Abstract:<p>The nematode <em>Caenorhabditis elegans</em> (<em>C. elegans</em>) is characterized by small size, a transparent body, and a simple structure. However, traditional techniques (e.g., sorting and behavioral analysis) used in <em>C. elegans</em> behavior research are cumbersome; in addition, obtaining large amount of information from experiments is challenging. Microfluidic chips can process single or multiple nematodes in a high-throughput manner through their automation and miniaturization, providing a new platform for nematode research. This review mainly focuses on different stimuli, such as heavy metals, chemicals, drugs, neurotoxins, and pathogens. Based on behavioral indicators, such as crawling speed, swing frequency, and crawling methods, the applications of microfluidics in toxicity evaluation, drug screening, and aging development are summarized. Lastly, challenges and potential opportunities associated with the use of microfluidic technology for <em>C. elegans</em> research include evaluating the joint effect of mixed endogenous pollutants, establishing an integrated evaluation system, and building a big data platform.</p>
