Identification and Expression Analysis of Cold-Regulated Genes in Vitis Amurensis Rupr. Cv. Zuoshan-1

Abstract:Summary Low temperatures in Winter, as well as cold spells in Spring, can cause severe damage to grapevines. Although cold-tolerance in grapevine is not fully understood, the genetic mechanisms responsible for cold-tolerance and cold-acclimatisation responses have been studied comprehensively in Arabidopsis thaliana, tomato, and rice. In order to identify cold-induced genes, and to analyse gene expression profiles during cold-acclimatisation, a cDNA library was constructed using suppression subtractive hybridisation from a clonal selection of wild Vitis amurensis ‘Zuoshan-1’ leaves exposed to 4ºC for 48 h. A total of 270 clones were picked at random and 176 were successfully sequenced. Of these, 127 sequences (16 genes) showed high homology to known Vitis vinifera ‘Pinot Noir’ sequences. The 16 genes were functionally annotated, and seven of them (genes encoding ORF61c, a cell wall-associated hydrolase, a kinase-like protein, a serine/threonine kinase, Rubisco activase, Rubisco large subunit, and a SQUAMOSA promoter-binding-like protein) were correlated with putative cold-hardiness genes in Arabidopsis and in woody perennial plants. Such genes have been described previously, with a wide range of biological functions including transcription, signal transduction, cell rescue, metabolism, and photosynthesis. Expression analysis revealed that all seven V. amurensis cv. Zuoshan-1 genes were upregulated by cold treatment, especially those genes associated with senescence.
