Abstract:Objective:An HPLC-MS/MS method for determination of tolterodine in human plasma was established and the pharmacokinetics of tolterodine fumarate/tartrate were studied in Chinese healthy volunteers.Methods: Tolterodine and the internal standard (buflomedil) were extracted by ether-n-hexane (3:1),then separated on an Hypurity C_(18)column,and were detected by a MS/MS detector.A single oral dose of 3.72 mg of tolterodine fumarate or 4 mg toherodine tartrate was administered to 12 (male 6,female 6) healthy volunteers in a randomized crossover study.In the multi-dose research,3.72 mg toherodine fumarate twice a day for 6 d were give to 12 (male 6,female 6) healthy volunteers.Tolterodine concentrations in plasma were determined by the newly developed HPLC-MS/ MS method,and the pharmacokinetics were studied.Results:The calibration curve was linear with the range 0.1 to 20.0 μg·L~(-1) (r=0.9999) ,the recovery of tolterodine from plasma was in the range of 102.4%-108.0% (n= 5),inter-day and intra-day RSD were 5.0%-10.6 % (n=5) and 9.2%-10.3 % (n=5) respectively.After a single oral of 3.72 mg or 4 rag,the concentration-time data was best fit to a one-compartment open model with first-order absorption kinetics.The C_(max) of tolterodine fumarate/tartrate were (8.2±5.5) μg·L~(-1) and (8.4± 5.7)μg·L~(-1),T_(max) were (1.8±0.5)h and (1.9±0.5)h,AUC_(0→12) were (45.2±32.3)μg·h·L~(-1) and (46.5 ±34.3)μg·h·L~(-1),AUC_(0→∞) were (55.6±42.5)μg·h·L~(-1) and (56.5±42.9 )μg·h·L~(-1),T_(1/2) were (3.8 ±1.7)h and (3.8±1.9 ) h,respectively.Results of statistics showed no significant differences between these pa- rameters of the two formulations,but C_(max),AUC_(0→12),AUC_(0→∞) between male and female volunteers.After multi-dose of tolterodine fumarate,the mean C_(min)~(ss) was (13.3±5.6)μg·L~(-1),T_(max)~(ss) was (1.7±0.3)h,AUC_(0→12)~(ss) was (68.3± 33.6)μg·h·L~(-1),C_(min)~(ss) was (1.8±1.5)μg·L~(-1),C_(av)(5.7±2.8)μg·L~(-1),DF was 2.1±0.6.These parame- ters have no significant differences between the two genders.Conclusions:The established HPLC-MS method is accuracy and sensible for tolterodine determination.The pharmacokinetics of tolterodine has no significant changes when the acid radical changes from tartrate to fumarate.The reason of the significant differences of parameters be- tween genders is related the EM and PM.No-significant accumulation is observed after given the volunteers 3.72 mg toherodine fumarate twice a day for 6 days.
