Abstract:This work revealed the importance of microbial succession to microbiota structure and metabolites. Multi-inoculations would promote deterministic assembly. It would facilitate the regulation of microbiota structure and metabolic profile. In addition, we established a model to predict final metabolites based on microbial genera related to microbial succession. This model was beneficial for optimizing the inoculation of the microbiota. This work would be helpful for controlling the spontaneous food fermentation and directionally improving the food quality.

What problem does this paper attempt to address?
The main problem that this paper attempts to solve is to control and predict the succession of microbial communities and their final metabolites in the fermentation process of Chinese Baijiu through microbial inoculation. Specifically, the research focuses on the following points: 1. **Influence of microbial inoculation on microbial community structure**: Researchers explored the influence of inoculating different microbial communities at different time points on the microbial community structure during Daqu fermentation. The results showed that both the inoculation time and the type of inoculated microbial community significantly affected the final microbial community structure. 2. **Influence of microbial community succession on final metabolites**: Researchers analyzed how the succession of microbial communities affects the final metabolites of Daqu fermentation. The results indicated that the succession mode of microbial communities (stochastic assembly vs. deterministic assembly) significantly affected the final metabolites, especially the deterministic assembly process had a more obvious impact on metabolites. 3. **Establishment of prediction models**: Based on the relative abundances of 27 microbial genera related to microbial community succession, researchers constructed an elastic - network model to predict the relative abundances of 121 metabolites. This model was verified by Procrustes analysis and showed high prediction accuracy (average Spearman correlation coefficient > 0.3). ### Key Findings - **Influence of microbial inoculation**: - The inoculation time and the type of inoculated microbial community significantly affected the microbial community structure during Daqu fermentation. - Multiple inoculations can promote deterministic assembly, thereby better controlling the succession of microbial communities. - **Relationship between microbial community succession and metabolites**: - Deterministic assembly is significantly correlated with the abundances of multiple metabolites, including alcohols, aldehydes and ketones, esters, organic acids, etc. - Stochastic assembly is only correlated with the Shannon index of metabolites and the abundances of phenolic compounds. - **Accuracy of prediction models**: - Based on 27 microbial genera related to microbial community succession, the constructed elastic - network model can accurately predict the relative abundances of 121 metabolites. - The verification results of the model on independent samples showed that 76.86% of metabolites were accurately predicted. ### Conclusion This research reveals the influence of microbial inoculation on the succession of microbial communities and final metabolites during Daqu fermentation, and provides a new strategy for optimizing the quality of food fermentation through directed microbial inoculation. This not only helps to control the spontaneous food fermentation process, but also provides a new method for improving the quality of fermented foods.
