Abstract:Seven new quinoxaline derivatives 3a-3g were synthesized through two steps convenient approach. Their structures were confirmed by Fourier transform infrared, proton nuclear magnetic resonance, and high-resolution mass spectrometry. The synthesized compounds 3a-3g showed good fluorescent intensity in dimethyl sulfoxide. They were also shown to possess good antifungal activities. The structure-activity relationship was studied by density functional theory calculations.

Abstract:Natural products are a source of many novel compounds with biological activity for the discovery of new pesticides and pharmaceuticals. Quinoxaline is a fused N-heterocycle in many natural products and synthetic compounds, and seven novel quinoxaline derivatives were designed and synthesized via three steps. Pesticidal activities of title quinoxaline derivatives were bioassayed. Most of these compounds had herbicidal, fungicidal, and insecticidal activities. The compounds 2-(6-methoxy-2-oxo-3-phenylquinoxalin-1(2H)-yl)acetonitrile (3f) and 1-allyl-6-methoxy-3-phenylquinoxalin-2(1H)-one (3g) were the most active herbicides and fungicides. Mode-of-action studies indicated that 3f is a protoprophyrinogen oxidase-inhibiting herbicide. Compound 3f also possessed broad-spectrum fungicidal activity against the plant pathogen Colletotrichum species. Some of these compounds also had insecticidal activity. Molecular docking and DFT analysis can potentially be used to design more active compounds.

Abstract:Flavonoids are a class of natural substances present in plants, fruits, vegetables, wine, bulbs, bark, stems, roots, and tea. Several attempts are being made to isolate such natural products, which are popular for their health benefits. Flavonoids are now seen as an essential component in a number of cosmetic, pharmaceutical, and medicinal formulations. Quercetin is the major polyphenolic flavonoid found in food products, including berries, apples, cauliflower, tea, cabbage, nuts, and onions that have traditionally been treated as anticancer and antiviral, and used for the treatment of allergic, metabolic, and inflammatory disorders, eye and cardiovascular diseases, and arthritis. Pharmacologically, quercetin has been examined against various microorganisms and parasites, including pathogenic bacteria, viruses, and Plasmodium, Babesia, and Theileria parasites. Additionally, it has shown beneficial effects against Alzheimer’s disease (AD), and this activity is due to its inhibitory effect against acetylcholinesterase. It has also been documented to possess antioxidant, antifungal, anti-carcinogenic, hepatoprotective, and cytotoxic activity. Quercetin has been documented to accumulate in the lungs, liver, kidneys, and small intestines, with lower levels seen in the brain, heart, and spleen, and it is extracted through the renal, fecal, and respiratory systems. The current review examines the pharmacokinetics, as well as the toxic and biological activities of quercetin.

Abstract:As a consequence of environmental quality changes as well as changes in our population's lifestyle, there is rapidly increasing variability and many so-called lifestyle disorders, allergies, and food intolerances (also known as non-allergic food hypersensitivity). Unhealthy eating practices, an inappropriate food composition with an excessive energy intake, a high intake of saturated fats, simple sugars, and salt, as well as an inadequate intake of fibre, vitamins, and substances with preventive effects (such as antioxidants), are some of the factors causing this detrimental phenomenon. Enhanced consumption of plant foods rich in valuable secondary metabolites such as phenolic acids and flavonoids with the benefit on human health, food research focused on these components, and production of foods with declared higher content of biologically active and prophylactic substances are some ways how to change and improve this situation. A unique class of hydroxylated phenolic compounds with an aromatic ring structure are called flavonoids. One unique subclass of flavonoids is quercetin. This phytochemical naturally takes place in fruits, vegetables, herbs, and other plants. Quercetin and its several derivates are considered to be promising substances with significant antidiabetic, antibacterial, anti-inflammatory, and antioxidant effects, which could also act preventively against cardiovascular disease, cancer, or Alzheimer's disease.

Abstract:Sulfated quercetin derivatives are important authentic standards for metabolic studies. Quercetin-3'-O-sulfate, quercetin-4'-O-sulfate, and quercetin-3-O-sulfate as well as quercetin-di-O-sulfate mixture (quercetin-7,3'-di-O-sulfate, quercetin-7,4'-di-O-sulfate, and quercetin-3',4'-di-O-sulfate) were synthetized by arylsulfotransferase from Desulfitobacterium hafniense. Purified monosulfates and disulfates were fully characterized using MS and NMR and tested for their 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS⁺) and N,N-dimethyl-p-phenylenediamine (DMPD) radical scavenging, Folin-Ciocalteau reduction (FCR), ferric reducing antioxidant power (FRAP), and anti-lipoperoxidant activities in rat liver microsomes damaged by tert-butylhydroperoxide. Although, as expected, the sulfated metabolites were usually less active than quercetin, they remained still effective antiradical and reducing agents. Quercetin-3'-O-sulfate was more efficient than quercetin-4'-O-sulfate in DPPH and FCR assays. In contrast, quercetin-4'-O-sulfate was the best ferric reductant and lipoperoxidation inhibitor. The capacity to scavenge ABTS+• and DMPD was comparable for all substances, except for disulfates, which were the most efficient. Quantum calculations and molecular dynamics simulations on membrane models supported rationalization of free radical scavenging and lipid peroxidation inhibition. These results clearly showed that individual metabolites of food bioactives can markedly differ in their biological activity. Therefore, a systematic and thorough investigation of all bioavailable metabolites with respect to native compounds is needed when evaluating food health benefits.

Abstract:Polyphenols, widespread in the kingdom of plants, are targeted in numerous chemical, biochemical, and clinical studies. Flavonoids make a particularly interesting group of polyphenols due to their diverse biological activities. In both in vivo and in vitro studies, they strongly demonstrate antioxidant, anti-inflammatory, anticancer, and anti-aggregation activities, and they are also known for tightening the capillary blood vessels. Due to their remarkable biological activities, flavonoids find a wide spectrum of practical applications and the botanical materials rich in flavonoids are omnipresent in traditional medicines, phytotherapy, pharmacotherapy, and cosmetics. Particularly attractive biological properties and, hence, considerable valor of the cosmetics are attributed to such flavonoids, as rutin, quercetin, naringenin, catechin, catechin hydrate, and apigenin. In our study, we selected a simple and cost-friendly thin-layer chromatographic technique to identify the aforementioned flavonoids, and also caffeic, ferulic, gallic, and coumaric acids, in the commercially available cosmetic raw materials (such as extracts derived from red wine, grape skins, pomegranate peels and juice, green tea, etc.) Quercetin and rutin proved to be the two flavonoids most frequently encountered in the examined cosmetic raw materials. The results of such investigations provide the phenolics-oriented evaluation of the quality of the cosmetic raw materials and, hence, better judgment on the applicability thereof, e.g., as components of specific cosmetics prepared to treat various skin problems.

Abstract:Quercetin is a flavonoid with a low molecular weight that belongs to the human diet's phenolic phytochemicals and nonenergy constituents. Quercetin has a potent antioxidant capacity, being able to capture reactive oxygen species (ROS), reactive nitrogen species (RNS), and reactive chlorine species (ROC), which act as reducing agents by chelating transition-metal ions. Its structure has five functional hydroxyl groups, which work as electron donors and are responsible for capturing free radicals. In addition to its antioxidant capacity, different pharmacological properties of quercetin have been described, such as carcinostatic properties; antiviral, antihypertensive, and anti-inflammatory properties; the ability to protect low-density lipoprotein (LDL) oxidation, and the ability to inhibit angiogenesis; these are developed in this review.

Abstract:Quercetin, a bioactive plant flavonoid, is an antioxidant, and as such it exhibits numerous beneficial properties including anti-inflammatory, antiallergic, antibacterial and antiviral activity. It occurs naturally in fruit and vegetables such as apples, blueberries, cranberries, lettuce, and is present in plant waste such as onion peel or grape pomace which constitute good sources of quercetin for technological or pharmaceutical purposes. The presented study focuses on the role of quercetin in prevention and treatment of dermatological diseases analyzing its effect at a molecular level, its signal transduction and metabolism. Presented aspects of quercetin potential for skin treatment include protection against aging and UV radiation, stimulation of wound healing, reduction in melanogenesis, and prevention of skin oxidation. The article discusses quercetin sources (plant waste products included), methods of its medical administration, and perspectives for its further use in dermatology and diet therapy.
Studies on antioxidative activity of some C-glycosylflavones

Abstract:Ten flavonoid C-glycosyl derivatives: orientin (1), isoorientin (2), vitexin (3), isovitexin (4), isovitexin 7,2"-di-O-glucoside (5), isovitexin 7-O-galactoside-2"-O-glucoside (6), two different 6,8-di-C-hexosylapigenins (7, 8), and two different 6-C-hexosyl-8-C-pentosylapigenins (9, 10) have been either produced from flavonoid fractions from Adonis vernalis L. (1, 2) and Crataegus species (3, 4), or isolated from Stellaria media (L.) Vill. (5-10) to study their antioxidative properties. These were found only for two compounds: orientin (1) and isoorientin (2).

Abstract:Quercetin, a naturally occurring bioflavonoid substance widely used in the nutraceutical and food industries, exists in various solid forms that can have different physicochemical properties, thus impacting this compound's performance in various applications. In this work, we will clarify the complex solid-form landscape of this molecule. Two elusive isostructural solvates of quercetin were obtained from ethanol and methanol. The obtained crystals were characterized experimentally, but the crystallographic structure could not be solved due to their high instability. Nevertheless, the desolvated structure resulting from a high-temperature treatment (or prolonged storage at ambient conditions) of both these two labile crystals was characterized and solved via powder X-ray diffraction and solid-state nuclear magnetic resonance (SSNMR). This anhydrous crystal structure was compared with another anhydrous quercetin form obtained in our previous work, indicating that, at least, two different anhydrous polymorphs of quercetin exist. Navigating the solid-form landscape of quercetin is essential to ensure accurate control of the functional properties of food, nutraceutical, or pharmaceutical products containing crystal forms of this substance.

Abstract:Quercetin and its derivatives are important metabolites that belong to the flavonol class of flavonoids. Quercetin and some of the conjugates have been approved by the FDA for human use. They are widely distributed among plants and have various biological activities, such as being anticancer, antiviral, and antioxidant. Hence, the biosynthesis of novel derivatives is an important field of research. Glycosylation and methylation are two important modification strategies that have long been used and have resulted in many novel metabolites that are not present in natural sources. A strategy for modifying quercetin in E. coli by means of glycosylation, for example, involves overexpressing respective glycosyltransferases (GTs) in the host and metabolic engineering for increasing nucleoside diphosphate sugar (NDP sugar). Still others have used microorganisms other than E. coli, such as Streptomyces sp., for the biotransformation process. The overall study of the structural activity relationship has revealed that modification of some residues in quercetin decreased one activity but increased others. This review summarizes all of the information mentioned above.

Abstract:OBJECTIVE:To study the flavonoids of Scutellaria baicalensis and their bioactivities. METHODS:Chromatographic methods were used to separate compounds, spectral methods were used to determine the structures and bioactivities of the flavonoids were screened in several models in vitro. RESULTS:Nine compounds were isolated and identified as baicalein (1), wogonin (2), oroxylin A (3), 5, 7, 2', 6'-tetrahydroxyflavone (4), viscidulin III (5), baicalin (6), wogonoside (7), oroxylin A-7-O-beta-D-glucuronide (8) and chrysin-6-C-alpha-L-arabinopyranosyl-8-C-beta-D-glucopyranoside (9). CONCLUSION:Baicalein had good anti-bacteria activity, and some compounds showed inhibiting activity against IL-1beta converting enzyme. The 13C NMR data of compounds 9 were assigned correctly by 2D nuclear magnetic resonance (NMR).

Abstract:Quercetin is a flavonoid compound widely present in plants and exhibits a variety of biological activities. Research on quercetin has shown its potential for medical application. In this research, we elucidate its antioxidant mechanism and the broad-spectrum antibacterial and antiparasite properties; summarise its potential application in antioncology and cardiovascular protection and anti-immunosuppression treatment; and demonstrate its ability to alleviate the toxicity of mycotoxins. This research is expected to offer some insights and inspirations for the further study of quercetin, its properties, and the scientific basis for its better application in clinical practice.

Some fluorescence properties of dimethylaminochalcone and its novel cyclic analogues

Abstract:This paper demonstrates the basic character (polarity, solubility, colour, absorption and fluorescence quantum yield) of synthetic dimethylaminochalcone (1) and its cyclic analogues measured in toluene, chloroform, dimethylsulfoxide and ethanol, which have been studied by absorption and fluorescence spectroscopy. The biologically active dye 4'-dimethylaminochalcone (1b) and its less flexible analogues 4-dimethylaminoindanone (2b), -tetralone (3b), and -benzosuberone (4b) are lipophilic molecules that displayed the best solubility in toluene and chloroform. The highest fluorescence and quantum yields of compounds 1 and 2 have been obtained in DMSO and chloroform. Quenching effect of fluorescence compounds (1-4) has been studied in the mixture of the most polar organic solvents DMSO and water. In the presence of water, fluorescence of compound 1 has been quenched the best from all studied chalcones and emission maxima of molecules 1-4 have been shifted to the longer wavelengths. Quenching effect of fluorescence by water was in order 1 > 2 > 3 > 4.

Abstract:In this paper we are reporting the results obtained regarding 3-(N-morpholinylcarbodithioate)-2-(4-methoxyphenyl)chroman-4-one, mainly by IR and NMR spectroscopy combined with Density Functional Theory calculations. The structural and vibrational characterization of this synthetic flavanone was carried out by performing quantum mechanical calculations in the ground state and by using several hybrid functionals (B3LYP, PBE1PBE, CAM-B3LYP and ωB97X-D) in conjunction with the same basis set, namely 6-311G(d,p). A complete assignment of all the absorption bands present in the vibrational spectrum has been performed. The theoretical 1H and 13C NMR chemical shifts (with respect to TMS) of both isomers of the investigated flavanone were obtained with four DFT-GIAO functionals, i.e. B3LYP, WP04, WC04 and PBE1PBE, in conjunction with the 6-311++G(d,p) and cc-pVDZ basis sets. The values of the theoretical 1H and 13C NMR chemical shifts have been compared with the experimental ones. The effect of different functionals and basis sets on the accuracy and precision in predicting the1H and 13C NMR chemical shifts was also assessed. The chemical potential and oral bioavailability of the studied compound was explored by computing several quantum molecular descriptors that characterize the electronic interactions between a compound and its biological receptor, such as the dipole moment, the energy of frontier molecular orbitals, the gap energy, the electronegativity, the chemical hardness/softness and the electrophilicity index. The druglikeness of the targeted compound was evaluated by using filters known to be efficient in estimating the potential of a molecule to be a bioavailable oral drug.

Abstract:Quercetin is one of the most common, naturally occurring flavonoids, structurally classified to the flavonol subfamily. This compound, found in many edible and medicinal plants either as a free or glycosidated form, has been scientifically exploited for many years, and one could hardly expect it could be a hero of some additional story. Commonly recognized as an anti-inflammatory agent, quercetin not only limits capillary vessel permeability by inhibiting hyaluronidase but also blocks cyclooxygenases and lipoxygenases. As a typical flavonoid, it is also known for its antioxidant effect, which was confirmed by many in vitro and in vivo studies. Throughout the years, numerous other activities were reported for quercetin, including antidiabetic, anti-proliferative, or anti-viral. Of note, recent data have revealed its potential role as a therapeutic agent for several central nervous system disorders. This review provides an overview of available experimental data on quercetin and its complexes with respect to central nervous system diseases, with a main focus on some aspects that were not discussed previously, such as anti-anxiolytic effects, anti-Huntington’s disease activity, or therapeutic potential in brain cancer. Moreover, quercetin’s protective role in some of these diseases is discussed, especially as an anti-neuroinflammatory agent. Bearing in mind the poor bioavailability of this compound, possible options that would enhance its delivery to the site of action are also presented.

Abstract:Flavonoids are ubiquitous in photosynthesising cells and are commonly found in fruit, vegetables, nuts, seeds, stems, flowers, tea, wine, propolis and honey. For centuries, preparations containing these compounds as the principal physiologically active constituents have been used to treat human diseases. Increasingly, this class of natural products is becoming the subject of anti-infective research, and many groups have isolated and identified the structures of flavonoids possessing antifungal, antiviral and antibacterial activity. Moreover, several groups have demonstrated synergy between active flavonoids as well as between flavonoids and existing chemotherapeutics. Reports of activity in the field of antibacterial flavonoid research are widely conflicting, probably owing to inter- and intra-assay variation in susceptibility testing. However, several high-quality investigations have examined the relationship between flavonoid structure and antibacterial activity and these are in close agreement. In addition, numerous research groups have sought to elucidate the antibacterial mechanisms of action of selected flavonoids. The activity of quercetin, for example, has been at least partially attributed to inhibition of DNA gyrase. It has also been proposed that sophoraflavone G and (-)-epigallocatechin gallate inhibit cytoplasmic membrane function, and that licochalcones A and C inhibit energy metabolism. Other flavonoids whose mechanisms of action have been investigated include robinetin, myricetin, apigenin, rutin, galangin, 2,4,2'-trihydroxy-5'-methylchalcone and lonchocarpol A. These compounds represent novel leads, and future studies may allow the development of a pharmacologically acceptable antimicrobial agent or class of agents.

Abstract:This comprehensive exploration delves into the multifaceted attributes of quercetin, a flavonoid with extensive health-promoting potential. The review navigates through its fundamental properties, encompassing its chemical structure, classification as a flavonoid, and its natural prevalence in various sources. Addressing solubility, stability, and bioavailability challenges, the investigation delves into innovative isolation techniques, including solvent extraction, solid-phase extraction, natural deep eutectic solvents, supercritical fluid extraction, microwave-assisted extraction, column chromatography, and high-performance thin-layer chromatography. Transitioning into pharmacological implications, the study unveils quercetin's roles in anti-inflammatory pathways, antioxidant effects, and immune modulation, reflecting its versatile significance in health management. The review highlights its impact on wound healing processes and its potential to mitigate arthritis, elucidating its holistic contributions. Culminating in an exploration of recent studies, the analysis underscores quercetin's remarkable anti-inflammatory and anti-arthritis activities, reflecting its substantial potential across various ailments. Abstract:The main goal of this systematic review on the flavonol class secondary metabolite quercetin is to evaluate and summarize the existing research on quercetin's potential health benefits, therapeutic properties, and effectiveness in disease prevention and treatment. In addition to evaluating quercetin's potential for drug development with fewer side effects and lower toxicity, this type of review attempts to collect scientific evidence addressing quercetin's roles as an antioxidant, anti-inflammatory, antibacterial, and anticancer agent. In the first part, we analyze various flavonoid compounds, focusing on their chemical structure, classification, and natural sources. We highlight their most recent biological activities as reported in the literature. Among these compounds, we pay special attention to quercetin, detailing its chemical structure, physicochemical properties, and process of biosynthesis in plants. We also present natural sources of quercetin and emphasize its health benefits, such as its antioxidant and anti-inflammatory effects. Additionally, we discuss methods to enhance its bioavailability, analyzing the latest and most effective delivery systems based on quercetin.
