Evaluation of the antiepileptic activity of hesperidin by fluorescence imaging

Abstract:Epilepsy is a clinical syndrome caused by highly synchronized abnormal discharges of neurons in the brain due to a variety of causes, and the clinical manifestations are characterized by seizures, transient, repetitive and stereotyped. Lipid droplets, as one of the important organelles in cells, have an important role in maintaining cellular functioning. Studies have shown that persistent epilepsy is associated with altered lipid droplet metabolism. Hesperidin as a natural product has been shown to be used in treatments related to neurological disorders. Therefore, studying the effect of hesperidin on cellular lipid droplets during epilepsy has become one of the priorities. Based on the problem, the near-infrared probe YCN was presented for evaluating the effect of hesperidin on epileptic cells. First, the probe YCN had good photostability, biocompatibility and excellent lipid droplet targeting ability. In addition, the probe YCN successfully detected changes of cell induced by honokiol, plumbagin and hesperidin, demonstrating the ability of the YCN probe to sensitively detect changes in the intracellular environment. Most importantly, YCN was resoundingly used to image changes of epileptic cells in situ and to explore the drug effects of hesperidin on epileptic cells, testifying significant antiepileptic activity of hesperidin at certain concentrations. The effectively application of probe YCN will provide a newfangled centering for the development and efficacy evaluation of antiepileptic drugs.
