Abstract:In order to obtain high purity nattokinase from the fermentation broth of Bacillus subtilis, some separation and purification techniques were explored. The optimized separation and purification process includes the following steps: removing cells by the centrifugation, 20%~60% saturation ammonium sulfate precipitation, gel filtration chromatography with Superdex 75 and ion-exchange chromatography with SP Sepharose Fast Flow. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) was used to examine the purification effect, and the results indicate that the final product obtained is homogeneous and has a molecular weight around 27.7 kD. The fibrinolytic activity of the nattokinase was measured by means of fibrin plate method. The purification factor and activity recovery of the nattokinase are 8.4 and 49%, respectively. The specific activities of purified nattokinase could reach 28530 IU·mg-1. The results demonstrate that the combination of bioseparation process of ammonium sulfate precipitation with gel filtration and ion exchange chromatography is suitable for the separation and purification of nattokinase from culture broth of Bacillus subtilis. Especially for lab scale, the process mentioned above can be easily performed as an efficient way to prepare pure nattokinase with high yield.
Separation of Immunoglobulin from Porcine Plasma with Mixed-Mode Adsorption Chromatography

Abstract:Mixed-mode adsorption chromatography is a new kind of bioseparation technology,which normally combines the electrostatic and hydrophobic interactions.It has a unique property of salt-independent adsorption that can adsorb the target protein efficiently under either high-salt or low-salt condition.In the present work mixed-mode adsorption was used to separate immunoglobulin from porcine plasma.Firstly different methods were compared to precipitate the fibrinogen from porcine plasma,and the precipitation with ammonium sulfate was optimized.Then the adsorption isotherm and kinetics of mixed-mode adsorbent for immunoglobulin were investigated,and the conditions of adsorption and desorption were determined.Finally the mixed-mode adsorption chromatography was used to separate immunoglobulin directly from the supernate of ammonium sulfate precipitation.The yield was about 11.5 mg·ml~(-1),and the purity of immunoglobulin reached about 77.4%with the image analysis of SDS-PAGE.The process developed in the present work provided a new way for the comprehensive utilization of porcine plasma resources.
Purification and Stability of an Antithrombus Enzyme from Bacillus Subtilis

Abstract:An antithrombus enzyme (ATE) was precipitated by (NH4)2SO4 or ethanol from supernatant of Bacillus subtilis culture broth then purified using ion exchange chromatography on CM-sepharose fast flow. The effects of ionic strength and pH value on protein adsorption, the gradient elution at different flow rates and step elution were examined respectively. The recovery yield of the optimised process was 74.5% with a purification factor 8.1. The ATE molecular weight was estimated as 30ku by SDS-PAGE. The experimental results showed that the enzyme was stable in the range of pH 7 to pH11, and temperature 25℃ to 37℃.
Development of an effective method for purifying trypsin using a recombinant inhibitor

Abstract:A trypsin affinity material was prepared by covalently immobilizing buckwheat trypsin inhibitor (BTI) on epichlorohydrin-activated cross-linked agarose gel (Selfinose CL 6 B). The optimal conditions for activating Selfinose CL 6 B were 15 % epichlorohydrin and 0.8 M NaOH at 40 °C for 2 h. The optimal pH for immobilizing BTI was 9.5. BTI-Sefinose CL 6 B showed a maximum adsorption capacity of 2.25 mg trypsin/(g support). The material also displayed good reusability, retaining over 90 % of its initial adsorption capacity after 30 cycles. High-purity trypsin was obtained from locust homogenate using BTI-Selfinose CL 6 B through one-step affinity chromatography. The molecular mass and Km value of locust trypsin were determined as 27 kDa and 0.241 mM using N-benzoyl-DL-arginine-nitroanilide as substrate. The optimal temperature and pH of trypsin activity were 55 °C and 9.0, respectively. The enzyme exhibited good stability in the temperature range of 30-50 °C and pH range of 4.0-10.0. BTI-Selfinose CL 6 B demonstrates potential application in the preparation of high-purity trypsin and the discovery of more novel trypsin from various species.
Preparation of High-purity Aprotininum by Crosslinked Chitosan Microsphere

Abstract:The trypsin was covalently linked with the crosslinked chitosan microsphere, which was prepared through inversing suspension crosslinking and was used to isolate and purify aprotininum from the extract of cattle lungs by affinity chromatography. The result showed that the activity of immobilized trypsin on the crosslinked chitosan microsphere was 1270 KIU·g-1, 52.4 %protein was coupled, and the activity recovery of trypsin was 50.8 %. The special activity of the prepared aprotininum was 5650 KIU·mg-1. The immobilized trypsin had the advantage of low no-special absorption, high strength, easy avtivation and very cheap.
Purification of Trypsin by Using AOT/Isooctane Reverse Micellar Extraction and Overcoming Denaturation and Precipitation in the Process

Abstract:Trypsin was purified from crude material of pig pancreas by AOT/isooctane reversed micellar system.The influence of the main operating parameters such as ethanol concentration in forward and backward extraction,pH,KCl and AOT concentration,temperature were investigated.Forward and backward extraction recovery of trypsin reached almost 90% and neared 100%,respectively.Finally,about 88% of total yield was obtained,and the specific activity of trypsin was increased to over 1800U/mg with purification factor of 5 folds more.In AOT-isooctane reverse micellar extraction system,denaturation or precipitation of proteins always occured due to strong electrostatic interaction between AOT-proteins molecules.It had been resolved by adding ethanol into reverse micellar system,and no denaturation was observed.Otherwise,the phase separation time was shortened significantly because of ethanol added.It was only 10 minutes or less to reach phases separation after forward and backward extraction.If this method can be applied in industry,efficiency will be greatly improved.

Abstract:Aprotinin is a 58 amino acid bovine protease inhibitor which inhibits a range of serine proteases including trypsin,chymotrypsin,plasmin,kallikrein and trombin.This single-chain polypeptide with the molecular mass of 6.5 kDa and the PI of 10.5 is crosslinked by three disulphide-bond bridges.It was initially isolated from bovine pancreas by Kunitz and Northrop.Aprotinin has been widely used in various fields.It can be used as inhibitor in protein purification procedure and tissue culture to prevent the degradation.In clinical use,because of its antifibrinolytic properties,aprotinin can be used in cardiac surgery to decrease postoperative blood loss and preserve platelet function during cardiopulmonary bypass.Due to the large requirement of aprotinin,the methods to produce aprotinin with high homogeneity and yields have been studied.Most of the reports regarding this topic are about purification of aprotinin from bovine tissues.But these purification procedures are complicated and costly.So the development of effective recombinant expression system is necessary.In this study,the deoxyoligonucleotide encoding aprotinin was synthesized according to the codon prefernce of E.coli.The DNA sequence was amplified by PCR and inserted into the expression vector pET28a between the NdeI and XhoI sites.The recombinant expression plasmid was then transformed into E.coli strain Rosetta(DE3),and recombinant aprotinin(r-aprotinin) was expressed as inclusion body by IPTG induction.The amount of r-aprotinin expressed accounted for 8%～10% of the total bacterial protein.After purification of inclusion body,renaturation and CM cation-exchange chromatography,2.4 mg r-aprotinin was obtained from one liter of shaking flask culture of E.coli with homogeneity greater than 90%.Kinetic studies demonstrated that r-aprotinin competitively inhibited the amidolytic activity of plsamin against small substrate S2251. The inhibition constant(Ki) was(0.6±0.3) nmol/L,which was similar to that of the native aprotinin.
Purification and biochemical characterization of a novel fibrinolytic enzyme, PSLTro01, from a medicinal animal Porcellio scaber Latreille

Abstract:A novel protease, named PSLTro01, with fibrinolytic and anticoagulant activity was isolated from Porcellio scaber Latreille and was purified by a combination of hollow fibre membrane molecular weight cut-off (MWCO), ammonium sulfate fractionation, gel filtration and ion-exchange chromatography. PSLTro01 is a single-chain protein with a molecular mass of 38,497 Da as estimated by non-reduced SDS-PAGE and MALDI-TOF MS spectrometry, and its N-terminal 15 amino acid sequence was determined as DINGGGATLPQPLYQ. PSLTro01 is stable in the range of 20-40 °C and pH 6.0-10.0, with a maximum fibrinolytic activity at 40 °C and pH 7.0. The PSLTro01-induced fibrinolytic activity was not influenced by K(+) or Na(+) but was slightly increased by Mg(2+) and completely inhibited by aprotinin and pepstatin A. Fibrin plate assays revealed that PSLTro01 could not directly degrade fibrin but was a plasminogen activator. PSLTro01 exhibited high specificity for the substrate S-2251 for plasmin, followed by S-2238 for thrombin and S-2444 for urokinase. Moreover, the fibrinogenolysis pattern of PSLTro01 was Aα-chains>Bβ-chains>γ-chain. Tail-thrombus of the enzyme treated group was significantly shorter than the physiological saline treated group and the thrombus decrement was correlated with the enzyme dose. PSLTro01 prolongs both thrombin time (TT) and activated partial thromboplastin time (APTT). These results indicate that PSLTro01 may have potential applications in the prevention and treatment of thrombosis.

Abstract:An extracellular alkaline protease produced by Bacillus licheniformis AP-1 was purified 76-fold, yielding a single 28 kDa band on SDS-PAGE. It was optimally active at pH 11 and at 60°C (assayed over 10 min). The protease was completely inhibited by phenylmethylsulfonyl fluoride and diodopropyl fluorophosphate, with little increase upon Ca 2+ and Mg 2+ addition.

Abstract:Trypsin from the pancreas of Tibetan sheep was purified to 7.4‐fold by ammonium sulphate and acetone precipitation, followed by Sephacryl S‐200 (Whatman, Maidstone, England) and Sephadex G‐75 (Whatman) gel filtration, with an 23.2‐fold increase in specific activity and 13.6% yield. The final enzyme preparation was nearly homogeneous on sodium dodecyl sulfate‐polyacrylamide gel electrophoresis (SDS‐PAGE) and the molecular weight of the enzyme was estimated to be approximately 27 kDa by SDS‐PAGE. Trypsin‐like enzymes had maximal activities at around pH 9.0 and 60C for the hydrolysis of Nα‐ρ‐Tosyl–L–arginine methyl ester hydrochloride (TAME). Trypsin was unstable above 60C and below pH 4.0, and was stabilized by calcium ions. Purified trypsin had a Michaelis–Menten constant (Km) and catalytic constant (Kcat) of 0.53 mM and 206 s−1, respectively, when TAME was used as the substrate. The specific trypsin inhibitors, soybean trypsin inhibited, Nα‐ρ‐tosyl‐L‐lysine chloromethyl ketone and phenyl methyl sulfonyl fluoride, strongly inhibited the activity of trypsin, while other protease inhibitors exhibited negligible inhibition. The result suggests that major proteinase in the pancreas of Tibetan sheep was trypsin‐like serine proteinase. Tibetan sheep is one of the unique livestock resources in western China. It was found that the trypsin extraction and purification from waste pancreas have the biological characteristics of the heat resistance, alkali resistance, high specific activity and high hydrolysis efficiency. It does not only serve as a better research tool enzyme, but is also widely applied in the food, pharmaceutical, textile and other industries. This paper provides the scientific research basis of the development of the waste pancreas for the Tibetan sheep, therefore reducing environmental pollution.

Isolation and characterization of the pig endometrial arylsulphatase A

Abstract:The pig endometrial arylsulphatase A was purified 3322-fold to a specific activity of 150 mumol/min per mg. The purification involved (NH4)2SO4 fractionation, chromatography on concanavalin A-Sepharose and DEAE-Sepharose, gel filtrations on Sephadex G-200 at pH 7.4 and 5, and a new preparative gel-electrophoresis technique. The homogeneous enzyme is a glycoprotein containing 20% carbohydrate. The purified enzyme has Mr about 120 000 and it contains subunits of Mr 63 000. The pig endometrial arylsulphatase A shows many properties in common with those of arylsulphatases A purified from other sources. The similarities include their low isoelectric points, the anomalous time-activity relationships, multi-pH optima, inhibition by SO3(2-), SO4(2-), phosphate ions, metal ions and nucleoside phosphates, pH- and ionic-strength-dependent polymerization and amino acid composition.

Abstract:Separation of biliproteins from Spirulina platensis extracts was accomplished by ion exchange chromatography on DEAE Sepharose Fast Flow, which appears superior to other three kinds of anion exchangers, i.e., ANX Sepharose 4 Fast Flow (high sub), ANX Sepharose 4 Fast Flow (low sub) and Q Sepharose Fast flow, with respect to dynamic capacity and purity of objective product. The electrophoretic titration curves of c-phycocyanin (CPC) and allophycocyanin (APC) show that these two proteins have similar charge properties and their electrophoretic mobilities are approaching, both of which varies little against pH. Separation of CPC and APC is done by chromatography on hydroxylapatite, which results in products of high purity and homogeneity demonstrated by IEF analysis.

Abstract:Acuthrombin-B, a thrombin-like enzyme from Agkistrodon acutus venom, has been isolated and purified to homogeneity by ion-exchange chromatography on DEAE-Sepharose, gel filtration on Sephacryl S-100 and fast performance liquid chromatography on DEAE-8HR. The protease is an acid protein (pI 6.0) consisting of two non-identical polypeptide chains (14.4 and 16 kDa) and there is no disulfide bond between the subunits. Its molecular weight is 27 kDa as estimated by gel filtration on Sephacryl S-100. The protease has arginine-esterase activity and hydrolyzes synthetic substrates such as p-toluenesulfonyl arginine methyl ester and alpha-N-benzoyl-L-arginine amide ethyl ester, and shows clotting activity with human fibrinogen, rabbit citrated plasma and human citrated plasma in vitro. The specific activity with human fibrinogen was estimated to be 230 NIH units mg-1. The protease is considered as a serine-type protease and contains metal ion(s) to some extent, as indicated by the fact that its clotting and arginine-esterase activities could be completely inhibited by PMSF and partially inhibited by the chelating agent EDTA, while the thrombin inhibitor heparin had no effect on its clotting activity towards rabbit citrated plasma. Acuthrombin-B crystals with a resolution limit of 2.06 A were obtained by conventional hanging-drop vapour diffusion. The crystals belong to space group P21 with unit-cell parameters a = 34.97, b = 53.58, c = 67.88 A, beta = 98.89 degrees and contain one molecule per asymmetric unit.
Isolation and purification of apolipoprotein H with heparin affinity column and preparation and identification of anti-ApoH

Abstract:Objective:To purify apolipoprotein H (apoH) from fresh human serum with heparin affinity column self-prepared and to prepare polyclonal antibody anti-ApoH.Methods:Affinity column was made by reaction of indirect reductive amination and operating conditions were optimized and their effects were investigated.The apolipoprotein H antigen was purified from human serum by methods of perchloric acid precipitation,ion exchange and affinity chromatography.The polyclonal antiserum against the antigen was obtained by immuning rabbits.Results:The amount of heparin immobilized on the synthesized gel was 5 mg/ml (medium).Molecular weight of the purified antigen was about 50 kD by SDS-PAGE and no other bands were seen,the percentages of 16 amino acids were consistent with that reported previously.In ELISA and dot-ELISA,the polyclonal antibody displayed a cross reaction with bovine serum albumin,while no such reaction was observed in result of Western blot under reduced and denatured condition.Conclusion:The self-prepared affinity column costs very low,but has excellent affinity performance and stable heparin linking,and high purified apoH can be obtained by using the reagent.Polyclonal antibody needs to be further purified before detecting serum apolipoprotein H.
Enrichment of Trypsin Inhibitor from Soybean Whey Wastewater Using Different Precipitating Agents and Analysis of Their Properties

Abstract:Recovering valuable active substances from the by-products of agricultural processing is a crucial concern for scientific researchers. This paper focuses on the enrichment of soybean trypsin inhibitor (STI) from soybean whey wastewater using either ammonium sulfate salting or ethanol precipitation, and discusses their physicochemical properties. The results show that at a 60% ethanol content, the yield of STI was 3.983 mg/mL, whereas the yield was 3.833 mg/mL at 60% ammonium sulfate saturation. The inhibitory activity of STI obtained by ammonium sulfate salting out (A-STI) was higher than that obtained by ethanol precipitation (E-STI). A-STI exhibited better solubility than E-STI at specific temperatures and pH levels, as confirmed by turbidity and surface hydrophobicity measurements. Thermal characterization revealed that both A-STI and E-STI showed thermal transition temperatures above 90 °C. Scanning electron microscopy demonstrated that A-STI had a smooth surface with fewer pores, while E-STI had a rough surface with more pores. In conclusion, there was no significant difference in the yield of A-STI and E-STI (p < 0.05); however, the physicochemical properties of A-STI were superior to those of E-STI, making it more suitable for further processing and utilization. This study provides a theoretical reference for the enrichment of STI from soybean whey wastewater.

Abstract:Lipase from Antarctic krill, with a molecular weight of 71.27kDa, was purified with ammonium sulfate precipitation and a series of chromatographic separations over ion exchange (DEAE) and gel filtration columns (Sephacryl S-100), resulting in 5.2% recovery with a 22.4-fold purification ratio. The optimal pH and temperature for enzyme activity were 8.0 and 45°C, respectively. Purified lipase had Km and Vmax values of 3.27 mmol L-1 and 2.4 Umg-1, respectively, using p-nitrophenyl laurate as the substrate. Li-pase activity was enhanced by adding Ca2+ and Mg2+ ions in the concentration ranges of 0–0.5 mmol L-1 and 0–0.3mmol L-1, respectively, while the activity was inhibited by a further increase in these ion concentrations. Fe3+ and Cu2+ ions showed obvious inhibitory effects on enzyme activity, and the inhibition rates were 71.8% and 53.3% when the ion concentrations were 0.5 mmol L-1.

Abstract:An enzymatic hydrolysis approach was proposed for the preparation of bioactive hydrolysate of scorpion Buthus martensii Karsch (BmK) protein. Results showed that the anticoagulant activity of the hydrolysates of BmK protein was closely related to both the enzyme type and the degree of hydrolysis. Alcalase AF showed to be the best enzymes for the hydrolysis. And the hydrolysis degree (DH) was closely related with the anticoagulant activity of the hydrolyzate. At a DH value of 18% with Alcalase AF, the hydrolyzate exhibited the highest activity. The hydrolysate was then separated and purified by consecutive chromatographic procedures, giving a novel anticoagulant peptide consisting of ten amino acids (MW: 1119.8Da) with its sequence of Val-Glu-Pro-Val-Thr-Val-Asn-Pro- His-Glu identified by MALDI-TOF/TOF MS. The negatively charged amino acids and hydrophobic amino acids may contribute to the anticoagulant activity of the prepared peptides. And the Val residue in N-terminal was also critical for the anticoagulant activity of the BmK peptide. Furthermore, the anticoagulant activity kept stable after in vitro digestive simulation. This research provided a promising bioprocessing route for production of anticoagulant peptides using BmK protein as a potentially valuable bioresource.
Characterisation of Ag1, the major species-specific contaminant of bovine crude heparin, and its identification as an aprotinin/heparin complex

Abstract:Heparin is a potent anticoagulant polysaccharide purified for decades from ruminants or porcine tissues. However, with the emergence of bovine spongiform encephalopathy (BSE), the source of pharmaceutical heparin is currently restricted to porcine intestinal mucosa. A major species-specific contaminant, called Ag1, has recently been identified in bovine crude heparin [Rivera et al., J. Pharm. Biomed. Anal., submitted] and used to develop an enzyme-linked immunosorbent assay (ELISA) for the species origin control of crude heparins [Levieux et al., J. Immunoassay, submitted]. In this report, we describe the different investigations, which were carried out to identify Ag1. This antigen was first localised by immunohistological studies essentially in the connective tissue of the bovine small intestine. After extraction from an intestinal extract by immuno-affinity chromatography, Ag1 was isolated as a single band by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). Ag1 was then partly sequenced and identified as an aprotinin/heparin complex. Aprotinin, also known as the bovine pancreatic trypsin inhibitor (BPTI), is present with heparin in mast cells, and is very resistant to heat, pH, chemical treatments and proteolytic digestion. The stability of Ag1 towards the different treatments performed during heparin extraction process allows this protein to remain in sufficient amounts in crude heparin and makes it an ideal target for the immunochemical control of the absence of bovine material in crude heparins.
Expression, Purification and Activity Measurement of Soluble BACE Protein in E.coli.

Abstract:To express BACE (beta-site APP cleaving enzyme), an aspartic protease related to Alzheimer's disease in E.coli to obtain the active protein after refolding and purification, the sequence for soluble form of BACE was inserted into prokaryotic expression vector pET11a. After expression in E.coli BL21(DE3), the protein was obtained as inclusion bodies, after refolding and purification. The proteolytic activity of the protein was measured by HPLC and MS. After refolding and purification, the protein exhibited beta-secretase activity by cleaving the synthetic peptide, which was designed according to the beta-secretase site at Swedish mutant of APP, and according Hanes method the kinetics constants for the synthetic peptide of BACE protein were measured. The study will facilitate BACE protein structure determination and inhibitor development efforts, which may lead to the evolution of promising and effective treatments for Alzheimer's disease.

Abstract:We purified and characterized xylanase from Bacillus sublitis JH-1. The crude enzyme was purified by ammonium sulfate fractionation,dialysis desalination and DEAE-Sepharose FF anion exchange chromatography.The specific activity was 75 899. 68 U/mg,and its subunit molecular weight was 3. 45× 104 determined by SDS-PAGE.The xylanase showed optimal activity at pH 6. 0.It retained more than 75%of its maximum activity at pH 6. 0 for 2 h,and the higher the pH,the faster the activity lost. The optimal temperature was 55℃ and it retained more than 70% of its maximum activity between 50 and 60℃ for 2 h. Metal ions Fe2 +, Mg2+, Ca2+, Zn2+, Ba2+ and low concentration ( 5 mmol/L) of Fe3+have obviously promoting effect on xylanase activity,whereas Mn2+ and high concentration(10 mmol/L) of Fe3+inhibited the enzyme.
