Abstract:We report an approach to the design of multilayered polyelectrolyte thin films (or 'polyelectrolyte multilayers', PEMs) that can be used to provide tunable control over the release of plasmid DNA (or multiple different DNA constructs) from film-coated surfaces. Our approach is based upon methods for the layer-by-layer assembly of DNA-containing thin films, and exploits the properties of a new class of cationic 'charge-shifting' polymers (amine functionalized polymers that undergo gradual changes in net charge upon side chain ester hydrolysis) to provide control over the rates at which these films erode and release DNA. We synthesized two 'charge-shifting' polymers (polymers 1 and 2) containing different side chain structures by ring-opening reactions of poly(2-alkenyl azlactone)s with two different tertiary amine functionalized alcohols (3-dimethylamino-1-propanol and 2-dimethylaminoethanol, respectively). Subsequent characterization revealed large changes in the rates of side chain ester hydrolysis for these two polymers; whereas the half-life for the hydrolysis of the esters in polymer 1 was ~200 days, the half-life for polymer 2 was ~6 days. We demonstrate that these large differences in side chain hydrolysis make possible the design of PEMs that erode and promote the surface-mediated release of DNA either rapidly (e.g., over ~3 days for films fabricated using polymer 2) or slowly (e.g., over ~1 month for films fabricated using polymer 1). We demonstrate further that it is possible to design films with release profiles that are intermediate to these two extremes by fabricating films using solutions containing different mixtures of these two polymers. This approach can thus expand the usefulness of these two polymers and achieve a broader range of DNA release profiles without the need to synthesize polymers with new structures or properties. Finally, we demonstrate that polymers 1 and 2 can be used to fabricate multilayered films with hierarchical structures that promote the sequential release of two different DNA constructs with separate and distinct release profiles (e.g., the release of a first construct over a period of ~3 days, followed by the sustained release of a second for a period of ~70 days). With further development, this approach could contribute to the design of functional thin films and surface coatings that provide sophisticated control over the timing and the order of the release of two or more DNA constructs (or other agents) of interest in a range of biomedical contexts.
A Novel Zwitterionic Copolymer with A Short Poly(Methyl Acrylic Acid) Block for Improving Both Conjugation and Separation Efficiency of A Protein Without Losing Its Bioactivity

Abstract:PEGylation of protein drugs has thus far been the most important method used in improving the stability and the circulation time, while lowering the immunogenicity of protein drugs. However, the loss of bioactivity of PEGylated protein drugs and the low recovery through the complicated separation process are the most challenging issues. To overcome these problems, a zwitterionic block copolymer poly(methyl acrylic acid-b-sulfobetaine methacrylate) (PMAA-b-PSBMA or abbreviated to PMS) has been chosen to modify a protein drug (uricase) since zwitterionic materials are superior to polyethylene glycol (PEG) in resistance to nonspecific protein adsorption, chemical stability and also low cytotoxicity. As indicated by the results, the conjugation between PMS and uricase could be achieved under 0.03 mg mL-1 of 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride (EDC·HCl) and 0.018 mg mL-1N-hydroxysuccinimide (NHS) through weak charge-induced adsorption of PMS on uricase, which is about 0.3% of the concentration for conventional protein-polymer conjugation. The conjugates, efficiently separated from the reaction solution and re-suspended in physiological solution through pH adjustment from pH 5.35 to 7.4, show about 133% of the original activity and better anti-trypsin digestion than the native uricase. These results demonstrate that the conjugation of a zwitterionic copolymer with a short poly(methyl acrylic acid) block could improve the stability of an enzyme without destructively affecting its bioactivity. This may open a new way for the delivery of therapeutic protein drugs.

Abstract:A series of well defined semitelechelic and mid-functionalized poly(poly(ethylene glycol) methyl ether methacrylate)s (poly(PEGMA)s) were synthesized through reversible addition-fragmentation chain transfer (RAFT) polymerization using thiazolidine-2-thione-functionalized chain transfer agents (CTAs). The thiazolidine-2-thione group was located either at the end or in the middle of polymer chains depending on the different structural CTAs. All polymers were fully analyzed by (1)H NMR spectroscopy and GPC, confirming their well-defined structures, such as predesigned molecular weights, narrow polydispersity indices, and high yield chain-end or chain-middle functionalization. The thiazolidine-2-thione functionality located at the end of or at the middle of the polymer chains can react with amine residues on protein surfaces, forming protein-polymer conjugates via amide linkages. The bioactivity of protein conjugates were subsequently tested using micrococcus lysodeikticus cell as substitute. The protein conjugations from the mid-functionalized polymer remained much more protein bioactivity comparing to their semitelechelic counterpart with similar molecular weights, indicating the steric hindrance of the mid-functionalized poly(PEGMA) s lead to the better selective conjugation to protein. The number of polymer chains on the protein surface was additionally evaluated by TNBS analysis, exhibiting that there are less mid-functionalized poly(PEGMA) s linked on the protein surface than the semitelechelic polymers, also supporting the hypothesis that the steric hindrance from branch-structural polymers results in the better reaction selectivity. This synthetic methodology is suitable for universal proteins, seeking a balance between the protein bioactivity and the protein protection by the covalent linkage with polymer, and exhibits promising potential for pharmaceutical protein conjugation.

Abstract:Various polymers have been tested for protein conjugation with a goal of bridging the complementary advantages of both components. However, many of these polymers, including the most well-established PEG, are nondegradable, which raises potential concerns on their cumulative chronic toxicity. Moreover, the immunogenicity of PEG has recently evoked considerable controversy. Synthetic polypeptides, on the other hand, are biomimetic polymers with tunable degradability, versatile side chain functionalities, unique secondary structures, and fascinating self-assembly behaviors. These properties have made them promising materials in protein modification for various applications. In this Topical Review, we summarize recent advances and list a number of interesting future directions in protein polypeptide conjugation, which we termed protein PEPylation.
A facile entrapment approach to construct PEGylated polyplexes for improving stability in physiological condition.

Abstract:PEGylated polyplexes had been proved to improve the stability of DNA complexes. However, the conjugation reaction might reduce the capacity of efficient DNA complexation. Herein we described an easy and favorable approach to construct PEGylated polyplexes via entrapping poly(ethylene glycol) cholesterol ether (CPEG) into polyplexes. It was of interest to find the addition sequence of CPEG had great effect on the stability of polyplexes in physiological salt concentration. The addition of CPEG into the formed PEI25k/DNA polyplexes had no effect to improve the stability. Whereas by the “CPEG first” method of adding CPEG and PEI25k mixture into the DNA solution, the PEI25k/CPEG/DNA polyplexes showed excellent anti-aggregation effect and enhanced transfection efficiency in physiological condition. The difference performance might be explained by the possibility of CPEG entrapment. By the “CPEG first” method, PEGylated polyplexes was constructed due to the hydrophobic interaction between the cholesterol group of CPEG and hydrophobic charged-compensated core. The PEG coating significantly improved the stability of polyplexes in physiological condition. This facile entrapment approach to prepare PEGylated polyplexes might have great potential in non-viral gene delivery research and application.
Protein Release from Biodegradable Polyhpma-Lysozyme Conjugates Resulting in Bioactivity Enhancement.

Abstract:A novel biodegradable thiazolidine-2-thione functional chain transfer agent was synthesized and employed as a reversible additional fragmentation chain transfer agent to prepare well-defined semitelechelic poly-N-(2-hydroxypropyl) methacrylamides (polyHPMAs) with predetermined molecular weights and narrow polydispersities. The protein reactive group, thiazolidine-2-thione, was located at the polymer chain ends fixed by biodegradable disulfide bonds. The functional polyHPMA chains were subsequently conjugated to protein (lysozyme) by exploiting reactions between the thiazolidine-2-thione functionality and amine residues on the protein surface to form covalent amide linkages. The in vitro bioactivities of the lysozyme-polyHPMA conjugates were assessed by using Micrococcus lysodeikticus cells as substrates. The lysozyme bioactivity was significantly reduced following the conjugation procedure. However, cleavage of the polymer chains from the bioconjugates (under reducing conditions) yielded free protein and a remarkable recovery of bioactivity. In vivo tests were performed by subcutaneous injection into mice and clearly demonstrated decreased proteolytic degradation for the protein-polymer conjugate when compared with native protein, indicating effective protein protection through a conjugation strategy. This bioreversible approach to conjugation allows for a balance to be made between protein protection and effective bioactivity maintenance.

Abstract:Modifications at polypeptide side chains have been found to affect conformational and properties,which is a vital strategy to prepare functional polypeptides.This contribution describes the synthesis of homo and co-polypeptides containing unsaturated bonds and thioether moieties,poly(S-allylcysteine)(PAC).The pendant vinyl groups provide an active binding site which are able to be further modified with cysteine by thiol-ene click reaction.The sulfoxide from thioether oxidation bestows the polypeptide water solubility and enzymatic catalytic sites.The self-assembly micelles of PEG-b-PAC are ruptured along with oxidation to release the Nile red payloads.Moreover,poly(S-allyl-cysteine sulfoxide),aka.polyalliin,is the oxidation product of PAC which undergoes side chain cleavage catalyzed by alliinase enzyme.The polypeptides are biocompati-ble as confirmed by cell viability assays and may find applications in drug delivery,biosensing and nanoreactor.

Abstract:Poly(ethylene glycol) (PEG) segments were introduced into a polyanhydride main chain by copolymerization of terminal-carboxylated poly(ethylene glycol) with diacidic monomers (sebacic acid and trimellitylimidoglycine). IR and 1H NMR spectroscopy confirmed the copolymer structures. DSC analysis showed that these polyanhydrides have low Tg and low crystallinity. In vitro degradation tests indicated that introducing PEG segments accelerated the degradation rate of these polymers and the degradation duration could be manipulated from 3 days to 3 weeks. The pH of the environment caused by the polymer degradation was lower than 5.0; therefore, the polyanhydrides could be used as components of a newly designed pulsed-release device for peptide and protein delivery. © 1999 Society of Chemical Industry

Abstract:Reversible conjugation of polymers to proteins is important for a variety of applications, for example to control protein activity. Light is often employed as an external trigger to allow for spatio and temporal control over release of a payload. In this report, we demonstrate preparation of photocleavable poly(polyethylene glycol) acrylate-lysozyme (pPEGA-Lys) conjugates via ortho -nitrobenzyl linkages. The conjugates were made by both grafting-to and grafting-from in order to compare and contrast the two synthetic approaches. First, a lysine-reactive ortho -nitrobenzyl atom transfer radical polymerization (ATRP) initiator was synthesized. For the grafting -to strategy, the initiator was employed in the ATRP of PEGA, and the subsequent polymer was conjugated to the lysine residues of lysozyme. For the grafting-from strategy, lysozyme was modified first with the photocleavable initiator, and the purified macroinitiator was then subjected to polymerization conditions to synthesize the protein-polymer conjugate. The polymer was cleaved from the protein via UV light, and activity before and after polymer removal was evaluated, showing 83% recovery. This work provides evidence that reversing conjugation is successful for activity modulation for ortho -nitrobenzyl linked protein-polymer conjugates.

Abstract:Poly(ethylene glycol) (PEG) is widely used for covalent conjugation with therapeutic molecules to prolong their biocirculation time. This effect can be further improved by increasing the molecular weight of PEG. However, because of the lack of effective degradation mechanisms, PEG with a molecular mass over 10 kDa is hardly excreted from the body, thus accumulating in the liver and kidneys which increases the risk of toxicity. Herein, biodegradable PEGs were synthesized by linking PEG backbones with ester bonds through the polycondensation of PEG diols and dicarboxylic acids. To overcome the difficulties in synthesis of high molecular weight products by traditional esterification strategies, a novel carboxyl-ester transesterification mechanism was developed to produce biodegradable PEGs with a molecular weight up to 112.6 kDa. It was confirmed that these biodegradable PEGs show comparable characteristics of nondegradable PEGs with similar molecular weights, but they can be metabolized after the cleavage of ester bonds.

Abstract:Application of proteins and peptides as human therapeutics is expanding rapidly as drug discovery becomes more prevalent. Conjugation of polymers to proteins can circumvent many problems and pegylation of proteins is now emerging as acceptable practice. This paper describes the synthesis of alpha-aldehyde-terminated poly(methoxyPEG)methacrylates from Cu(I) mediated living radical polymerization (Mn = 11 000, 22 000 and 32 000; PDi < 1.15), and their efficient conjugation to lysozyme, as a model protein. This offers an attractive and flexible alternative to linear poly(ethylene glycol) opening up the possibility of using the full power of living radical polymerization.

Abstract:A one-step, mild procedure based on coaxial electrospinning was developed for incorporation and controlled release of two model proteins, BSA and lysozyme, from biodegradable core-shell nanofibers with PCL as shell and protein-containing PEG as core. The thickness of the core and shell could be adjusted by the feed rate of the inner dope, which in turn affected the release profiles of the incorporated proteins. It was revealed that the released lysozyme maintained its structure and bioactivity. The current method may find wide applications for controlled release of proteins and tissue engineering.
A simple methodology for the synthesis of heterotelechelic protein-polymer-biomolecule conjugates

Abstract:A synthetic protocol for the preparation of hetero-biofunctional protein-polymer conjugates is described. A chain transfer agent, SS-bis (alpha,alpha'-dimethyl-alpha "-acetic acid) trithiocarbonate was functionalized with alpha,omega-pyridyl disulfide (PDS) groups, Subsequently, one of the PDS groups was covalently attached to bovine serum albumin (BSA) at the specific free thiol group on the cysteine residue through a disulfide linkage. The second PDS group remained intact, as it was found to be inaccessible to further BSA functionalization. The BSA-macro-reversible addition-fragmentation chain transfer (RAFT) agent was then used to prepare BSA-polymer conjugates via in situ polymerization of oligo (ethyleneglycol) acrylate and N-(2-hydroxypropyl) methacrylamide using an ambient temperature initiator, 4,4'-azobis [2,9-imidazolin-2-ethyl)propane] dihydrochloride in an aqueous medium. Sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) confirmed that the in situ polymerization occurred at the protein surface where the RAFT agent was attached and the molecular weights of the BSA-polymer conjugates were found to increase concomitantly with monomer conversion and polymerization time. After polymerization the remaining terminal PDS groups were then utilized to attach thiocholesterol and a flurophore, rhodamine B to the protein-polymer conjugates via disulfide coupling. UV-Vis and fluorescence analyses revealed that similar to 80% of the protein conjugates were found to retain integral PDS end groups for further attachment to free thiol-tethered precursors. Dna Polyplexes Formed Using Pegylated Biodegradable Hyperbranched Polymers

Abstract:A novel PEGylated biodegradable hyperbranched PEG-b-PDMAEMA has been synthesized. The low toxicity, small molecular weight PDMAEMA chains were crosslinked using a biodegradable disulfide-based dimethacrylate (DSDMA) agent to yield higher molecular weight hyperbranched polymers. PEG chains were linked onto the polymer surface, masking the positive charge (as shown by Zeta potential measurements) and reducing the toxicity of the polymer. The hyperbranched structures were also cleaved under reducing conditions and analyzed, confirming the expected component structures. The hyperbranched polymer was mixed with DNA and efficient binding was shown to occur through electrostatic interactions. The hyperbranched structures could be reduced easily, generating lower toxicity oligomer chains.

Abstract:The challenge in the synthesis of protein-polymer conjugates for biological applications is to synthesize a stoichiometric (typically 1: 1) conjugate of the protein with a monodisperse polymer, with good retention of protein activity, significantly improved pharmacokinetics and increased bioavailability, and hence improved in vivo efficacy. Here we demonstrate, using myoglobin as an example, a general route to grow a PEG-like polymer, poly(oligo(ethylene glycol) methyl ether methacrylate) [poly(OEGMA)], with low polydispersity and high yield, solely from the N-terminus of the protein by in situ atom transfer radical polymerization (ATRP) under aqueous conditions, to yield a site-specific (N-terminal) and stoichiometric conjugate (1:1). Notably, the myoglobin-poly(OEGMA) conjugate [hydrodynamic radius (R-h): 13 nm] showed a 41-fold increase in its blood exposure compared to the protein (R-h: 1.7 nm) after IV administration to mice, thereby demonstrating that comb polymers that present short oligo(ethylene glycol) side chains are a class of PEG-like polymers that can significantly improve the pharmacological properties of proteins. We believe that this approach to the synthesis of N-terminal protein conjugates of poly(OEGMA) may be applicable to a large subset of protein and peptide drugs, and thereby provide a general methodology for improvement of their pharmacological profiles.

Abstract:To render protein drugs more suitable for clinical treatment, PEGylation has been widely used to ameliorate their inherent deficiencies, such as poor stability, rapid elimination in the bloodstream, and high immunogenicity. While increasingly PEGylated protein drugs have been approved by the FDA, the non-degradability of PEG and the emergence of anti-PEG antibodies after injection raise concerns about their cumulative chronic toxicity and long-term therapeutic efficacy. Zwitterionic polymer, with a unique structure containing equal amounts of positively charged and negatively charged groups, shows a different hydration behavior to PEG, which may be a superior PEG alternative for protein conjugation. In this concept review, a series of features beyond that of PEGylated protein exhibited by protein-zwitterionic polymer conjugate are discussed and some suggestions are presented for their future direction.
