Role of 5-Aza-2'-deoxycytidine in Breast Cancer Cells CDH4 Promoter Demethylation

Abstract:Background and purpose:Cadherin(CDH),as a class of important cell adhesion molecule(CAM),is significantly decreased expressed in the malignant carcinomas through the pathogenesis of the abnormal methylation of gene promoter region.The purpose of this study was to investigate the recovery of CDH4 expression and function in human breast adenocarcinoma cell line MCF-7 induced by 5-aza-2'-deoxycytidine(5-Aza-CdR) after the demethylation of CDH4 gene promoter region.Methods:In the present study,methylation-specific PCR(MSP) was used to assess the role of 5-Aza-CdR in the methylation of CDH4 promoter in the MCF-7 cell lines,which was treated with different concentration of 5-Aza-CdR.We mapped the expression of CDH4 the MCF-7 cell lines by RT-PCR pro-and posto-treatment of 5-Aza-CdR.The survival of MCF-7 cells were detected by MTT assay and the migration of MCF-7 cells was also assessed by the scratch assay exposed to 5-Aza-CdR or vehicle control.Results:Loss of CDH4 expression could be restored by the methyltransferase inhibitor 5-aza-2'-deoxycytidine in the MCF-7 cell lines,which is proportional to the concentration of 5-Aza-CdR.The survival rates were significantly decreased in MCF-7 cell lines treated with 5-Aza-CdR of the different concentrations(10,20 and 40 μmol/L) compared with control group(concentration of 0 μmol/L,both P0.05).Restored expression of CDH4 in the MCF-7 cell lines inhibited cell proliferation,colony formation and cell migration.Conclusion:Methylation of aberrant promoter region can silence CDH4 gene in MCF-7 cell.5-Aza-CdR played an important role in the recovery expression of CDH4 and the inhibitation of cell proliferation and migration.This suggests that the restoration of CDH4 expression may be one of the mechanisms for the biological behavioral changes of MCF-7 cell.
