Abstract:In order to provide a reference for the further study of microbial contamination in the pork production process. Microbial contamination of pigs came from three slaughterhouses were detected,and critical control points in the progress of hog slaughter and processing were analyzed. The results showed that microbial contamination existed in the entire slaughter and processing progress,including shower and assassination bloodletting,separation of the internal organs,chopping boards,workshop environment,personal hygiene of the operators,etc.,which should be paid more attention to. The results indicated that reasonable protection measures should be carried out,disinfection awareness of the operators should be improved,and regular disinfection should be ruled under the condition of continuous operation.

Abstract:Pork is one of the most common foods causing microbial foodborne diseases. Since pork directly enters the market after slaughtering, the control of microbial contamination in the slaughtering processes is the key to ensuring the quality and safety of pork. The contamination level of Escherichia coli , a health-indicator bacterium, can reflect the risk level of potential pathogens. In order to assess the E. coli exposure risk of pork during slaughtering and to identify the key control points, we established an E. coli quantitative exposure assessment model for swine-slaughtering processes in slaughterhouses of different sizes. The model simulation data indicated the E. coli contamination pattern on the surfaces of swine carcasses during slaughtering. The changes in E. coli contamination were analyzed according to the simulation data of each slaughtering process. It was found that the number of E. coli after trimming in big and small slaughterhouses increased to the maximum values for the whole processes, which were 3.63 and 3.52 log 10 CFU/100 cm 2 , respectively. The risk contribution of each slaughtering process to the E. coli contamination on the surface of terminal swine carcasses can be determined by correlation analysis. Because the absolute value of correlation coefficient during the trimming process was maximum (0.49), it was regarded as the most important key control point. This result can be further proved via the multilocus sequence typing of E. coli . The dominant sequence type before trimming processes was ST10. ST1434 began to appear in the trimming process and then became the dominant sequence type in the trimming and pre-cooling processes. The model can provide a theoretical basis for microbial hygiene supervision and risk control in swine-slaughtering processes.

Abstract:Chilled beef is inevitably contaminated with microorganisms, starting from the very beginning of the slaughter line. A lot of studies have aimed to improve meat safety and extend the shelf life of chilled beef, of which some have focused on improving the decontamination effects using traditional decontamination interventions, and others have investigated newer technologies and methods, that offer greater energy efficiency, lower environmental impacts, and better assurances for the decontamination of beef carcasses and cuts. To inform industry, there is an urgent need to review these interventions, analyze the merits and demerits of each technology, and provide insight into 'best practice' to preserve microbial safety and beef quality. In this review, the strategies and procedures used to inhibit the growth of microorganisms on beef, from slaughter to storage, have been critiqued. Critical aspects, where there is a lack of data, have been highlighted to help guide future research. It is also acknowledge that different intervention programs for microbiological safety have different applications, dependent on the initial microbial load, the type of infrastructures, and different stages of beef processing.

Abstract:Meat spoilage (MS) is a complex microbial ecological process involving multiple specific microbial interactions and is vulnerable to numerous events. As consumer demand for meat products increases, the quality and safety of meat products gain much attention. MS is detrimental to people's health and leads to the waste of meat products which will cause huge losses during production, storage, transportation, and marketing. This mini-review summarizes the diversity of spoilage microorganisms in livestock, poultry, and aquatic products, and the approaches to inhibit MS. This would facilitate the targeted development of technologies against MS, to extend meat's shelf life, and effectively diminish food waste and economic losses.

Abstract:A total of 37 samples of feces,water and soil were collected from 10 different types of livestock and poultry breeding farms in Jiangsu Province for determination of population of Fecal coliform and Samonella therein.Meanwhile drug resistance of the Escherichia coli isolated from wastewater and effect of animal excreta on population of antibiotic tolerant bacteria in soil and water were analyzed.Results show that of the 10 livestock and poultry farms studied,9 were found discharging animal feces directly into the soil and water without any pre-treatment,and their discharges significantly exceeded the criterion for population of Fecal coliform,and 19% of the fecal samples was found to have Samonella.The count of Fecal coliform in the soils applied with fresh manure was well above 105 g-1.All the Escherichia coli isolated from the wastewater showed tolerance to a number of drugs and one isolate was even able to tolerate 10 kinds of antimicrobial.The count of tetracycline or streptomycin resistant bacteria in the soil or water amended with fresh manure was much higher than in common soil or water.

Comprehensive Reviews in Food Science and Food Safety

Abstract:Microorganisms play a crucial and unique role in fish and fish product safety. The presence of human pathogens and the formation of histamine caused by spoilage bacteria make the control of both pathogenic and spoilage microorganisms critical for fish product safety. To provide a comprehensive and updated overview of the involvement of microorganisms in fish and fish product safety, this paper reviewed outbreak and recall surveillance data obtained from government agencies from 1998 to 2018 and identified major safety concerns associated with both domestic and imported fish products. The review also summarized all available literature about the prevalence of major and emerging microbial safety concerns, including Salmonella spp., Listeria monocytogenes, and Aeromonas hydrophila, in different fish and fish products and the survival of these pathogens under different storage conditions. The prevalence of antibiotic‐resistant bacteria (ARB) and antibiotic‐resistant genes (ARGs), two emerging food safety concerns, is also reviewed. Pathogenic and spoilage microorganisms as well as ARB and ARGs can be introduced into fish and fish products in both preharvest and postharvest stages. Many novel intervention strategies have been proposed and tested for the control of different microorganisms on fish and fish products. One key question that needs to be considered when developing and implementing novel control measures is how to ensure that the measures are cost and environment friendly as well as sustainable. Over the years, regulations have been established to provide guidance documents for good farming and processing practices. To be more prepared for the globalization of the food chain, harmonization of regulations is still needed.

Abstract:The objective of this study was to investigate the microbial contamination levels (aerobic bacteria plate count [APC], coliforms, Escherichia coli, Staphylococcus aureus, and Listeria monocytogenes) in mixed pressed ham and cooked sausage. A total of 180 samples were collected from factories with and without hazard analysis critical control point (HACCP) systems at four steps: after chopping (AC), after mixing (AM), cooling after the first heating process, and cooling after the second heating process. For ham, APCs and coliform and E. coli counts increased when ingredients were added to the meat at the AC step. Final product APC was 1.63 to 1.85 log CFU/g, and coliforms and E. coli were not detected. S. aureus and L. monocytogenes were found in nine (15.0%) and six (10.0%) samples, respectively, but only at the AC and AM steps and not in the final product. Sausage results were similar to those for ham. The final product APC was 1.52 to 3.85 log CFU/g, and coliforms and E. coli were not detected. S. aureus and L. monocytogenes were found in 29 (24.2%) and 25 (20.8%) samples at the AC and AM steps, respectively, but not in the final product. These results indicate that the temperature and time of the first and second heating are of extreme importance to ensure the microbiological safety of the final product regardless of whether a HACCP system is in place. Microorganism contamination must be monitored regularly and regulations regarding sanitization during processing should be improved. Education regarding employee personal hygiene, environmental hygiene, prevention of cross-contamination, ingredient control, and step-by-step process control is needed to reduce the risk of food poisoning.

Abstract:Microbiological contamination changes the qualitative and quantitative profile of food, which makes it an important issue of food safety systems. Finished products with components of animal origin or combined vegetable and livestock raw materials are especially prone to microbiological contamination. This review features the most common sources of microbiological contamination and its prevention in finished products. The review covered 20 years of English and Russian scientific articles and standards indexed in Scopus, ScienceDirect, PubMed, and eLIBRARY.RU. The descriptors included microbiological contamination, food raw materials, and ready-to-eat products. The analysis involved some older publications, provided they possessed a high scientific relevance and/or a high citation index. The search criteria concentrated on the detection of microbiological contaminants in food raw materials and food products in Russia and abroad. The percentage of detections for individual groups of microorganisms was calculated as opposed to their total number. The data obtained can be summarized as follows. Regular industrial monitoring can minimize the risk of microbiological contamination. Complex finished products have a higher risk and require heat treatment. Composite products that cannot be succumbed to additional heat treatment need new technologies that minimize microbiological contamination, e.g., intensive cooling, shock freezing, electromagnetic processing, protective food coatings, etc. Even a small amount of animal raw materials may cause extra risk of contamination with Listeria monocytogenes, Salmonella enteritidis, etc. A mix of different raw materials changes the microbiological profile of the finished product. As a result, finished products have to be monitored throughout the entire shelf-life cycle, regardless of the percentage of animal raw material in the formulation.

Abstract:Poultry and livestock contribute a significant amount of energy and protein to the human diet. However, a main concern is the undesirable microbial population from the poultry and livestock system that may cause diseases in both humans and animals. Electrolyzed water has shown broad-spectrum sanitization efficiency and gained considerable popularity. This review aims to provide an overview of the use of electrolyzed water under different environment conditions for sanitization in livestock and poultry systems. Electrolyzed water serves as an excellent antimicrobial and anti-biofilm agent against various pathogenic, disease-producing, and undesirable microbes in every step of the livestock and poultry sectors from farm to fork. The synergistic effects of the oxidation reduction potential, pH, and chlorine concentration of electrolyzed water enable its potential and prompt mechanism for controlling and eliminating various microorganisms. Compared to other conventional disinfectants, electrolyzed water has been shown to be more efficient and effective. As an environmentally-friendly sanitizer, electrolyzed water has been applied in livestock and poultry production and processing sectors such as animal drinking water, feed decontamination, house sanitation, transportation system, slaughterhouse sanitation, meat-processing industry decontamination, sanitization of milk-processing industries, hatchery and eggshell decontamination. This review is aimed to discuss the microbial control in livestock and poultry industry by applying electrolyzed water as a novel sanitizer.

Abstract:Meat production is a complex system, continually receiving animals, water, air, and workers, all of which serve as carriers of bacteria. Selective pressures involved in different meat processing stages such as antimicrobial interventions and low temperatures, may promote the accumulation of certain residential microbiota in meat cutting facilities. Bacteria including human pathogens from all these sources can contaminate meat surfaces. While significant advancements have been made in enhancing hygienic standards and pathogen control measures in meat plants, resulting in a notable reduction in STEC recalls and clinical cases, STEC still stands as a predominant contributor to foodborne illnesses associated with beef and occasionally with pork. The second-and third-generation sequencing technology has become popular in microbiota related studies and provided a better image of the microbial community in the meat processing environments. In this article, we reviewed the potential factors influencing the microbial ecology in commercial meat processing facilities and conducted a meta-analysis on the microbiota data published in the last 10 years. In addition, the mechanisms by which bacteria persist in meat production environments have been discussed with a focus on the significant human pathogen E. coli O157:H7 and generic E. coli , an indicator often used for the hygienic condition in food production.

Abstract:The slaughtering environment is crucial for the food hygiene and safety of poultry products. Despite the global dominance of industrial processing, live bird slaughtering in wet markets persists due to cultural, religious, and economic reasons. This study aims to reveal the correlation between hygiene scales in wet markets and bacterial contamination levels on broiler carcasses, with a particular focus on pathobiont transmission risks and microbiome characteristics. Wet markets were categorized based on home-made ratings and the Hygiene and Biosecurity Assessment Tool (HABT). The study assessed total aerobic bacterial (TAB) levels, food spoilage and hygiene indicators (Pseudomonas and E. coli), foodborne pathogen Salmonella, and the microbiome of broiler carcasses, intestinal contents, and slaughtering facilities. Comparative analyses were conducted between market and industrial processing environments. TAB levels on broiler carcasses showed a significant negative correlation with hygiene scores, indicating that both HABT and home-made rating tools effectively assess and improve processing hygiene. Industrial processing consistently reduced bacterial contamination compared to wet markets. Although Salmonella spp. prevalence was lower in market-processed carcasses, the study identified significant cross-transmission of pathobionts and variations in bacterial composition with hygiene improvements. Notably, the microbiome analysis revealed overlaps in amplicon sequence variants (ASVs) between carcasses and contamination vectors, highlighting pathobiont transmission risks. The present study confirmed the scales of hygiene standards among wet markets reflect bacterial contamination on broiler carcasses. Enhancing slaughter practices to meet industrial hygiene standards is essential for reducing the transmission of foodborne pathogens and pathobionts, and improving food safety.

Abstract:The article presents the data of the analysis of the control strategy of food zoonoses (Campylobacter, E.coli O157, Enterobacteriacae, Listeria, Salmonella, Enterococcus) on the basis of chemical and physical decontamination methods at the stage of poultry processing in the conditions of slaughter enterprises of the EU. The factors of transmission of pathogens that determine the risks of contamination of poultry carcasses during the technological processes of transportation and slaughter of poultry, nutrition, cooling and heat treatment of carcasses are analyzed. It also analyzes scientific developments to reduce the risk to human health, depending on measures to reduce microbiological contamination by pathogens of broiler meat zoonoses. The article presents data on the effective strategy poultry food control zoonoses in the European Union based on the methods of reducing microbial contamination of poultry carcasses during processing. We conducted information by analyzing statistics and materials and reports published in national and international journals, study and systematization of scientific literature, the official reports of the International Program of WHO for the control and supervision of zoonoses in Europe, ESFA (European Agency for Safety food), the Center for disease control in the USA, documents regulating controls zoonosis poultry in the European Union. Continuous monitoring of food zoonoses pathogens of poultry is effective. Collecting information on the use of antimicrobials in zoonoses control programs in poultry is important. The data on the effective control bacterial pathogens at transportation poultry and during the slaughter process are: exposure without food, sanitary treatment of cages, identification the party with an high level of microbiological contamination, fixing the cloaca and prevent faecal contamination of carcasses, plan of slaughter at the slaughterhouse (defining positive poultry). We have also analyzed the effective control measures of the bacterial contamination of poultry carcasses in terms of companies engaged in the slaughter and processing of poultry.

Abstract:In a changing world where sustainability has become a must, the food system has been identified as one of the weakest links where there is a need for innovative solutions to address longstanding and novel challenges related to food safety and quality. The current food system is facing enormous pressure to tackle environmental sustainability issues while guaranteeing safety and diversifying production. In the last years, the possibility to exploit naturally occurring competitive mechanisms, which regulate the homeostasis of microbial ecosystems, has attracted a lot of attention and interest. More specifically, in the food transformation sector, bacteriocins and bacteriocinogenic microorganisms have long been considered an innovation tool attracting the attention of the researchers and industries due to their potential application in the meat sector. This paper presents an analysis of the opportunities and the challenges of the use of bioprotection, also called biopreservation, in the food system as sustainable strategy to be implemented in meat production. Specifically methodologies to identify bacteriocinogenic strains, the effect of processing conditions on the expression of bacteriocin genes and the efficacy of bacteriocin producing strains to inhibit not only target microorganisms but whole spoilage consortia will be discussed.

Abstract:Background Microbial meat spoilage is responsible for huge losses during production, distribution, and storage of meat products, accounting for approximately 21% of total food losses. A thorough understanding of the characteristics of microorganisms related to meat spoilage is the necessary prerequisite to prevent spoilage processes, and develop innovative preservative approaches and strategies of meat and meat products. Scope and approach This review has discussed the drawbacks of existing definition of dominant spoilage organisms determining the process of meat spoilage, and the microbial sources of contamination along the specific procedures of meat primary and further processing. Moreover, the spoilage phenotypes of meat and the mechanisms of spoilage bacteria have been highlighted in this review, and the future perspectives, being benefit to break through the current barriers of meat spoilage, have also been covered. Key findings and conclusions A re-definition of dominant spoilage organisms related to meat spoilage has been concluded, based on the spoilage strength of microbial isolates in situ of meat. The contamination sources are ascribed to the specific processing procedures of chilled meat and meat products, and the microbial biofilm formed on the contact surfaces. Insight into the spoilage determinants including complete genomes of isolates, intracellular metabolic pathways, and exoenzymes secreted by bacteria, could deeply illustrate the molecular mechanisms of microbial spoilage of meat and meat products during storage.

Abstract:Reducing the initial bacteria number from meat and extending its shelf life are crucial factors for ensuring product safety and enhancing economic benefits for enterprises. Currently, controlling enzyme activity and the microbial survival environment is a common approach to reducing the rate of deterioration in raw meat materials, thereby achieving the goal of bacteria reduction during storage and preservation. This review summarizes the commonly used technologies for reducing bacteria in meat, including slightly acidic electrolyzed water (SAEW), organic acids, ozone (O3), ultrasound, irradiation, ultraviolet (UV), cold plasma, high-pressure processing (HPP), and biological bacterial reduction agents. This review outlines the mechanisms and main features of these technologies for reducing bacteria in meat processing. Additionally, it discusses the status of these technologies in meat storage and preservation applications while analyzing associated problems and proposing solutions. The aim is to provide valuable references for research on meat preservation technology.
Optimization of PCR Assays of Major Foodborne Pathogens and Their Application in Investigation of Pathogen Contamination in Chilled Pork

Abstract:To optimize the PCR identification systems of major foodborne pathogens for investigating into their prevalence in chilled pork,we compared 5 assay formats of Salmonella spp(including S.typhimurium and S.enteritidis) and 6 assay formats of Escherichia coli O157:H7 reported in the literature.With two optimized assay formats,one for S.typhimurium and S.eneritidis and the other for E.coli,and with the PCR method set up in our laboratory for Listeria monocytogenes,we investigated the prevalence of L.monocytogenes,Salmonella spp and E.coli O157:H7 in 50 fresh pork samples and 150 chilled pork samples collected from Hangzhou markets.The results showed that the overall rate of pathogen contamination in the 200 samples was 21%.The incidence in chilled pork samples was higher than that in fresh pork samples(24.7% vs 10%).The incidence of L.monocytogenes was 2% in fresh pork samples and 14.7% in chilled pork samples.The incidence of Salmonella spp in chilled pork samples was also higher than that in fresh pork samples(11.3% vs 8.0%);No strain of E.coli O157:H7 was found.There were 4 strains of S.typhimurium,all from chilled pork samples.This research suggests the potential risk of major foodborne pathogens in chilled pork,probably due to contamination of the production line.

Intensive artificial selection and high energy input into the diet of many commercial animals for the last decade has significantly increased the daily gain of body weight and shortened the raising period, but unexpectedly decreased the meat quality. The gastrointestinal tract of animals harbors a diverse and complex microbial community that plays a vital role in the digestion and absorption of nutrients, immune system development, pathogen exclusion, and meat quality. Fatty acid composition and oxidative stress in adipose and muscle tissue influences meat quality in livestock and poultry. Recent studies showed that nutraceuticals are receiving increased attention, which could alter the intestinal microbiota and regulate the fat deposition and immunity of hosts to improve their meat quality. Understanding the microbiota composition, the functions of key bacteria, and the host-microbiota interaction is crucial for the development of knowledge-based strategies to improve both animal meat quality and host health. This paper reviews the microorganisms that affect the meat quality of livestock and poultry. A greater understanding of microbial changes that accompany beneficial dietary changes will lead to novel strategies to improve livestock and poultry meat product quality.

Abstract:Livestock and poultry breeding environments suffer from serious microbial aerosol pollution, posing a significant challenge to maintaining healthy animal rearing. This study reviewed the sources, pollution status, hazards, pathogenic mechanisms, and mitigation measures of microbial aerosols in livestock and poultry breeding settings, based on research conducted over the past two decades. Notably, the study analyzed the distribution characteristics of aerosol components in various animal houses, with a focus on identifying the main factors affecting these characteristics and the molecular mechanisms by which they damage the animal immune system. Quantitative analysis revealed varying concentrations of bacterial and fungal aerosols in different animal houses, with poultry houses often exhibiting higher concentrations. The dominant bacterial and fungal species varied across different animal houses, emphasizing the complex composition of microbial aerosols. Furthermore, antibiotic-resistant bacteria and genes, particularly those resistant to tetracycline, are prevalent in these environments, challenging disease prevention and control efforts. Thus, the infection source must be controlled through isolation measures and proper waste management. Proper disinfectant use, responsible antibiotic stewardship, biosecurity measures, and alternative disease prevention strategies should be implemented. Future research should focus on developing monitoring technologies for pathogenic microorganisms, implementing purification technologies, and investigating the immune-damaging mechanisms of microbial aerosols. By addressing these areas, we can further understand microbial aerosols in livestock and poultry environments and develop effective strategies to mitigate their harmful effects. This review contributes to the sustainable development of animal farming to ensure the health and welfare of animals.

Abstract:The results of many studies prove the microbiota of the surfaces of the production environment can be a source of food contamination. Environmental monitoring allows to identify problem areas in the enterprise and take corrective actions to eliminate them. This work is conducted to the analysis of the microbiota of abiotic objects selected in the area of close proximity to food products at a pork processing plant by sequencing the 16S RNA gene. The phylum Proteobacteria (from 37.7 to 73.6%), Firmicutes (from 0.14 to 18.6%), Bacteroidota and Actinobacteriota were the dominant components of the microbial communities of the meat processing enterprise. Bacteria of the genus Pseudomonas were found in all samples, the number of readings of these bacteria ranged from 1.90% to 28.76% of the total number of readings. Bacteria of the genus Brochotrix were found in samples from 0.02% to 2.75%. The identification of this phylum indicates the potential presence of pathogenic microorganisms and spoilage microorganisms at production facilities, which can negatively affect the quality and safety of food products.
