Abstract:Karst tiankengs, as one of the most magnificent negative topographies, are capable of forming a bank for species diversity conservation easily. More than 300 karst tiankengs have been discovered and identified worldwide. Given its treacherous terrain, although original karst tiankeng were identified as species refuges, the broader distribution of degraded karst tiankeng has not been systematically studied. Our study area comprised the degraded karst tiankeng cluster immersed in the fragmented karst forests of Yunnan, China. Fifty-eight plant samples were selected from karst tiankengs and surface. We compared species composition, and analyzed diversity indices and similarity coefficients to verify the isolation effect of karst tiankengs on floras. The results indicated that: (1) In the degraded karst tiankeng, there were 24 families, 37 genera and 48 species in the tree layer and 27 families, 43 genera and 49 species in the shrub layer. Outside the degraded karst tiankengs, 20 families, 31 genera and 39 species were in the tree layer, and the shrub layer included 26 families, 44 genera and 55 species. (2) The species composition reached significant differences within and outside degraded karst tiankeng ( p < 0.05) based on the analysis of variance (ANOVA). (3) In the degraded karst tiankeng, species richness/diversity in trees were higher than those in the shrub layer, while at the surface, shrubs had higher richness and lower diversity than trees by Alpha-diversity index. And for Beta-diversity index, species similarity among degraded karst tiankengs (0.215) was extremely dissimilar, which was even lower than the contrast within and outside the degraded karst tiankengs (0.272). (4) Shared species ranged from 1 to 5 among the four habitats, with high variability in plant species across the habitat matrices. Through a comparative analysis of systematic biodiversity methods, we found that the degraded karst tiankengs, an independent type of karst tiankeng, are the unreported refugia. Species records in degraded karst tiankeng cluster will contribute to plant diversity conservation and resource management, and to the linkage with broader China's karst floras. Karst tiankeng botanical habitats possess not only biodiversity value for in situ conservation, but will further support the ecological recovery of surface flora. While its mechanism needs to be further revealed.

Abstract:The karst tiankeng is a special and grand negative terrain on the surface, that maintains a unique ecosystem. However, knowledge about bacterial and fungal communities in karst tiankengs is still limited. Therefore, soil samples from five karst tiankengs were collected and subjected to high-throughput sequencing of 16S rRNA and ITS genes, and multivariate statistical analysis. The results showed abundant and diversified bacterial and fungal communities in karst tiankeng. The bacterial communities were dominated by Proteobacteria and Acidobacteria , and the fungal communities were dominated by Ascomycota and Basidiomycota . Statistical analysis revealed significant differences in bacterial and fungal communities among the five karst tiankengs, which may indicate that the distribution of bacterial and fungal communities was driven by separate karst tiankengs. The co-occurrence network structure was characterized by highly modularized assembly patterns and more positive interactions. The keystone taxa were mainly involved in nutrient cycling and energy metabolism. The null model analysis results showed that the stochastic process, especially dispersal limitation, tended to be more important in controlling the development of bacterial and fungal communities in karst tiankeng. The bacterial community structure was significantly associated with soil properties (SWC, TN, AN, and BD), while the fungal community structure was significantly associated with soil properties (SWC and TP) and plant diversity. These results can expand our knowledge of the karst tiankeng microbiome.

Abstract:Karst tiankeng are important biodiversity conservation reservoirs. However, the unique habitats of karst tiankeng affect microbial community structure remained poorly understood. In this study, we collected soil samples from karst tiankeng (TK) and karst land (KL) and subjected to high-throughput sequencing. Based on the classification of the total, abundance, and rare taxa for bacteria and fungi, a multivariate statistical analysis was carried out. The results revealed that bacterial community Shannon diversity and Pielou's evenness were highest in TK. The rare taxa were ubiquitous in all soil samples, while the higher Shannon diversity of the abundant taxa of TK may be related to the habitat preferences of species and niche differentiation. The community composition of bacterial and fungal sub-communities exhibited significant dissimilarity between TK and KL. The redundancy analysis further demonstrated that abundant taxa were environmentally more constrained than rare taxa. The bacterial and fungal networks of KL were more complex than TK. The keystones of the network transforms may suggest their significant role in the ecological function of the karst tiankeng ecosystem. This study represents the first reports of the characteristics of bacterial and fungal communities in karst tiankeng.

Abstract:Two new Cordyceps -like species, Perennicordyceps zongqii and Purpureocillium zongqii , isolated from a larva and soil, are introduced. Morphological comparisons and phylogenetic analyses based on multigene datasets ( ITS , LSU , RPB2 and TEF ) support the establishment of the new species. Moreover, new species in the families Polycephalomycetaceae and Ophiocordycipitaceae were introduced into Tiankeng and the valley for the first time. Further attention needs to be paid to the diversity of other Cordyceps -like fungi in the special eco-environment of the karst region.

Abstract:An investigation of Trichoderma biodiversity involving a large-scale environmental gradient was conducted to understand the Trichoderma distribution in China. A total of 3,999 isolates were isolated from forestry, grassland, wetland and agriculture ecosystems, and 50 species were identified based on morphological characteristics and sequence analysis of genetic markers. Trichoderma harzianum showed the largest proportion of isolates and the most extensive distribution. Hypocrea semiorbis, T. epimyces, T. konilangbra, T. piluliferum, T. pleurotum, T. pubescens, T. strictipilis, T. hunua, T. oblongisporum and an unidentified species, Trichoderma sp. MA 3642, were first reported in China. Most Trichoderma species were distributed in Jilin and Heilongjiang Provinces in northeast China and the fewest were distributed in Qinghai Province. Based on the division of ecological and geographic factors, forestry ecosystems and low-altitude regions have the greatest species biodiversity of Trichoderma.

Abstract:Yunnan is exceedingly rich in higher fungi (Ascomycota and Basidiomycota). Given that the number of fungi (including lichens) occurring in a given area is, as Hawksworth suggested, roughly six times that of local vascular plants, a total of approximately 104,000 fungal species would be expected in Yunnan. However, to date only about 6000 fungal species, including roughly 3000 species of higher fungi, have been reported from the province. Although studies on Yunnan's fungi started in the late nineteenth century, significant progress has been made only in the last forty-five years. Over the first twenty-five years of this period, studies on fungal diversity in this area have largely been about taxonomy based on morphological characters and partially on geographical distribution. Over the past twenty years, the combination of both morphological and molecular phylogenetic approaches has become the preferred method to help understand the diversity and evolution of higher fungi. This review focuses on our current knowledge of how geological, geographical, and ecological factors may have contributed to the diversity patterns of higher fungi in Yunnan. Based on this knowledge, three aspects for future studies are suggested.

Abstract:Karst tiankengs are rare natural habitats, having a local microclimate different from surrounding regions. A contrast study on plant communities at the inside and outside of the primitive tiankeng was carried out by performing the qualitative analysis of species compositions of arborous and shrub layers. We found that plant communities in the primitive tiankeng belong to the subtropical moist evergreen broad-leaf forest, whereas those outside the tiankeng belong to subtropical semi-moist needle-broadleaved mixed forest. Trapped habitat of primitive karst tiankeng protects the plant communities significantly different from those in external karst ecosystems, so karst tiankeng has the great value in the plant species protection. Although the trapped habitat decreases plant species abundance inside tiankeng to some extent, highly diverse shrub species are present in the inside-tiankeng plant communities, and the primitive karst tiankeng plays an important role in the plant diversity protection. The primitive karst tiankeng is an important refugia for plant not only as a plant species protection library, but a plant diversity protection library. When implementing measures for the reduction of damages to biodiversity due to global climate changes and human activities, more attention should be paid to the primitive karst tiankeng as a small ecological refugia and biodiversity protection library.

Unique Habitat of Karst Tiankengs Changes the Taxonomy and Potential Metabolism of Soil Microbial Communities

Abstract:Microbial communities in karst ecosystems have been extensively studied. However, in a class of deep-lying habitats with unique climates (karst tiankeng), the structure and ecological functions of microorganisms receive little attention, which is essential for understanding the biogeochemistry of karst tiankeng. Herein, microorganisms from inside (ITK) and outside (OTK) karst tiankengs were analyzed by high-throughput sequencing and multivariate statistical analysis. The results showed that the structure and function of soil bacterial communities inside and outside karst tiankengs were significantly different. The ITK microbial communities presented significantly higher Shannon diversity due to the abundant nutrients in karst tiankeng soil. Random molecular ecological network analysis revealed that the ITK network was simpler and more vulnerable and may be susceptible to environmental changes. More positive links within the network indicate that microorganisms adapt to the karst tiankeng through synergies. The keystones in karst tiankeng were mainly involved in the decomposition of soil organic matter and carbon/nitrogen cycles. Although soil total phosphorus and available potassium regulate microbial community structure variation, dispersal limitation is the predominant ecological process within the microbial community in karst tiankeng. In addition, the functional profiles of the microbial communities reveal that some human diseases (such as infectious diseases) exist in OTK. Collectively, these findings have enhanced our understanding of microbial interactions, ecological functions, and community composition processes in karst tiankeng ecosystems. IMPORTANCE Constrained by the trapped terrain, a unique ecosystem has formed in karst tiankeng. Soil microorganisms are essential for the formation and maintenance of ecosystems, but soil microbial ecology research in karst tiankeng is still lacking. In this study, representative habitats inside and outside karst tiankeng were selected to study the taxonomy and potential metabolism of soil microbial communities. The results show that the unique habitat of karst tiankeng reshapes the composition, structure, and function of soil microbial communities. Our results contribute to enhancing our understanding of sustainable recovery strategies in fragile ecosystems and understanding the biodiversity value of karst tiankeng under climate change.

Abstract:Abstract Background Karst tiankengs serve as a reservoir of biodiversity in the degraded karst landscape areas. However, the microbial diversity of karst tiankengs is poorly understood. Here, we investigated the composition and function of the microbial community in a karst tiankeng. Results We found that habitat differences inside and outside the karst tiankeng changed the composition and structure of the soil microbial communities, and the dominant phyla were Proteobacteria , Actinobacteria , Chloroflexi and Acidobacteria. The Shannon–Wiener diversity of microbial communities inside and outside the tiankeng was significantly different, and it was higher inside the tiankeng (IT). Venn and LEfSe analysis found that the soil microbial communities inside the tiankeng had 640 more endemic species and 39 more biomarker microbial clades than those identified outside of the tiankeng (OT)..Functional prediction indicated that soil microorganisms in outside the tiankeng had a high potential for carbohydrate metabolism, translation and amino acid metabolism. There were biomarker pathways associated with several of human diseases at both IT and OT sites. Except for auxiliary activities (AA), other CAZy classes had higher abundance at IT sites, which can readily convert litter and fix carbon and nitrogen, thereby supporting the development of underground forests. The differences in microbial communities were mainly related to the soil water content and soil total nitrogen. Conclusions Our results provide a metagenomic overview of the karst tiankeng system and provide new insights into habitat conservation and biodiversity restoration in the area.

Abstract:Towards assessing the biodiversity and biogeography of Trichoderma, we have analyzed the occurrence of Trichoderma species in soil and litter from four areas in China: North (Hebei province), South-East (Zhejiang province), West (Himalayan, Tibet) and South-West (Yunnan province). One hundred and thirty five isolates were grouped according to tentative morphological identification. A representative 64 isolates were verified at the species level by the oligonucleotide barcode program TrichO Key v.1.0 and the custom BLAST server TrichoBLAST, using sequences of the ITS1 and 2 region of the rRNA cluster and from the longest intron of the tef1 (translation elongation factor 1-α) gene. Eleven known species (Trichoderma asperellum, T. koningii, T. atroviride, T. viride, T. velutinum, T. cerinum, T. virens, T. harzianum, T. sinensis, T. citrinoviride, T. longibrachiatum) and two putative new species (T. sp. C1, and T. sp. C2), distinguished from known species both by morphological characters and phylogenetic analysis, were identified. A significant difference in the occurrence of these species was found between the North (Hebei) and South-West (Yunnan) areas, which correlates with previously reported species distributions in Siberia and South-East Asia, respectively. As in previous studies, T. harzianum accounted for almost half of the biodiversity; although, in this study, it was exclusively found in the North, and was predominantly represented by an ITS1 and 2 haplotype, which has so far been rarely found elsewhere. This study therefore reveals a North–South gradient in species distribution in East Asia, and identifies Northern China as a potential center of origin of a unique haplotype of T. harzianum.
Diversity and Co-Occurrence Patterns of Soil Bacterial and Fungal Communities of Chinese Cordyceps Habitats at Shergyla Mountain, Tibet: Implications for the Occurrence

Abstract:Chinese Cordyceps is a well-known medicinal larva-fungus symbiote distributed in the Qinghai-Tibetan Plateau and adjacent areas. Previous studies have involved its artificial cultivation but commercial cultivation is difficult to perform because the crucial factors triggering the occurrence of Chinese Cordyceps are not quite clear. The occurrence of Chinese Cordyceps is greatly affected by the soil environment, including the soil’s physicochemical and microecological properties. In this study, the effects of these soil properties on the occurrence of Chinese Cordyceps were investigated. The results show that the physicochemical properties, including easily oxidizable organic carbon (EOC), soil organic carbon (SOC), humic acid carbon (HAC), humin carbon (HMC), and pH, might be negatively related to the occurrence of Chinese Cordyceps, and soil water content (SWC) might be positively related. Several soil physicochemical parameters (pH, SOC, HMC, HAC, available potassium (APO), available phosphorus (APH), microbial biomass carbon (MBC), and the ratio of NH4+ to NO3- (NH4+/NO3-)) and microbial properties interact and mix together, which might affect the occurrence of Chinese Cordyceps. Soil microbial community structure was also a possible factor, and a low level of bacterial and fungal diversity was suitable for the occurrence of Chinese Cordyceps. The intra-kingdom network revealed that a closer correlation of the bacterial community might help the occurrence of Chinese Cordyceps, while a closer correlation of the fungal community might suppress it. The inter-kingdom network revealed that the occurrence rate of Chinese Cordyceps might be negatively correlated with the stability of the correlation state of the soil habitat. In conclusion, this study shows that soil physicochemical properties and microbial communities could be greatly related with the occurrence of Chinese Cordyceps. In addition, soil physicochemical properties, the level of bacterial and fungal diversity, and correlations of bacterial and fungal communities should be controlled to a certain level to increase the production of Chinese Cordyceps in artificial cultivation.

Abstract:Karst tiankengs are oases in degraded karst landscapes and act as repositories for biodiversity conservation; however, knowledge about the bacterial and fungal structure and function of the karst tiankeng ecosystems is limited. This study investigated the microbial communities in three different tiankeng (nondegraded, moderately degraded, and heavily degraded tiankeng) by Illumina NovaSeq sequencing. We found that the degradation of karst tiankeng can lead to changes in microbial community structure and functions, while there are differences in bacterial and fungal responses. There were significant differences in bacterial and fungal community composition and beta diversity in the three tiankeng soils. Random molecular ecological network analysis results indicated that a more complex and stable bacterial network existed in nondegraded tiankeng, while more complex fungal networks existed in moderately degraded tiankeng. The keystones of Proteobacteria, Actinobacteria, Acidobacteria, Ascomycota, and Basidiomycota played essential roles in maintaining soil function and stability. The functional profiles revealed that tiankeng habitat changes may affect microbial survival strategies, such as increasing gene abundance associated with the carbon cycle. To our knowledge, this is the first report on bacterial and fungal communities in different degrees of karst tiankeng, which provides crucial insights into our understanding of the microbial communities' structure and potential function in karst tiankeng ecosystems.
Comparison of bacterial and fungal diversity and network connectivity in karst and non-karst forests in southwest China

Abstract:Microbial communities contribute to sustaining the function of terrestrial ecosystems and are influenced by soil type and climate gradients. The effects of karst and non-karst soils on bacterial and fungal profiles for seven climate gradients were assessed to better understand bacterial and fungal diversity and community composition in response to soil type with changes in soil physicochemical properties under different temperatures and precipitations. Bacterial and fungal abundance, diversity, and community composition differed between karst and non-karst forests. Bacterial and fungal richness, Shannon index, and bacterial abundance in karst forests were higher than non-karst forests, but the fungal abundance was lower. Mean annual temperature was negatively correlated with bacterial diversity in the karst forest and fungal abundance in karst and non-karst forests. The community composition of bacteria and fungi differed among these two soil types. The karst forest had greater connectivity among bacterial and fungal communities than non-karst forests. The bacterial members of Acidobacteria, Proteobacteria, Actinobacteria, and fungal groups of Ascomycota and Basidiomycota were mainly connected with other taxa in the network, implying that taxa were associated with highly functional potential. The relative abundance of Actinobacteria and Ascomycota was higher in karst than in non-karst forests. Proteobacteria and Basidiomycota showed the opposite results. A random forest and multiple regression tree analyses revealed that soil properties, specifically pH, calcium, and total nitrogen, were the main factors influencing the variation in bacterial and fungal profiles between karst and non-karst forests. This study provides novel evidence that the abundant microbial taxa were kinless hubs in co-occurrence patterns. Controlling complex networks of species interactions may contribute to improving soil nutrient processes rather than microbial diversity, enhancing our understanding of developing sustainable recovery strategies in fragile ecosystems.

Variations of the biodiversity and carbon functions of karst forests in two morphologically different sites in southwestern China

Abstract:Abstract Knowledge of the biodiversity and carbon (C) functions of karst forests is scarce. This study comprehensively compared the species diversity and floristic characteristics, biomass and its allocation, leaf and soil C and nitrogen (N) concentrations, and photosynthetic capacity of dominant tree species between peak clump depression (PCD)-type and plateau surface (PS)-type karst forests on the basis of two large plots (i.e., 1 and 2 ha, respectively) in southwestern China. Results showed that PCD-type karst forest exhibits higher biodiversity and more tropical family and genus types than PS-type karst forest. These two types of karst forest presented similar total biomass, but PCD-type karst forest allocated more biomass to supporting roots and less biomass to absorbing roots. PS-type karst forest had higher C/N ratios in leaves and soils than PCD-type karst forest. Deciduous tree species in PS-type karst forest had low net photosynthetic rates, resulting in lower net photosynthetic rate in PS-type karst forest than in PCD-type karst forest. Species richness and C storage in the karst morphologies would be considerably enhanced if degraded vegetation in different types of karst area could be successfully restored to forests according to respective morphological and vegetation features. A comprehensive understanding of the biodiversity and C functions of karst vegetation is essential to biodiversity conservation, regional C storage estimation, vegetation management and restoration, and potential global change mitigation.

Through fungi culture, morphological characterization, and ITS sequence analysis, we identified a total of 565 isolates belonging to 405 species across 133 genera. The diversity indices viz. H′, D, and J were evaluated for fungal diversity across the lakes. Interestingly, although diversity indices were highest during summer and at Yangzonghai Lake, no significant differences in fungal community diversity were observed between seasons and regions. Water variables were analyzed and indicated that changes with rapid fluctuations in temperature, pH, and dissolved oxygen likely influence fungal diversity. These findings significantly contribute to our understanding of fungal communities within plateau lake ecosystems, thereby aiding in managing and conserving vital aquatic resources.

Two Novel Species of Kirschsteiniothelia from Sichuan Province, China

Abstract:Sichuan Province is recognized as one of the richest biodiversity hotspots of plants and animals in the world. However, fungal diversity in this region has been largely overlooked, prompting us to investigate it further. During a long-term survey of fungal diversity, two morphologically similar hyphomycetous fungi were collected. Multi-locus phylogenetic analysis and morphological examinations confirmed that these two collections are new to the genus Kirschsteiniothelia, designated as K. pini sp. nov. and K. sichuanensis sp. nov. Both species showed significant differences in sequence identities of the ITS barcode. Additionally, Kirschsteiniothelia sichuanensis, collected from decaying branches of an unidentified woody plant, differs from the phylogenetically close species K. extensum by having shorter conidia. Kirschsteiniothelia pini, collected from decaying branches of Pinus sp., differs from phylogenetically close species K. aquatica by having shorter and wider conidia and from K. cangshanensis by having shorter conidiophores. The study provided detailed descriptions, photographs of the new species, and an updated phylogeny for Kirschsteiniothelia.
Diversity and Composition of Fungicolous Fungi Residing in Macrofungi from the Qinling Mountains

Abstract:Sporocarps of macrofungi support other diverse fungal species that are termed fungicolous fungi. However, the external environmental factors that affect the diversity and composition of fungicolous fungal communities remains largely unknown. In this study, the diversities, composition, and trophic modes of fungicolous fungal communities residing in host macrofungi from diverse habitats in the Qinling Mountains were analyzed. Additionally, the number of carbohydrate-active enzymes (CAZymes) encoded by saprophytic, pathogenic, and symbiotic fungi was also quantified and compared. The results revealed that the diversity and composition of fungicolous fungal communities varied with months of collection and the habitats of host fungi, and saprophytic fungi were more abundant on wood than on the ground. Meanwhile, it was also found that saprophytic fungi possessed higher abundances of cell-wall-degrading enzymes than pathogenic or symbiotic fungi. Based on the above findings, it was hypothesized that the greater abundance of saprophytic fungi on wood compared to the ground may be due to their possession of a more diverse array of enzymes capable of degrading wood cell walls, thereby allowing for more efficient nutrient acquisition from decaying wood.

Abstract:A single specimen of Cordyceps,collected from Huaping Nature Reserve in Guilin of Guangxi Zhuang Autonomous Region,was identified as Cordyceps cardinalis,a new species record in China.The anamorph of C.cardinalis belongs to the genus Isaria and the fungal characteristics were described first in China as well.Meanwhile,Paecilomyces loushanensis was recombined to Isaris loushanensis.

Abstract:Macrofungi are essential in forest ecological functioning. Their distribution and diversity are primarily impacted by vegetation, topography, and environmental factors, such as precipitation and temperature. However, the composition and topographical changes of the macrofungi between the eastern mountainous area and central plains of Jilin Province are currently unknown. For this study, we selected six investigational sites representing three different topographical research sites in Jilin Province to assess macrofungal diversity, and applied a quadrat sampling method. Macro- and micro-morphological characteristics combined with the molecular method were used to identify the collected macrofungi. Meanwhile, selected meteorological data were obtained for statistical analysis. As a result, 691 species were identified, of which Agarics were the most common, accounting for 60.23%, while the Cantharelloid fungi were the least common (0.91%). Furthermore, most of the shared genera (species) were saprophytic. The α diversity showed that the species diversity and richness in Longwan National Forest Park (B2) were the highest at the genus level. The mycorrhizal macrofungi proportion revealed that Quanshuidong Forest Farm (A1) was the healthiest. Finally, species composition similarity decreased with the transition from mountainous to hilly plains. We concluded that the occurrence of macrofungi was most influenced by vegetation. The air humidity, precipitation, and wind velocity were also found to significantly impact the occurrence of macrofungi. Finally, the mycorrhizal:saprophytic ratios and species similarity decreased with the transition from the mountainous area to the plains. The results presented here help elucidate the macrofungi composition and their relationship with environmental factors and topography in Jilin Province, which is crucial for sustainable utilization and future conservation.
