Effects of cholesterol on osteogenic differentiation of human ostosarcoma MG63 cells

Abstract:Objective To investigate the effects of cholesterol on osteogenic differentiation of human ostosarcoma MG63 cells and the underlying mechanisms.Methods MG63 cells were cultured in medium with 0, 12.5, 25, and 50 μg/mL of cholesterol for 24, 48, and 72 h,to investigate the inhibition ratio using MTT. The cell lysate were collected for the ALP, SOD and MDA testing.3) The expression of ALP, ColA1, BMP2 and Cbfa1 genes in MG63 cells after the treatment of cholesterol was measured by the qRT-PCR assay. Results Free cholesterol inhibited the proliferation of MG63 with different concentrations and different times. Cholesterol treatment reduced the activity of both ALP and SOD, while increased the level of MDA. Cholesterol reduced the expression of ALP,Collagen I,BMP2 and Cbfa1 compared with the control group. Conclusion Free cholesterol in vitro experiments inhibited the proliferation of osteoblasts, which may involved in hypercholesterolaemia induced the increased risk of osteoporosis.
