Abstract:A facile and efficient domino cyclization between 3-aminocoumarins and arylglyoxal monohydrates in the presence of para-toluenesulfonic acid (p-TSA) as a promoter was employed for the synthesis of two libraries of chromeno[3,4-b]pyrrol-4(3H)-one derivatives. Simple workup procedures were used to obtain the products in moderate to excellent yields. The solvents toluene and ethanol served important roles in the selective formation of the pyrrolocoumarin derivatives characterized by their different substitution patterns.

Abstract:Herein, we report unprecedented Ni-catalyzed chemodivergent annulations to produce the 3,4-dihydrocoumarins and 2H-chromenes that are important attractive building blocks in many biologically active compounds. By the introduction of dcype ligand, a range of 3,4-dihydrocoumarins can be afforded with good to excellent yield and wide functional group tolerance via chemoselective intramolecular tan-dem isomerization/esterification. In the absence of ligand, a useful synthesis of 2H-chromenes has been developed through a nickel-catalyzed direct O-allylic substitution with good to excellent efficiency.(c) 2023 Elsevier Inc. Many compounds possessing this structure are nutraceuticals, pharmaceutical nutrients. Therefore, benzopyran and chromane scaffolds are important building blocks in organic synthesis and many efforts have been made to set up efficient methods for their synthesis. In particular, asymmetric methods are of great importance, being natural products, and generally chiral substances. This review aims to cover literature in the range 2017–first half of 2019.

Abstract:Selective synthesis of chiral bridged (hetero)bicyclic scaffolds via asymmetric C-H activation constitutes substantial challenges due to the multiple reactivities of strained bicyclic structures. Herein, we develop the domino transformations through an unprecedented cobalt-catalyzed enantioselective C-H activation/nucleophilic [3 + 2] annulation with symmetrical bicyclic alkenes. The methods offer straightforward access to a wide range of chiral molecules bearing [2.2.1]-bridged bicyclic cores with four and five consecutive stereocenters in a single step. Two elaborate salicyloxazoline (Salox) ligands were synthesized based on the rational design and mechanistic understanding. The well-defined chiral pockets generated from asymmetric coordination around the trivalent cobalt catalyst direct the orientation of bicyclic alkenes, leading to excellent enantioselectivity.

Abstract:Asymmetric diastereodivergent catalysis is a powerful strategy for accessing different stereoisomers of a molecule, which are important in the discovery of biologically and pharmacologically active candidates. While the asymmetric diastereodivergent synthesis of acyclic molecules has seen recent rapid development, the corresponding asymmetric diastereodivergent cycloaddition reactions have received comparatively less attention. Herein, we report the asymmetric diastereodivergent synthesis of chromanes with C2,C3-contiguous tetrasubstituted stereocenters via chiral phosphoric acid (CPA)-catalyzed [4 + 2] cycloadditions of ortho-quinone methides with γ-silyl-substituted allenones. A wide range of C3-epimeric chromanes were generated with high to excellent diastereoselectivities and enantioselectivities, starting from the same substrates but with modifications of the CPA catalysts. The diverse derivatizations of these chiral chromane stereoisomers into complex scaffolds illustrate the utility and value of this method.

Abstract:A domino annulation/oxidation of heterocyclic ketene aminals (HKAs) and 2-aminochalcones has been developed for the selective synthesis of poly-substituted benzo[f]imidazo[2,1-a][2,7]naphthyridines and 3-azaheterocyclic substituted 2-arylquinolines. These reactions proceed well under mild conditions without any additives. Plausible mechanisms for such a polycyclic ring system assembly were also proposed. Moreover, benzo[f]imidazo[2,1-a][2,7]naphthyridine 3g displayed a fluorescence effect, demonstrating the potential applications in organic optical materials.

The reaction exhibits a broad substrate scope under a metal-free catalytic condition.

Abstract:Quinone imines are important derivatives of quinones with a wide range of applications in organic synthesis and the pharmaceutical industry. The attack of nucleophilic reagents on quinone imines tends to lead to aromatization of the quinone skeleton, resulting in both the high reactivity and the unique reactivity of quinone imines. The extreme value of quinone imines in the construction of nitrogen- or oxygen-containing heterocycles has attracted widespread attention, and remarkable advances have been reported recently. This review provides an overview of the application of quinone imines in the synthesis of cyclic compounds via the domino annulation reaction.

Abstract:Herein, we report the rhodium-catalyzed domino arylation/cyclization reaction of unactivated internal alkynes to afford the optically active indenols in excellent yields (up to 92%) and extremely high enantioselectivities (up to 99% ee) in the presence of new amide substituted chiral diene ligands. Notably, a total of 21 new chiral diene ligands are synthesized, which are potential choices for other asymmetric reactions.

Abstract:A novel chiral spirocyclic amide (SPA)-derived triazolium organocatalyst has been designed and demonstrated to effect asymmetric homo- and heterodialkylations of various bisoxindoles, enabling enantioselective construction of vicinal all-carbon quaternary stereocenters. These reactions feature excellent enantioand diastereoselectivities (up to 99% ee and >20:1 dr) as well as good to high yields (up to 89% over two steps). As an application of this methodology, the first asymmetric total synthesis of (-)-chimonanthidine has been achieved.

Abstract:An efficient method for the asymmetric Diels-Alder cycloaddition of 2'-hydroxychalcones with acyclic or cyclic dienes has been successfully developed. The Diels-Alder cycloaddition is mediated by a chiral boron complex with VANOL, affording the corresponding products in high yields and with excellent diastereo- and enantioselectivities. This reaction enabled the enantioselective construction of cyclohexene skeletons crucial for the total synthesis of a number of Diels-Alder-type natural products (-)-nicolaioidesin C, (-)-panduratine A, (-)-kuwanon I, (+)-kuwanon J, and (-)-brosimones A and B.

Abstract:An enantioselective [5 + 1] annulation of α-hydroxyl aryl ketones with cyclohexadienone-tethered enones via chiral dinuclear zinc catalysts is reported. A Brønsted base and Lewis acid cooperative activation model is the critical factor for the chirality transfer process, giving access to a wide array of enantioenriched bicyclic pyrans scaffolds bearing two oxa-quaternary carbon centers with good yields and high optical purities. Notably, this reaction offers a new reaction mode of α-hydroxyl ketones as C,C-bisnucleophiles in catalytic asymmetric transformation.

Abstract:The bicyclo[3.3.1]nonane ring represents an attractive yet challenging synthetic targets in the field of organic synthesis, especially in a stereocontrolled fashion. Over the past two decades, organocatalysis has emerged as an enabling technology for the facile construction of enantioenriched molecules with remarkably increasing complexity and diversity, and a myriad of chiral architectures containing bicyclo[3.3.1]nonane units were elegantly assembled through organocatalytic asymmetric transformations. This review summarizes recent advancements on this topic (mainly from 2010 to 2023), emphasizing the reaction types such as desymmetrization and cascade reactions of different prochiral starting materials toward various chiral bicyclo[3.3.1]nonanes as well as their aza, oxa and thio derivatives. Meanwhile, the synthetic strategy, reaction mechanism, substrate scope and synthetic applications of these catalytic reactions are also discussed. We hope that this review will motivate further development and application of organocatalytic asymmetric synthesis of bicyclo[3.3.1]nonane frameworks.

Abstract:Atropisomers featuring restricted rotation around a single bond have emerged as important structural motifs due to their prevalence in drug discovery, materials science and catalyst design. In recent years, the catalytic asymmetric synthesis of atropisomers has hold significant fascination from chemists. While traditional six-membered axially chiral skeletons are commonly studied, methodologies directed towards less abundant five-membered heterobiaryls are relatively rare. In particular, axially chiral azole-based skeletons are essential components of five-membered heterocyclic framework and have received considerable interest. This is the focus of this review, which summarizes the recent achievements in atroposelective preparation of azole derivatives, by emphasizing mechanistic insights and synthetic applications. We hope that this review will encourage chemists worldwide to promote a continued development of the field in the future.

Abstract:The merger of photoredox and transition-metal catalysis has evolved as a robust platform in organic synthesis over the past decade. The stereoselective 1,4-functionalization of 1,3-enynes, a prevalent synthon in synthetic chemistry, could afford valuable chiral allene derivatives. However, tremendous efforts have been focused on the ionic reaction pathway. The radical-involved asymmetric 1,4-functionalization of 1,3-enynes remains a prominent challenge. Herein, we describe the asymmetric three-component 1,4-dialkylation of 1,3-enynes via dual photoredox and chromium catalysis to provide chiral allenols. This method features readily available starting materials, broad substrate scope, good functional group compatibility, high regioselectivity, and simultaneous control of axial and central chiralities. Mechanistic studies suggest that this reaction proceeds through a radical-involved redox-neutral pathway.
