Anomalous concentration dependences observed here may be reconciled by considering the role of phase transfer processes that mediate concentrations in each phase. Abstract:Copper-catalyzed radical transformations establish a powerful toolkit to construct versatile complex organic compounds. The copper-mediated bond formation step of radicals plays a critical role in controlling chemo- and stereoselectivity of copper-catalyzed radical transformation reactions. This bond formation step involves three possible pathways: ion-type bond formation, radical substitution, and reductive elimination. This review highlights the recent advances in theoretical studies on mechanisms and controlling models on the selectivity of Cu-mediated radical-involved bond formation, providing a general mechanistic comprehension of this key elementary step in copper catalysis.

Abstract:Organic radicals consisting of light elements exhibit a low spin-orbit coupling and weak hyperfine interactions with a long spin coherence length, which are crucial for future applications in molecular spintronics. However, the synthesis and characterization of these organic radicals have been a formidable challenge due to their chemical instability arising from unpaired electrons. Here, we report a direct imaging of the surface chemical transformation of an organic monoradical synthesized via the monodehydrogenation of a chemically designed precursor. Bond-resolved scanning tunneling microscopy unambiguously resolves various products formed through a complex structural dissociation and rearrangement of organic monoradicals. Density functional theory calculations reveal detailed reaction pathways from the monoradical to different cyclized products. Our study provides unprecedented insights into complex surface reaction mechanisms of organic radical reactions at the single molecule level, which may guide the design of stable organic radicals for future quantum technology applications.
A radical-polar crossover approach to complex nitrogen heterocycles via the triplet state

Abstract:The transition from radical to ionic reactivity is a key design feature of many photochemical reactions, enabling complex transformations not possible under either mechanistic regime alone. Ground state alkenes are common substrates in existing methods of this type, serving as radical acceptors to generate open-shell intermediates from which the radical–polar crossover (RPC) event is oxidatively or reductively triggered by a photocatalyst. Here, we describe an alternative RPC mechanism proceeding via an alkene triplet diradical. In this transformation, an iodine radical liberated during a homolytic aromatic substitution step functions as a single electron oxidant to generate an iminium electrophile that can be intercepted en route to complex natural product-like amines. An enantioselective variant of the reaction, enabled by an oxidatively installed sulfinyl leaving group, points to the generality of this underdeveloped pattern of diradical reactivity, paving the way to other triplet-state reactions that incorporate both one and two electron bond-forming processes.

Abstract:Radical/radical cross-coupling reactions represent an efficient and straightforward approach for the construction of chemical bonds and accordingly have drawn increasing attention over the past decades. In order to achieve synthetically useful transformations, a persistent radical should be coupled with a transient radical in accordance with the persistent radical effect (PRE). However, known transient radicals outnumber by far the known types of free persistent radicals, which limits the widespread application of the PRE, until today. Thus, the development of efficient cross-coupling reactions between transient radicals has been in focus, and meanwhile transition-metal catalysis has been successfully implemented to artificially prolong radical lifetimes, allowing their utilization in formal radical/radical cross-couplings. Complementary research in the field recently uncovered that organocatalytically generated NHC-derived ketyl radicals are a type of catalytically generated free persistent radicals. NHC-catalyzed radical transformations of aldehydes and carboxylic acid derivatives have enabled the disclosure of an ever-increasing number of interesting reactions, which are different from traditional NHC-catalyzed ionic processes, offering otherwise inaccessible activation modes. These discoveries have opened a door to NHC organocatalysis for the manipulation of radical reactions. Due to its obvious potential in synthetic organic chemistry, it is timely to provide a perspective on this emerging field.

Abstract:Most organic radicals possess short lifetimes and quickly undergo dimerization or oxidation. Here, we report on the synthesis by radical templation of a class of air- and water-stable organic radicals, trapped within a homo[2]catenane composed of two rigid and fixed cyclobis (paraquat-p-phenylene) rings. The highly energetic octacationic homo[2] catenane, which is capable of accepting up to eight electrons, can be configured reversibly, both chemically and electrochemically, between each one of six experimentally accessible redox states (0, 2+, 4+, 6+, 7+, and 8+) from within the total of nine states evaluated by quantum mechanical methods. All six of the observable redox states have been identified by electrochemical techniques, three (4+, 6+, and 7+) have been characterized by x-ray crystallography, four (4+, 6+, 7+, and 8+) by electron paramagnetic resonance spectroscopy, one (7+) by superconducting quantum interference device magnetometry, and one (8+) by nuclear magnetic resonance spectroscopy.

Abstract:One of the most common reactions of diazo compounds with alkenes is cyclopropanation, which occurs through metal carbene or free carbene intermediates. Alternative functionalization of alkenes with diazo compounds is limited, and a methodology for the addition of the elements of Z-CHR2 (with Z = H or heteroatom, and CHR2 originates from N2═CR2) across a carbon–carbon double bond has not been reported. Here we report a novel reaction of diazo compounds utilizing a radical-mediated addition strategy to achieve difunctionalization of diverse alkenes. Diazo compounds are transformed to carbon radicals with a photocatalyst or an iron catalyst through PCET processes. The carbon radical selectively adds to diverse alkenes, delivering new carbon radical species, and then forms products through hydroalkylation by thiol-assisted hydrogen atom transfer (HAT), or forms azidoalkylation products through an iron catalytic cycle. These two processes are highly complementary, proceed under mild reaction conditions, and show high functional group tolerance. Furthermore, both transformations are successfully performed on a gram-scale, and diverse γ-amino esters, γ-amino alcohols, and complex spirolactams are easily prepared with commercially available reagents. Abstract:Radical transformations are powerful in organic synthesis for the construction of molecular scaffolds and introduction of functional groups. In radical difunctionalization reactions, the radicals in the first functionalized intermediates can be relocated through resonance, hydrogen atom or group transfer, and ring opening. The resulting radical intermediates can undertake the following paths for the second functionalization: (1) couple with other radical groups, (2) oxidize to cations and then react with nucleophiles, (3) reduce to anions and then react with electrophiles, (4) couple with metal-complexes. The rearrangements of radicals provide the opportunity for the synthesis of 1,3-, 1,4-, 1,5-, 1,6-, and 1,7-difunctionalization products. Multiple ways to initiate the radical reaction coupling with intermediate radical rearrangements make the radical reactions good for difunctionalization at the remote positions. These reactions offer the advantages of synthetic efficiency, operation simplicity, and product diversity.

Abstract:Radical reactions play an important role in modern organic synthetic chemistry. The generation of carbon radicals from the homolytic cleavage of carbon–metal bonds has been widely studied as a fundamental step in organometallic chemistry. However, the implementation of this phenomenon in catalysed cascade reactions, that is, an organometallic-radical relay, is a highly challenging undertaking due to transient radicals generated from these thermodynamically disfavoured endergonic processes. Here we disclose a set of catalytic alkene 1,1-difunctionalization reactions conceptually based on an organometallic-radical relay. This strategy enables a diversity of sp 3 / sp 2 fragments and aryl groups to be simultaneously introduced to the same carbon of both terminal and internal alkenes with outstanding chemo- and regioselectivity. This study provides a concept for the generation of functionalized benzylic radicals, which are difficult to access by classical protocols, from non-radical feedstock alkenes and arylboronic acids.

Abstract:The radical difunctionalization of unsaturated hydrocarbons serves as an efficient means to rapidly construct molecular skeletons and synthesize‐highly valuable compounds. In this transformational process, diverse positions within unsaturated hydrocarbons are sequentially functionalized by a single radical precursor reagent, thereby achieving highly efficient and selective transformations that promote atom economy. Furthermore, this approach minimizes the generation of numerous byproducts stemming from cross‐coupling reactions between reactants, owing to the reduction of the number of participating components. Therefore, this review provides an in‐depth analysis of the radical difunctionalization of unsaturated hydrocarbons utilizing a single functional reagent in recent decades. The discussion is based on eight different radical classes (carbon‐, nitrogen‐, phosphine‐, oxygen‐, sulfur‐, selenium‐, tellurium‐, and chlorine‐centered radicals), emphasizing the mechanism of specific radical difunctionalization. It also analyzes in detail the regulation of key factors such as regional selectivity, and provides unique insights into reaction transformations.

Abstract:Since Friedrich Wohler's groundbreaking synthesis of urea in 1828, organic synthesis over the past two centuries has predominantly relied on the exploration and utilization of chemical reactions rooted in two-electron heterolytic ionic chemistry. While one-electron homolytic radical chemistry is both rich in fundamental reactivities and attractive with practical advantages, the synthetic application of radical reactions has been long hampered by the formidable challenges associated with the control over reactivity and selectivity of high-energy radical intermediates. To fully harness the untapped potential of radical chemistry for organic synthesis, there is a pressing need to formulate radically different concepts and broadly applicable strategies to address these outstanding issues. In pursuit of this objective, researchers have been actively developing metalloradical catalysis (MRC) as a comprehensive framework to guide the design of general approaches for controlling over reactivity and stereoselectivity of homolytic radical reactions. Essentially, MRC exploits the metal-centered radicals present in open-shell metal complexes as on-electron catalysts for homolytic activation of substrates to generate metal-entangled organic radicals as the key intermediates to govern the reaction pathway and stereochemical course of subsequent catalytic radical processes. Different from the conventional two-electron catalysis by transition metal complexes, MRC operates through one-electron chemistry utilizing stepwise radical mechanisms.

Abstract:Here, we demonstrate that the relationship between reactivity and thermodynamics in radical ligand transfer chemistry can be understood if this chemistry is dissected as concerted ion-electron transfer (cIET). Namely, we investigate radical ligand transfer reactions from the perspective of thermodynamic contributions to the reaction barrier: the diagonal effect of the free energy of the reaction, and the off-diagonal effect resulting from asynchronicity and frustration, which we originally derived from the thermodynamic cycle for concerted proton-electron transfer (cPET). This study on the OH transfer reaction shows that the three-component thermodynamic model goes beyond cPET chemistry, successfully capturing the changes in radical ligand transfer reactivity in a series of model Fe III –OH⋯(diflouro)cyclohexadienyl systems. We also reveal the decisive role of the off-diagonal thermodynamics in determining the reaction mechanism. Two possible OH transfer mechanisms, in which electron transfer is coupled with either OH − and OH + transfer, are associated with two competing thermodynamic cycles. Consequently, the operative mechanism is dictated by the cycle yielding a more favorable off-diagonal effect on the barrier. In line with this thermodynamic link to the mechanism, the transferred OH group in OH − /electron transfer retains its anionic character and slightly changes its volume in going from the reactant to the transition state. In contrast, OH + /electron transfer develops an electron deficiency on OH, which is evidenced by an increase in charge and a simultaneous decrease in volume. In addition, the observations in the study suggest that an OH + /electron transfer reaction can be classified as an adiabatic radical transfer, and the OH − /electron transfer reaction as a less adiabatic ion-coupled electron transfer.

Abstract:Propargylic alcohols are regarded as a class of readily available and versatile synthons in organic synthesis. Due to their unique bifunctional character, the transformation of propargylic alcohols has been widely utilized as a powerful strategy for the construction of various functionalized frameworks over the past few decades. Recently, the radical transformation of propargylic alcohols has attracted considerable attention in synthetic chemistry. This review summarises recent progress in the radical transformation of propargylic alcohols. Based on the different mechanisms of radical formation underlying these transformations, this review has been divided into four parts: (1) transition‐metal catalyzed radical transformation; (2) photo‐induced radical transformation; (3) electroc‐induced radical transformation; and (4) metal‐free or oxidant‐mediated radical transformation of propargylic alcohols.

Supramolecular Free Radicals: Fabrication, Modulation and Functions

Abstract:Modulating the activity of radicals is of great importance for the applications in radical-based materials and radical-mediated reactions. To this end, we have proposed a new concept of "supramolecular free radicals", which refers to the free radicals stabilized or activated through supramolecular approaches. Based on the host-guest chemistry of cucurbiturils (CB), we have fabricated three kinds of supramolecular free radicals to modulate the activity and realize diverse functions. Firstly, radical anions can be stabilized by the steric effect and electrostatic effect of CB. As a result, we have constructed a highly efficient near-infrared photothermal conversion system, which displays selective antibacterial performance. Secondly, owing to the electrostatic effect of CB, radical cations can be activated to induce a significant acceleration of Fenton oxidation reaction. Thirdly, by taking advantage of the dynamic nature of host-guest interactions, we can endow the reaction intermediate with adaptive reactivity, which greatly improves the catalytic efficiency of alcohol oxidation. It is highly anticipated that this series of research opens a new horizon in supramolecular materials and supramolecular catalysis.

Abstract:Based on the strategy of polarity reversal in the generation of free radicals derived from diazo compounds, photocatalyzed multicomponent reactions (MCRs) of nitrogen aromatic heterocycles, alkenes, and diazo compounds form functionalized derivatives in good to high yields and exacting regioselectivities. The carbon radicals generated from the acceptor diazo compounds are electrophilic, and their selective additions with alkenes provide nucleophilic radicals, which enable the further rapid assembly with various heteroarenes. A delicate balance has been achieved between the activation of heteroarenes through protonation and the decomposition of diazo compounds by the same acid. This multicomponent Minisci reaction shows high functional group tolerance, especially in the incorporation of biologically active molecules. Detailed mechanistic studies that include photophysical measurements elaborate this radical cascade reaction. Furthermore, this transformation provides new opportunities for versatile reactions of diazo compounds in radical cascade multicomponent coupling reactions.

Abstract:Abstract Oxygen-centered radicals (R1O•) are reactive intermediates in organic synthesis, with versatile synthetic utilities in processes such as hydrogen-atom transfer (HAT), β-fragmentation, radical addition to unsaturated carbon–carbon bonds, and rearrangement reactions. In this review, we focus on recent advances in the generation and transformation of oxygen-centered radicals, including (alkyl-, α-oxo-, aryl-) carboxyl, alkoxyl, aminoxyl, phenoxyl, and vinyloxyl radicals, and compare the reactivity of oxygen-centered radicals under traditional reaction conditions with their reactivity under visible-light-induced reaction conditions.
