Abstract:
Background

Major depressive disorder (MDD) has been shown to be related to immune inflammation and the complement system. After the administration of hUC-MSCs, the depression-like and anxiety-like phenotypes, neuronal histopathology, synaptic-related protein expression and inflammatory index of the mice were assessed. Microglial M1/M2 polarization and the expression of C3a in astrocytes and C3aR in microglia was detected by immunofluorescence co-localization. Then, CUMS mice were injected with a C3aR antagonist, and the expression of C3a and C3aR and microglial polarization were observed.

Results

Based on the sucrose preference and tail suspension tests, hUC-MSCs ameliorated the depression-like behaviors of CUMS mice. Additionally, the anxiety-like behaviors of CUMS mice in the open-field and plus-maze tests were improved after the administration of hUC-MSCs. hUC-MSCs altered microglia polarization by alleviating complement C3a-C3aR signaling activation, which decreased pro-inflammatory factor levels and increased anti-inflammatory factor levels, alleviating neuronal damage and synaptic deficits.

Conclusion

hUC-MSCs have therapeutic effects on anxiety-like and depressive-like phenotypes caused by CUMS. They can alter the polarization of microglia by inhibiting C3a-C3aR signaling to reduce neuroinflammation.

What problem does this paper attempt to address?
The main problem that this paper attempts to solve is to explore how human umbilical cord mesenchymal stem cells (hUC - MSCs) can improve chronic unpredictable mild stress (CUMS) - induced depressive symptoms by regulating microglial cell polarization mediated by complement C3 signaling. Specifically, the research focuses on the following points: 1. **The relationship between depression and immune inflammation**: The paper points out that major depressive disorder (MDD) is related to abnormal immune - inflammatory pathways, especially the role of the complement system therein. Researchers hope to verify through experiments whether hUC - MSCs can treat depression by regulating neuroinflammation. 2. **Regulation of microglial cell polarization**: Microglial cells are the main immune effector cells in the central nervous system and play a key role in the stress response. When activated, microglial cells will change from the resting branched morphology to the phagocytic amoeboid morphology and can be polarized into M1 or M2 types. M1 - type microglial cells release pro - inflammatory mediators, while M2 - type microglial cells produce anti - inflammatory factors. The paper aims to explore how hUC - MSCs affect the polarization process of microglial cells. 3. **The interaction between astrocytes and microglial cells**: The interaction between astrocytes and microglial cells has an important impact on neuroinflammation. Researchers observed through experiments how hUC - MSCs affect the polarization of microglial cells by regulating the C3a protein secreted by astrocytes and the expression of its receptor C3aR on microglial cells. 4. **The role of the C3a - C3aR signaling pathway**: The paper pays special attention to the role of the C3a - C3aR signaling pathway in the pathogenesis of depression. By administering a C3aR antagonist, researchers further verified the importance of the C3a - C3aR signaling pathway in hUC - MSCs improving CUMS - induced depressive symptoms. In summary, the core problem of this paper is to verify through experiments whether hUC - MSCs can change the polarization state of microglial cells by regulating the C3a - C3aR signaling pathway, thereby alleviating CUMS - induced depressive symptoms. This research not only helps to understand the pathogenesis of depression, but also may provide a theoretical basis for the development of new treatment methods.
