Determination of polysaccharides from sagittaria by method of H2SO4-Phenol

Abstract:Objective To establish a method to measure the content of sagittaria polysaccharide. Methods Water extract - alcohol precipitation method was to extract sagittaria polysaccharide,and test the content of polysaccharides from sagittaria with the method of H2SO4 - Phenol,wavelength was read at 490 nm. Results There was a good linear relationship between 20~100 μg, r2=0.9928, average recovery was 104%, and RSD was 4.72(n = 5), the colour of the test solution is stable within 2 h. Conclusion H2SO4- Phenol method is sensitive, reproducible and suitable for the determination of the polysaccharide content of sagittaria.
