Abstract:Glutenin plays an important role in influencing the elasticity and breadmaking quality of dough.As is well known,the structure of organic molecule determines its function.Therefore,the knowledge about the structure and composition of glutenin laid the foundation for the research on flour quality.In this paper,advances in the research of characteristics of glutenin structure,which were obtained with the help of techniques such as protein isolation,polypeptide synthesis,mass spectrometry,spectroscopy analysis and scanning tunneling microscopy,were reviewed.Meanwhile,the applications of protein structure prediction and modeling methods in the research of glutenin structure were also reviewed in the paper.On this basis,the modes of glutenin inter-and intra-disulfide bond formation were discussed finally.

Abstract:Abstract Vital wheat gluten plays an important role in the food industry, especially in baking to help standardize dough properties and improve bread volume. However, a fundamental characterization of a wide variety of vital gluten samples is not available so far. This would be necessary to relate compositional characteristics to the production process. Therefore, we analyzed the content of crude protein, starch, lipids and ash, oil and water absorption capacity, particle size distribution, gluten protein composition and spectroscopic properties of 39 vital gluten samples from 6 different suppliers. Principle component analysis of all analytical parameters revealed that the samples from one specialized vital gluten manufacturer had a different composition and a greater variability compared to all other samples from wheat starch producers. While the composition of vital gluten samples from the same manufacturer was similar and the score plot showed a cluster formation for samples from three suppliers, the variability over all samples was comparatively low. The samples from the other suppliers were too similar altogether so that it was hardly possible to identify clear differences, also related to functionality.

Abstract:Wheat flour can form dough with a three-dimensional viscoelastic structure that is responsible for gas holding during fermentation and oven-rise, creating a typical fixed, open-cell foam structure of bread after baking. As the major components of dough, the continuous reticular skeleton formed by gluten proteins and the concentrated starch granules entrapped in gluten matrix predominantly determine dough rheological behaviors and bread qualities. This review surveys the latest literatures and draws out a conclusion from a plethora of information related to the filling effects of starch granules on gluten matrix and the cross-linking mechanisms between gluten proteins and starch granules, which is of great significance to provide sufficient scientific knowledge for development of bread with satisfactory attributes and quality control of end products.

Abstract:The quality of wheat flour is largely determined by the properties of gluten proteins. Chemical components that influence gluten proteins are used as flour improvers in the flour industry. In this study, tannins which are natural occurring polyphenols were found to improve dough mixing properties. The effects of tannins on the physicochemical and structural properties of gluten proteins were examined, and the results showed that tannins promoted the non-covalent interactions among gluten proteins, although they induced SH/SS interchange reactions in the dough. The β-turn and α-helix conformations were increased, whereas the β-sheet conformation was decreased in dough containing tannins as detected by FTIR. Moreover, the addition of tannins promoted the aggregation of gluten proteins, modified the microstructure of gluten networks, and improved the mixing properties. The positive effects of tannins on dough properties implied the potential of tannins as a new flour improver.

Abstract:Gluten is a key component that allows wheat flour to form a dough, and it is also a byproduct of the production of wheat starch. As a commercial product, wheat gluten is increasingly used in the food-related industry because of its versatile functional properties and wide range of sources. Wheat gluten is manufactured industrially on a large scale through the Martin process and batter process and variants thereof. Gliadin and glutenin impart cohesiveness and elasticity properties, respectively, to wheat gluten. The formation of gluten networks and polymers depends mainly on covalent bonds (disulfide bonds) and noncovalent bonds (ionic bonds, hydrogen bonds, and hydrophobic interactions). The multifunctional properties (viscoelasticity, gelation, foamability, etc.) of wheat gluten are shown by rehydration and other processing techniques. Wheat gluten has been widely used in wheat-based products, food auxiliary agents, food packaging, encapsulation and release of food functional ingredients, food adsorption and heat insulation materials, special purpose foods, and versatile applications. In the future, wheat gluten protein will be used as an important raw material to participate in the development and preparation of various food and degradable materials, and the application potential of wheat gluten in food-related industries will be massive. This review summarizes the main manufacturing processes, composition, and structure of gluten protein, and the various functional properties that support its application in the food and related industries. The Additive Interactions Between High-Molecular-weight Glutenin Subunits and Tannic Acid Improve the Wheat Quality

Abstract:Wheat gluten proteins, especially high-molecular-weight glutenin subunits (HMW-GS), are the main contributor to flour processing quality. Tannic acid (TA) consisting of a central glucose unit and ten gallic acid molecules is a phenolic acid that improves the processing quality. However, the underlying mechanism of TA's improvement remains largely unknown. Here, we showed that TA's improving effects on gluten aggregation, dough-mixing and bread-making properties were directly associated with the kinds of HMW-GS expressed in wheat seeds in HMW-GS near-isogenic lines (NILs). We established a biochemical framework, elucidated the additive effects of HMW-GS-TA interaction and discovered that TA cross-linked specifically with wheat glutenins but not gliadins, and reduced gluten surface hydrophobicity and SH content depending on the kinds of expressed HMW-GS in the wheat seeds. We also demonstrated that hydrogen bonds play an essential role in TA-HMW-GS interactions and improvement of wheat processing quality. Additionally, the effects of TA on the antioxidant capacity and on nutrient (protein and starch) digestibility were also investigated in the NILs of HMW-GS. TA increased antioxidant capacity but did not affect the digestion of starches and proteins. Our results revealed that TA more effectively strengthened wheat gluten in the presence of more HMW-GS kinds, highlighting TA's potential as an improver toward healthy and quality bread and demonstrating that manipulating hydrogen bonds was a previously overlooked approach to improve wheat quality.

Abstract:Avenin-like b proteins may contribute to the viscoelastic properties of wheat dough via inter-chain disulphide bonds, due to their rich cysteine residues. In order to clarify the effect of the avenin-like b proteins on the functional properties of wheat flour, the functional and biochemical properties of wheat flour were analyzed in three transgenic wheat lines overexpressing the - gene using the sodium dodecyl sulfate sedimentation (SDSS) test, Mixograph and size exclusion-high performance liquid chromatography (SE-HPLC) analysis. The results of the SDSS test and Mixograph analysis demonstrated that the overexpression of avenin-like b proteins in transgenic lines led to significantly increased SDSS volume and improved flour mixing properties. The results of SE-HPLC analysis of the gluten proteins in wheat flour demonstrated that the improvement in transgenic line flour properties was associated with the increased proportion of large polymeric proteins due to the incorporation of overexpressed avenin-like b proteins into the glutenin polymers. These results could help to understand the influence and mechanism of avenin-like b proteins on the functional properties of wheat flour.

Abstract:The gluten proteins distribute heterogeneously in the wheat grain, but the characteristics of gluten protein fractions in different layers of the starchy endosperm and their effects on the textural quality of the Chinese steamed bread have not been well understood. The bran and endosperm of wheat grains from six cultivars were peeled from the outer to the inside layer to obtain five fractions (F1 ∼ F5), whose relative weight of the remaining grain around 80%, 67%, 54%, 41% and 28% of the whole grain weight, respectively. It is found that wet gluten content (WGC) of flour basically decreased from F1 to F5, but the gluten index (GI) did not change obviously. For the dough made from F1 to F5 flours, their developing time (DT), stability time (ST), extension area (EA) and extensibility (E) decreased, but the weakening degree (WD) and resistance to extension/extensibility (R/E) increased. These dough rheological results suggested that the rheological properties of dough mainly depended on the gluten content for a given variety of wheat. Moreover, the hardness and chewiness of steamed bread made from F1 to F5 flours increased, but the cohesiveness and resilience decreased, suggesting that F1 flour is more suitable for manufacturing steamed bread with improved textural quality. These findings are useful for quality improvement of steamed bread by selecting the suitable wheat flour fraction.

Formation of macromolecules in wheat gluten/starch mixtures during twin-screw extrusion: effect of different additives

Abstract:Background: Wheat gluten comprises a good quality and inexpensive vegetable protein with an ideal amino acid composition. To expand the potential application of wheat gluten in the food industry, the effect of different additives on the physicochemical and structural properties of wheat gluten/starch mixtures during twin-screw extrusion was investigated. Results: Macromolecules were observed to form in wheat gluten/starch mixtures during twin-screw extrusion, which may be attributed to the formation of new disulfide bonds and non-covalent interactions, as well as Maillard reaction products. Additionally, the water retention capacity and in vitro protein digestibility of all extruded wheat gluten/starch products significantly increased, whereas the nitrogen solubility index and free sulfhydryl group (SH) content decreased, during twin-screw extrusion. Secondary structural analysis showed that α-helices disappeared with the concomitant increase of antiparallel β-sheets, demonstrating the occurrence of protein aggregation. Microstructures suggested that the irregular wheat gluten granular structure was disrupted, with additive addition favoring transformation into a more layered or fibrous structure during twin-screw extrusion. Conclusion: The findings of the present study demonstrate that extrusion might affect the texture and quality of extruded wheat gluten-based foods and suggest that this process might serve as a basis for the high-value application of wheat gluten products. © 2017 Society of Chemical Industry.

Abstract:The qualities of wheat dough are influenced by the high-molecular-weight glutenin subunits (HMW-GS), a critical component of wheat gluten protein. However, it is still unknown how HMW-GS silencing affects the aggregation characteristics of dough. Two groups of near-isogenic wheat were used to study the effects of HMW-GS silencing on dough aggregation characteristics, dough texture characteristics, and dough microstructure. It was observed that the content of gliadin in LH-11 strain significantly increased compared to the wild-type (WT). Additionally, the amount of glutenin macropolymer and the glutenin/gliadin both decreased. The aggregation characteristics and rheological characteristics of the dough in LH-11 strain were significantly reduced, and the content of β-sheet in the dough was significantly reduced. The HMW-GS silencing resulted in a reduction in the aggregation of the gluten network in the dough, which related to the alteration of the secondary and microstructure of the gluten.

Abstract:A set of high-molecular-weight glutenin subunit (HMW-GS) deletion lines were used to investigate the influences of HMW-GS on wheat gluten, and dough properties were investigated using a set of HMW-GS deletion lines. Results showed that HMW-GS deletion significantly decreased the dough stability time, as well as viscoelastic moduli (G' and G″), compared with the wild type, where the deletion of x-type HMW-GSs (Ax1d, Bx7d, and Dy12d) decreased more than y-type HMW-GSs (By8d and Dy12d). The deletion of HMW-GS significantly decreased HMW-GS contents and increased α-/γ-gliadin contents. A proteomic study showed that the HMW-GS deletion down-regulated the HMW-GS, β-amylase, serpins, and protein disulfide isomerase and up-regulated the LMW-GS, α/γ-gliadin, and α-amylase inhibitor. Meanwhile, HMW-GS deletion significantly decreased contents of β-turn and β-sheet. In addition, less energetically stable disulfide conformations (trans-gauche-gauche and trans-gauche-trans) were abundant in HMW-GS deletion lines. Furthermore, analysis of five HMW-GSs based on amino acid sequences proved that Dx2 and Bx7 had a more stable structure, followed by Ax1, then Dy12, and finally By8.

Abstract:Introduction: Steamed bread (SB) is a daily food in many countries in the world, but the relationship between HMW-GS and the quality of SB remain unclear. Methods: This study investigated the effects of 12 subunit combinations on the characteristics of SB, including volume, physical properties, and sensory evaluation, combined with the microstructure and dough rheological properties. Results: The locus effect results showed, volume and physical properties of SB were Glu-D1 > Glu-B1 > Glu-A1 , while sensory scores Glu-B1 > Glu-D1 > Glu-A1 . According to individual subunit effects, subunit 1 at Glu-A1 locus, 7+8 and 7+9 subunits at Glu-B1 locus, and 2+10 and 5+12 subunits at Glu-D1 locus were significantly superior to other subunits in physical indices like volume, chewiness, glueyness, and sensory scores, and were less affected by moisture. The effect of subunits combination is mainly affected by subunits, and the combination of superior subunits tends to make SB quality better. The subunit combinations (1, 7+8, 5+12), (N, 7+9, 2+10) and (1, 7+9, 5+12) had better physical properties indexes, sensory scores, dense, uniform and delicate micro-pore structure, and smaller thickness wall. Discussion: The results showed that protein content, wet gluten content and stability time were the main factors affecting the volume and physical properties of SB. The protein content, wet gluten content and stability time of flour in 7+8, 7+9, 2+10 and 5+12 subunits were higher. Therefore, the quality of SB containing these subunits was found better.

Abstract:The unique properties of wheat reside primarily in its gluten-forming storage proteins. Their intrinsic viscoelastic behavior is responsible for the characteristics of different wheat-based foods and for the use of wheat gluten proteins in different food products. Wheat-based food processing generally develops and sets the gluten protein network. Heat-induced gluten aggregation proceeds through cross-linking within and between its protein fractions. Prominent reactions include sulfhydryl (SH) oxidation and SH-disulfide (SS) interchange, which lead to SS cross-links. Other covalent bonds are also formed. Gluten functionality can be (bio-) chemically impacted. We focus on bread making, in which gluten proteins contribute to dough properties, bread loaf volume, and structure, and on pasta production, in which gluten proteins generate the desired cooking quality. Furthermore, it is speculated that the structure and texture of soft wheat products are also, at least to some degree, shaped by the heat-induced changes in the gluten protein fraction.

Abstract:High-molecular-weight glutenin subunits (HMW-GSs) are storage proteins present in the starchy endosperm cells of wheat grain. Encoding the synthesis of HMW-GS, the Glu-1 loci located on the long arms of group 1 chromosomes of the hexaploid wheat (1A, 1B, and 1D) present multiple allelism. In hexaploid wheat cultivars, almost all of them express 3 to 5 HMW-GSs and the 1Ay gene is always silent. Though HMW-GSs are the minor components in gluten, they are crucial for dough properties, and certain HMW-GSs make more positive contributions than others. The HMW-GS acts as a “chain extender” and provides a disulfide-bonded backbone in gluten network. Hydrogen bonds mediated by glutamine side chains are also crucial for stabilizing the gluten structure. In most cases, HMW-GSs with additional or less cysteines are related to the formation of relatively more or less interchain disulfide bonds and HMW-GSs also affect the gluten secondary structures, which in turn impact the end use qualities of dough.

Abstract:BACKGROUND: The effects of tannic acid, which is present in many plants, on the structure of gluten proteins and the properties of dough and bread were studied. Tannic acid was added at levels of 0.01, 0.02 and 0.03 g kg(-1) during the dough-making process.RESULTS: The added tannic acid acted negatively on disulfide bond formation but interacted with gluten proteins via other covalent bonds, as detected by UV spectroscopy and dynamic rheometry. Rheological properties and texture of the bread were measured by farinograph, extensograph and texture profile analyser. Texture analysis indicated little change in adhesiveness and resilience of the bread at all three levels of tannic acid compared with the control, but changes in hardness and chewiness of the bread made with added tannic acid indicated that tannic acid could delay bread staling.CONCLUSION: The effect of tannic acid on flour and dough is different from that of other flour redox agents. It breaks down disulfide bonds but also has positive effects on dough properties and bread quality. Disulfide bonds are commonly considered to be the most important factor affecting changes in the quality of bread. However, this study presents the new concept that other covalent bonds can also improve the quality of flour and bread and uses this property to investigate new, safe and efficient flour additives. Abstract:Physicochemical properties of wheat grains with largest kernel thickness always was lowest than the other sections, examination of microstructure of wheat grains can help us understand this phenomena. Two varieties of wheat, soft white winter wheat Yangmai 11 and hard white winter wheat Zhengmai 9023, were fractionated into five sections by kernel thickness. Then the fractionated wheat grains in 2.7–3.0 mm section were separated into three fractions by kernel specific density sequentially. Microstructure of the fractured surface were evaluated at different scale level to two varieties wheat with different kernel thickness and specific density by using environmental scanning electron microscopy. Compactness and size of endosperm cell tended to decrease with decreasing wheat kernel thickness and specific density. Protein matrix tended to increase with decreasing wheat kernel thickness and specific density. Size of starch granules and proportion for different type starch granules also varied with different wheat kernel thickness and specific density. Those microstructure properties of the fractured surface, formation of endosperm cells, protein matrix and starch granules were close related to rheological properties and pasting properties of wheat grains.
