Abstract:Fifteen new water-soluble alkaloids were obtained from the fresh herbs of Portulaca oleracea L. The structures of 15 alkaloids 1−15 were established according to spectroscopic data, and the stereoconfigurations were determined based on experimental and calculated electronic circular dichroism (ECD) data and single crystal X-ray diffraction. Alkaloids 1−15 were found to display good anti-inflammatory activity at 10 μM and could significantly reduce the interleukin-6 (IL-6) and nitric oxide (NO) levels induced by lipopolysaccharide (LPS) in RAW 264.7 macrophages.

Abstract:A new alkaloid, identified as 1-benzyl-2-nitroso-1,2,3,4-tetrahydroisoquinoline-6,7-diol, named oleraisoquinoline ( 1 ), and five organic acids and two esters, identified as 5-(hydroxymethyl)furan-2-carboxylic acid ( 2 ), 1 H -pyrrole-2,5-dicarboxylic acid ( 3 ), (7 E ,10 E )-octadeca-7,10-dienoic acid ( 4 ), (10 E ,13 E )-octadeca-10,13-dienoic acid ( 5 ), (7 E ,10 E )-hexadeca-7,10-dienoic acid ( 6 ), methyl tridecanoate ( 7 ) and methyl (9 E ,12 E )-octadeca-9,12-dienoate ( 8 ), were isolated from Portulaca oleracea L., among which compounds 2 and 4 ‒ 7 were isolated for the first time. Abstract:Eleven compounds were obtained from Portulaca oleracea L., including two novel ketone alkaloids, ( 1 , 2 ), 4-hydroxy-3-methoxybenzamide ( 3 ) (isolated for the first time), β-adenosine ( 4 ), oleracrylimide A and B ( 5 , 6 ), oleracein H, C, D, Q and A ( 7 – 11 ). The two novel ketone alkaloids were identified as 5-acetyl-5-methylcyclopent-2-ene-1-carboxamide ( 1 ), named oleraciamide H, and (2 R,3S,4R,5R)-5-((R)-1,2-dihydroxyethyl)-3,4-dihydroxytetrahydrofuran-2-yl glycinate ( 2 ), named oleracone Q by spectroscopic methods, including 1D, 2D NMR and compound fingerprints. Additionally, their anti-inflammatory activities were tested via RAW 264.7 cells induced by LPS and found that they could significantly inhibit the release of IL-1 β and TNF- α .

Their structures were determined by spectroscopic methods, including UHPLC-ESI-Q-TOF/MS, 1D and 2D NMR spectra. Both Olerapyrimidine and Olerapyridine at 20 μM could inhibit the inflammatory factors, IL-1β and TNF-α in the RAW 264.7 cells induced by LPS.

Abstract:Two new alkaloids identified as 2-(((S,Z)-1-(1H-azirin-1-yl)-5-methylhex-1-en-3-yl)oxy)-6-(hydroxymethyl)tetrahydro-2H-pyran-3,4,5-triol and (S,Z)-1-(1H-azirin-1-yl)-5-methylhex-1-en-3-ol, named olerazirin A ( 1 ), olerazirin B ( 2 ), together with five known alkaloids, identified as cyclo (L-Val-L-Ala) ( 3 ), cyclo-(glycyl-L-leucine) ( 4 ), cyclo-(Gly-Phe) ( 5 ), cyclo (Ser-Phe) ( 6 ), (3S,6S)-3-[(1R)-1-hydroxyethyl]-6-(phenylmethyl)-2,5-piperazinedione ( 7 ) were obtained from Portulaca oleracea L. using a range of chromatographic techniques, 1D and 2D NMR, and high-resolution electrospray ionisation time-of-flight mass spectroscopic methods, in which the compounds 3 – 7 were isolated from P. oleracea for the first time. In addition, the results showed that the compounds 1 and 2 have anti-inflammatory activities and compounds 1 – 3 and 5 – 7 exhibit the anticholinesterase activities.

Abstract:Phytochemical investigation on the stems of Alstonia scholaris (L.) R. Br. led to the isolation and identification of 30 natural products, including sixteen alkaloids (1–16), nine triterpenoids (17–25), two phenols (26–27), and three lignans (28–30). Their structures were established by comprehensive spectroscopic analyses together with the comparison with reported data. Compound 1 was a new indole alkaloid. Compound 24 was firstly isolated from Apocynaceae family, compounds 22–23 and 26–30 were obtained from the genus Alstonia for the first time, and this is the first report of the presence of 11 and 16 in A. scholaris. The chemotaxonomic significance of these isolated compounds was also discussed.

Abstract:Eight pairs of dihydrohomoisoflavonoids (1-8), including four pairs of enantiomeric aglycones [(R,S)-portulacanones B (1) and C (2) and (R,S)-oleracones C (3) and Q (4)] and four pairs of epimeric glycosides [portulacasides A-D and epiportulacasides A-D (5-8)], were obtained from Portulaca oleracea L. Among them, (R,S)-oleracone Q (4) and four pairs of epimeric glycosides (5-8) were reported for the first time. The 50% EtOH fraction from the 70% EtOH extract prevented HepG2 human liver cancer cell damage induced by N-acetyl-p-aminophenol (APAP), and the cell survival rate was 62.3%. Portulacaside B (6a), which was isolated from the 50% EtOH fraction, exhibited hepatoprotective and anti-inflammatory effects. The compound increased the survival rate of APAP-damaged HepG2 human liver cancer cells from 40.0% to 51.2% and reduced nitric oxide production in RAW 264.7 macrophages, resulting in an inhibitory rate of 46.8%.

Abstract:Acidic compounds were enriched from a water decoction of Portulaca oleracea using 717 anion exchange resin column chromatography. A total of 22 compounds including 9 catecholamine derivatives, of which six were rare sulfonic acid derivatives, and 9 nitro derivatives, were further isolated through various column chromatographic methods, and their structures were elucidated by interpreting their spectroscopic data and ECD calculations. Among them, 16 compounds were isolated from P. oleracea for the first time, 8 of which were undescribed compounds and four compounds were natural products. Pharmacological screening indicated that cis-3-(3-nitro-4-hydroxyphenyl)-methyl acrylate exhibited anti-inflammatory activity, measured as inhibition of nitric oxide production in LPS-stimulated RAW264.7 macrophage cells, with an EC50 value of 18.0 μM, The compounds showed only weak anti-microbial activity with (2R)-(+)-2-chloro-3-(3-nitro-4-hydroxyphenyl)-propionic acid methyl ester inhibiting Candida albicans with a MIC of 256 μg/mL, and 3-methoxy-4,5-dinitrophenol inhibiting Shigella sonnei with a MIC of 512 μg/mL.

Abstract:A novel skeleton alkaloid was obtained from Portulaca oleracea L., which was identified as 10,11-dihydroxybenzo[5',6'] pentaleno[1',2':3,4]pyrrolo[2,1-b]oxazol-7(11bH)-one, named oleracone M, and its structure was determined using UHPLC-ESI-QTOF/MS, 1D NMR and 2D NMR spectroscopy, and circular dichroism. Then the bioactivities of the compound were investigated including the anti-inflammatory, anti-acetylcholinesterase and antioxidant activities. The results showed that the novel skeleton alkaloid exhibited the potent effect on inhibiting the secretion of IL-1β at 10 μM, anticholinesterase activity with IC50 value of 49.58 μM, and antioxidant activity with IC50 value of 66.43 μM.

Abstract:Eight alkaloids were isolated from the thin sulfuric acid extracts of the fresh roots of Stephania dentifolia by aluminum oxide, silica and Sephadex LH-20 column chromatography methods. Based on the spectroscopic analysis and chemical evidence, the structures of these alkaloids were identified as sinoacutine (1), sinomenine (2), cephamonine (3), tetrahydropalmatine (4), capaurine (5), stepharanine (6), (+)-stepharine (7) and palmatine (8). All compounds were obtained from this plant for the first time.

Abstract:Objective To study the chemical constituents of the alkaloids from the root of Stephania tetrandra.Methods The compounds were isolated by chromatography on silica gel,Sephadex LH-20 column and identified by spectroscopic analysis.Results Seven compounds were isolated and their structures were identified as tetrandrine(1),fangchinoline(2),2′-N-chloromethyltetrandrine(3),oxofangchirine(4),fenfangjine D hydrochloride(5),(+)-dicentrine(6),tazopsine(7).Conclusions Compound 7 is firstly isolated from the genus Stephania.Compound 6 is isolated from Stephania tetrandra for the first time.

Abstract:Phytochemical investigation of the root tubers of Lindera aggregata resulted in the isolation of five new sesquiterpene lactones, linderagalactones A-E (1-5), along with eight known sesquiterpenoids, 3-eudesmene-1beta,11-diol, hydroxylindestenolide, strychnistenolide, hydroxyisogermafurenolide, atractylenolide III, linderane, neolinderalactone, and linderalactone. The structures and relative configurations of 1-5 were determined by spectroscopic methods, especially HRESIMS and 2D NMR techniques. The absolute configurations of 1-4 were defined by comparison of quantum chemical TDDFT calculated and experimental ECD spectra. Linderagalactone A (1) is a halogenated sesquiterpene lactone possessing a unique rearranged carbon skeleton. Linderagalactone E (5), linderane, hydroxylindestenolide, and linderalactone showed hepatoprotective activity against H2O2-induced oxidative damages on HepG2 cells with EC(50) values of 67.5, 167.0, 42.4, and 98.0 microM, respectively.

Abstract:Eight compounds were isolated from the methanol extract of the roots and aerial parts of Anemone rivularis Buch. -Ham. ex DC. Abstract:A phytochemical investigation on the roots and stems of Litsea cubeba led to the isolation of seven isoquinolone alkaloids. By spectroscopic analysis and comparison of their 1H and 13C-NMR data with those in literatures, these alkaloids were identified as (+)-norboldine (1), (+)-boldine (2), (+)-reticuline (3), (+)-laurotetanine (4), (+)-isoboldine (5), (+)-N-methyl-laurotetanine (6), and berberine (7), respectively. Among them, 7 was isolated from the genus for the first time. The evaluation of these compounds showed weak anti-inflammatory activity against NO production in RAW 267.4 and BV-2 cells.

Abstract:Six previously undescribed rigidly monoterpenoid indole alkaloids, alstolactines F–K ( 1 − 6 ), were isolated from Alstonia scholaris . Among them, a pair of cage-like epimers, 1 and 2 , featuring a rare 6/5/6/6/7 ring system, represent the first example of C 5 →C 20 -olide, while compound 3 possesses unique degraded C 18 and C 19 . The structures of the isolates were established by multiple spectroscopic analyses, quantum computational chemistry methods, and X-ray diffraction. Furthermore, the expression levels of proteins including NLRP3, TLR4, P-p65, NF-ĸB, Notch-2, IL-18, P-p38, and p38 in LPS-induced human normal hepatocyte (LO2) cells could be significantly downregulated by compounds 1 − 6 , which showed potent anti-inflammatory bioactivity.

Abstract:One new laurane-type sesquiterpene, 10-hydroxyepiaplysinol (1), along with two known related sesquiterpenes, epiaplysinol (2) and ar-bisabol-9-en-7,11-diol (3), and one carotenoid metabolite, loliolide (4), were obtained from the red alga Laurencia tristicha. Their structures were elucidated based on comprehensive spectroscopic data analysis. Of the known compounds, 4 was isolated from the genus Laurencia for the first time. Additionally, the antioxidant activities of eleven laurane-type sesquiterpenes from L. tristicha were evaluated.

Abstract:Four compounds were isolated from the ethanolic extract of the bark of Alseodaphnehainanensis Merr. , The structures were identified as: a neolignan eusiderin A [(7R, 8R)-3,4,5,3′-tetram-ethox-△8’，9’-8-o-4’，7-o-5’lignan]（I）two benzylisoquinoline alkaloids (6,7-dimethoxyisoquinolinyl)-(4′-methoxyphenyl) methanone (Ⅱ), and ( 6, 7-methylenedioxyisoquinolinyl )-( 4′-methoxyphenyl ) methanone (Ⅲ), and 4-hydroxy-3-methoxy benzoic acid ( Ⅳ ) on the basis of HR-SIMS，1HNMR，13CNMRand 2D-NMR spectroscopic analysis. Compounds Ⅰ～Ⅲ were obtained from the Alseodaphne genus forthe first time.
