Abstract:The past decade has witnessed a rapidly growing interest in the biological basis of human mate choice. Here we review recent studies that demonstrate preferences for traits which might reveal genetic quality to prospective mates, with potential but still largely unknown influence on offspring fitness. These include studies assessing visual, olfactory and auditory preferences for potential good-gene indicator traits, such as dominance or bilateral symmetry. Individual differences in these robust preferences mainly arise through within and between individual variation in condition and reproductive status. Another set of studies have revealed preferences for traits indicating complementary genes, focussing on discrimination of dissimilarity at genes in the major histocompatibility complex (MHC). As in animal studies, we are only just beginning to understand how preferences for specific traits vary and inter-relate, how consideration of good and compatible genes can lead to substantial variability in individual mate choice decisions and how preferences expressed in one sensory modality may reflect those in another. Humans may be an ideal model species in which to explore these interesting complexities.

Despite this fundamental property of female mate choice, very few loci have been identified that contribute to mate choice and reproductive isolation. We used a combination of population genetics, quantitative complementation tests, and behavioral assays to demonstrate that and contribute to female mate choice, and could contribute to partial reproductive isolation between populations of . Our study is among the first to identify genes that contribute to female mate preference in this historically important system, where female preference is an active premating barrier to reproduction. The identification of loci that are primarily known for their roles in neurodevelopment provides intriguing questions of how female mate preference evolves in populations via changes in sensory system and higher learning brain centers.

Abstract:Males in many species show courtship and mating preferences for certain females over others when given the choice. One of the most common targets of male mate choice in insects is female body size, with males preferring to court and mate with larger, higher-fecundity females and investing more resources in matings with those females. Although this preference is well-documented at the species level, less is known about how this preference varies within species and whether there is standing genetic variation for male mate choice within populations. We used hemiclonal analysis in the fruit fly, , to test for heritable genetic variation in pre- and post-copulatory components of male mate choice for large females. We found additive genetic variation for both forms of male choice: males from different hemiclone lines varied in the strength of their courtship preferences for large females and the degree to which they extended matings with large females. Although males from hemiclone lines with stronger courtship preferences for large females were more likely to mate with those females, there was no genetic correlation between pre- and post-copulatory components of male mate choice, suggesting that they are under independent genetic control. Genetic variation in male mate choice may be widespread, potentially impacting the fitness of both sexes and the adaptive evolution of populations.

Abstract:Visual preferences are important drivers of mate choice and sexual selection, but little is known of how they evolve at the genetic level. In this study, we took advantage of the diversity of bright warning patterns displayed by Heliconius butterflies, which are also used during mate choice. Combining behavioral, population genomic, and expression analyses, we show that two Heliconius species have evolved the same preferences for red patterns by exchanging genetic material through hybridization. Neural expression of regucalcin1 correlates with visual preference across populations, and disruption of regucalcin1 with CRISPR-Cas9 impairs courtship toward conspecific females, providing a direct link between gene and behavior. Our results support a role for hybridization during behavioral evolution and show how visually guided behaviors contributing to adaptation and speciation are encoded within the genome.

Abstract:BackgroundIdentifying the sources of variation in mating interactions between males and females is important because this variation influences the strength and/or the direction of sexual selection that populations experience. While the origins and effects of variation in male attractiveness and ornamentation have received much scrutiny, the causes and consequences of intraspecific variation in females have been relatively overlooked. We used cytogenetic cloning techniques developed for Drosophila melanogaster to create “hemiclonal” males and females with whom we directly observed sexual interaction between individuals of different known genetic backgrounds and measured subsequent reproductive outcomes. Using this approach, we were able to quantify the genetic contribution of each mate to the observed phenotypic variation in biologically important traits including mating speed, copulation duration, and subsequent offspring production, as well as measure the magnitude and direction of intersexual genetic correlation between female choosiness and male attractiveness.ResultsWe found significant additive genetic variation contributing to mating speed that can be attributed to male genetic identity, female genetic identity, but not their interaction. Furthermore we found that phenotypic variation in copulation duration had a significant male-associated genetic component. Female genetic identity and the interaction between male and female genetic identity accounted for a substantial amount of the observed phenotypic variation in egg size. Although previous research predicts a trade-off between egg size and fecundity, this was not evident in our results. We found a strong negative genetic correlation between female choosiness and male attractiveness, a result that suggests a potentially important role for sexually antagonistic alleles in sexual selection processes in our population.ConclusionThese results further our understanding of sexual selection because they identify that genetic identity plays a significant role in phenotypic variation in female behaviour and fecundity. This variation may be potentially due to ongoing sexual conflict found between the sexes for interacting phenotypes. Our unexpected observation of a negative correlation between female choosiness and male attractiveness highlights the need for more explicit theoretical models of genetic covariance to investigate the coevolution of female choosiness and male attractiveness.

Abstract:1.Although vertebrates have been reported to gain higher reproductive outputs by choosing mates, few studies have been conducted on threatened species. However, species recovery should benefit if natural mate choice could improve reproductive output (i.e., pair performance related to offspring number, such as increased clutch size, numbers of fertilized egg and fledglings). We assessed the evidence for major histocompatibility complex (MHC)‐based mate preference in the endangered crested ibis (Nipponia nippon), and quantified the impacts of such choice on reproductive output.2. We tested the hypothesis that crested ibis advertise "good genes" through external traits, by testing whether nuptial plumage characteristics and body morphology mediate mate choice for underlying genetic MHC variation.3. We found differences between males and females in preferred MHC genotypes, external traits used in mate choice, and contributions to reproductive outputs. Females preferred MHC‐heterozygous males, which had darker [i.e., lower total reflectance and ultraviolet (UV) reflectance] nuptial plumage. Males preferred females lacking the DAB*d allele at the MHC classIIDAB locus, which had higher average body mass. DAB*d‐free females yielded heavier eggs and more fledglings, while MHC‐heterozygous males contributed to more fertilized eggs and fledglings. Fledging rate was highest when both parents had the preferred MHC genotypes (i.e., MHC‐heterozygous father and DAB*d‐free mother). Comparisons showed that free‐mating wild and seminatural pairs yielded more fertilized eggs and more fledglings, with a higher fledging rate, than captive pairs matched artificially based on pedigree.4. Conservation programs seldom apply modern research results to population management, which could hinder recovery of threatened species. Our results show that mate choice can play an important role in improving reproductive output, with an example in which an endangered bird selects mates using UV visual capability. Despite the undoubted importance of pedigree‐based matching of mates in conservation programs, we show that free‐mating can be a better alternative strategy.This article is protected by copyright. Abstract:Abstract Traits under divergent ecological selection that also function during mating can be important in maintaining species boundaries. Few studies have considered mutual mate choice, where both males and females base mating decisions on the same trait. Wing colouration in Heliconius butterflies evolved as a warning signal but also functions as a mating cue. We investigated the contribution of visual preference to assortative mating in an aposematic butterfly Heliconius cydno in the context of reproductive isolation with its sympatric, visually distinct relative Heliconius melpomene. Heliconius cydno have conspicuous white bands on their forewings, whereas those of H. melpomene are red in colour. We predicted that both sexes of H. cydno contributed to assortative mating by exhibiting visual preference towards conspecific wing colouration. We analysed published and new data from preference experiments, in which males were presented with conspecific and H. melpomene females. We also recorded female responses and mating outcomes in choice experiments, involving conspecific males with either the original white or artificially painted red forewing bands. Both sexes of H. cydno responded more positively towards the conspecific colouration, and males strongly preferred females of its own colours. In contrast, male colouration did not predict mating outcomes in female choice experiments. As courtships are initiated by males in butterflies, our findings suggest that female visual preference might be of secondary importance in H. cydno. Our data also suggest that the contribution of visual preference to reproductive isolation might be unequal between H. cydno and its sympatric relative H. melpomene.

Abstract:Selection can facilitate diversification by inducing character displacement in mate choice traits that reduce the probability of maladaptive mating between lineages. Although reproductive character displacement (RCD) has been demonstrated in two-taxa case studies, the frequency of this process in nature is still debated. Moreover, studies have focused primarily on visual and acoustic traits, despite the fact that chemical communication is probably the most common means of species recognition. Here, we showed in a large, mostly sympatric, butterfly genus, a strong pattern of recurrent RCD for predicted male sex pheromone composition, but not for visual mate choice traits. Our results suggest that RCD is not anecdotal, and that selection for divergence in male sex pheromone composition contributed to reproductive isolation within the Bicyclus genus. We propose that selection may target olfactory mate choice traits as a more common sensory modality to ensure reproductive isolation among diverging lineages than previously envisaged.

Abstract:Abstract Background Genes of the major histocompatibility complex (MHC) are an important component of the vertebrate immune system and play a significant role in mate choice in animal populations. However, the MHC genetic targets of female mate choice have not been clearly identified, and whether female mate choice is based on neutral genetic characteristics remains an open question. Here, we focus on the effects of morphological traits and genetic similarity among individuals in MHC class IIB (MHC IIB) exon 2 on mating in a sexually dimorphic songbird that exhibits social monogamy with extra-pair paternity (EPP). Methods We sequenced 64 parent–offspring triads sampled over a 3-year period using two MHC class II loci to detect disassortative mating in the Yellow-rumped Flycatcher ( Ficedula zanthopygia ). Results We found that MHC similarity in social pairs was lower than that in random pairs. Extra-pair mate choice according to MHC IIB was observed, in which females’ extra-pair mates had fewer MHC alleles than their within-pair mates, but there was no significant band-sharing between extra-pair sires and potential extra-pair mates. However, the interaction between the MHC diversity of females and that of the social males affected the occurrence of EPP. Conclusions Our results support the “optimality hypothesis” of MHC-based social and extra-pair choice. Female choice probably maintains a certain level of MHC diversity in offspring in the Yellow-rumped Flycatcher.

Abstract:Few major histocompatibility complex (MHC)-based mate choice studies include all MHC genes at the inter-individual, sperm-egg, and mother-fetus recognition levels. We tested three hypotheses of female mate choice in a 17-year study of the giant panda (Ailuropoda melanoleuca) while using ten functional MHC loci (four MHC class I loci: Aime-C, Aime-F, Aime-I, and Aime-L; six MHC class II loci: Aime-DRA, Aime-DRB3, Aime-DQA1, Aime-DQA2, Aime-DQB1, and Aime-DQB2); five super haplotypes (SuHa, SuHaI, SuHaII, DQ, and DR); and, seven microsatellites. We found female choice for heterozygosity at Aime-C, Aime-I, and DQ and for disassortative mate choice at Aime-C, DQ, and DR at the inter-individual recognition level. High mating success occurred in MHC-dissimilar mating pairs. No significant results were found based on any microsatellite parameters, suggesting that MHCs were the mate choice target and there were no signs of inbreeding avoidance. Our results indicate Aime-DQA1- and Aime-DQA2-associated disassortative selection at the sperm-egg recognition level and a possible Aime-C- and Aime-I-associated assortative maternal immune tolerance mechanism. The MHC genes were of differential importance at the different recognition levels, so all of the functional MHC genes should be included when studying MHC-dependent reproductive mechanisms.

Abstract:To investigate whether female mate choice could be directed at male genetic quality, male chemical signals and progeny fitness were studied in the red flour beetle (Tribolium castaneum). Differences among males in the attractiveness of their pheromone to females were statistically significant. Developmental time of progeny was significantly heritable, indicating that some males have "good genes" for this trait. There was no statistically significant correlation between progeny fitness and male attractiveness. These results do not support the hypothesis that in this species the evolution of female preferences for male pheromone is adaptive.

Abstract:The concept of "genetic coupling" in mate recognition systems arose in the 1960s as a potential mechanism to maintain coordination between signals and receivers during evolutionary divergence. At its most basic it proposed that the same genes might influence trait and preference, and therefore mutations could result in coordinated changes in both traits. Since then, the concept has expanded in scope and is often used to include linkage or genetic correlation between recognition system components. Here we review evidence for genetic coupling, concentrating on proposed examples of a common genetic basis for signals and preferences. Mapping studies have identified several examples of tight genetic linkage between genomic regions influencing signals and preferences, or assortative mating. Whether this extends as far as demonstrating pleiotropy remains a more open question. Some studies, notably of Drosophila, have identified genes in the sex determination pathway and in pheromonal communication where single loci can influence both signals and preferences. This may be based on isoform divergence, in which sex- and tissue-specific effects are facilitated by alternative spicing, or on regulatory divergence. Hence it is not clear that such examples provide compelling evidence of pleiotropy in the sense that "magic mutations" could maintain trait coordination. Rather, coevolution may be facilitated by regulatory divergence but require different mutations or coevolution across isoforms. Reconsidering the logic of genetic coupling, it may be that pleiotropy could actually be less effective than linkage if distinct but associated variants allow molecular coevolution to occur more readily than potentially "unbalanced" mutations in single genes. Genetic manipulation or studies of mutation order effects during divergence are challenging but perhaps the only way to disentangle the role of pleiotropy versus close linkage in coordinated trait divergence.

Abstract:BACKGROUND:Sympatric sister species provide an opportunity to investigate the genetic mechanisms and evolutionary forces that maintain species boundaries. The persistence of morphologically and genetically distinct populations in sympatry can only occur if some degree of reproductive isolation exists. A pair of sympatric sister species of Primulina (P. depressa and P. danxiaensis) was used to explore the genetic architecture of hybrid male sterility.RESULTS:We mapped one major- and seven minor-effect quantitative trait loci (QTLs) that underlie pollen fertility rate (PFR). These loci jointly explained 55.4% of the phenotypic variation in the F2 population. A Bateson-Dobzhansky-Muller (BDM) model involving three loci was observed in this system. We found genotypic correlations between hybrid male sterility and flower morphology, consistent with the weak but significant phenotypic correlations between PFR and floral traits.CONCLUSIONS:Hybrid male sterility in Primulina is controlled by a polygenic genetic basis with a complex pattern. The genetic incompatibility involves a three-locus BDM model. Hybrid male sterility is genetically correlated with floral morphology and divergence hitchhiking may occur between them.
The Major Histocompatibility Complex and Mate Choice: Inbreeding Avoidance and Selection of Good Genes

Abstract:It has been known for decades that MHC genes play a critical role in the cellular immune response, but only recent research has provided a better understanding of how these molecules might affect mate choice. Original studies in inbred mouse strains revealed that mate choice was influenced by MHC dissimilarity. Detection of MHC differences between individuals in these experiments was related to olfactory cues, primarily in urine. Recent studies in humans have shown an analogous picture of MHC-based mating. Taken together, these findings could support either the hypothesis of MHC-based inbreeding avoidance or the hypothesis of MHC-related avoidance of reproductive failure, since studies in mice, humans and pigtailed macaques have shown that parental sharing of certain MHC alleles correlates with frequent spontaneous abortion or prolonged intergestational intervals. Data from many mammalian species clearly demonstrate that reproductive failure occurs as a result of inbreeding. Therefore, MHC similarity might serve as an indicator of genome-wide relatedness. In contrast, increased fitness due to the presence of individual MHC alleles in a pathogenic environment could explain MHC-based selection of currently good genes. Specifically, the physical condition of long-living animals depends on the ability to respond to immunological challenge and an individual's MHC alleles determine the response, since, unlike the T cell receptors, MHC alleles are not somatically recombined. Therefore, sexual selection of condition-dependent traits during mate choice could be used to select successful MHC alleles, thereby providing offspring with a higher relative immunity in their pathogenic environment.
Cryptic Mate Preference in Male Bicyclus anynana Butterflies

Abstract:While male mate choice has received sparse attention in comparison to female choice, it occurs often in insects. In addition, male insects may preferentially allocate sperm and ejaculate in response to female quality. Previous research indicates that male Bicyclus anynana butterflies can learn mate preference through prior exposure to females, though naïve males mate randomly. It is unclear whether this preference learning may also influence male sperm and ejaculate allocation after mate selection, or whether males have cryptic mate preference for female wing patterns independent of preference learning. Here we test whether B. anynana males adjust their sperm and ejaculate allocation in response to a learned preference. We also assess whether males exhibit an innate cryptic preference and adjust their sperm and ejaculate in response to female wing pattern. We compared number of eggs laid by females and spermatophore (male butterfly ejaculate) weight in four no-choice treatments: naïve male butterflies (having no prior exposure to females), paired with a 2 or 0-spot female, and experienced male butterflies (having a previous three-hour interaction with a 0-spot female), paired with a 2 or 0-spot female. All females used were naturally 2-spot females, 0-spot females had artificially blocked spots. We found that 0-spot females laid significantly more eggs than 2-spot females, independent of male experience. There was no effect of female phenotype or male experience on spermatophore weight. Our findings suggest that male B. anynana have an innate cryptic preference for 0-spot females, which has been shown in other studies to only be seen as a pre-copulatory preference when enhanced by early experience.

Abstract:Genes of the major histocompatibility complex (MHC) play important roles in vertebrate immunocompetence. MHC genes thus offer females indirect benefits to mate choice through the production of offspring of an optimal MHC genotype. Females may choose males with specific MHC haplotypes, dissimilar MHC genotypes, MHC heterozygous males or MHC‐diverse males. We tested these four alternatives for both female social and paternal choice in wild golden snub‐nosed monkeys (Rhinopithecus roxellana) by examining overall genetic variability (via microsatellites) and four MHC‐genes (DRB1, DRB2, DQA1 and DQB1). Monte Carlo randomization tests showed that MHC dissimilarity was favored for social choice (males to which females were socially affiliated) and intermediate MHC dissimilarity was favored in paternal choice (fathers of offspring). No evidence of inbreeding avoidance was found for either social or paternal mates. We found that MHC heterozygotes, higher microsatellite multi‐locus heterozygosity and higher microsatellites diversity were favored for social mates, and higher microsatellite diversity was favored for paternal mates. Independent of male age, we found that the formation of male‐female social pairings is significantly predicted by compatibility based on the sharing of MHC haplotypes. However, we found no evidence of independent genetic effects on the duration of male‐female social pairings, male social status (achieving OMU leader male status or not), the number of females with which individual leader males paired, the likelihood of potential male‐female pairings producing offspring, or whether males fathered offspring or not. Overall, our findings suggest different genetic factors are involved in social and paternal choice in R. roxellana.

Abstract:Abstract The total strength of sexual selection on males depends on the relationship between various components of pre- and post-copulatory fitness. Misalignment between male and female interests creates inter-locus sexual conflict, where the fitness of one sex is increased at the expense of the other. Although rarely considered, mating behaviours can also be genetically correlated between males and females, creating intra-locus sexual conflict, where beneficial alleles in one sex are costly when expressed in the other sex. How inter- and intra-locus sexual conflicts operate on the expression of mating behaviours remains little understood. Here, we study male attractiveness, mating latency and copulation duration in two populations of the polyandrous Drosophila serrata . Univariate analyses show little genetic variance in mating latency, and that males, but not females, contribute to copulation duration genetic variance. Further, multivariate analyses revealed little covariance between the studied traits. However, analyses considering male and female contribution in a single framework supported genetic contributions from both sexes for mating behaviours and complex patterns of between sexes correlations. Finally, our study did not find any association between those mating behaviours and fitness component, specifically (i) no phenotypic covariance between male attractiveness and mating latency and, (ii) longer copulations did not result in the production of more offspring. With no detectable fitness benefits in any sexes for shorter mating latency or longer copulation duration, our results do not support the presence of inter-nor intra-locus sexual conflict for these mating traits.

Abstract:Mate choice and sexual displays are widespread in nature, but their evolutionary benefits remain controversial. Theory predicts these traits can be favored by runaway sexual selection, in which preference and display reinforce one another due to genetic correlation; or by good genes benefits, in which mate choice is advantageous because extreme displays indicate a well-adapted genotype. However, these hypotheses are not mutually exclusive, and the adaptive benefits underlying mate choice can themselves evolve. In particular, examining how and why sexual displays become indicators of good genes is challenging in natural systems. Here, we use experimental evolution in "digital organisms" to demonstrate the origins of condition-dependent indicator displays following their spread due to a runaway process. Surprisingly, handicap-like costs are not necessary for displays to become indicators of male viability. Instead, a pleiotropic genetic architecture underlies both displays and viability. Runaway sexual selection and good genes benefits should thus be viewed as interacting mechanisms that reinforce one another.

Abstract:Mating and reproduction are central to natural selection, and decisions associated with one’s choice of mate can have significant fitness consequences. From an evolutionary perspective, attraction functions to direct one’s attention and energy toward pursuing, mating with, and retaining individuals who display traits that contribute to greater survival and reproductive success. Humans are theorized to possess a suite of psychological mechanisms that facilitate the identification of such individuals. One trait that humans have potentially evolved to be attracted to is genetic dissimilarity or diversity in the major histocompatibility complex, which is argued to promote greater immunocompetence and pathogen resistance and, hence, health in one’s mate and putative offspring. Another trait is bilateral symmetry, which is theorized to function as a cue to a potential mate’s genetic quality and ability to withstand developmental stressors. Yet another trait is sexual dimorphism. Women are theorized to be attracted to masculinity in men, which is theorized to function as a reliable signal of underlying genetic quality. In contrast, men are theorized to be attracted to femininity in women, which is argued to signal their reproductive viability. Importantly, evolutionary perspectives propose that many attraction mechanisms evolved to adaptively adjust to local conditions. Thus, when faced with high pathogen prevalence, people have heightened preferences for symmetry, which indicates having good genes and thus, greater ability to withstand disease. As another example, when potential mates of the other sex are in relative abundance, people tend to be more selective in their mate choice and exaggerate their preferences for other-sex mates with sex-typical traits. Additionally, near peak fertility, women may have evolved to increase their preferences for masculinity in men, which signals underlying genetic quality. In addition to having psychological mechanisms that facilitate the identification of potential mates, humans may have also evolved psychological mechanisms that adaptively increase the motivation to allocate attention and energy toward pursuing viable mates that have been identified. Both sets of psychological mechanisms are necessary to successful mate selection, and likely operate in tandem. In this regard, dopamine may be centrally involved in driving behaviors associated with attraction and mate pursuit. Finally, recent studies have shown that the evidence for some of the hypothesized attraction preferences is not conclusive; future scholarship will profit from more careful research design and robust methodology.
