Abstract:Many recent research studies have reported indirect methods for the detection and quantification of OH radicals generated during photocatalysis. The short lifespan and high reactivity of these radicals make indirect detection using probes such as coumarin a more viable quantification method. Hydroxyl radical production is commonly monitored using fluorescence spectroscopy to determine the concentration of the compound 7-hydroxycoumarin, which is formed from hydroxyl radical attack on coumarin. There are, however, a number of additional hydroxylated coumarins generated during this process, which are less amenable to detection by fluorescence spectroscopy. Consequently, limitations and inaccuracies of this method have previously been reported in the literature. As an alternative approach to those previously reported, this work has developed an electrochemical screening method using coumarin as a OH radical trap, that is capable of in situ monitoring of not only 7-hydroxycoumarin, but all the main mono-hydroxylated products formed. As a result, this technique is a more representative and comprehensive method for the quantification of OH radicals produced by photocatalysts using coumarin as a probe molecule. Moreover, the electroanalytical method provides a portable, rapid, sensitive, and accurate in situ method for the monitoring of OH radical formation without the need for sample preparation.

Abstract:The detection and quantification of hydroxyl radicals (HO•) generated by low-temperature plasmas (LTPs) are crucial for understanding their role in diverse applications of plasma radiation. In this study, the formation of HO• in the irradiated aqueous phase is investigated at various plasma parameters, by probing them indirectly using the coumarin molecule. We propose a quantification methodology for these radicals, combining spectrophotometry to study the coumarin reaction with hydroxyl radicals and fluorimetry to evaluate the formation yield of the hydroxylated product, 7-hydroxycoumarin. Additionally, we thoroughly examine and discuss the impact of pH on this quantification process. This approach enhances our comprehension of HO• formation during LTP irradiation, adding valuable insights to plasma's biological applications.

Abstract:Hydroxyl radical(∙OH) is one of the strongest oxidizing species, which can react with a variety of organic and inorganic chemicals. Although ∙OH is widely used for degradation of environmental pollutants, detection of ∙OH remains a major challenge due to its high reactivity and short lifetime, especially online detection. In this study, a novel method for online detection of ∙OH by flow oxidization chemiluminescence using coumarin as probe was established. Because coumarin is known as a classic fluorescence probe for ∙OH detection, the measurement using new method can be evaluated by traditional method using high performance liquid chromatography combined with fluorescence detection. Off-line detection showed that chemiluminescence signal obtained using new method was in good agreement with traditional method, indicating the reliability of using chemiluminescence signal to quantify ∙OH concentration. Based on this new method, the dynamic production of ∙OH during the processes of TiO2 photocatalysis, homogeneous and heterogeneous Fenton reactions were successfully measured at real time. Considering the flexibility and sensitivity of this new method, it is expected to become a widely used ∙OH online monitoring method in both natural and engineering systems.

Abstract:The infrared absorption spectra of the carbonyl stretching vibrations of 6-methylcoumarin (6MC) have been investigated in CCl4/ROH mixtures (CCl4/C2H5OH, CCl4/n-C3H7OH, CCl4/i-C3H7OH, and CCl4/t-C5H11OH). Two types of carbonyl stretching vibration bands for 6MC are found with the change of the mole fraction of the aprotic solvent CCl4(XCCl4) in binary solvent mixtures. The dependencies of the frequencies of carbonyl stretching vibrations υ(CO) on XCCl4 allow a distinction and assignment of all species resulting from the solvent–solute interactions. Linear correlations between the υ(CO) of each species and XCCl4 are found. The influence on the transformation of some species caused by the self-associated alcohols is discussed.
Coumarin fluorometry to quantitatively detectable OH radicals in ultrasound aqueous medium

Abstract:When ultrasound (US) was exposed to aqueous coumarin solution in air atmosphere, the UV-visible and fluorescence spectra of the probe were measured at different US exposure times. The US exposure was carried out at 43 kHz and 500 kHz with different out-put power. It was found that the 500 kHz US produced umbelliferone fluorescence, while the 43 kHz US had no fluorescence. In addition, the coumarin absorbance at 270 nm maximum was decreased with in cases of the US exposure time. In contrary, the fluorescent intensity of umbelliferone at 460 nm increased with increasing of US exposure time. This exhibited that the coumarin probe was converted to umbelliferone by the US exposure, when the 500 kHz US was operated. This was facted that the coumarin framework was caused with addition of OH groups which was generated by the 500 kHz US. Therefore, the umbelliferone fluorescent became a probe to estimate OH radical in US medium. Furthermore, the chemo-fluorometry showed that the emission maximum of the formed umbelliferone could probe the bulk pHs in the US aqueous medium.

Abstract:Irradiation of coumarin-3-carboxylic acid in acetonitrile and methanol solutions at 355 nm results in complex multistep photochemical transformations, strongly dependent on the solvent properties and oxygen content. A number of reaction intermediates, which themselves undergo further (photo)chemical reactions, were identified by steady-state and transient absorption spectroscopy, mass spectrometry, and NMR and product analyses. The triplet excited compound in acetonitrile undergoes decarboxylation to give a 3-coumarinyl radical that traps molecular oxygen to form 3-hydroxycoumarin as the major but chemically reactive intermediate. This compound is oxygenated by singlet oxygen, produced by coumarin-3-carboxylic acid sensitization, followed by a pyrone ring-opening reaction to give an oxalic acid derivative. The subsequent steps lead to the production of salicylaldehyde, carbon monoxide, and carbon dioxide as the final products. When 3-coumarinyl radical is not trapped by oxygen in degassed acetonitrile, it abstracts hydrogen from the solvent and undergoes triplet-sensitized [2 + 2] cycloaddition. The reaction of 3-coumarinyl radical with oxygen is largely suppressed in aerated methanol as a better H-atom donor, and coumarin is obtained as the primary product in good yields. Because coumarin derivatives are used in many photophysical and photochemical applications, this work provides detailed and sometimes surprising insights into their complex phototransformations.Graphical abstract

Dual-emissive fluorescence measurements of hydroxyl radicals using a coumarin-activated silica nanohybrid probe

Abstract:This work reports a novel dual-emissive fluorescent probe based on dye hybrid silica nanoparticles for ratiometric measurement of the hydroxyl radical (˙OH). In the probe sensing system, the blue emission of coumarin dye (coumarin-3-carboxylic acid, CCA) immobilized on the nanoparticle surface is selectively enhanced by ˙OH due to the formation of a coumarin hydroxylation product with strong fluorescence, whereas the emission of red fluorescent dye encapsulated in the silica nanoparticle is insensitive to ˙OH as a self-referencing signal, and so the probe provides a good quantitative analysis based on ratiometric fluorescence measurement with a detection limit of 1.65 μM. Moreover, the probe also shows high selectivity for ˙OH determination against metal ions, other reactive oxygen species and biological species. More importantly, it exhibits low cytotoxicity and high biocompatibility in living cells, and has been successfully used for cellular imaging of ˙OH, showing its promising application for monitoring of intracellular ˙OH signaling events.

Abstract:As part of this study, the mechanisms of the antioxidant activity of previously synthesized coumarin–trihydrobenzohydrazine derivatives were investigated: (E)-2,4-dioxo-3-(1-(2-(2′′,3′′,4′′-trihydroxybenzoyl)hydrazineyl)ethylidene)chroman-7-yl acetate (1) and (E)-2,4-dioxo-3-(1-(2-(3′′,4′′,5′′-trihydroxybenzoyl)hydrazineyl)ethylidene)chroman-7-yl acetate (2). The capacity of the compounds to neutralize HO• was assessed by EPR spectroscopy. The standard mechanisms of antioxidant action, Hydrogen Atom Transfer (HAT), Sequential Proton Loss followed by Electron Transfer (SPLET), Single-Electron Transfer followed by Proton Transfer (SET-PT), and Radical Adduct/Coupling Formation (RAF/RCF) were examined using the QM-ORSA methodology. It was estimated that the newly synthesized compounds, under physiological conditions, exhibited antiradical activity via SPLET and RCF mechanisms. Based on the estimated overall rate constants (koverall), it can be concluded that 2 exhibited a greater antiradical capacity. The obtained values indicated a good correlation with the EPR spectroscopy results. Both compounds exhibit approximately 1.5 times more activity in comparison to the precursor compound used in the synthesis (gallic acid).

Abstract:This study examined the antiradical activity of three synthesized coumarin derivatives: ( E )-3-(1-((2-hydroxyphenyl)amino)ethylidene)-2,4-dioxochroman-7-yl acetate ( A 1 -OH ), ( E )-3-(1-((3-hydroxyphenyl)amino)ethylidene)-2,4-dioxochroman-7-yl acetate ( A 2 -OH ), and ( E )-3-(1-((4-hydroxyphenyl)amino)ethylidene)-2,4-dioxochroman-7-yl acetate ( A 3 -OH ) against HOO•/O2•− radical species. The investigation included electron spin resonance (ESR) measurements and a DFT kinetic study. Thermodynamic and kinetic parameters of antiradical mechanisms—Formal Hydrogen Atom Transfer ( f -HAT), Radical Adduct Formation (RAF), Sequential Proton Loss followed by Electron Transfer (SPLET), and Single-Electron Transfer followed by Proton Transfer (SET-PT)—were evaluated using the Quantum Mechanics-based test for Overall Free Radical Scavenging Activity (QM–ORSA) under physiological conditions. ESR results indicated antiradical activity decreased in the sequence A 1 –OH (58.7%) > A 2 –OH (57.5%) > A 3 –OH (53.1%). Kinetic analysis revealed the f -HAT mechanism dominated HOO • inactivation. A newly formulated Sequential Proton Loss followed by Radical Adduct Formation (SPL-RAF) mechanism described interactions with O 2 •− . The activity toward O 2 •− was A 2 –OH (1.26 × 10 6 M −1 s −1 ) > A 3 –OH (7.71 × 10 5 M −1 s −1 ) > A 1 –OH (4.22 × 10 5 M −1 s −1 ). Molecular docking and dynamics studies tested inhibitory capability against enzymes producing reactive species: Lipoxygenase (LOX), Myeloperoxidase (MPO), NAD(P)H oxidase (NOX), and Xanthine Oxidase (XOD). Affinity to enzymes decreased in the order: XOD > LOX > NOX > MPO.

Abstract:Coumarins 3‐aldehyde were synthesized under mild conditions with 3‐phenylpropiolates and 1,3‐dioxolane via tandem visible light‐catalyzed cascade radical cyclization and acidic hydrolysis process in one pot. New synthetic method of coumarins 3‐aldehyde via visible light stimulation and oxidation promotion cascade radical cyclization/hydrolysis process is reported. this reaction involves a two‐step procedure, the 1,3‐dioxolane radical is initiated in situ from 1,3‐dioxolane via the excited Ru(bpy)3Cl2 and oxidant TBHP, subsequently an addition to triple bond of various alkynes produces vinyl radicals, the radical intermediates can smoothly convert to coumarin 3‐aldehyde via tandem cyclization/acidic hydrolysis process in good to excellent yields. Furthermore, the synthetic application valuable of coumarin 3‐aldehyde is also evaluated in this work.

An “On-Off” Ratio Photoluminescence Sensor Based on Catalytically Induced PET Effect by Fe3O4 NPs for the Determination of Coumarin

Abstract:Herein, using Fe3O4 nanoparticles (Fe3O4 NPs) as a magnetic artificial peroxidase, an "on-off ' ratiometric photoluminescence sensor with high-sensitivity and high-selectivity for coumarin was constructed based on photoinduced electron transfer (PET) between 7-hydroxycoumarin and rhodamine B (RB). The results showed that Fe3O4 NPs catalyzed H2O2 to generate nucleophilic group center dot OH, which attacked the active site of coumarin and produced strong fluorescent 7-hydroxycoumarin molecules. Then, the fluorescence of RB was quenched with 7-hydroxycoumarin through the PET effect. The ratio signal generated in the above process was used for the quantitative detection of coumarin. Under optimized conditions, the linear range 0.5-25 mg/L was acquired for coumarin with the detection limit of 0.016 mg/L. This method had excellent selectivity and the recovery rate was 81.8%-106.8% with the relative standard deviation less than 5.6%, so it can be used for the quantitative analysis of coumarin in complex matrix samples.

Abstract:The organic compound 7–hydroxy–4–methyl coumarin, also known as 4–methylumbelliferone or hymecromone, belongs to the lactone family. The compound's molecular geometry, the bond angle, and the bond length of its dimers were determined by theoretical calculations using the software packages Gaussian 09W and Multiwfn 3.8. The most stable form of the 7–hydroxyl–4–methyl coumarin molecule is found to be a syn-ht dimer with an energy of −1223.1466 (a.u.).VEDA 4 software was used to determine vibrational assignments and PED values to gain insight into the structural details of these dimers. The vibrations of dimers formed from the same monomer showed remarkable differences. To support the chemical reactivity of the dimers, the HOMO–LUMO energy gap, MEP, and ALIE maps were analyzed. The anti- hh dimer was found to be highly polarizable and reactive in both solvent and gaseous media. In addition, Fukui functions were used to analyze the electrophilic (E +) and nucleophilic (Nu-) sites. The electronic excitation and UV–vis spectra were predicted using the time–dependent density functional theory (DFT) method. RDG-NCI and AIM analyses were utilized for predicting weak interactions between dimer molecules. Electronic parameters such as density of states (DOS) analysis, transition density matrix (TDM), and energy levels of electrons and holes were also analyzed. Based on the theoretical analysis, it is evident that this particular molecule exhibits a propensity for dimerization, with the resulting dimers showing considerable potential for advancement in various fields, particularly in the field of polymers and materials.

Abstract:Many organic and medicinal chemists are interested in the coumarin compounds due to their potential for use in pharmaceuticals and other industries. Coumarin compounds are a significant class of biologically active chemicals. Here, we outline the steps forproducing coumarin derivatives quickly and effectively from commercially accessible substrates via multicomponent condensation processes involving pyrone and benzene derivatives. These substances were examined for their potential anti-tumor effects, including their ability to block carbonic anhydrase, activate the cell apoptosis protein, and coumarin and its derivatives are the primary oral anticoagulants. Candida albicans, Aspergillus fumigatus, and Fusarium solani are three distinct fungus species that are resistant to the antifungal properties of synthetic coumarins. Long known are the coumarins' antibacterial due to its biological activities against some kinds of bacteria such as Zeitschrift für Naturforschung C, as well as their antifilarial, antiulcerogenic, anti-inflammatory, and antioxidant effects for its capacity to inhibit their growth activities. Coumarin has long been valued as a major raw element in the fragrance sector due to its distinct sweet aroma and stability. When mixed with organic essential oils like lavender, citrus, rosemary, and oak moss, it is used as an odour-enhancer to produce a long-lasting impact. It has been utilized as a bleaching or brightening agent in detergents when it revealed a remarkable brightening effect with excellent overall stability on fibers of synthetic origin. Materials such as liquid crystalline, organic/inorganic composites, as well as light harvesting, energy transferring compounds, and electro-optics materials are all employed as a dye in organic photo-redox catalysis and as potent photo-sensitizers reagent. These materials polymerize epoxy-silicones, under near-UV and visible light. Coumarin has also found useful as metal-free sensitizers for solar cells.

Abstract:Self-cleaning materials are widely applied, but the available methods for determining their photocatalytic activity are time consuming. A simple analysis method was proposed to evaluate rapidly the photocatalytic activity of self-cleaning materials. This method is based on monitoring of a highly fluorescent product generated by the self-cleaning materials after illumination. Under UV irradiation, holes photo-induced on the surface of self-cleaning materials can oxidize water molecules (or hydroxide ions) adsorbed on the surface to produce hydroxyl radicals, which then quantitatively oxidize coumarin to highly fluorescent 7-hydroxycoumarin. It was observed that the fluorescence intensity of photo-generated 7-hydroxycoumarin at 456 nm (excited at 346 nm) linearly increased with irradiation time, and the fluorescence intensity at a given irradiation time was linearly proportional to the photocatalytic activity of self-cleaning materials. Consequently, the photocatalytic activity of self-cleaning materials was able to be probed simply by using this new method, which requires an analysis time of 40 min, being much less than 250 min required for a dye method.

Abstract:The constant need for high-performance aniline- or phenol-based fluorophores suitable for the construction of activity-based fluorescent probes, led us to study both synthesis and photophysics of under-explored C3-N-methylpyridinium-4-yl substituted 7-(dialkylamino)/7-hydroxycoumarins. Indeed, in the field of photoactive organic molecules, the positively charged N-alkylpyridinium-4-yl groups are often used as acceptor units to dramatically impact spectral features through promoting intramolecular charge transfer (ICT) process. They are also known as effective water-solubilizing and cell organelle (mitochondria) targeting moieties. The poor fluorescence efficiency of cationic 7-hydroxycoumarin derivatives in aqueous physiological conditions was highlighted and rationalized by the predominance of a neutral quinonoid form in such buffer medium. The ability of the excited singlet state (S1) of this neutral species to undergo intersystem crossing (ISC) to triplet state (T1) was partly supported by phosphorescence measurements of singlet oxygen (1O2). We took advantage of superior green-emissive properties of 7-(diethylamino)-3-(N-methylpyridinium-4-yl)coumarin to successfully design and validate a novel small-molecule fluorescent probe for the detection of alkaline phosphatase (ALP), based on the "covalent-assembly" principle. In last part of this study, a practical use of ortho-formylated 7-hydroxy-3-(N-methylpyridinium-4-yl)coumarin was considered with the synthesis of a Fe(III)-salen complex known to act as effective "molecular disassembly" probe for fluorogenic sensing pyrophosphate (PPi) ions in aqueous media, and through the promising approach recently proposed by the Zelder group (Chimia 2020, 74, 252-256).

Abstract:Benzopyrone is a popular fluorescent scaffold, but how chemical modifications affect its properties is less understood. We investigated this using halogenated 7‐hydroxycoumarin, unsubstituted 4‐methylumbiliferone, and ortho‐chloro and bromo substitutions on the phenolic ring. Experimental charge density data and computational methods revealed that halogenation at the ortho position significantly reduced quantum yield (QY). Specifically, 7‐hydroxycoumarin (1) had a QY of 70%, while ortho‐chloro (2) and ortho‐bromo (3) had QYs of 61% and 30%, respectively. Experimental data showed that all probes excited similarly, but the electrostatic potential and dipole moments indicated that 2 and 3 dissipated excitation energy more easily due to charge separation. The heavy‐atom effect of Cl and Br did not fully explain the QY reductions, suggesting other radiative decay processes were involved. By incorporating spin‐orbit coupling (SOC) effects, we estimated intersystem crossing (ISC) and phosphorescence rates, providing theoretical QYs of 78% for 1, 59% for 2, and 15% for 3. The large deviation for 3 was attributed to its higher SOC potential. Our findings indicate that 3's reduced QY results from a mix of SOC‐induced ISC and charge dissipation, while 2's reduction is primarily due to charge separation. Further studies are needed to validate this approach with other scaffolds.

Abstract:Photo-sensitive probes based on coumarin deserve extensive attention because of their high biocompatibility, quantum yield, and stability. The synthesis of coumarin and its derivatives is mainly based on four classical methods: Perkin synthesis, Knoevenagel synthesis, Wittig synthesis, and Pechmann synthesis. As everyone knows, fluorescent probes consist of a recognition group, a bridging group, and a luminescent group. Mechanism of fluorescent probes can be roughly summarized into four models: Chelation-induced enhanced fluorescence (CHEF), intramolecular charge transfer (ICT), photoinduced electron transfer (PET), and aggregation-induced luminescence (AIE), which can instruct the design of coumarin-based photo-sensitive probes. Since the fluorescence-based probe was first reported, probes based on coumarin have found widespread application in analytical chemistry, proving to be a selective, efficient, and convenient tool in ion monitoring, molecular monitoring, cell imaging, drug delivery, and many other fields. In this critical review, the synthetic routes of these compounds and the design for fluorescent probes were reported. Furthermore, the recent researches on the employment of coumarin in analytical chemistry were also introduced. Finally, the future research trends and application foreground of coumarin derivatives were forecasted herein.

Development of a Quinolinium/Cobaloxime Dual Photocatalytic System for Oxidative C–C Cross-Couplings via H 2 Release

Abstract:Designing molecular photocatalysts for potent photochemical reactivities ranks among the most challenging but rewarding endeavors in synthetic photochemistry. Herein, we document a quinoline-based organophotoredox catalyst, 2,4-bis(4-methoxyphenyl)quinoline (DPQN2,4‑di‑OMe ), that could be assembled via the facile aldehyde–alkyne–amine (A3) couplings. Unlike the reported photocatalysts, which impart their photoreactivities as covalently linked entities, our mechanistic studies suggested a distinct proton activation mode of DPQN2,4‑di‑OMe . Simply upon protonation, DPQN2,4‑di‑OMe could reach a highly oxidizing excited state under visible-light irradiation (E*1/2 = +1.96 V vs a standard calomel electrode, SCE). On this basis, the synergistic merger of DPQN2,4‑di‑OMe and cobaloxime formulated an oxidative cross-coupling platform, enabling the Minisci alkylation and various C–C bond-forming reactions with a diverse pool of radical precursors in the absence of chemical oxidants. The catalytic loading of DPQN2,4‑di‑OMe could be minimized to 0.025 mol % (TON = 3360), and a polymer-supported photocatalyst, DPQN2,4-di-OR@PS, was prepared to facilitate catalyst recycling (at a 0.50 mmol % loading and up to five times without significant loss of photosynthetic efficiency).

Abstract:3-Alkylated coumarins have many applications in medicinal chemistry, however, methods access to such structures are still limited. Herein, we report a site-selective photocatalytic deaminative alkylation of coumarins utilizing pyridinium-activated aliphatic primary amines as alkylation reagents. The protocol was highlighted by its mild reaction conditions, operational simplicity, and broad functional group compatibility. Moreover, this strategy enables late-stage modification of some pharmaceuticals and natural products, thus providing an appealing approach to valuable molecules in medicinal chemistry.
