Abstract:An efficient stereocontrolled total synthesis of (+)-biotin (1) has been achieved via the intermediacy of Roche's lactone 5 starting from cis-1,3-dibenzyl-2-imidazole-4,5-dicarboxylic acid (2). The bifunctional cinchona alkaloid-derived squaramide-promoted enantioselective alcoholysis was utilizing as a tool for the construction of two contiguous stereocenters of C-3a and C-6a in biotin molecular with excellent enantioselectivity. In addition, the 4-carboxybutyl side chain was assembled by first using C4+C1 approach via a novel tricyclic thiophanium salt intermediate.

Abstract:A practical asymmetric total synthesis of (+)-biotin based upon the chiral bifunctional sulfamide 8-promoted enantioselective anhydride desymmetrization has been achieved via the key chiral intermediacy of (3aS,6aR)-1,3-bis(4-methoxybenzyl)tetrahydro-4H-thieno[3,4-d]imidazole-2,4(1H)-dione (4). In addition, the installation of the 4-carboxybutyl side chain at C-4 position of 4 was efficiently introduced by an improved C4+C1 strategy.

Abstract:The highly enantioselective total synthesis of (+)-biotin 1 via the Hoffmann–Roche lactone–thiolactone strategy has been achieved starting from cis-1,3-dibenzyl-2-imidazolidone-4,5-dicarboxylic acid 2 with an overall yield of 35%. Two contiguous stereogenic centers at C-3a and C-6a were established through a rapid cinchona alkaloid-based sulfonamide-mediated enantioselective alcoholysis of meso-cyclic anhydride 3 to afford (4S,5R)-cinnamyl hemiester 4h, the direct precursor to (3aS,6aR)-lactone 5 with high enantioselectivity. A one-pot installation of the 4-carboxybutyl side chain was accomplished by a Fukuyama coupling reaction of (3aS,6aR)-thiolactone 6 with the organozinc reagent prepared from ethyl 5-bromopentanoate.

Abstract:AIM: To investigate feasible method for total synthesis of d-biotin on a large scale. METHODS AND RESULTS: cis-1,3-Dibenzyl-5-[(1S, 2S)-(+)-threo-1-hydroxymethyl-2-(p-nitrophenyl)-2-hydroxyethyl]-tetrahydro-4H- pyrro[3,4-d]imidazole-2,4,6-trione (3) prepared from (1S;2S)-(+)-threo-(p-nitro-phenyl)-2-amino-1, 3-propanediol and cis-1,3-dibenzyl-tetrahydro-4H-furo[3, 4-d]imidazole-2, 4, 6-trione. (2) by melting condensation was converted to (3aS, 6aR)-1, 3-dibenzyl-tetrahydro-4H-furo[3,4-d]imidazole-2, 4(1H)-dione(6) via highly stereoselective reduction, hydrolytic lactonization. Compound 6 was subjected to sulfurization, Grignard reaction, reduction to give (3aS, 6aR)-1, 3-dibenzyl-4-hydroxy-4-(3-ethoxypropyl)-tetrahydro-4H-thieno [3, 4-d] imidazole-2 (3H)-one (9). Compound 9 underwent dehydration, reduction, cleavage-cyclization, debenzylation in four steps procedure to lead to the formation of (3aR, 8aS, 8bS)-2-oxodecahydro-imidazo[4, 5-c] thieno [1, 2-a] thiolium bromide (12), which was transformed into d-biotin by condensation/ring openning, hydrolysis and decarboxylation in an overall yield of 25.7% from compound 2. CONCLUSION: A practical and cost-effective process for the manufacture of d-biotin was developed.

Abstract:An efficient and practical total synthesis of d-biotin is described. (4S,5R)-cis-1,3-dibenzyl-5-methoxycarbonyl-2-oxo-imidazoline-4-carboxylic acid, prepared from 1, 3-dibenzylimidazolidone-2-cis-4, 5-dicarboxylic acid via dehydration, monoesterification, optical resolution was subjected to reduction-cyclization, sulfuration to produce (3aS,6aR)-1,3-dibenzyl-tetrahydro-4H-thieno[3,4-d]imidazol-2, 4(1H)-dione(7). Compound 7 via Grignard reaction, dehydration, reduction, cleavage-cyclization in one-pot procedure led to (3aR, 8aS, 8bS)-1, 3-dibenzyl-2-oxo-decahydromidazo[4, 5-c]-thieno[1, 2-a]thiolium bromide(ii), which was converted to d-biotin via condensation, ring-opening, hydrolysis, decarboxylation, debenzylation in an overall yield of 14.5%.

Abstract:Chiral organosilanes are of great value in asymmetric synthesis, functional materials, and medicinal chemistry. Compared with single-silyl compounds, bis(silyl) ones are understudied because of the lack of the efficient synthetic protocols. The development of efficient synthetic approaches to access bis(silyl) compounds is highly desirable for studying their basic properties and potential utilities. Here, a cobalt-catalyzed sequential double hydrosilylation of aliphatic alkynes was developed to synthesize highly enantioenriched gem-bis(silyl)alkanes. This protocol used simple aliphatic alkynes and silanes to construct valuable chiral gem-bis(silyl)alkanes. The control experiments, isotopic labeling experiments, kinetic studies, and density functional theory calculations were conducted to elucidate the reaction mechanism. The synthetic versatility of gem-bis(silyl)alkanes was demonstrated by the synthesis of chiral organosilanols, α-hydroxysilanes through selective C–Si bond transformation and hydrosilylation of alkynes to construct chiral silanes containing adjacent C-stereocenter and Si-stereocenter.

Abstract:A novel and convenient method for the stereoselective synthesis of d-biotin 1 starting from the commercially available cis-1,3-dibenzyl-2-imidazolidone-4,5-dicarboxylic acid 2 has been developed. The key features of this synthesis include the enantioselective reduction of a meso-cyclic imide, mediated by a chiral oxazborolidine catalyst, derived from (1S,2S)-(+)-threo-1-(4-nitrophenyl)-2-amino-1,3-propanediol and the direct introduction of a C5 side chain to the (3aS,6aR)-thiolactone through a modified di-Grignard reaction. Enantioselectivities of 98% in the oxazborolidine-catalyzed asymmetric reduction process have been achieved.

Abstract:An efficient and highly enantioselective synthesis of d-biotin from the known cis-1,3-dibenzyl-2-imidazolidone-4,5-dicarboxylic acid (5) was accomplished in 48% overall yield. The key reactions in the sequence involve the catalytic enantioselective reduction of meso-cyclic imide 6 using polymer-supported chiral oxazoborolidine, derived from (S)-alpha,alpha-diphenylprolinol and polymer-bound sulfonyl chloride, and the installation of the C-5 side chain at C4 in the thiolactone 9 via a Ni/C-catalyzed Fukuyama coupling reaction.

Abstract:A practical chemoenzymatic method for the asymmetric total synthesis of d-biotin (1) starting from the commercially available cis-1,3-dibenzyl-2-imidazolidone-4,5-dicarboxylic acid (2) has been developed. The key step of the synthesis is the highly enantioselective hydrolysis of meso-dicarboxylic esters by a polymer-supported pig liver esterase and introduction of a formyl group at the C-4 position in 4 via a Grignard reaction. The polymer-supported PLE can be recovered quantitatively from the reaction mixture by simple filtration and reused without significant loss of activity.

Abstract:Achieving the preparation of enantiomerically enriched S-stereogenic compounds is a long-standing objective in stereoselective synthesis, owing to the fundamental importance and broad applications of these chiral scaffolds in various fields. Despite recent significant advancements, catalyst-controlled stereoselective synthesis of S-stereogenic compounds remains to be a considerable challenge, particularly by means of small-molecule catalysts. Herein, we disclosed an organocatalytic strategy for highly practical and enantioselective sulfinylation of alcohols and amines through the activation of sulfinates by forming mixed sulfinic anhydrides. Tuning the structure of the reactive species with sterically congested moieties, a simple, naturally occurring quinine catalyst effectively controls the chemo- and enantioselectivity over the nucleophilic S–O and S–N bond constructions, affording a wide range of chiral sulfinyl derivatives with excellent optical purities. Notably, the protocol readily facilitates the coupling with various natural products and commercial drugs that could offer an attractive strategy for the late-stage diversification of important biologically intriguing molecules.

Abstract:The control of chemo-, regio-, diastereo-, and enantioselectivity is a central theme in organic synthesis. The capability to obtain the full set of stereoisomers of a molecule would significantly enhance the efficiency for the synthesis of natural product analogues and creation of chiral compound libraries for drug discovery. Despite the tremendous progress achieved in the field of asymmetric synthesis in the past decades, the precise control of both relative and absolute configurations in catalyst-controlled reactions that create multiple stereocenters remains a significant synthetic challenge. We report here the development of a catalyst-controlled hydroboration with hitherto unattainable selectivity. The Rh-catalyzed hydroboration of a, beta-unsaturated carbonyl compounds with pinacolborane proceeds with high levels of regio-, diastereo-, and enantioselectivities to provide a hydroboration product with two vicinal stereocenters. Through the appropriate choice of substrate geometry (E or Z) and ligand enantiomer (S or R), all the possible diastereoisomers are readily accessible. The boron-containing products underwent many stereospecific transformations, thus providing a strategy for collective stereodivergent synthesis of diverse valuable chiral building blocks.
