Formation of EGCG oxidation self-assembled nanoparticles and its antioxidant activity in vitro and hepatic REDOX regulation activity in vivo

Abstract:(-)-Epigallocatechin-3-gallate (EGCG) is a major polyphenol in tea with various health-promoting effects. It is easily autoxidized into complex polymers and deactivated due to the presence of multiple phenolic hydroxyl structures. Currently, there is a lack of clarity regarding the morphology and biological activity of complex EGCG polymers. The present study demonstrated that EGCG autoxidation self-assembles into nanoparticles (ENPs) that exhibit antioxidant activity in vitro and hepatic REDOX homeostasis regulation activity in vivo. We identified the formation of ENPs during EGCG autoxidation process and revealed the intermolecular interaction forces that maintain the stability of nanoparticles. Like EGCG, ENPs exhibit excellent in scavenging ROS and hydroxyl radical in vitro as well as in regulating hepatic REDOX activity, suggested by regulate liver redox enzymes including thioredoxin reductase (TrxR), thioredoxin (Trx), glutathione reductase (GR), glutaredoxin (Grx), and glutathione S-transferase (GST) in vivo. Moreover, ENPs can also activate the NRF2 antioxidant-responsive element pathway to exert a detoxification effect at high dose. Impressively, unlike EGCG, ENPs do not cause liver damages at low dose and kept better liver biosafety at high dose through extend self-assembly form large particles, which supported by uninfluenced of liver damage biomarkers, including serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), liver γ-phosphorylated histone 2AX (γ-H2AX) , and P53-related genes (Thbs, MDM2, P53, Bax). Collectively, these experiments reveal that with adequate biosafety and regulation of hepatic REDOX activity in vivo, ENPs have great potential for antioxidant application or food additive as an EGCG substitute to overcome the instability and liver toxicity of EGCG.
