Abstract:Despite the wide success of antibiotics in modern medicine, the treatment of bacterial infections still faces critical challenges, especially due to the rapid emergence of antibiotic resistance. As a result, local antimicrobial treatment aimed at enhancing drug concentration at the site of infection while avoiding systemic exposure is becoming increasingly attractive, as it may alleviate resistance development. Meanwhile, therapeutic nanoparticles, especially liposomes, polymeric nanoparticles, dendrimers, and inorganic nanoparticles, are gaining traction to improve the therapeutic efficacy with many applications specifically focused on local antimicrobial treatment. This review highlights topics where nanoparticle-based strategies hold significant potential to advance treatment against local bacterial infections, including (1) promoting antibiotic localization to the pathogen, (2) modulating drug-pathogen interaction against antibiotic resistance, and (3) enabling novel anti-virulence approaches for ‘drug-free’ antimicrobial activity. In each area, we highlight the innovative antimicrobial strategies tailored for local applications and review the progress made for the treatment of bacterial infections.

Abstract:Diseases caused by pathogens continue to threaten the health of people around the world, especially with the emergence of drug resistance. Therefore, there is an urgent need to develop new strategies that can replace antibiotic therapy. Novel nanomaterials represented by two-dimensional nanomaterials (2DNMs) have great application prospects in the field of antibacterial due to their tunable band gaps, special crystal structures, and limitations on carrier migration and thermal diffusion. In this review, several representative 2DNMs such as graphene, graphitized carbonitride and transition-metal dichalcogenides are reviewed and summarized including their research basis, progress, and prospects in the field of antibacterial. It starts with the introduction of the material, focusing on summarizing its inherent antibacterial properties and light-induced antibacterial properties. Specific strategies to optimize the antibacterial properties of 2DNMs are discussed. Finally, the challenges facing current antimicrobial 2DNMs are discussed and future prospects for the field are proposed based on the combination of photocatalysis and other germicidal technologies.

Abstract:Nano‐antibacterial materials are important in the antibacterial and antibiofilm field because of their excellent physical and chemical properties. This review focuses on new physicochemical antibacterial strategies and versatile antibacterial nanomaterials, especially the mechanism and types of 2D antibacterial nanomaterials. This review also provides guidance on the development direction of antibiotic‐free disinfections in the antibacterial field in the future. Bacterial infection continues to be an increasing global health problem with the most widely accepted treatment paradigms restricted to antibiotics. However, the overuse and misuse of antibiotics have triggered multidrug resistance of bacteria, frustrating therapeutic outcomes, and leading to higher mortality rates. Even worse, the tendency of bacteria to form biofilms on living and nonliving surfaces further increases the difficulty in confronting bacteria because the extracellular matrix can act as a robust barrier to prevent the penetration of antibiotics and resist environmental damage. As a result, the inability to eliminate bacteria and biofilms often leads to persistent infection, implant failure, and device damage. Therefore, it is of paramount importance to develop alternative antimicrobial agents while avoiding the generation of bacterial resistance to prevent the large‐scale growth of bacterial resistance. In recent years, nano‐antibacterial materials have played a vital role in the antibacterial field because of their excellent physical and chemical properties. This review focuses on new physicochemical antibacterial strategies and versatile antibacterial nanomaterials, especially the mechanism and types of 2D antibacterial nanomaterials. In addition, this advanced review provides guidance on the development direction of antibiotic‐free disinfections in the antibacterial field in the future.

Abstract:The emergence and global spread of bacterial resistance to currently available antibiotics underscore the urgent need for new alternative antibacterial agents. Recent studies on the application of nanomaterials as antibacterial agents have demonstrated their great potential for management of infectious diseases. Among these antibacterial nanomaterials, carbon-based nanomaterials (CNMs) have attracted much attention due to their unique physicochemical properties and relatively higher biosafety. Here, a comprehensive review of the recent research progress on antibacterial CNMs is provided, starting with a brief description of the different kinds of CNMs with respect to their physicochemical characteristics. Then, a detailed introduction to the various mechanisms underlying antibacterial activity in these materials is given, including physical/mechanical damage, oxidative stress, photothermal/photocatalytic effect, lipid extraction, inhibition of bacterial metabolism, isolation by wrapping, and the synergistic effect when CNMs are used in combination with other antibacterial materials, followed by a summary of the influence of the physicochemical properties of CNMs on their antibacterial activity. Finally, the current challenges and an outlook for the development of more effective and safer antibacterial CNMs are discussed.

Abstract:In recent years, antibiotic-resistant bacterial strains have spread rapidly and widely around the world. New antibacterial drugs have emerged to develop. With the development of biological nanotechnology, two-dimensional nanomaterials have become a very promising alternative for the treatment of antibiotic-resistant bacteria. Based on the recent literature, the structural features and antibacterial applications of graphene materials (GMs), transition-metal dichalcogedes (TMDs), layered double hydroxides (LDHs) and MXenes were elaborated, the antibacterial mechanisms of those materials were discussed, such as physical/mechanical damage, lipid extraction, oxidative stress, and photothermal/photodynamic effects, etc. Finally, the antibacterial research progress and challenges of two-dimensional nanomaterials were prospected: (1) 2D materials has a unique space structure and excellent biocompatibility, so it can be used as an ideal carrier for antibacterial drugs; (2) the material has photodynamic and photothermal bactericidal effects, so it has a strong potential to cure local skin infections; (3) it can be made into antibacterial coatings to achieve simple in-situ disinfection applying to sterile medical equipment in the future.

Abstract:Clinically, multidrug resistant (MDR) bacteria caused by the abuse of antibiotics and the formation of biofilms are the two major obstacles to the treatment of bacterial infections. The utilization of noble metal materials has wide prospects to therapy pathogenic bacterial infections as the substitutes of antibiotics. Noble metal nanoparticles (NMNPs), especially gold (Au), silver (Ag), and platinum (Pt), have been attaching great attentions in antimicrobial field, owing to their multiple bactericidal properties, simple synthesis methods, good photo-responsive performance, etc. NMNPs can kill bacteria and eradicate biofilms by disrupting cell membrane potential and stability, binding to biological macromolecules (DNA or enzymes). Additionally, some NMNPs can produce hyperthermia locally or radical oxygen species (ROS). The underlying mechanism is that these NMNPs can be excited under light with different wavelength to exhibit photothermal and photocatalytic effect, aroused by the surface plasmon resonance of activated electrons. In this review, NMNPs and their functionalized nanocomposites for combating pathogenic bacterial infections are reviewed. Herein, the synthetic methods and antibacterial mechanisms of noble metal nanomaterials are discussed. Abstract:Nanomaterials and nanotechnologies have been expected to provide innovative platforms for addressing antibacterial challenges, with potential to even deal with bacterial infections involving drug-resistance. The current review summarizes recent progress over the last 3 years in the field of antibacterial nanomaterials with a photothermal conversion effect. We classify these photothermal nanomaterials into four functional categories: carbon-based nanoconjugates of graphene derivatives or carbon nanotubes, noble metal nanomaterials mainly from gold and silver, metallic compound nanocomposites such as copper sulfide and molybdenum sulfide, and polymeric as well as other nanostructures. Different categories can be assembled with each other to enhance the photothermal effects and the antibacterial activities. The review describes their fabrication processes, unique properties, antibacterial modes, and potential healthcare applications.

Abstract:Carbon nanomaterials like graphene, carbon nanotubes, fullerenes and the various forms of diamond have attracted great attention for their vast potential regarding applications in electrical engineering and as biomaterials. The study of the antibacterial properties of carbon nanomaterials provides fundamental information on the possible toxicity and environmental impact of these materials. Furthermore, as a result of the increasing prevalence of resistant bacteria strains, the development of novel antibacterial materials is of great importance. This article reviews current research efforts on characterizing the antibacterial activity of carbon nanomaterials from the perspective of colloid and interface science. Building on these fundamental findings, recent functionalization strategies for enhancing the antibacterial effect of carbon nanomaterials are described. The review concludes with a comprehensive outlook that summarizes the most important discoveries and trends regarding antibacterial carbon nanomaterials.

Abstract:The understanding of the interactions between biological systems and nanoengineered devices is crucial in several research fields, including tissue engineering, biomechanics, synthetic biology and biomedical devices. This review discusses the current knowledge of the interactions between bacteria and abiotic nanostructured substrates. First, the effects of randomly organized nanoscale topography on bacterial adhesion and persistence are described. Second, the interactions between microorganisms and highly organized/ordered micro- and nano-patterns are discussed. Finally, we survey the most promising approaches for the fabrication of silver polymeric nanocomposites, which have important applications as antimicrobial materials. The advantages, drawbacks and limitations of such nanotechnologies are critically discussed in view of potential future applications.

Abstract:Nanomaterial‐based antibacterial agents emerge as promising solutions to combat bacteria and inhibit their proliferation. However, selective elimination of specific bacteria remains crucial. This review examines nanomaterial‐based antibacterial agents and their mechanisms, including hydrophobic and electrostatic interactions, protein and nucleic acid leakage, selective labelling and killing, extracellular and intracellular ROS generation, and damage to inner and outer membranes, all contributing to selective antibacterial activity. Bacterial infections continue to threaten public health due to limitations in rapid and accurate diagnostic techniques. While broad‐spectrum antibiotics offer empirical treatment, their overuse has fuelled the emergence of antimicrobial resistance (AMR) pathogens, posing a critical global public health challenge. In this critical scenario, nanomaterial‐based antibacterial agents emerge as a promising solution to combat bacteria and inhibit their proliferation. However, selective elimination of pathogenic bacteria is paramount. This review highlights recent advancements in developing nanomaterials for selective antibacterial activity. We categorize these agents based on their mode of action, exploring how they selectively interact with bacteria and their potential antibacterial mechanisms. This review offers crucial insights for researchers exploring the potential of nanotechnology to address the growing threat of AMR.

Abstract:Infections caused by multidrug-resistant (MDR) bacteria are becoming a serious threat to public health worldwide. With an ever-reducing pipeline of last-resort drugs further complicating the current dire situation arising due to antibiotic resistance, there has never been a greater urgency to attempt to discover potential new antibiotics. The use of nanotechnology, encompassing a broad range of organic and inorganic nanomaterials, offers promising solutions. Organic nanomaterials, including lipid-, polymer-, and carbon-based nanomaterials, have inherent antibacterial activity or can act as nanocarriers in delivering antibacterial agents. Nanocarriers, owing to the protection and enhanced bioavailability of the encapsulated drugs, have the ability to enable an increased concentration of a drug to be delivered to an infected site and reduce the associated toxicity elsewhere. On the other hand, inorganic metal-based nanomaterials exhibit multivalent antibacterial mechanisms that combat MDR bacteria effectively and reduce the occurrence of bacterial resistance. These nanomaterials have great potential for the prevention and treatment of MDR bacterial infection. Recent advances in the field of nanotechnology are enabling researchers to utilize nanomaterial building blocks in intriguing ways to create multi-functional nanocomposite materials. These nanocomposite materials, formed by lipid-, polymer-, carbon-, and metal-based nanomaterial building blocks, have opened a new avenue for researchers due to the unprecedented physiochemical properties and enhanced antibacterial activities being observed when compared to their mono-constituent parts. This review covers the latest advances of nanotechnologies used in the design and development of nano- and nanocomposite materials to fight MDR bacteria with different purposes. Our aim is to discuss and summarize these recently established nanomaterials and the respective nanocomposites, their current application, and challenges for use in applications treating MDR bacteria. In addition, we discuss the prospects for antimicrobial nanomaterials and look forward to further develop these materials, emphasizing their potential for clinical translation.

Abstract:With the overuse or abuse of antibiotics, drug-resistant bacteria increase rapidly. It is urgent to develop effective antimicrobial agents to overcome the problem of drug-resistant bacteria. At present, there has been a huge academic focus on the development of nanomaterials with broad-spectrum antibacterial ability to fight drug-resistant bacteria. Moreover, the porous materials play a unique role in loading various antibacterial materials. In this review, the recent research progresses of porous materials for antibacterial application are discussed according to the classification of microporous (zeolites, MOFs, COFs, CMPs), mesoporous (mesoporous silica nanomaterials, non-silicon-based mesoporous materials) and macroporous materials. Besides, the antibacterial mechanisms of porous materials and their challenges and outlook for future applications are also discussed in this review. The antibacterial focus and expectations for the future of these porous materials are summarized to facilitate the development of porous material manufacturing techniques and fight bacterial infections.

Abstract:Highlights • 1) Fabrication of 2D antibacterial nanomaterials with prominent properties to exhibit antibacterial mechanisms. • 2) Associated challenges of nanotoxicity and dysbiosis are discussed in detail with major changes in the gut microbiome. 3) Issues with nanotoxicity data availability, curation, and mechanistic perception can be resolved by ML approaches. The number of multi-drug-resistant bacteria has increased over the last few decades, which has caused a detrimental impact on public health worldwide. In resolving antibiotic resistance development among different bacterial communities, new antimicrobial agents and nanoparticle-based strategies need to be designed foreseeing the slow discovery of new functioning antibiotics. Advanced research studies have revealed the significant disinfection potential of two-dimensional nanomaterials (2D NMs) to be severed as effective antibacterial agents due to their unique physicochemical properties. This review covers the current research progress of 2D NMs-based antibacterial strategies based on an inclusive explanation of 2D NMs' impact as antibacterial agents, including a detailed introduction to each possible well-known antibacterial mechanism. The impact of the physicochemical properties of 2D NMs on their antibacterial activities has been deliberated while explaining the toxic effects of 2D NMs and discussing their biomedical significance, dysbiosis, and cellular nanotoxicity. Adding to the challenges, we also discussed the major issues regarding the current quality and availability of nanotoxicity data. However, smart advancements are required to fabricate biocompatible 2D antibacterial NMs and exploit their potential to combat bacterial resistance clinically. Abstract:Present worldwide difficulties in healthcare and the environment have motivated the investigation and research of novel materials in an effort to find novel techniques to address the current challenges and requirements. In particular, the use of nanomaterials has demonstrated a significant promise in the fight against bacterial infections and the problem of antibiotic resistance. Metal nanoparticles and carbon-based nanomaterials in particular have been highlighted for their exceptional abilities to inhibit many types of bacteria and pathogens. In order for these materials to be as effective as possible, synthetic techniques are crucial. Therefore, in this review article, we highlight some recent developments in the design and synthesis of various nanomaterials, including metal nanoparticles (e.g., Ag, Zn, or Cu), metal hybrid nanomaterials, and the synthesis of multi-metallic hybrid nanostructured materials. Following that, examples of these materials’ applications in antimicrobial performance targeted at eradicating multi-drug resistant bacteria, material protection such as microbiologically influenced corrosion (MIC), or additives in construction materials have been described.

Robust Antibacterial Activity of Tungsten Oxide (WO3-X) Nanodots

Abstract:Antibacterial agents are an important tool in the prevention of bacterial infections. Inorganic materials are attractive due to their high stability under a variety of conditions compared to organic antibacterial agents. Herein tungsten oxide nanodots (WO3-X), synthesized by a simple one-pot synthetic approach, was found to exhibit efficient antibacterial capabilities. The analyses with colony-forming units (CFU) showed excellent antibacterial activity of WO3-X against both gram-negative E. coli (Escherichia coli) and gram-positive S. aureus (Staphylococcus aureus) strains. The scanning electron microscopy (SEM) and transmission electron microscopy (TEM) images revealed clear damage to the bacterial cell membranes, which was further confirmed by molecular dynamics simulations. Additionally, exposure to simulated sunlight was found to further increase germicidal activity of WO3-X nanodots – a 30-minute exposure to sunlight (combining 50 μg/mL WO3-X nanodots) showed a 70% decrease in E. coli viability compared to without exposure. Electron spin resonance spectroscopy (ESR) was used to elucidate the underlying mechanism of this photocatalytic activity through the generation of hydroxyl radical species. Cell counting kit-8 (CCK-8) and the live/dead assay were further employed to evaluate the cytotoxicity of WO3-X nanodots on eukaryotic cells, which demonstrated their general biocompatibility. In all, our results suggest WO3-X nanodots have considerable potential in antibacterial applications, while also being biocompatible at large.

Abstract:The rise in drug resistance in pathogenic bacteria greatly endangers public health in the post-antibiotic era, and drug-resistant bacteria currently pose a great challenge not only to the community but also to clinical procedures, including surgery, stent implantation, organ transplantation, and other medical procedures involving any open wound and compromised human immunity. Biofilm-associated drug failure, as well as rapid resistance to last-resort antibiotics, necessitates the search for novel treatments against bacterial infection. In recent years, the flourishing development of nanotechnology has provided new insights for exploiting promising alternative therapeutics for drug-resistant bacteria. Metallic agents have been applied in antibacterial usage for several centuries, and the functional modification of metal-based biomaterials using nanotechnology has now attracted great interest in the antibacterial field, not only for their intrinsic antibacterial nature but also for their ready on-demand functionalization and enhanced interaction with bacteria, rendering them with good potential in further translation. However, the possible toxicity of MNPs to the host cells and tissue still hinders its application, and current knowledge on their interaction with cellular pathways is not enough. This review will focus on recent advances in developing metallic nanoparticles (MNPs), including silver, gold, copper, and other metallic nanoparticles, for antibacterial applications, and their potential mechanisms of interaction with pathogenic bacteria as well as hosts.

Abstract:The dramatic increase of microbial resistances against conventional available antibiotics is a huge challenge to the effective treatment of infectious disease and thus becoming a daunting global threat of major concern, which necessitates the development of innovative therapeutics. Nanomaterial-based antimicrobial strategies have emerged as novel and promising tools to combat lethal bacteria and recalcitrant biofilm, featuring the abilities to evade existing drug resistance-related mechanisms. In this review, recent advances in "state-of-the-art" nanosystems which acting either as inherent therapeutics or nanocarriers for the precise delivery of antibiotics, are comprehensively summarized. Those nanosystems can effectively accumulate at the infectious sites, achieve multifunctional synergistic antibacterial efficacy, and provide controlled release of antibiotics in response to endogenous or exogenous stimulus (e.g., low pH, enzymes, or illumination). Especially, the nanoplatform that integrated with photothermal/photodynamic therapy (PTT/PDT) can enhance the bacterial destruction and biofilm penetration or ablation. In addition, nanoparticle-based approaches with enzymatically promoting bacterial killing, anti-virulence, and other mechanisms were also involved. Overall, this review provides crucial insights into the recent progress and remaining limitations of various antimicrobial nanotherapeutic strategies, and enlightens the further developments in this field simultaneously, which eventually benefiting public health.

Abstract:Biofilm-associated infections pose a significant challenge in healthcare, constituting 80% of bacterial infections and often leading to persistent, chronic conditions. Conventional antibiotics struggle with efficacy against these infections due to the high tolerance and resistance induced by bacterial biofilm barriers. Two-dimensional nanomaterials, such as those from the graphene family, boron nitride, molybdenum disulfide (MoS2), MXene, and black phosphorus, hold immense potential for combating biofilms. These nanomaterial-based antimicrobial strategies are novel tools that show promise in overcoming resistant bacteria and stubborn biofilms, with the ability to circumvent existing drug resistance mechanisms. This review comprehensively summarizes recent developments in two-dimensional nanomaterials, as both therapeutics and nanocarriers for precision antibiotic delivery, with a specific focus on nanoplatforms coupled with photothermal/photodynamic therapy in the elimination of bacteria and penetrating and/or ablating biofilm. This review offers important insight into recent advances and current limitations of current antibacterial nanotherapeutic approaches, together with a discussion on future developments in the field, for the overall benefit of public health.

Abstract:The technique of antimicrobial therapy action is to stop or slow the growth of bacteria that can kill people, animals, and crops. The most widely used antibacterial agents are antibiotics. Even though these antimicrobial medications are quite effective, there are still certain barriers or challenges in using them effectively. To solve these issues, new antimicrobial drug molecules that don't have side effects or resistance are needed. These days, antimicrobial drugs placed in nanosized vehicles, or nanomedicine, made of different metal and metallic oxides as well as of polymer, carbon or lipid-based may be used to address these issues with conventional therapy and delivery techniques. This review focuses on the importance of nanotechnology in antimicrobial therapy, nanoparticles (NPs) used in this therapy, their mode of action, and the recent advancement in nanotechnology for antimicrobial therapy.
