Abstract:A new Schiff-base fluorescence probe CS containing benzimidazole and coumarin fluorophores was synthesized and characterized by NMR, IR and ESI-MS. Fluorescence and absorption spectra show the unique changes in the presence of Hg2+ and Cu2+. Hg2+ induces a remarkable fluorescence enhancement and results in mercury-promoted hydrolysis of a mercury-CS complex to convert the imine group to the aldehyde group. Cu2+ leads to a colorimetric change from colorless to pale yellow and causes formation of a copper-CS complex. Thus, CS can act as a fluorescent chemodosimeter for Hg2+ and a naked-eye chemosensor Cu2+. The cell imaging experiments for Hg2+ show a slow enhancement of a bright blue fluorescence with increasing incubation time, while Cu2+ instantly quenches the fluorescence and then the fluorescence intensity has no further change by extending the incubation time. Therefore, the probe CS shows its potential application for simultaneous differential detection of Hg2+ and Cu2+ in living cells based on the fluorescence response time. (C) 2016 Elsevier B.V. Abstract:Cu（Ⅱ） detection is important because it plays crucial role in several biological processes and ecological systems.Fluorescent techniques have attracted more and more attention in Cu（Ⅱ） detection.In this report,we contribute a novel strategy to use fluorescence spectroscopy for Cu（Ⅱ） specific detection.The specificity relies on the fact that,of the many metal cations,only Cu（Ⅱ） can catalyze the hydrolyzation of a-amino acid ester.The novelty originates from the unique aggregation-induced emission（AIE） property of the fluorescent label.We designed a model a-amino acid ester（TPE-Ala） constructed with alanine and tetraphenylethene-functionalized methanol（TPE-methanol）.In comparison with the precursor TPE-Ala, TPE-methanol has lower solubility and is easy to form aggregates in water,thereby displaying a higher fluorescent response.Thus,the Cu（Ⅱ） catalyzed hydrolyzation can be monitored by recording the fluorescence enhancement and fluorescent detection Cu（Ⅱ） is rationally achieved.

Abstract:In this work, an AIE-active tetraphenylethene-based Schiff base fluorescent probe 3 with a large Stokes shift (247 nm) was designed and synthesized. It was found that the aggregated probe 3 exhibited very high selectivity and anti-interference ability for Cu2+ in PBS buffer (70% fw) through a fluorescence "turn-off" strategy. Job's plot and NMR analysis indicated the two phenolic hydroxyl groups of the benzene ring and the N atom (-CH=N-) on probe 3 interacted with Cu2+ ions in a 1:1 stoichiometric ratio. A comprehensive analysis of the Stern-Volmer and binding constant indicated a rather strong interaction between probe 3 and Cu2+ ions. Probe 3 illustrated excellent sensitivity toward Cu2+ under ppb level (4.5 nM) and achieved more than 95% recovery in river, lake and tap water toward estimation of Cu2+ ions in the analytical applications. Moreover, probe 3 was able to realize bioimaging of HepG2 cells and be quenched by intracellular Cu2+ ions, making it promising as a sensitive Cu2+ sensor for organisms.

Abstract:A novel activity-based Cu2+ fluorescent probe featuring multidentate binding sites was synthesized. It functions through chelation with Cu2+, which in turn specifically triggers hydrolysis of the probe to release a near-infrared emission with AIE + ESIPT properties. The probe was found to be capable of ratiometric imaging of Cu2+ in living HeLa cells.

Abstract:Detection of Cu(II) is very important in disease diagnose, biological system detection and environmental monitoring. Previously, we found that the product TPE-CS prepared by attaching the chromophores of tetraphenylethylene (TPE) to the chitosan (CS) chains showed excellent fluorescent properties. In this study, we tried to use TPE-CS for detecting Cu(II) because of the stable complexation of CS with heavy metals and the luminosity mechanism of the Restriction of Intramolecular Rotations (RIR) for aggregation-induced emission (AIE)-active materials. The fluorescence intensity changed when TPE-CS was contacted with different metal ions, to be specific, no change for Na + , slightly increase for Hg 2+ , Pb 2+ , Zn 2+ , Cd 2+ , Fe 2+ , Fe 3+ due to the RIR caused by the complexation between CS and metal ions. However, for Cu 2+ , an obvious fluorescence decrease was observed because of the Photoinduced-Electron-Transfer (PET). Moreover, we found that the quenched FL intensity of TPE-CS was proportional to the concentration of Cu(II) in the range of 5 μmol/L to 100 μmol/L, which provided a new way to quantitatively detect Cu(II). Besides, TPE-CS has excellent water-solubility as well as absorbability (the percentage of removal, R = 84%), which is an excellent detection probe and remover for Cu(II).

Abstract:Fluorescent sensor has demonstrated to be a facile and effective method to detect heavy metal ions owing to its unique characteristics such as simplicity, ease of operation and cost-effectiveness. In this work, a novel fluorescent probe with aggregation-induced emission (AIE) feature that contains an amide ligand base on the tetraphenylethylene (TPE) dye was designed and successfully synthesized. The utilization of the AIE-active fluorescent probe (named as FDPA) for detection of Cu2+ in aqueous solution has also been examined. We demonstrated that the resultant AIE-active molecule displays amphiphilic property and can self-assemble in aqueous solution with remarkable enhancement of fluorescent intensity owing to its AIE feature. The detection limit of probe for Cu2+ determination is 6.11 × 10-9 M and shows excellent selectivity in the presence of competitive ions. This work provides a useful route to overcome the fluorescence quenching of conventional fluorescent probes in aqueous solution and an elegant way to prepare fluorescent probes with better fluorescence properties.

Abstract:The excessive accumulation of Cu2+ in the human body and the natural environment will endanger human health and break the balance of the ecosystem. Therefore, the development of a low-cost and rapid method to detect Cu2+ is of great significance to protect human health and the environment. In this study, we used electron-rich triphenylamine as the electron donor and AIE active core, coupled with the electron acceptor imine nitrogen, and further connected to the ESIPT active salicylaldimine unit, detection based on the ESIPT system a new AIE fluorescent probe 5-(diethylamino)-2-((E)-(((E)-4-(diphenylamino)benzylidene)hydrazono)methyl)phenol (DDP) was synthesized. DDP has strong fluorescence and can coordinate specifically with Cu2+ to form a copper complex and cause fluorescence quenching, thereby achieving the purpose of detecting Cu2+. The EDTA titration experiment was carried out on the DDP–Cu2+ solution, and the experimental results proved that the combination of DDP and Cu2+ is reversible. The interaction between DDP and Cu2+ rapidly forms a copper complex, which is approximately completed within 6 s. The detection limit of the probe is as low as 5.7 × 10–8 M and it shows good stability in a weakly alkaline and weakly acidic environment, which provides necessary conditions for the detection of Cu2+ in a living environment. It is applied to the detection of Cu2+ in river water and lake water, showing a good linear relationship, indicating that DDP has the ability to detect Cu2+ in real water samples. Therefore, DDP is expected to become an effective tool for detecting Cu2+ in the environment and organisms.

Abstract:A new fluorescent molecular probe 1, derived from 3-(1-aminoiminethyl)-4-hydroxycoumarine and 2-formyl-3,5-dimethyl-1H-pyrrole by a simple Schiff base condensation, was designed, synthesised and accurately characterized. The aggregation induced ratiometric emission (AIRE) active probe 1 showed selective interaction with Cu2+ in both 1:1 and 9:1 H2O/DMSO solutions (v:v, buffered by 10 mM HEPES at pH = 7.4). However, the sensing mechanism was found to be different in each media. In H2O/DMSO 1:1 solution, 1 exhibited a coumarin emission at 525 nm and the addition of Cu2+ results in a coordination complex under strong Photoinduced Electron Transfer (PET) process. Conversely, TEM studies revealed that probe 1 organizes itself as mono disperse aggregates in H2O/DMSO 9:1 solution with the emission at 585 nm, and further disassembly of aggregates was induced by the addition of Cu2+, accomplishing with the fluorescence quenching. Abstract:The fluorescent probe L, based on naphthalimide-modified coumarin, was designed, synthesized, and characterized, which could recognize Cu2+ from other cations selectively and sensitively in HEPES buffer (10 mM, Ph = 7. 4)/CH3CN (1:4, V/V). When the probe L interacted with Cu2+, the color and the fluorescent intensity changed obviously and it provided the naked-eye detection for Cu2+. The recognition mode between them was achieved by Job's plot, IR, MS, SEM, and 1HNMR. In addition, test strips made from L could still interact with Cu2+ in tap water effectively. The limit of detection (LOD) of L was 3.5 × 10−6 M. Additionally, the density functional theory (DFT) calculation method was used to analyze the action mechanism of L toward Cu2+. Importantly, the fluorescent probe L could demonstrate favorable selectivity toward Cu2+ in Caenorhabditis elegans. Thus, L was considered to have some potential for application in bioimaging.

Abstract:A novel fluorescence chemical sensor-based probe 1-{[(E)-(2-aminophenyl)azanylidene]methyl}naphthalen-2-ol (AMN) was designed and synthesized, which performed a "naked eye" detection ability toward Cu2+ and Co2+ based on aggregation-induced emission (AIE) fluorescence strategy. It has sensitive detection ability for Cu2+ and Co2+. In addition, the color changed from yellow-green to orange under the sunlight, realizing the rapid identification of Cu2+/Co2+, which has the potential of on-site visual detection under the "naked eye". Moreover, different "on" and "off" fluorescence expressions were exhibited under excessive glutathione (GSH) in AMN-Cu2+ and AMN-Co2+ systems, which could be employed to distinguish Cu2+ from Co2+. The detection limits for Cu2+ and Co2+ were measured to be 8.29 × 10−8 M and 9.13 × 10−8 M, respectively. The binding mode of AMN was calculated to be 2:1 by Jobs' plot method analysis. Ultimately, the new fluorescence sensor was applied to detect Cu2+ and Co2+ in real samples (tap water, river water, and yellow croaker), and the results were satisfying. Therefore, this high-efficiency bifunctional chemical sensor platform based on "on–off" fluorescence detection will provide significant guidance for the advance development of single-molecule sensors for multi-ion detection.

Abstract:A novel coumarin-derived Schiff base fluorescent probe was reported to detect Cu 2+ ions in human cells. It was synthesized by the reaction between 7-(diethylamino)coumarin-3-carbohydrazide and pyridoxal 5′-phosphate. The probe has good stability and it exhibited obviously decreasing fluorescent response at λ 490 nm upon addition of Cu(II) ions. The fluorescent probe showed high selectivity for Cu(II) ions compared to other metal ions. The probe has been successfully applied to detect Cu(II) ions in human cells.

Abstract:By combination of the AIEE-active fluorophore and coordination binding group, a new selective and sensitive probe (1) for Hg2+ detection has been synthesized. 1 exhibits weak fluorescence in dilute solution, but exhibits dramatically enhanced emission in aggregation. In the solid state, a remarkable red-shifted emission and longer PL lifetime of 1 are present, which is caused by the formation of excimers due to the pi-pi stacking interactions of the coumarin groups. This AIEE effect of 1 is utilized to design probe for the detection of Hg2+ in a luminescence "turn-on" mode. The functionalization of the AIEE-active molecule with chelating group provides the potential binding sites for Hg2+. The selective coordination-induced aggregation bursts the emission. 1 has been successfully applied to detect Hg2+ in aqueous and living cells. (C) 2015 Elsevier B.V. All rights reserved.

Abstract:The fluorescent probe has become an important method for the detection of heavy metal ions. In the present work, a new and simple fluorescent probe, Cu-P, for detecting copper ion (Cu^2+) was designed and synthesized. The probe has shown high sensitivity and selectivity toward Cu^2+. The detection limit was 13 nM (based on the 3o^-/slope). A significant color change from yellow to pink was observed; thus, the probe Cu-P could serve as a “naked-eye” indicator for Cu^2+. Furthermore, the proposed probe was used to detect Cu^2+ in real water and soil extract samples, with the result being satisfactory. Therefore, our proposed probe would provide a promising method for the detection of Cu^2+ in the environment.

Abstract:In this paper, a novel coumarin-derived fluorescent probe NY was designed and synthesized. NY displayed a significant ratiometric fluorescence response towards Cu 2+ in PBS buffer (10 mM, pH = 7.4), with the emission wavelength blue-shifted from 580 to 495 nm, and a fluorescence change from orange to green was evident under a 365 nm UV light. Meanwhile, NY had the advantages of high selectivity, short response time (5 min), low detection limit (1.3 × 10 -8 M) and large binding constant (1.45 × 10 5 M -1 ) towards Cu 2+ . The binding mechanism between NY and Cu 2+ was elucidated by FT-IR, 1 H NMR titration, TOF-MS and Job's plot analysis. In addition, NY was successfully employed in the detection of Cu 2+ within environmental water and vegetable samples with satisfactory results. Laser confocal microscopy imaging results showed that NY could easily penetrate HeLa cells membrane to target mitochondria and image Cu 2+ in living cells. Furthermore, NY demonstrated mechanochromic properties by exhibiting orange-red fluorescence when subjected to mechanical grinding.

Abstract:A near-infrared (NIR) fluorescence probe for copper(II) ions (Cu2+) has been synthesized and demonstrated that it could selectively and sensitively react with Cu2+ through specific coordination reaction. ESI-Mass and Job's plot confirmed a stoichiometric ratio of 2-1 between the probe molecules and Cu2+. This NIR fluorescence probe shows a fast response, high sensitivity and anti-interference performance for Cu2+ and has been successfully applied for the detection of Cu2+ in aqueous solution. The detection limit was calculated as low as 14 nM. It has been further demonstrated that the probe exhibited good cell permeability and could be applied for monitoring the fluctuation of Cu2+ in living cells by NIR fluorescence imaging. Abstract:Constructed on the benzothiazole-oxanthracene structure, a fluorescent probe RBg for Cu+ was designed under the ESIPT mechanism and synthesized by incorporating amide bonds as the connecting group and glyoxal as the identifying group. Optical properties revealed a good sensitivity and a good linear relationship of the probe RBg with Cu+ in the concentration range of [Cu+] = 0–5.0 μmol L−1. Ion competition and fluorescence-pH/time stability experiments offered further possibilities for dynamic Cu+ detection in an aqueous environment. HRMS analysis revealed a possible 1:1 combination of RBg and Cu+. In addition, colorimetric Cu+ detection and lysosome-targeted properties of the probe RBg were analyzed through RBg-doped PVDF nanofiber/test strips and RBg-Mito/Lyso trackers that were co-stained in living HeLa cells, enabling the probe's future applications as real-time detection methods for dynamic Cu+ tracking in the lysosomes and Cu+ detection under diversified conditions.
