Abstract:In this chapter, we will introduce the methods for the generation of transgenic rabbits. These methods are those used for transgenic mice but with some modifications and refinements. We will review all possible methods for rabbit transgenesis with emphasis on microinjection method because it is the most popular method so far.

Abstract:Transgenic rabbits have been found to be excellent animal models for biomedical research.Rabbits expressing human genes have been used as models for cardiovascular disease,AIDS,and cancer researches.Transgenic rabbits have provided new special insights into the mechanisms regulating the development of human atherosclerosis.The most commonly used method for creating transgenic rabbits is pronuclear microinjection.Here,we describe the protocol of creating transgenic rabbits currently used in our laboratory,and briefly review the present and future use of transgenic rabbits in biomedical research.

Abstract:Transgenic rabbits are widely used as a model organism for biomedical research, and the transgenic rabbit system is especially valuable because it fills an important niche between laboratory mice and larger domesticated mammals. In order to describe the current status and development trends of the use of transgenic rabbits in biomedical research precisely, we performed a quantitative analysis of the published data, collected by searching biomedical databases. Currently, there are about 217 papers related to transgenic rabbits, originating from 22 countries. The number of publications has slowly increased over time, reaching its peak in 2004 and 2007. Approximately one third of the publications come from the USA, and one quarter come from Japan. The USA, Japan and France were the top three producers of publications related to transgenic rabbits. These publications mainly focused on cardiovascular disease (CVD) and the study of therapeutic protein bioreactors. Approximately 19 transgenic rabbit lines have been established for the study of CVD, and 20 recombinant proteins have been produced from transgenic rabbit milk or blood. The remaining publications largely focused on virology and immunology, diabetes mellitus, cancer, and genetics. These publications provide new insights into the mechanisms responsible for the development of human disease and shed light on the management of some genetic disorders. Thus, this quantitative review of the literature reveals that transgenic rabbits play an increasingly important role in biomedical research.
Create transgenic rabbits by microinjecting human apoA- II gene into fertilized eggs

Abstract:Objective: To create transgenic rabbits by microinjecting human apolipoprotein A- II (apoA- II) gene into one-cell embryos, to study apoA- II gene function on plasma lipoprotein metabolism and atherosclerosis. Methods: Superovulation and synchronization of estrus were induced in female Japanese White Rabbits by injecting hormone, then mating with male. After collected the fertilized eggs, the human apoA- II gene was microinjected into the male pronucleus of eggs. The injected eggs were transferred into recipient female rabbits. Last, extract DNA from the new borns ear and determine whether the newborns were transgenic by polymerase chain reaction (PCR) or Southern blot analysis. Results: A total of 822 embryos with microinjection of human apoA II gene were implanted into 28 recipient rabbits. The number of surviving newborns was 37.3 transgenic positive surviving founders were found with human apoA- II.

Abstract:While the pronuclear embryos are microinjected by the current microinjection system, its genetransfer rate of foreign DNA is usually low due to its relatively unsatisfied diaphaneity and inconvenient operation. In addition, its uncovered structure with microdrop is also easily contaminated. In this study, a new horizontal microinjection system has been established to solve these problems. Because the average time per pronuclear embryo by microinjection has been significantly shortened with the horizontal system, the cleavage rate and 16-cell rate of those microinjected embryos increased markedly. Furthermore, with only a half -dose of DNA in the current system, by the horizontal system under×100 magnification both the survival rate of pronuclear embryos following microinjection and the integration frequency of exogenous DNA injected into embryos also increased. With the horizontal microinjection system, the fusion gene of pMT-hIGF-1 was utilized as foreign DNA. According to the result of Southern blot assay, among 24 founders there were 5 rabbits integrated with foreign DNA.

Abstract:Somatic cell nuclear transfer (SCNT) is a technology that enables differentiated somatic cells to acquire a totipotent state, thus making it of great value in developmental biology, biomedical research, and agricultural applications. Rabbit cloning associated with transgenesis has the potential to improve the applicability of this species for disease modeling, drug testing, and production of human recombinant proteins. In this chapter, we introduce our SCNT protocol for the production of live cloned rabbits.

Abstract:The rabbit is a classic animal model for biomedical research, but the production of gene targeted transgenic rabbits had been extremely challenging until the recent advent of gene editing tools. More than fifty gene knockout or knock-in rabbit models have been reported in the past decade. Gene edited (GE) rabbit models, compared to their counterpart mouse models, may offer unique opportunities in translational biomedical research attributed primarily to their relatively large size and long lifespan. More importantly, GE rabbit models have been found to mimic several disease pathologies better than their mouse counterparts particularly in fields focused on genetically inherited diseases, cardiovascular diseases, ocular diseases, and others. In this review we present selected examples of research areas where GE rabbit models are expected to make immediate contributions to the understanding of the pathophysiology of human disease, and support the development of novel therapeutics.

Abstract:Somatic cell nuclear transfer (SCNT) shows great value in the generation of transgenic animals, protection of endangered animals, and stem cell therapy. The combination of SCNT and gene editing has produced a variety of genetically modified animals for life science and medical research. Rabbits have unique advantages as transgenic bioreactors and human disease models; however, the low SCNT efficiency severely impedes the application of this technology. The difficulty in SCNT may be attributable to the abnormal reprogramming of somatic cells in rabbits. This review focuses on the abnormal reprogramming of cloned mammalian embryos and evaluates the progress and prospects of rabbit somatic cell cloning.

Abstract:AbstractThe rabbit is a common animal model that has been employed in studies on various human disorders and the generation of genetically modified rabbit lines is highly desirable. Female rabbits have been successfully cloned from cumulus cells and the somatic cell nuclear transfer (SCNT) technology is well established. The present study generated hypoxanthine phosphoribosyltransferase (HPRT) gene knockout rabbits using recombinant adeno-associated virus-mediated homologous recombination and SCNT. Gene trap strategies were employed to enhance the gene targeting rates. The male and female gene knockout fibroblast cell lines were derived by different strategies. When male HPRT knockout cells were used for SCNT, no live rabbits were obtained. However, when female HPRT+/− cells were used for SCNT, live, healthy rabbits were generated. The cloned HPRT+/− rabbits were fertile at maturity. We demonstrate a new technique to produce gene-targeted rabbits. This approach may also be used in the genetic manipulation of different genes or in other species.

Abstract:A high efficient and simple transgenic technology on mice and rabbits to transfect spermatozoa with exogenous DNA/DMSO complex to obtain transgenic offspring, which is namely called DMSO-sperm mediated gene transfer (SMGT). Mouse sperm could be either directly transfected via injection into testis or cultured in vitro with the plasmed DNA containing the enhanced green fluorescent protein (EGFP) that could be expressed in the embryos and offspring. Then, 36 living transgenic rabbits were produced using the same technology, and the transgenic ratio of 56.3% was detected using PCR and Southern blot. As the controls, the transgenic ratios of 39.6% and 47.8% have also been tested using the liposomes mediated technology of Tfx-50 Reagent or Lipefectamin-2000, respectively. The results show that the female transgenic rabbits, as the mammary gland bioreactor models, could express the human tissue plasminogen activator mutant (htPAm) in their mammary cells when they are adult.

Abstract:Rabbits are one of the most used experimental animals for investigating the mechanisms of human cardiovascular disease and lipid metabolism because they are phylogenetically closer to human than rodents (mice and rats). Cholesterol-fed wild-type rabbits were first used to study human atherosclerosis more than 100 years ago and are still playing an important role in cardiovascular research. Furthermore, transgenic rabbits generated by pronuclear microinjection provided another means to investigate many gene functions associated with human disease. Because of the lack of both rabbit embryonic stem cells and the genome information, for a long time, it has been a dream for scientists to obtain knockout rabbits generated by homologous recombination-based genomic manipulation as in mice. This obstacle has greatly hampered using genetically modified rabbits to disclose the molecular mechanisms of many human diseases. The advent of genome editing technologies has dramatically extended the applications of experimental animals including rabbits. In this review, we will update genetically modified rabbits, including transgenic, knock-out, and knock-in rabbits during the past decades regarding their use in cardiovascular research and point out the perspectives in future.
Transgene Expression of Enhanced Green Fluorescent Protein in Cloned Rabbits Generated from in Vitro-Transfected Adult Fibroblasts

Abstract:Live rabbits have previously been generated through nuclear transfer using adult cells as nuclear donors. We demonstrated in this study that transfected adult rabbit fibroblasts are also capable of supporting full-term development. The fibroblasts were transfected with a pEGFP-C1 plasmid using lipofectamine™ 2000, and the transgenic cells were derived from conditioned medium. The transgenic fibroblasts were cultured until confluent and then serum-starved prior to be used as nuclear donors. After nuclear transfer and activation, 22% (12/55) of the transgenic cloned embryos developed to the blastocyst stage. A total of 114 embryos at the 4- to 8-cell stage were transferred to the oviducts of 8 pseudo-pregnant mothers; 5 of these animals became pregnant, and 3 of the 5 mother rabbits carried the pregnancy to term. Caesarean section was performed on the 3 pregnant mothers, yielding 4 kits, one of which has survived for more than 9 months. Green fluorescence could be detected in the toenails of the living cloned rabbit and the offspring from the living cloned rabbit under ultraviolet light. DNA analyses confirmed that all 4 cloned rabbits were genetically identical to the transgenic donor cells, and that they all carried the EGFP gene. The present study demonstrated that transgenic rabbits can be generated through nuclear transfer. These results may facilitate future developments in the genetic engineering of rabbits.

Abstract:Objective To establish a transgenic animal method suitable for our laboratory by optimizing the factors affecting the procedure of transgenic urotesin Ⅱ(U Ⅱ) rabbits creation.Methods U Ⅱ transgenic rabbits were created by microinjection method.We compared the follicle-stimulating hormone(FSH) administration protocol and the pregnant mare serum gonadotropin(PMSG) administration protocol in collection of more zygotes.Results The results showed that the FSH administration protocol was better than the PMSG administration protocol in collection of more zygotes.We successfully established the U Ⅱ transgenic rabbits model by microinjection method.Conclusion We have successfully created UⅡ transgenic rabbits by microinjection method and provided a powerful tool for studies on U Ⅱ biological function and its relationship with diseases.

Abstract:Immunodeficient mice have been used predominantly in biomedical research. Realizing that large animal species may have an enhanced ability to predict clinical outcome relative to mice, we worked to develop immunodeficient rabbits by CRISPR/Cas9. We first demonstrated that multiplex embryo transfer efficiently produced multiple lines of single-gene mutant (SGM) founders. Embryos microinjected with single sgRNA targeting FOXN1 , RAG2 , IL2RG or PRKDC were pooled for embryo transfer. As few as three recipients were used to produce twenty SGM founders for four genes. We then demonstrated the powerful multiplex targeting capacity of CRISPR/Cas9. First, two genes on the same chromosome were targeted simultaneously, resulting in three RAG1/RAG2 double-gene mutant (DGM) founders. Next we microinjected forty-five embryos each with five sgRNAs targeting FOXN1 , RAG1 , RAG2 , IL2RG and PRKDC , and transferred them to two recipients. Five founders were produced: one SGM, two DGM, one triple-gene mutant and one quadruple-gene mutant. The present work demonstrates that multiplex embryo transfer and multiplex gene targeting can be used to quickly and efficiently generate mutant rabbit founders. Four lines of SGM (e.g. FOXN1 , RAG2 , IL2RG , and PRKDC ) immunodeficient rabbits, as well as multigenic mutant immunodeficient rabbits have been produced. These animals may prove useful for biomedical research.

Abstract:The rabbit is a classical animal model in biomedical research. The development of genetically modified rabbit models has been severely impeded because of the lack of pluripotent stem cells and the extremely low efficiency of somatic cell nuclear transfer. With the rapid development of novel genome-editing techniques, such as ZFNs, TALENs, and CRISPR/Cas9, genetic engineering of rabbits has developed rapidly. Genetically modified rabbit models are playing increasingly important roles in the field of biomedicine. In this review, we discuss the development of genetically modified rabbit models and their applications in human disease research to provide a theoretical reference for promoting the development of human disease animal models.

Abstract:Rabbits have played a significant role in both livestock production and the advancement of reproductive scientific research. Their unique biological traits, including induced ovulation and a reproductive process that closely mirrors that of humans, have been pivotal in their use as a model. Moreover, their body size is perfectly aligned with the 3Rs principles: Replacement, Reduction, and Refinement. Consequently, techniques for gamete collection and embryo recovery, followed by their use in artificial insemination or embryo transfer, are characterized by being minimally invasive. However, refining in vitro fertilization and embryo culture techniques continues to present challenges. The incorporation of cutting‐edge genomic editing tools, such as CRISPR/Cas9, has reestablished rabbits as essential models in genetic and biomedical research, driving scientific progress. This review aims to describe the most effective reproductive biotechnologies for both male and female rabbits and how these methodologies are in line with the 3Rs principles—Replacement, Reduction, and Refinement—highlighting their significance in conducting ethical research.

Abstract:<span>Transgenic animals incorporating human antibody genes are extremely attractive for drug development because they obviate subsequent antibody humanization procedures required for therapeutic translation. Transgenic platforms have previously been established using mice, but also more recently rats, chickens, and cows and are now in abundant use for drug development. However, rabbit-based antibody generation, with a strong track record for specificity and affinity, is able to include gene conversion mediated sequence diversification, thereby enhancing binder maturation and improving the variance/selection of output antibodies in a different way than in rodents. Since it additionally frequently permits good binder generation against antigens that are only weakly immunogenic in other organisms, it is a highly interesting species for therapeutic antibody generation. We report here on the generation, utilization, and analysis of the first transgenic rabbit strain for human antibody production. Through the knockout of endogenous IgM genes and the introduction of human immunoglobulin sequences, this rabbit strain has been engineered to generate a highly diverse human IgG antibody repertoire. We further incorporated human CD79a/b and Bcl2 (B-cell lymphoma 2) genes, which enhance B-cell receptor expression and B-cell survival. Following immunization against the angiogenic factor BMP9 (Bone Morphogenetic Proteins 9), we were able to isolate a set of exquisitely affine and specific neutralizing antibodies from these rabbits. Sequence analysis of these binders revealed that both somatic hypermutation and gene conversion are fully operational in this strain, without compromising the very high degree of humanness. This powerful new transgenic strategy will allow further expansion of the use of endogenous immune mechanisms in drug development.</span>

Abstract:Zinc-finger nucleases and transcription activator-like effector nucleases are novel gene-editing platforms contributing to redefine the boundaries of modern biological research. They are composed of a non-specific cleavage domain and a tailor made DNA-binding module, which enables a broad range of genetic modifications by inducing efficient DNA double-strand breaks at desired loci. Among other remarkable uses, these nucleases have been employed to produce gene knockouts in mid-size and large animals, such as rabbits and pigs, respectively. This approach is cost effective, relatively quick, and can produce invaluable models for human disease studies, biotechnology or agricultural purposes. Here we describe a protocol for the efficient generation of knockout rabbits using transcription activator-like effector nucleases, and a perspective of the field.
Production of Transgenic Rabbit Embryos Through Intracytoplasmic Sperm Injection

Abstract:SummaryThe objective of this study was to test if intracytoplasmic sperm injection (ICSI)-mediated gene transfer was an effective method in the production of transgenic rabbit embryos. Rabbit sperm diluted in different media with various pH were treated by freezing without cryoprotectant, and their ability for DNA uptake was determined. In these experiments using production of transgenic rabbit embryos by ICSI, exogenous genes at three concentrations and of two conformation types were used. The rate of DNA association to the sperm seen by rhodamine-tagged DNA encoding green fluorescent protein (GFP) was 90.0%, 92.7%, 91.0%, 91.7%, and 92.3%, respectively in TCM199, DM, DPBS, CZB, and HCZB media. The DNA attachment to sperm was not affected by media pH within the range of 5.4–9.4 (p > 0.05). Expression of GFP first occurred at the 2-cell stage and continued to blastocyst formation. DNA concentration (between 5, 10, and 20 ng/μl) or conformation (linear and circular) had no effect on the production rate of transgenic embryos. These results indicated that genetically modified rabbit blastocysts can be efficiently produced by ICSI technique.
