in the presence of PTSA H-shuttle are responsible for the moderate barriers. In addition, careful study of the solvent effect indicates the polar solvents which are capable of hydrogen bonding, can promote the catalytic reactions. Mechanistic insights gained by this theoretical study have not only rationalized the experimental observations well but also have implications for new reaction development.

Abstract:Metal-catalyzed alkene arylalkoxylation is a powerful complexity-building strategy for the synthesis of oxygen heterocycles from simple γ-unsaturated alcohols, but only a few examples of catalytic enantioselective methods exist. Herein, an efficient palladium-catalyzed enantioselective arylalkoxylation of γ-hydroxyalkenes with aryl halides is reported. The salient features of this transformation include a remarkable broad substrate scope, mild reaction conditions, and good functional group tolerance, delivering a series of chiral tetrahydrofurans containing a tertiary or quaternary stereocenter in good yields with up to 95% ee. The Xu10 ligand with a suitable side-arm was responsible for the high reactivity and good enantioselectivity of this transformation.

Abstract:Highly regio- and enantioselective allylic alkylation has been achieved enabled by the merger of photoredox and palladium catalysis. In this dual catalytic process, alkyl radicals generated from 4-alkyl-1,4-dihydropyridines act as the coupling partners of the π-allyl palladium complexes. The generality of this method has been illustrated through the reaction of a variety of allyl esters with 4-alkyl-1,4-dihydropyridines. This mechanistically novel strategy expands the scope of the traditional Pd-catalyzed asymmetric allylic alkylation reaction and serves as its alternative and potential complement.

Abstract:A catalytic 1,2‐oxyhalogenation method that converts non‐conjugated internal alkynes into tetrasubstituted alkenes with high regio‐ and stereoselectivity is described. Mechanistically, the reaction involves a Pd II /Pd IV catalytic cycle that begins with a directed oxypalladation step. The origin of regioselectivity is the preference for formation of a six‐membered palladacycle intermediate, which is facilitated by an N,N‐bidentate 2‐(pyridin‐2‐yl)isopropyl (PIP) amide directing group. Selectivity for C(alkenyl)−X versus −N (X=halide) reductive elimination from the Pd IV center depends on the identity of the halide anion; bromide and iodide engage in C(alkenyl)−X formation, while intramolecular C(alkenyl)−N reductive elimination occurs with chloride to furnish a lactam product. DFT calculations shed light on the origins of this phenomenon.

Abstract:Primary alkyl halides have broad utility as fine chemicals in organic synthesis. The direct halogenation of alkenes is one of the most efficient approaches for the synthesis of these halides. Internal alkenes, in particular mixtures of isomers from refineries, constitute readily available and inexpensive feedstock and are the most attractive starting materials for this synthesis. However, the hydrohalogenation of alkenes generally affords branched alkyl halides; there are no catalytic methods to prepare linear alkyl halides directly from internal alkenes, let alone from a mixture of alkene isomers. Here we report the remote oxidative halogenation of alkenes under palladium catalysis via which both terminal and internal alkenes yield primary alkyl halides efficiently. Engineering pyridine-oxazoline ligands by introducing a hydroxyl group is essential for achieving excellent chemo- and regioselectivity. The catalytic system is also good for the mixture of alkene isomers generated from dehydrogenation of alkanes, providing a window to investigate the high-value utilization of inexpensive alkanes.

Abstract:A palladium-catalyzed reaction of aryl iodide-tethered alkenes with homoallyl alcohol is reported, providing a convenient and efficient approach to C(sp3)-allylation product. The cascade process involves intramolecular Heck/retro-allylation. This new approach has good functional group compatibility, broad substrate scope and high efficiency. This domino reaction further proved that homoallyl alcohol is a convenient and efficient allylation source.

Abstract:A palladium-catalyzed direct alkenation of thiophenes and furans has been developed in the presence of AgOAc and pyridine. A variety of olefinic substrates such as acrylates, acrylamides, and acrylonitrile can perform the direct oxidative coupling reactions with various thiophenes and furans to give the mono-alkenylated products in good yields. In most cases, the (E)-isomers were isolated as the major products.

Abstract:The palladium-catalyzed “regioreversed” α-hydroalkenylation and -hydroalkynylation of conjugated alkenes is re-ported. The unconventional selectivity of this reaction is achieved through the utilization of trans-spanning bidentate phosphine ligands that stabilize the proposed trans-Pd(II)Ln(R)(H) (R = alkenyl or alkynyl) intermediate. The α-hydrofunctionalization methods tolerate a variety of electron-deficient alkenes including α,β-unsaturated amides, esters and vinyl pyridines. A range of different alkenyl/alkynyl bromides are demonstrated to be effective coupling partners in the reaction. The phosphine-ligated palladium catalyst operates under mild weakly basic or neutral conditions and is resistant to air and moisture, giving rise to practical methodology for construction of β,γ-unsaturated carbonyl compounds.

Abstract:Palladium-catalyzed C-H activation reactions have attracted the attention of organic researchers due to their unique high selectivity, broad functional group tolerance, and high efficiency, and they are widely used in natural products and asymmetric synthesis. Here, we report an example of enantioselective C-H alkenylation between β-alkyl phenylethylamine compounds and styrenes with Boc-L-lle-OH as the ligand and nosylamide as the directing group. This reaction is applicable to styrene containing various electron-deficient and electron-donating substitutions and may be utilized for the synthesis of benzoazepine compounds.
