Abstract:Heat shock protein 90 (HSP90) is a highly conserved molecular chaperone that plays important roles in modulating multiple cellular stress responses. In this study, full-length complementary DNA of hsp90 from the giant spiny frog Quasipaa spinosa (Qs) was first isolated and designated as Qshsp90. This gene consisted of 2801 nucleotides with a complete open reading frame of 2172 nucleotides encoding a putative protein of 724 amino acids. Sequence analysis revealed that QsHSP90 shared high similarity with other known HSP90 and belonged to a vertebrate HSP90β subtype. Tissue distribution analysis showed that Qshsp90 was present in all of the test tissues, and its expression in liver, skin and kidney was significantly up-regulated after heat shock for 3 or 6 h, while skin showed a prompt and durable response to such heat stress. Qshsp90 transcripts in kidney and liver also markedly increased in pathogen infection models such as Aeromonas hydrophila and polyinosinic polycytidylic acid. These results indicate that Qshsp90 may play critical roles in coping mechanisms for thermal stress, and may be involved in the immune response.

Abstract:Heat shock proteins (Hsps) are conserved molecules whose main role is to facilitate folding of other proteins. Most Hsps are generally stress-inducible as they play a particularly important cytoprotective role in cells exposed to stressful conditions. Initially, Hsps were generally thought to occur intracellulary. However, recent work has shown that some Hsps are secreted to the cell exterior particularly in response to stress. For this reason, they are generally regarded as danger signaling biomarkers. In this way, they prompt the immune system to react to prevailing adverse cellular conditions. For example, their enhanced secretion by cancer cells facilitate targeting of these cells by natural killer cells. Notably, Hsps are implicated in both pro-inflammatory and anti-inflammatory responses. Their effects on immune cells depends on a number of aspects such as concentration of the respective Hsp species. In addition, various Hsp species exert unique effects on immune cells. Because of their conservation, Hsps are implicated in auto-immune diseases. Here we discuss the various metabolic pathways in which various Hsps manifest immune modulation. In addition, we discuss possible experimental variations that may account for contradictory reports on the immunomodulatory function of some Hsps.

Abstract:Heat shock proteins (HSPs) are a family of proteins produced by cells in response to exposure to stressful conditions. In addition to their role as chaperones, they also play an important role in the cardiovascular, immune, and other systems. Normal bone tissue is maintained by bone metabolism, particularly by the balance between osteoblasts and osteoclasts, which are physiologically regulated by multiple hormones and cytokines. In recent years, studies have reported the vital role of HSPs in bone metabolism. However, the conclusions remain largely controversial, and the exact mechanisms are still unclear, so a review and analyses of previous studies are of importance. This article reviews the current understanding of the roles and effects of HSPs on bone cells (osteoblasts, osteoclasts, and osteocytes), in relation to bone metabolism.

Abstract:Interest in the heat shock proteins (HSPs), as a natural physiological toolkit of living organisms, has ranged from their chaperone function in nascent proteins to the remedial role following cell stress. As part of the defence system, HSPs guarantee cell tolerance against a variety of stressors, including exercise, oxidative stress, hyper and hypothermia, hyper and hypoxia and improper diets. For the past couple of decades, research on functional foods has revealed a number of substances likely to trigger cell protection through mechanisms that involve the induction of HSP expression. This review will summarize the occurrence of the most easily inducible HSPs and describe the effects of dietary proteins, peptides, amino acids, probiotics, high-fat diets and other food-derived substances reported to induce HSP response in animals and humans studies. Future research may clarify the mechanisms and explore the usefulness of this natural alternative of defense and the modulating mechanism of each substance.

Abstract:Experts from around the world review the current field of the immunobiology of heat shock proteins, and provide a comprehensive account of how these molecules are spearheading efforts in the understanding of various pathways of the immune system. This one-stop resource contains numerous images to both help illustrate the research on heat shock proteins, and better clarify the field for the non-expert. Heat shock proteins (HSPs) were discovered in 1962 and were quickly recognized for their role in protecting cells from stress. Twenty years later, the immunogenicity of a select few HSPs was described, and for the past 30 years, these findings have been applied to numerous branches of immunology, including tumor immunology and immunosurveillance, immunotherapy, etiology of autoimmunity, immunotherapy of infectious diseases, and expression of innate receptors. While HSPs can be used to manipulate immune responses by exogenous administration, they appear to be involved in initiation of de novo immune responses to cancer and likely in the maintenance of immune homeostasis.

Abstract:The heat shock response is a general property of all living organisms. In the detrimental environment, different kinds of organisms have the same molecular response, that is the inhibition of normal gene expression and the expression of heat shock genes. Heat shock proteins exist in cells and mainly acts as molecular chaperones, however, the syntheses of heat shock protein significantly increase at heat shock or other stress conditions. Heat shock proteins act as molecular chaperones to regulate protein folding, translocation and assembly, and they clear denatured proteins away or make them renaturation to stabilize cell structure. The synthesis of the heat shock proteins is correlated with cell thermotolerance. With the progress of research in this field, the application of heat shock proteins in gene engineering and medicine or relevant fields is becoming more and more prospective.

Abstract:Heat shock proteins (HSPs) are molecular chaperones produced in response to oxidative stress (OS). These proteins are involved in the folding of newly synthesized proteins and refolding of damaged or misfolded proteins. Recent studies have been focused on the regulatory role of HSPs in OS and ischemia/reperfusion injury (I/R) where reactive oxygen species (ROS) play a major role. ROS perform many functions, including cell signaling. Unfortunately, they are also the cause of pathological processes leading to various diseases. Biological pathways such as p38 MAPK, HSP70 and Akt/GSK-3β/eNOS, HSP70, JAK2/STAT3 or PI3K/Akt/HSP70, and HSF1/Nrf2-Keap1 are considered in the relationship between HSP and OS. New pathophysiological mechanisms involving ROS are being discovered and described the protein network of HSP interactions. Understanding of the mechanisms involved, e.g., in I/R, is important to the development of treatment methods. HSPs are multifunctional proteins because they closely interact with the antioxidant and the nitric oxide generation systems, such as HSP70/HSP90/NOS. A deficiency or excess of antioxidants modulates the activation of HSF and subsequent HSP biosynthesis. It is well known that HSPs are involved in the regulation of several redox processes and play an important role in protein-protein interactions. The latest research focuses on determining the role of HSPs in OS, their antioxidant activity, and the possibility of using HSPs in the treatment of I/R consequences. Physical exercises are important in patients with cardiovascular diseases, as they affect the expression of HSPs and the development of OS.

Abstract:Heat shock proteins (Hsp) are a class of stress-inducible proteins that mainly act as molecular protein chaperones. This chaperone activity is diverse, including assisting in nascent protein folding and regulating client protein location and translocation within the cell. The main proteins within the Hsp family, particularly Hsp70 and Hsp90, also have a highly diverse and numerous set of protein clients, which when combined with the high expression levels of Hsp proteins (2%-6% of total protein content) establishes these molecules as "central regulators" of cell protein physiology. Among the client proteins, Hsps regulate numerous signal-transduction and receptor-regulatory kinases, and indeed directly regulate some receptors themselves. This also makes the Hsps, particularly Hsp90, central regulators of signal-transduction machinery, with important impacts on endogenous and drug ligand responses. Among these roles, Hsp90 in particular acts to maintain mature signaling kinases in a metastable conformation permissive for signaling activation. In this review, we will focus on the roles of the Hsps, with a special focus on Hsp90, in regulating receptor signaling and subsequent physiologic responses. We will also explore potential means to manipulate Hsp function to improve receptor-targeted therapies. Overall, Hsps are important regulators of receptor signaling that are receiving increasing interest and exploration, particularly as Hsp90 inhibitors progress toward clinical approval for the treatment of cancer. Understanding the complex interplay of Hsp regulation of receptor signaling may provide important avenues to improve patient treatment.

Effects of Chronic Stress on Behavior and Heat Shock Protein 70 Expression in Immune Cells

Abstract:Chronic stress can suppress organisms' resistance to adverse stimuli in the environment. The stress responses occur at both the system and single-cell levels. Although many studies have investigated the neuroendocrine mechanisms underlying the effects of stress on health, the characteristics of cellular stress response are still poorly understood. One manner in which cells resist damage and/or death induced by stress is to synthesize a highly conserved set of intracellular proteins, termed heat shock protein (HSP). In particular, the 70-kDa HSP (HSP70) is essential for cellular recovery after stress as well as survival and maintenance of normal cellular function. It has been demonstrated that the expression of high levels of HSP70 is associated with an increased resistance of cells to challenges that would otherwise lead to cell injury and/or death. Therefore, the intensity of an HSP70 response to stress is an important biomarker of how the cell tolerates stress damage. This study aimed to determine whether prior chronic stress inhibited the synthesis of HSP70 in immune cells in the peripheral blood and spleen of rats exposed to an acute hot water bath. Twenty-eight rats were randomly divided into 2 groups with 14 rats in each group: the control group (C) and the chronic stress group (CS). CS rats were exposed to chronic unpredictable stress for 4 weeks to induce depressive-like behavior. The controls were stress-free. Thereafter, 8 rats in each group were exposed to acute heat stress (C-heat shock and CS-heat shock) to induce whole-body hyperthermia (maintaining the core temperature at 41℃ for 25 minutes).The other rats remained undisturbed in their home cages (C-no heat shock and CS-no heat shock). All rats were decapitated 6 hours after the completion of the treatments. The blood and spleen were collected and the level of HSP70 expression in the leukocytes was measured using flow cytometry. It was found that CS rats showed a decreased cellular HSP70 expression following the heat treatment, while a significant increase in the synthesis of HSP70 was observed in all types of immune cells from C rats. However, in CS rats, such HSP70 changes were only detected in the monocytes and granulocytes. Further, the increasing degree of heat-induced cellular HSP70 expression in CS rats was less pronounced than that observed in C rats. These results demonstrated that chronic stress decreased heat-induced HSP70 response in immune cells. Further, they suggested that the deficit of HSP70 might be involved in the suppressive modulation of the immune function induced by chronic stress.

Abstract:As molecular chaperones, heat-shock proteins (HSPs) function to limit protein aggregation, facilitate protein refolding and chaperone other proteins. Under conditions of cellular stress, intracellular HSP levels increase in order to provide cellular protection and maintain homeostasis. Evidence exists that the HSP family may be secreted into the circulation via lipid raft-mediated, granule-mediated or exosome-mediated exocytosis in haematopoietic and tumour cells. Extracellular HSPs exert immunomodulatory activities and play an important role in innate immune activation against pathogen infection. Membrane-bound Hsp70 in tumour cells or released chaperone-tumour associated antigen complex represent a target structure for the cytolytic attack by natural killer cells or T lymphocytes. Cellular stresses induce stress granule formation to evade detrimental cellular effects, mediating preconditioning phenotype. Therefore, induction of cellular stress tolerance by preconditioning (e.g., heat shock) might be potential therapeutic targets.

Abstract:The heat shock response (HSR) is a highly conserved cellular pathway that is responsible for stress relief and the refolding of denatured proteins [1]. When a host cell is exposed to conditions such as heat shock, ischemia, or toxic substances, heat shock factor-1 (HSF-1), a transcription factor, activates the genes that encode for the heat shock proteins (Hsps), which are a family of proteins that work alongside other chaperones to relieve stress and refold proteins that have been denatured (Burdon, 1986) [2]. Along with the refolding of denatured proteins, Hsps facilitate the removal of misfolded proteins by escorting them to degradation pathways, thereby preventing the accumulation of misfolded proteins [3]. Research has indicated that many pathological conditions, such as diabetes, cancer, neuropathy, cardiovascular disease, and aging have a negative impact on HSR function and are commonly associated with misfolded protein aggregation [4,5]. Studies indicate an interplay between mitochondrial homeostasis and HSF-1 levels can impact stress resistance, proteostasis, and malignant cell growth, which further support the role of Hsps in pathological and metabolic functions [6]. On the other hand, Hsp activation by specific small molecules can induce the heat shock response, which can afford neuroprotection and other benefits [7]. This review will focus on the modulation of Hsps and the HSR as therapeutic options to treat these conditions.
Revealing the potential regulatory relationship between HSP70, HSP90 and HSF genes under temperature stress

Abstract:Heat Shock Protein (HSPs) gene family members play fundamental roles in different environmental stress tolerances, protect the structure and function of cells, and perform a significant task in cellular homeostasis. In this study, we conducted a genome-wide identification, evolutionary relationship analysis and gene expression analysis of the HSP70, HSP90, and HSF gene families in Ruditapes philippinarum. We identified 83 RpHSP70, 6 RpHSP90, and 3 RpHSF genes in R. philippinarum. The structural characteristics, chromosomal localization, and the gene structure map were constructed to reveal the characteristics of protein structures. Furthermore, the expression profiling of transcriptome data showed the expression pattern of HSP70, HSP90 and HSF genes in Manila clam from different populations, and under high and low temperature stress. In addition, we performed protein-protein interaction network analysis between HSP70, HSP90, and HSF gene family which enabled us to recognize the regulatory relationship between the two HSP gene families and the HSF gene family. Furthermore, the predicted sub-cellular location revealed a diversified subcellular distribution of HSP70, HSP90, and HSF proteins, which may be directly or indirectly associated with functional diversification under heat stress condition.

Abstract:Heat shock proteins (HSPs) are in charge of proteostasis and buffer the stresses. Malfunction of HSPs lead to many diseases. In this review, we discuss the current understanding about the functions and structures of the major HSP families, the HSP network in proteostasis and their roles in diseases, and the prospects of these proteins as potential targets for therapeutic intervention. The heat shock proteins (HSPs) are ubiquitous and conserved protein families in both prokaryotic and eukaryotic organisms, and they maintain cellular proteostasis and protect cells from stresses. HSP protein families are classified based on their molecular weights, mainly including large HSPs, HSP90, HSP70, HSP60, HSP40, and small HSPs. They function as molecular chaperons in cells and work as an integrated network, participating in the folding of newly synthesized polypeptides, refolding metastable proteins, protein complex assembly, dissociating protein aggregate dissociation, and the degradation of misfolded proteins. In addition to their chaperone functions, they also play important roles in cell signaling transduction, cell cycle, and apoptosis regulation. Therefore, malfunction of HSPs is related with many diseases, including cancers, neurodegeneration, and other diseases. In this review, we describe the current understandings about the molecular mechanisms of the major HSP families including HSP90/HSP70/HSP60/HSP110 and small HSPs, how the HSPs keep the protein proteostasis and response to stresses, and we also discuss their roles in diseases and the recent exploration of HSP related therapy and diagnosis to modulate diseases. These research advances offer new prospects of HSPs as potential targets for therapeutic intervention.

Abstract:Cardiovascular disease is one of the leading causes of mortality worldwide, primarily driven by atherosclerosis, a chronic inflammatory condition contributing significantly to fatalities. Various biological determinants affecting cardiovascular health across different age and sex groups have been identified. In this context, recent attention has focused on the potential therapeutic and preventive role of increasing circulating levels of heat shock protein 27 (plasma HSP27) in combating atherosclerosis. Plasma HSP27 is recognized for its protective function in inflammatory atherogenesis, offering promising avenues for intervention and management strategies against this prevalent cardiovascular ailment. Exercise has emerged as a pivotal strategy in preventing and managing cardiovascular disease, with literature indicating an increase in plasma HSP27 levels post-exercise. However, there is limited understanding of the impact of exercise on the release of HSP27 into circulation. Clarifying these aspects is crucial for understanding the role of exercise in modulating plasma HSP27 levels and its potential implications for cardiovascular health across diverse populations. Therefore, this review aims to establish a more comprehensive understanding of the relationship between plasma HSP27 and exercise.

Abstract:Emerging evidence indicates that heat shock proteins (HSPs) are critical regulators in normal neural physiological function as well as in cell stress responses. The functions of HSPs represent an enormous and diverse range of cellular activities, far beyond the originally identified roles in protein folding and chaperoning. HSPs are now understood to be involved in processes such as synaptic transmission, autophagy, ER stress response, protein kinase and cell death signaling. In addition, manipulation of HSPs has robust effects on the fate of cells in neurological injury and disease states. The ongoing exploration of multiple HSP superfamilies has underscored the pluripotent nature of HSPs in the cellular context, and has demanded the recent revamping of the nomenclature referring to these families to reflect a re-organization based on structure and function. In keeping with this re-organization, we first discuss the HSP superfamilies in terms of protein structure, regulation, expression and distribution in the brain. We then explore major cellular functions of HSPs that are relevant to neural physiological states, and from there we discuss known and proposed HSP impacts on major neurological disease states. This review article presents a three-part discussion on the array of HSP families relevant to neuronal tissue, their cellular functions, and the exploration of therapeutic targets of these proteins in the context of neurological diseases.

Abstract:Heat shock proteins (HSPs) are highly conserved stress proteins known as molecular chaperones, which are considered to be cytoplasmic proteins with functions restricted to the intracellular compartment, such as the cytoplasm or cellular organelles. However, an increasing number of observations have shown that HSPs can also be released into the extracellular matrix and can play important roles in the modulation of inflammation and immune responses. Recent studies have demonstrated that extracellular HSPs (eHSPs) were involved in many human diseases, such as cancers, neurodegenerative diseases, and kidney diseases, which are all diseases that are closely linked to inflammation and immunity. In this review, we describe the types of eHSPs, discuss the mechanisms of eHSPs secretion, and then highlight their functions in the modulation of inflammation and immune responses. Finally, we take cancer as an example and discuss the possibility of targeting eHSPs for human disease therapy. A broader understanding of the function of eHSPs in development and progression of human disease is essential for developing new strategies to treat many human diseases that are critically related to inflammation and immunity.

Abstract:Key PointsHeat shock transcription factors (HSFs) have broad roles in stress resistance that encompass protection from protein misfolding, inflammation and environmental insults.Structural biology studies reveal a new model for how HSFs bind to their target DNA sequence, whereby the DNA-binding domain of HSFs is exposed to the solvent. In this model, the different HSF isoforms would expose biochemically distinct surfaces, which can be subjected to differential regulation by protein–protein interactions and post-translational modifications to modulate abundance and activity of HSFs.Accordingly, HSFs activate and repress genes that modulate metabolism, survival and proliferation in a context-dependent manner.HSF1 stability and function are compromised in neurodegenerative diseases caused by protein misfolding, and this dysfunction contributes to disease progression.Signalling pathways in cancer cells and tumour stroma activate HSF1 through mechanisms distinct from protein misfolding stress.

HEAT-SHOCK PROTEINS, MOLECULAR CHAPERONES, AND THE STRESS RESPONSE: Evolutionary and Ecological Physiology

Abstract:▪ Abstract Molecular chaperones, including the heat-shock proteins (Hsps), are a ubiquitous feature of cells in which these proteins cope with stress-induced denaturation of other proteins. Hsps have received the most attention in model organisms undergoing experimental stress in the laboratory, and the function of Hsps at the molecular and cellular level is becoming well understood in this context. A complementary focus is now emerging on the Hsps of both model and nonmodel organisms undergoing stress in nature, on the roles of Hsps in the stress physiology of whole multicellular eukaryotes and the tissues and organs they comprise, and on the ecological and evolutionary correlates of variation in Hsps and the genes that encode them. This focus discloses that (a) expression of Hsps can occur in nature, (b) all species have hsp genes but they vary in the patterns of their expression, (c) Hsp expression can be correlated with resistance to stress, and (d) species' thresholds for Hsp expression are correlated with levels of stress that they naturally undergo. These conclusions are now well established and may require little additional confirmation; many significant questions remain unanswered concerning both the mechanisms of Hsp-mediated stress tolerance at the organismal level and the evolutionary mechanisms that have diversified the hsp genes.

Effect of exercise, heat stress, and hydration on immune cell number and function

Abstract:Purpose: The purpose of this study was to determine the effect of thermal stress and hydration status on immune function during exercise. Methods: Ten trained men completed four cycle ergometer rides at 55% VO2peak under the following conditions: EN (euhydrated neutral; 22 degrees C, 30% RH), DN (dehydrated neutral), EH (euhydrated hot; 38 degrees C, 45% RH), and DH (dehydrated hot). During EN and EH, a carbohydrate/electrolyte beverage was consumed at a rate matching sweat loss, and during DN and DH, no fluid was ingested. Blood samples were drawn pre- and postexercise, and at 2 and 24 h of recovery. Cell counts were determined by automated counting and flow cytometry. Neutrophil activity was assessed as superoxide production, lymphocyte function was determined via PHA-stimulated mitogenesis, and natural killer (NK) cell activity was measured with a 51Cr-release assay. Cortisol was assayed via RIA. Results: Lymphocytes proliferation was depressed 2 h after exercise in all conditions (P < 0.05); however, when expressed on a per cell basis, function was greater in the DH and EH conditions. NK activity (max x 10(3) cells) was greater post compared with preexercise in all conditions (EH = 25.5 +/- 16.8, DH = 26.2 +/- 10.5, EN = 19.3 +/- 11.0, and DN = 16.5 +/- 8.7) but was not different between conditions. Leukocyte, neutrophil, lymphocyte, and NK cell counts were also elevated postexercise with the former two remaining elevated 2 h postexercise in the EH and DH conditions. Cortisol was greater postexercise in EH (22.1 +/- 1.3) and DH (27.7 +/- 1.3) compared with EN (17.8 +/- 2.1) and DN (18.9 +/- 1.6 microg x dL(-1). Conclusion: Euhydration did not affect cell number or function when compared with a dehydrated state; however, the hot environment caused more severe disturbances in these measures compared with a neutral environment.
