Abstract:The paper studies the factors influencing the collagen extraction from pigs' tendon by using the pepsin, including pH and enzyme dosage. The result showed that the enzymic hydrolysis was the best when the ratio of pepsin to tendons was1∶10 and the concentration of acetic acid was 0.5mol/L.

Abstract:During the extraction, it is important to choose the class of enzyme. But, in order to ensure the biologic activity, purification and next work, the result of extraction is more influenced by enzyme and its quantity. The influence of pepsin and ficin on the collagen picking up is studied, and the result show that the efficiency and outcome are the best when the qualities ratio of pepsin to cattle tendons is 0.1, and the ratio of ficin to cattle tendons is 0.01.

Abstract:Enzymatic treatment to improve the extraction of collagen from chicken bone powder was explored in this paper.The raw materials was decalcified by 1.0mol/L HCl for 2 days,defatted by hexane and then hydrolyzed by pepsin under the following optimal conditions:enzyme dosage of 150U/g,reaction time of 120min,pH value of 1.7,and reaction temperature of 37℃.For the extraction of collagen from the enzymatic hydrolyzate,the best liquid/solid ratio and extraction time were 6:1 and 5h respectively.Under these conditions,the extraction rate and protein content of the collagen from poultry bone were of 18.4% and 54%,respectively.

Abstract:In the extraction of collagens from mammalian tissues, the free pepsin used in the acid–pepsin extraction system is hard to recycle, and there is a risk of enzyme protein contamination in the extracted collagen products, which limits their applications. To solve this problem, an immobilized pepsin was successfully prepared via the covalent crosslinking of glutaraldehyde using a 3-aminopropyltriethoxysilane (APTES) surface modified silica clay as the support. The immobilized pepsin was applied for the extraction of collagen from bovine hide. The optimal immobilization process involves incubating pepsin with an initial concentration of 35 mg mL −1 and glutaraldehyde with 5% activated APTES modified silica clay at 25 °C for 60 min, by which the loading amount of pepsin was 220 mg g −1 and the activity of the immobilized pepsin was 4.2 U mg −1 . The collagen extracted using acetic acid and the immobilized pepsin method retained its complete triple helix structure. This research thus details an effective separation method using pepsin for extraction of collagen via an acetic acid–enzyme method, where the extracted collagen may be a candidate for use in biomaterial applications.

Abstract:The different effects of three acid solutions in the extraction of pork skin collagen were discussed in this study.By the experiment of single factor,the main points influencing the extraction percentage were analyzed.Furthermore,the extraction by the three acid solutions was compared It was indicated that the citric acid solution acted best,the acetic acid solution took the second place,and the hydrochloric acid solution gained the worst effect when the extraction percentage of pork skin collagen was used as standard.

Abstract:The enzymatic method of collagen peptides from squid skin was studied. One-factor-at-a-time and orthogonal array methods were used to investigate the optimization of enzymatic preparation of collagen peptides from squid skin. The optimum extraction conditions:firstly,alcalase hydrolyzed for 4 h under the conditions of 47.5℃,pH 10.5 and enzyme concentration 0.3%,then papain combined flavorzyme (1∶2,quality ratio) hy-drolyzed for 4.5 h under the conditions of 50℃,pH 7.5 and enzyme concentration 0.4%,then the peptides max-imum DH was 28.89%,the DPPH·scavenging rate was 88.24%and recovery ratio of peptides was 49.74%,in addition,the content of the collagen peptides had a molecular weight distribution of 180 Da-2 000 Da was 84.56%. Result showed that the qualified peptides could be made under the optimized conditions.

Abstract:Collagens were extracted from bovine skin by acid method, the effects of the ration of material to liquor, temperature and extract time were studied. The optimum extraction conditions were obtained as follows: 0.5mol/ L acetic acid as extraction media, the ration of material to liquor 1∶30, and extraction at 32℃ for 6 h. Collagens by isoelectric precipitation and purified were then analyzed by SDS-PAGE and DSC. Comparing with type Ⅰ collagen from Sigma, the extractives were identified as the typical type Ⅰ collagen. In addition, the CD and fibril formation experiment confi rmed that the extracted collagens retain its original triple helical structure.

Abstract:In this study, neutral proteinase, alkaline proteinase, and papain were used to hydrolyze fresh pig skin respectively. The effects of pH value ,temperature, enzyme content and substrate content on hydrolysis were studied by the single factor test, and orthogonal design, by assessing the parameter ‐ degree of hydrolysis (DH), was adopted to determine the optimum condition of hydrolysis. The optimal hydrolysis condition of neutral proteinase was: pH 6.5, 45°C, enzymatic content 0.5%, and substrate content 15%; while The optimal hydrolysis condition of alkaline proteinase was: pH 11, 55°C, enzymatic content 0.5%, and substrate content 35%; and the most suitable hydrolysis parameters of papain were: pH 5, temperature of 60°C, 0.9% of enzyme content and 30% of substrate content. The ability of hydrolysis was sequenced as: alkaline proteinase > papain > neutral proteinase. On the basis of the optimum condition of each single proteinase, any two of the proteinases were chosen to hydrolyze fresh pig skin by two‐step‐hydrolysis. The results showed that any combination of the enzymes has a higher DH value than its single component and the adding sequence inflects DH value. Overall, the two‐step‐hydrolysis of papain plus alkaline shows the highest DH value (22.31%). Study on Enzymatic Hydrolysis Process of Fish Collagen and Antihypertensive Peptides of Collagen

Abstract:Single-factor and orthogonal experiments were employed to optimize the preparation process of collagen polypeptide through the hydrolysis of the fish collagen by pepsin.The angiotensin converting enzyme (ACE) inhibitory activity of marine and freshwater fish collagen was determined by high performance liquid chromatography to characterize its blood pressure-lowering function.It showed that the optimal hydrolysis condition of pepsin was emzyme-to-substrate ratio of 1 ∶ 125,substrate concentration 3％,temperature 37 ℃,initial pH2.5 and reaction time 12 h while DH was 19.17％.The results showed that the supernatant of enzymatic hydrolysis of marine fish collagen and freshwater fish collagen possessed higher ACE inhibitory activity,while the IC50 of their hydrolysates to ACE were 1.34 mg/mL and 1.52 mg/mL.The hydrolysate was separated by Amicon Ultra-4 Centrifugal Filter Devices with the molecular weight lower than 3 ku.The inhibitory activities of the two fractions was determined.The results showed that the IC50 of CP-Ⅰ (≤3 ku) and CP-Ⅲ (≤3 ku) were 0.43 mg/mL and 0.60 mg/mL.While the IC50 of CP-Ⅱ (≥3 ku)and CP-Ⅳ(≥3 ku)were 1.78 mg/mL and 1.69 mg/mL.

Abstract:BACKGROUND: Sturgeon (Acipenser sturio Linnaeus) skin contains high amount of nutrients including unsaturated fatty acids and collagen. A pepsin-assisted extraction procedure was developed and optimized for the extraction of collagen from Chinese sturgeon (Acipenser sturio Linnaeus) skins.OBJECTIVE: To determine the optimum conditions with the maximum yield of the pepsin-soluble collagen (PSC) extraction.MATERIALS AND METHODS: The conditions of the extraction were optimized using response surface methodology. The Box-Behnken design was used to evaluate the effects of the three independent variables (extraction time, enzyme concentration, and solid-liquid ratio) on the PSC yield of the sturgeon skin.RESULTS: THE OPTIMAL CONDITIONS WERE: solid-liquid ratio of 1:11.88, enzyme concentration of 2.42%, and extraction time of 6.45 h. The maximum yield of 86.69% of PSC was obtained under the optimal conditions. This value was not significantly different from the predicted value (87.4%) of the RSM (P < 0.05).CONCLUSION: The results of this study indicated that the production of PSC from sturgeon skin is feasible and beneficial. The patterns of sodium dodecyl sulfate-polyacrylamide gel electrophoretic patterns (SDS-PAGE) indicated that the sturgeon skin contains type I collagen, which is made of α-chain and β-chain. The infrared spectra of the collagens also indicated that pepsin hydrolysis does not affect the secondary structure of collagen, especially triple-helical structure.

Abstract:Type I collagen is commonly recognized as the gold standard biomaterial for the manufacturing of medical devices for health-care related applications. In recent years, with the final aim of developing scaffolds with optimal bioactivity, even more studies focused on the influence of processing parameters on collagen properties, since processing can strongly affect the architecture of collagen at various length scales and, consequently, scaffolds macroscopic performances. The ability to finely tune scaffold properties in order to closely mimic the tissues' hierarchical features, preserving collagen's natural conformation, is actually of great interest. In this work, the effect of the pepsin-based extraction step on the material final properties was investigated. Thus, the physico-chemical properties of fibrillar type I collagens upon being extracted under various conditions were analyzed in depth. Correlations of collagen structure at the supramolecular scale with its microstructural properties were done, confirming the possibility of tuning rheological, viscoelastic and degradation properties of fibrillar type I collagen.

Abstract:The collagen was extracted successfully from solid waste of leather industry under 4-35℃.The principle and method of extraction was investigated. The optimal extraction condition is confirmed: Ca(OH)2 concentration is 4.6%, enzyme concentration is 2‰, temperature is 30℃ and time is 72h. The extracted collagen was characterized by SDS-PAGE and IR spectrum.

Abstract:As the collagen protein market increasingly grows, it becomes a priority to explore a economic, stable , and environmentally-friendly extraction method to reali ze the high-value transformation of pigskin. In thi s paper, the structure of collagen protein is introduced first. Then five methods, which extract collagen protein f rom pigskin or a kind of leather industry waste-chrome shavings, are reviewed. These methods include the salting out me thod, alkali method, acid method, enzyme method and hybrid metho d. They are contrasted in several ways, and thus pr ovide evidence for better extracting collagen protein.

Process of Extracting Collagen from Bone by Compund-enzyme Stepping Hydrolysis

Abstract:With initial hydrolysis product of bone collagen as the research object and the degree of hydrolysis as the determining index,the course of the depth-hydrolysis was detected by the formol titration method.Enzymatic hydrolysis conditions of neutral protease and flavor protease were determined by analyzing the effects on the degree of hydrolysis.Orthogonal experimental results showed that the optimum conditions were as follows:material ratio of 1:750,initiative pH value of 6.5,hydrolysis 3h by neutral protease and then hydrolysis 2h by flavor protease.The degree of hydrolysis can attain 63.6% under these conditions.

Abstract:In the present study, response surface methodology was performed to investigate the effects of extraction parameters on pepsin-solubilised collagen (PSC) from the skin of the giant croaker Nibea japonica. The optimum extraction conditions of PSC were as follows: concentration of pepsin was 1389 U/g, solid-liquid ratio was 1:57 and hydrolysis time was 8.67 h. Under these conditions, the extraction yield of PSC was up to 84.85%, which is well agreement with the predict value of 85.03%. The PSC from Nibea japonica skin was then characterized as type I collagen by using sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). The fourier transforms infrared spetroscopy (FTIR) analysis revealed that PSC maintains its triple-helical structure by the hydrogen bond. All PSCs were soluble in the pH range of 1.0-4.0 and decreases in solubility were observed at neutral or alkaline conditions. All PSCs had a decrease in solubility in the presence of sodium chloride, especially with a concentration above 2%. So, the Nibea japonica skin could serve as another potential source of collagen.

Abstract:Collagen is a type of structural protein that constructs skin, teeth, bones, and muscles. Collagen hydrolysate is a product resulting from the hydrolysis of polypeptides that can be carried out enzymatically and chemically. This study aimed to obtain collagen hydrolysate using different enzymes and to determine its antioxidant content. The stages of the research carried out were characterization of collagen, hydrolysis of collagen with different enzymes, and characterization of collagen hydrolysate. The collagen used has a protein content of 95.75% considered as high protein content. The amino acid content of collagen was dominated by glutamic acid, which was 7.06%. Meanwhile, the pepsin-hydrolyzed collagen contained 7.81% of glutamic acid, and pepsin-hydrolyzed and trypsin-hydrolyzed collagen was 6.13% and 6.76%, respectively. TT the highest degree of hydrolysis was obtained from papain enzyme treatment 54.98%.

Abstract:The present study used acetic acid, sodium hydroxide, and pepsin extract acid-soluble collagen (ASC), alkali-soluble collagen (ALSC), and pepsin-soluble collagen (PSC) from the bones of spent-hens, and the effects of three extraction methods on the characteristics, processing properties, antioxidant properties and acceptability of chicken bone collagen were compared. The results showed that the extraction rates of ASC, ALSC and PSC extracted from bones of spent-hens were 3.39%, 2.42% and 9.63%, respectively. The analysis of the amino acid composition, sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), Fourier transform infrared spectroscopy (FTIR), and ultraviolet full spectrum showed that the collagen extracted by the three methods had typical collagen characteristics and stable triple-helix structure, but the triple helical structure of PSC is more stable, and acid and alkaline extraction seems to have adverse effects on the secondary structure of chicken bone collagen. Differential scanning calorimetry (DSC) and scanning electron microscopy (SEM) scanning showed that PSC had higher thermal stability and more regular, loose, and porous microstructure. In addition, PSC has good processing properties, in vitro antioxidant activity, and organoleptic acceptability. Therefore, enzymatic hydrolysis was still one of the best methods to prepare collagen from bones of spent-hens, and enzyme-soluble collagen has wider application prospects in functional food and medicine and also provides an effective way for the high-value comprehensive utilization of waste chicken bone by-products.

Abstract:We evaluated the optimal conditions for extraction of pepsin-soluble collagen from the skin of Chinese giant salamander (Andrias davidianus) using a Box-behnken design experiment. The concentration of pepsin, sol-vent-to-solid ratio, and enzyme extraction time were the test factors and the extraction rate of collagen was the target parameter. We used response surface methodology to describe the influence of the three factors on the ex-traction rate of collagen and developed aquadratic polynomial regression model to optimize the extraction condi-tions. The order of influence of the factors was:solvent-to-solid ratio>enzyme extraction time>pepsin amount. The interaction between the solvent-to-solid ratio and enzyme extraction time had a significant effect on the extraction rate of collagen, while other interactions had no significant effect. The optimal conditions predicted by the regres-sion model for extraction were slightly adjusted to meet actual production. The specific parameters were: pepsin concentration was 16.5%, solvent-to-solid ratio was 15 mL/g, and enzyme extraction time was 29 h. Under these conditions the extraction rate was 66.99%, which was similar to the theoretical value (67.83%) predicted by the regression model.
Comparison of the Structural Characteristics of Native Collagen Fibrils Derived from Bovine Tendons Using Two Different Methods: Modified Acid-Solubilized and Pepsin-Aided Extraction

Abstract:Native collagen fibrils (CF) were successfully extracted from bovine tendons using two different methods: modified acid-solubilized extraction for A-CF and pepsin-aided method for P-CF. The yields of A-CF and P-CF were up to 64.91% (±1.07% SD) and 56.78% (±1.22% SD) (dry weight basis), respectively. The analyses of both amino acid composition and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) confirmed that A-CF and P-CF were type I collagen fibrils. Both A-CF and P-CF retained the intact crystallinity and integrity of type I collagen’s natural structure by FTIR spectra, circular dichroism spectroscopy (CD) and X-ray diffraction detection. The aggregation structures of A-CF and P-CF were displayed by UV–Vis. However, A-CF showed more intact aggregation structure than P-CF. Microstructure and D-periodicities of A-CF and P-CF were observed (SEM and TEM). The diameters of A-CF and P-CF are about 386 and 282 nm, respectively. Although both A-CF and P-CF were theoretically concordant with the Schmitt hypothesis, A-CF was of evener thickness and higher integrity in terms of aggregation structure than P-CF. Modified acid-solubilized method provides a potential non-enzyme alternative to extract native collagen fibrils with uniform thickness and integral aggregation structure.
