Abstract:Somatic mutations drive cancer immune evasion. Zhuang et al. find SPPL3 as crucial for sensitivity to NK-mediated killing. SPPL3 deletion increases N-glycan complexity, which impairs NK receptor binding. B3GNT2 is responsible for N-glycan extensions. Inhibition of this pathway restores cancer cell sensitivity, highlighting a potential strategy to improve cancer immunotherapy.
