Experimental study of Ping-tang recipe improves glucose and Lipid metabolism in high fat diet-induced obese rats

Abstract:Objective To investigate the effects and mechanism of Ping-tang recipe (PTF) on Glucose and lipid metabolism in high-fat induced obese rats. Methods Through high-fat diet to build fat rat model, rats in the high-fat diet were randomly allocated into three groups (10 rats each group): control, high fat diet with treatment of low dose (0.42g/kg.d) and high dose (0.42g/kg.d)PTF respectively, and another 10 rats were given normal diet as the normal group. After 8 weeks of treatment, Insulin sensitivity were tested by insulin tolerance test (ITT) and oral glucose tolerance test (OGTT), Serum chemistry were analyzed by an automatic chemistry analyzer. Fat tissue morphology were tested by hematoxylin and eosin (H&E) stain, The mRNA change of AMPK, leptin and TNF-αwere analyzed by real-time PCR. Results After 8 weeks of PTF treatment, Rats treated with low-dose or high-dose PTF had less epididymal and perirenal adipose tissue compared with the HFD control group(P<0.05). fat cell volume decreased and insulin resistance was improved. PTF also decreased serum triacylglycerol (TG) and low density lipoprotein (LDL) level. The expression of AMPK-α2 were increased, and the adipokines including leptin and TNF-α were decreased (P<0.05). Conclusion PTF is effective in reduce visceral fat content, improve insulin resistance, improve glucose and lipid metabolism, the mechanism in part through regulating AMPK signaling and adipokines expression.
